Epac Proteins and Calmodulin as Possible Arrhythmogenesis Trigger in Alcoholic Cardiomyopathy.

The expression of Epac proteins (exchange protein directly activated by cAMP) and calmodulin (CaM) was assessed by the content of the corresponding mRNA in biopsy specimens of cardiac atrium, left ventricle, and thoracic aorta of rats with alcoholic cardiomyopathy. In the myocardium, overexpression of Еpac1, Ерас2, and СаМ mRNA was found. The content of Epac2 mRNA in the left ventricle was elevated by 2.9 times (p=0.000001), in the left atrium by 3.2 times (p=0.00001), in the right atrium by 3 times (p=0.00001). In contrast to the myocardial tissue, the content of CaM mRNA in the thoracic aort

The calmodulin intergenic spacer as molecular target for characterization of Leishmania species.

BACKGROUND: Human leishmaniasis is a neglected disease caused by parasites of the genus Leishmania. Clinical aspects of this disease can vary significantly, reflecting the wide range of parasites in the genus Leishmania. Knowing accurately the Leishmania species infecting humans is important for clinical case management and evaluation of epidemiological risk. Calmodulin is an essential gene in trypanosomatids that modulates the calcium metabolism in various cellular activities. Despite its strong conservation in trypanosomatids, it has been recently observed that its untranslated regions (UTR)

Activity-dependent modulation of endocytosis by calmodulin at a large central synapse.

Although Ca(2+)/calmodulin has been suggested to play a role during endocytosis, it remains unknown if binding of Ca(2+) to calmodulin is essential for initiating endocytosis or if this interaction only has a modulatory effect on endocytosis. In this study, using time-resolved capacitance measurements at the rat calyx of Held synapse, the role of calmodulin in endocytosis was examined. Our results demonstrate that blocking calmodulin with an inhibitory peptide, which interfers with the binding of calmodulin to downstream targets, slowed the rate of endocytosis, but only when accompanied by hig

The use of ESI-MS to probe the binding of divalent cations to calmodulin.

Proteins have evolved with distinct sites for binding particular metal ions. This allows metalloproteins to perform a myriad of specialized tasks with conformations tailor-made by the combination of its primary sequence and the effect on this of the ligated metal ion. Here we investigate the selectivity of the calcium trigger protein calmodulin for divalent metal ions. This ubiquitous and highly abundant protein exists in equilibrium between its apo and its holo form wherein four calcium ions are bound. Amongst its many functions, calmodulin modulates the calcium concentration present in cells

Calcium dependence of the interaction between calmodulin and anthrax edema factor.

Edema factor (EF), a toxin from Bacillus anthracis (anthrax), possesses adenylyl cyclase activity and requires the ubiquitous Ca2+-sensor calmodulin (CaM) for activity. CaM can exist in three major structural states: an apo state with no Ca2+ bound, a two Ca2+ state with its C-terminal domain Ca2+-loaded, and a four Ca2+ state in which the lower Ca2+ affinity N-terminal domain is also ligated. Here, the interaction of EF with the three Ca2+ states of CaM has been examined by NMR spectroscopy and changes in the Ca2+ affinity of CaM in the presence of EF have been determined by flow dialysis. Asymmetric calmodulin distribution in the hypothalamus: role of sexual differentiation in the rat.

The Ca2+/calmodulin (CaM) system plays important roles both in hypothalamic sexual differentiation and in the progesterone-induced facilitation of lordosis behavior in the adult rat. We recently showed sex-dependent differences in rat hypothalamic CaM levels, both in newborn and in adult animals. Here, we evaluated the presence of left-right hypothalamic asymmetries in CaM concentration in male and female rats, as well as the changes induced on these parameters by neonatal (1 h after birth) subcutaneous administration of tamoxifen (200 microg/rat) or testosterone (30 microg/rat). CaM was measu

A high affinity acceptor for phospholipase A2 with neurotoxic activity is a calmodulin.

