Magnesium-calcium signalling in rat parotid acinar cells: effects of acetylcholine.

This study investigated the effects of extracellular Mg(2+) ([Mg(2+)](o)) on basal and acetylcholine (ACh)-evoked amylase secretion and intracellular free Ca(2+) ([Ca(2+)](i)) in rat parotid acinar cells. In a medium containing 1.1 mM [Mg(2+)](o), ACh evoked significant increases in amylase secretion and [Ca(2+)](i). Either low (0 mM) or elevated (5 and 10 mM) [Mg(2+)](o) attenuated ACh-evoked responses. In a nominally Ca(2+) free medium, elevated [Mg(2+)](o) attenuated basal and ACh-evoked amylase secretion and [Ca(2+)](i). In parotid acinar cells incubated with either 0, 1.1, 5 or 10 mM [Mg(

Lipid rafts, the sarcoplasmic reticulum and uterine calcium signalling: an integrated approach.

The pathways involved in Ca2+ signalling in the uterus remain incompletely understood, impairing our ability to prevent preterm and difficult labours. In this review we focus on two elements in the pathway of Ca2+ signalling that have recently emerged as playing important roles: membrane lipid rafts and the sarcoplasmic reticulum. We examine the evidence for lipid rafts in the uterus and discuss their functional role. We suggest that the increases in cytosolic [Ca2+] and contractility that occur with raft disruption are due, at least in part, to effects on large conductance Ca2+-activated K+ (

The development of signal transduction pathways during epididymal maturation is calcium dependent.

Capacitation has been correlated with the activation of a cAMP-PKA-dependent signaling pathway leading to protein tyrosine phosphorylation. The ability to exhibit this response to cAMP matures during epididymal maturation in concert with the ability of the spermatozoa to capacitate. In this study, we have addressed the mechanisms by which spermatozoa gain the potential to activate this signaling pathway during epididymal maturation. In a modified Tyrode's medium containing 1.7 mM calcium, caput spermatozoa had significantly higher [Ca2+]i than caudal cells and could not tyrosine phosphorylate

The role of intracellular calcium signaling in premature protease activation and the onset of pancreatitis.

The exocrine pancreas synthesizes and secretes large amounts of digestive proteases as inactive precursor zymogens. Under physiological conditions a variety of cellular defense mechanisms protect the pancreatic acinar cell against a premature and intracellular activation of these zymogens. When these defenses fail, pancreatic autodigestion is initiated and acute pancreatitis can develop. A number of experimental observations suggest that extra- as well as intracellular calcium concentrations play an important part in the initiation of pancreatic protease activation, but the intracellular signa

The human calmodulin interactome reveals functional diversity linking calcium signaling to transcriptional regulation.

Calcium-calmodulin signaling represents a central mechanism through which intracellular calcium fluctuations coordinate diverse cellular processes. Despite the identification of numerous calmodulin binding proteins, a systems level understanding of how these interactions integrate into global signaling networks and influence disease relevant pathways remains incomplete. Here, we constructed a comprehensive human calmodulin interactome by integrating experimentally validated protein-protein interactions from the IntAct and BioGRID databases, identifying 919 calmodulin associated proteins. Tuina ameliorates sleep disturbances in PCPA-treated rats through Piezo1-mediated calcium signaling.

OBJECTIVES: Insomnia is a prevalent sleep disorder that severely impairs physical and mental health. Clinical research has shown that Tuina, a form of therapeutic massage, can improve sleep quality and restore normal sleep patterns, yet the underlying biological mechanism remains unclear. This study aimed to determine whether Tuina improves sleep-related behaviors by modulating the Piezo1-calcium signaling pathway through mechanical stimulation of the nervous system. MATERIALS AND METHODS: A rat model of sleep disturbances was induced using PCPA. Rats received daily Tuina on the dorsal region

Pathological calcium overload impairs skeletal muscle regeneration through sustained NFAT-dependent proliferative signaling.

Skeletal muscle regeneration requires coordinated myoblast proliferation and differentiation. Calcium dysregulation is commonly observed in aging and disease-associated muscle weakness; however, its impact on myogenic progression remains unclear. In this study, we investigated the mechanism by which intracellular calcium overload impairs myogenic differentiation and muscle regeneration. Transcriptomic analysis of aged human muscle revealed coordinated activation of calcium signaling and proliferation pathways. In vitro calcium exposure increased myoblast proliferation under differentiation con

Stigmatic papilla cells secrete jasmonic acid to promote pollen germination through calcium signaling.

Double fertilization begins with signal communication between pollen grains and the pistil stigma, a process that includes pollen adhesion, hydration, germination, and penetration. While several molecular regulators governing pollen hydration and penetration have been characterized, the molecules that mediate pollen germination on the stigma remain largely unelucidated. In this study, we identified jasmonic acid (JA), a maternal factor secreted by stigmatic papilla cells, which facilitates pollen germination by elevating cytosolic Ca2+ levels in pollen grains. We found that pollen grains exhib

Calcium signaling pathway implicates a shared genetic basis between psychiatric and cardiovascular diseases.

Psychiatric and cardiovascular diseases (CVDs) are frequently comorbid and are interconnected through the brain-heart axis. However, the underlying shared genetic etiology remains unknown in East Asians. To address this critical gap, we conducted a genome-wide pairwise trait pleiotropy study by leveraging genome-wide association studies of three major psychiatric disorders (schizophrenia [SCZ], bipolar disorder [BIP], major depressive disorder [MDD]) and ten cardiovascular traits (including eight CVDs) in East Asians. We identified genetic overlaps across seven disease pairs, such as SCZ with

Store-operated calcium signaling modulators improve a dendritic cell-based vaccine against melanoma.

Cruz Cobo et al. identify small molecules that reprogram dendritic cells toward an immuno-stimulating secretory phenotype, revealing endoplasmic reticulum calcium signaling as a druggable axis to boost vaccine-based and T cell-mediated cancer immunotherapies. The study links broad changes in calcium-induced secretion to improved immune activation and therapeutic potency.

Biallelic MCUR1 nonsense mutation associated with vacuolar myopathy and altered mitochondrial calcium signaling.

During muscle contraction, increased influx of calcium from the myocyte cytosol into the mitochondrial matrix through the mitochondrial calcium uniporter (MCU) links calcium homeostasis with high ATP provision. The MCU is located at the inner mitochondrial membrane and one of its structural components, the mitochondrial calcium uniporter regulator 1 (MCUR1), promotes its activity. Although MCUR1 function has been studied in cell models, mutations have not yet been associated with human disease. Here, we present a patient with proximal muscle weakness and atrophy, showing histological features

Neutrophils insert elastase into hepatocytes to regulate calcium signaling in alcohol-associated hepatitis.

Neutrophil infiltration occurs in a variety of liver diseases, but it is unclear how neutrophils and hepatocytes interact. Neutrophils generally use granule proteases to digest phagocytosed bacteria and foreign substances or neutralize them in neutrophil extracellular traps. In certain pathological states, granule proteases play a destructive role against the host as well. More recently, nondestructive actions of neutrophil granule proteins have been reported, such as modulation of tissue remodeling and metabolism. Here, we report a completely different mechanism by which neutrophils act nonde

Metformin regulates cellulase production in Trichoderma reesei via calcium signaling and mitochondrial function.

BACKGROUND: Trichoderma reesei is renowned for its cellulase-producing ability and is used for biofuel production from lignocellulose. In plants and fungi, cellulase production is induced by cellulose and suppressed by glucose; however, whether metformin can enhance cellulase production and mitochondrial function in T. reesei remains unclear. Metformin reduces blood glucose levels by inhibiting hepatic gluconeogenesis; therefore, it is worth investigating whether metformin transmission modulates cellulase biosynthesis in T. reesei. RESULTS: Metformin increased cellulase production and the tra

Regulation of cellulase production via calcium signaling in Trichoderma reesei under PEG8000 stress.

In this study, the effect of polyethylene glycol 8000 (PEG8000) stress on cellulase biosynthesis in Trichoderma reesei CICC2626 via calcium signaling was investigated, and a plausible mechanism by which intracellular Ca2+ regulates the transcription of cellulase genes was proposed. The results indicated that the total cellulase (filter paper-hydrolyzing activity [FPase]), endoglucanase (carboxymethyl cellulase activity [CMCase]), and β-glucosidase activities of the strain were 1.3-, 1.2-, and 1.3-fold higher than those of the control (no PEG8000 addition) at a final concentration of 1.5% (w/v)

Autonomous Calcium Signaling in Human and Zebrafish Podocytes Controls Kidney Filtration Barrier Morphogenesis.