One of the high affinity binding proteins for ammodytoxin C, a snake venom presynaptically neurotoxic phospholipase A(2), has been purified from porcine cerebral cortex and characterized. After extraction from the membranes, the toxin-binding protein was isolated in a homogenous form using wheat germ lectin-Sepharose, Q-Sepharose, and ammodytoxin-CH-Sepharose chromatography. The specific binding of (125)I-ammodytoxin C to the isolated acceptor was inhibited to different extents by some neurotoxic phospholipases A(2), ammodytoxins, bee venom phospholipase A(2), agkistrodotoxin, and crotoxin; bu

Molecular dynamics simulation of the calmodulin-trifluoperazine complex in aqueous solution.

Trifluoperazine (TFP) has been widely studied in relation to its mode of binding and its inactivation of calmodulin (CaM). Most studies in solution have indicated that CaM has two high-affinity binding sites for TFP. The crystal structure of the 1:4 CaM-TFP complex (CaM-4TFP) shows that three TFP molecules bind to the C-domain of CaM, and that one TFP molecule binds to the N-domain. In contrast, the crystal structure of the 1:1 CaM-TFP complex (CaM-1TFP) shows that one TFP molecule binds to the C-domain. It has been thought that the binding of one TFP molecule to the C-domain is followed by bi

A putative role for calmodulin in the activation of Neurospora crassa chitin synthase.

The possible role of calmodulin in cell wall formation and chitin synthesis was studied in Neurospora crassa by examining the effects of anti-calmodulin agents on protoplast regeneration and possible associations between chitin synthase and calmodulin related proteins in microsomal isolates. Protoplast regeneration was inhibited by trifluoperazine (> 20 microM), an anticalmodulin agent. Chitin synthase activity in microsomes was associated with that of calmodulin-dependent protein kinase and inhibited by trifluoperazine (100 microM). In vitro activity of chitin synthase was enhanced upon inclu

The interaction of a new anti-tumour drug, KAR-2 with calmodulin.

1. KAR-2 (3"-(beta-chloroethyl)-2",4"-dioxo-3,5" -spiro-oxazolidino-4-deacetoxy-vinblastine) is a semisynthetic bis-indol derivative, with high anti-microtubular and anti-tumour activities but with low toxicity. KAR-2, in contrast to other biologically active bis-indols (e.g. vinblastine) did not show anti-calmodulin activity in vitro (enzyme kinetic, fluorescence anisotropy and immunological tests). 2. Direct binding studies (fluorescence resonance energy transfer, circular dichroism) provided evidence for the binding of KAR-2 to calmodulin. The binding affinity of KAR-2 to calmodulin (dissoc

Molecular cloning and analysis of the cDNA for an auxin-regulated calmodulin gene.

An auxin-regulated calmodulin cDNA (arCaM) were isolated by differential screening from a mung bean (Vigna radiata) cDNA library. The expression of the arCaM transcript in the etiolated mung bean hypocotyl was examined by RNA gel blot analysis. The arCaM transcript was induced depending on indole-3-acetic acid (IAA) concentrations. An increase in level of the arCaM transcript upon treatment of hypocotyl segments with 10 microM IAA was detected after 1 h and a maximum level was detected at 2 h. Induction of the arCaM transcript occurred upon treatment with 10 microM 2,4-dichlorophenoxyacetic ac

Calcium and calmodulin inhibit phosphorylation of a novel auditory nerve protein.

The growing use of cochlear prosthetic devices and demonstrations of direct ototoxic insult to spiral ganglion neurons make it imperative to gain an understanding of intracellular biochemical regulation in primary sensory neurons. Calcium and calmodulin regulate many aspects of neuronal cellular physiology through stimulation of protein kinase activity. We have previously demonstrated the presence of calmodulin-dependent protein kinase substrates in the guinea pig modiolus and, additionally, the presence of two proteins (12 kDa and 81 kDa, designated as p12 and p81) whose phosphorylation is bl

Effect of calmodulin antagonists on the growth and graviresponsiveness of primary roots of maize.

We examined the effect of calmodulin (CaM) antagonists applied at the root tip on root growth, gravity-induced root curvature, and the movement of calcium across the root tip and auxin (IAA) across the elongation zone of gravistimulated roots. All of the CaM antagonists used in these studies delayed gravity-induced curvature at a concentration (1 micromole) that did not affect root growth. Calmodulin antagonists (> or = 1 micromole) inhibited downward transport of label from 45Ca2+ across the caps of gravistimulated roots relative to the downward transport of 45Ca2+ in gravistimulated roots wh

Modulation of calmodulin properties by amiodarone and its major metabolite desethylamiodarone.