BACKGROUND: Podocytes are critical to maintaining the glomerular filtration barrier, and mutations in nephrotic syndrome genes are known to affect podocyte calcium signaling. However, the role of calcium signaling during podocyte development remains unknown. METHODS: We undertook live imaging of calcium signaling in developing podocytes, using zebrafish larvae and human kidney organoids. To evaluate calcium signaling during development and in response to channel blockers and genetic defects, the calcium biosensor GCaMP6s was expressed in zebrafish podocytes. We used electron microscopy to eva

Calcium-sensing receptor regulates intestinal dipeptide absorption via Ca2+ signaling and IKCa activation.

Although absorption of di- and tripeptides into intestinal epithelial cells occurs via the peptide transporter 1 (PEPT1, also called solute carrier family 15 member 1 (SLC15A1)), the detailed regulatory mechanisms are not fully understood. We examined: (a) whether dipeptide absorption in villous enterocytes is associated with a rise in cytosolic Ca2+ ([Ca2+ ]cyt ), (b) whether the calcium sensing receptor (CaSR) is involved in dipeptide-elicited [Ca2+ ]cyt signaling, and (c) what potential consequences of [Ca2+ ]cyt signaling may enhance enterocyte dipeptide absorption. Reducing Astrocyte Calcium Signaling In Vivo Alters Striatal Microcircuits and Causes Repetitive Behavior.

Astrocytes tile the central nervous system, but their functions in neural microcircuits in vivo and their roles in mammalian behavior remain incompletely defined. We used two-photon laser scanning microscopy, electrophysiology, MINIscopes, RNA-seq, and a genetic approach to explore the effects of reduced striatal astrocyte Ca2+ signaling in vivo. In wild-type mice, reducing striatal astrocyte Ca2+-dependent signaling increased repetitive self-grooming behaviors by altering medium spiny neuron (MSN) activity. The mechanism involved astrocyte-mediated neuromodulation facilitated by ambient GABA

Dysfunctional Calcium and Glutamate Signaling in Striatal Astrocytes from Huntington's Disease Model Mice.

UNLABELLED: Astrocytes tile the entire CNS, but their functions within neural circuits in health and disease remain incompletely understood. We used genetically encoded Ca(2+)and glutamate indicators to explore the rules for astrocyte engagement in the corticostriatal circuit of adult wild-type (WT) and Huntington's disease (HD) model mice at ages not accompanied by overt astrogliosis (at approximately postnatal days 70-80). WT striatal astrocytes displayed extensive spontaneous Ca(2+)signals, but did not respond to cortical stimulation, implying that astrocytes were largely disengaged from co

Aberrant calcium signaling by transglutaminase-mediated posttranslational modification of inositol 1,4,5-trisphosphate receptors.

The inositol 1,4,5-trisphosphate receptor (IP3R) in the endoplasmic reticulum mediates calcium signaling that impinges on intracellular processes. IP3Rs are allosteric proteins comprising four subunits that form an ion channel activated by binding of IP3 at a distance. Defective allostery in IP3R is considered crucial to cellular dysfunction, but the specific mechanism remains unknown. Here we demonstrate that a pleiotropic enzyme transglutaminase type 2 targets the allosteric coupling domain of IP3R type 1 (IP3R1) and negatively regulates IP3R1-mediated calcium signaling and autophagy by lock

Calcium signaling regulates ventricular hypertrophy during development independent of contraction or blood flow.

In utero interventions aimed at restoring left ventricular hemodynamic forces in fetuses with prenatally diagnosed hypoplastic left heart syndrome failed to stimulate ventricular myocardial growth during gestation, suggesting chamber growth during development may not rely upon fluid forces. We therefore hypothesized that ventricular hypertrophy during development may depend upon fundamental Ca(2+)-dependent growth pathways that function independent of hemodynamic forces. To test this hypothesis, zebrafish embryos were treated with inhibitors or activators of Ca(2+) signaling in the presence or