Long-term amiodarone therapy is invariably associated with some side effects. Although its mechanism of action, as an antiarrhythmic drug is well understood, the side effect profile of amiodarone is not yet established. To determine possible mechanisms, the interaction of amiodarone and its major metabolite desethylamiodarone with calmodulin was investigated, since calmodulin is known to regulate Ca2+ transport, cell proliferation and the enzymes involved in signal transduction and nucleotide metabolism. The interaction between the drugs and calmodulin was studied by monitoring intrinsic tyros

Involvement of calcium, calmodulin and protein phosphorylation in morphogenesis of Candida albicans.

N-Acetyl-D-glucosamine-induced germ tube formation in Candida albicans at 37 degrees C was accompanied by an increase in the rate of protein phosphorylation. The calmodulin antagonist trifluoperazine and the Ca2+ ionophore A23187, which inhibited germ tube formation, also reduced the rate of phosphorylation. The rate of phosphorylation was also reduced when cells were incubated at 25 degrees C, which favoured yeast-phase growth. Two-dimensional SDS-PAGE analysis of phosphoproteins from germ-tube-forming and yeast cells revealed two germ-tube-specific and three yeast-specific phosphoproteins. G

Effects of calmodulin antagonists on radiation-induced lipid peroxidation in microsomes.

Rat liver microsomes were irradiated with gamma-rays at a dose rate of 1.31 Gys-1. The extent of lipid peroxidation, measured in terms of malondialdehyde (MDA) formed, increased with radiation dose. The presence of calmodulin antagonists during irradiation decreased lipid peroxidation. The order of their protective efficiency was: chlorpromazine (CPZ) greater than promethazine (PMZ) greater than trimeprazine (TMZ). Their protective effect was diminished in the presence of ferrous (Fe2+) ions and was restored on addition of EDTA. However, calmodulin antagonists considerably inhibited radiation-

Is calmodulin inhibition involved in shape transformations induced by amphiphiles in erythrocytes?

Potent antihaemolytic and shape transforming amphiphilic compounds were studied for their ability to inhibit calmodulin-activated phosphodiesterase activity. Some echinocytogenic and stomatocytogenic amphiphiles were potent calmodulin inhibitors. The most potent echinocytogenic and stomatocytogenic amphiphiles, however, had no or only weak inhibitory effect. Our results show that there is no causal relationship between the ability of amphiphiles to induce antihaemolysis or shape transformations in erythrocytes and their ability to inhibit calmodulin-activated phosphodiesterase activity, and it

Analysis of the calcium-dependent interaction of calmodulin with bovine serum albumin.

An air-driven ultracentrifuge has been used to investigate the calcium-dependent association between calmodulin and bovine serum albumin. Procedures were described which allowed the interaction to be analyzed to yield the equilibrium constant. At low ionic strength (25 mM Tris-HCl, pH 7.5, pCa 6.68, 9 degrees C) the equilibrium constant for the interaction was estimated to be 2.1 X 10(4) M-1, while at high ionic strength (25 mM Tris-HCl, pH 7.5, 150 mM KCl, pCa 6.68, 9 degrees C) the value was 4.5 X 10(3) M-1. Under similar conditions, calmodulin was also found to interact with beta-lactoglobu

Identification and characterization of calmodulin-binding proteins in islet secretion granules.

The binding of 125I-calmodulin to intact secretion granules and protein gel blots of secretion granules from pancreatic islet tissue was examined. Binding of 125I-calmodulin to intact secretion granules was Ca2+-dependent and inhibited by the calmodulin inhibitors trifluoperazine and calmidazolium. Binding was inhibited by excess (200 nM) unlabeled calmodulin, but not by parvalbumin, a Ca2+-binding protein which has little sequence homology to calmodulin. In order to study the binding of calmodulin to specific secretion granule proteins, secretion granules were solubilized, and the solubilized
