Cancer-Associated fibroblasts regulate the development of cholangiocarcinoma through IL-6/STAT3/AKR1C3 signaling axis.

Cancer-associated fibroblasts (CAFs) are the dominant component of the tumor microenvironment (TME), which contributes to tumor progression. Aldo-keto reductase 1 family member C3 (AKR1C3) has been correlated with the development of various kinds of cancers. Nevertheless, the effect and mechanism of CAFs on AKR1C3 in cholangiocarcinoma (CCA) remain unelucidated. Q-PCR assay and IHC were conducted to detect the expression of AKR1C3 in CCA tissues. Subsequently, CCK8 assay, colony formation, crystal violet assay, apoptosis assay, glucose uptake, and lactate production assay were performed to inv

FOXO1 Is Required for Growth and Viability of Cancer-Associated Fibroblasts in Human Breast Carcinomas.

Carcinoma-associated fibroblasts (CAFs), frequently present in the stroma of human breast carcinomas, influence tumor characteristics. A forkhead box protein O1 (FOXO1) transcription factor is a critical mediator of the cellular responses to oxidative stress in various cell types. However, the roles of FOXO1 in CAFs have been poorly understood. Here, we show more intense FOXO1 staining in stromal fibroblasts in human breast cancers compared to those in non-cancerous regions. Notably, stronger stromal FOXO1 staining is significantly associated with poorer outcomes in 237 breast cancer patients.

The Origin of Cancer-Associated Fibroblasts (CAFs) in Brain Metastases: Seven Hypotheses and Current Evidence.

Brain metastases have traditionally been investigated primarily with regard to interactions between tumor cells, the surrounding brain microenvironment, and associated inflammatory responses. By contrast, the presence of a true intratumoral stromal compartment in brain metastases has received much less attention, partly because the normal brain parenchyma lacks classical interstitial fibroblasts. However, recent histological, transcriptomic, and spatial studies suggest that at least a subset of carcinoma brain metastases contains fibroblast-like or cancer-associated fibroblast-like cells assoc

H3K18 lactylation in cancer-associated fibroblasts drives malignant pleural effusion progression via TNFR2+ Treg recruitment.

Tumor necrosis factor receptor 2-positive regulatory T (TNFR2+ Treg) cells, the most suppressive subset of Treg cells, are enriched in malignant pleural effusion (MPE), contributing to disease progression. However, the underlying mechanisms responsible for their accumulation remain unclear. Here we demonstrate that the C-X-C motif chemokine ligand 16 (CXCL16)/C-X-C chemokine receptor type 6 (CXCR6) axis plays a critical role in recruiting TNFR2+ Treg cells to MPE, with cancer-associated fibroblasts serving as the primary source of CXCL16. Mechanistically, under the hypoxic conditions prevailin

Nicotinamide N-methyltransferase negatively regulates metastasis-promoting property of cancer-associated fibroblasts in lung adenocarcinoma.

BACKGROUND: Recurrence and metastasis remain significant challenges in lung adenocarcinoma (LUAD) after radical resection. The mechanisms behind the recurrence and metastasis of LUAD remain elusive, and deregulated cellular metabolism is suspected to play a significant role. This study explores the metabolic and epigenetic regulation mediated by nicotinamide N-methyl transferase (NNMT) in LUAD. METHODS: Untargeted metabolomic analyses were performed to detect metabolism irregularities. Single-cell RNA sequencing (RNA-seq) databases and multiplex immunofluorescence analysis were used to identi

The role of transcription factors in the crosstalk between cancer-associated fibroblasts and tumor cells.

BACKGROUND: Transcription factors (TFs) fulfill a critical role in the formation and maintenance of different cell types during the developmental process as well as disease. It is believed that cancer-associated fibroblasts (CAFs) are activation status of tissue-resident fibroblasts or derived from form other cell types via transdifferentiation or dedifferentiation. Despite a subgroup of CAFs exhibit anti-cancer effects, most of them are reported to exert effects on tumor progression, further indicating their heterogeneous origin. Synergistic Chemoimmunotherapy Augmentation via Sequential Nanocomposite Hydrogel-Mediated Reprogramming of Cancer-Associated Fibroblasts in Osteosarcoma.

In osteosarcoma, immunotherapy often faces hurdles posed by cancer-associated fibroblasts (CAFs) that secrete dense extracellular matrix components and cytokines. Directly removing CAFs may prove ineffective and even promote tumor metastasis. To address this challenge, a sequential nanocomposite hydrogel that reshapes CAF behavior is developed, enhancing tumor-infiltrating T-cells in osteosarcoma. The approach utilizes an injectable blend of carboxymethyl chitosan and tetrabasic polyethylene glycol, forming a hydrogel for controlled release of a potent CAF suppressor (Nox4 inhibitor, Nox4i) an

Pancreatic tumor eradication via selective Pin1 inhibition in cancer-associated fibroblasts and T lymphocytes engagement.

Cancer associated fibroblasts (CAFs) support tumors via multiple mechanisms, including maintaining the immunosuppressive tumor microenvironment and limiting infiltration of immune cells. The prolyl isomerase Pin1, whose overexpression in CAFs has not been fully profiled yet, plays critical roles in tumor initiation and progression. To decipher effects of selective Pin1 inhibition in CAFs on pancreatic cancer, here we formulate a DNA-barcoded micellular system (DMS) encapsulating the Pin1 inhibitor AG17724. DMS functionalized with CAF-targeting anti-FAP-α antibodies (antiCAFs-DMS) can selective

Biejiajian pill inhibits progression of hepatocellular carcinoma by downregulating PDGFRβ signaling in cancer-associated fibroblasts.

ETHNOPHARMACOLOGICAL RELEVANCE: Biejiajian pill (BJJP) is a canonical formula that is clinically used to treat chronic liver disease, especially to decrease the incidence of hepatocellular carcinoma (HCC). However, the mechanisms underlying the prevention of HCC progression by BJJP remain unclear. AIM OF THE STUDY: This study aimed to determine whether BJJP inhibits HCC progression by downregulating platelet-derived growth factor receptor beta (PDGFRβ) signaling in cancer-associated fibroblasts (CAFs) in a mouse model of diethylnitrosamine (DEN)/carbon tetrachloride (CCl4)-induced HCC. Mechanism of Cancer-Associated Fibroblasts-Derived NEAT1 Promoting Pancreatic Cancer Cell Stemness via the miR-101-3p/KDM3A Axis.

Pancreatic cancer (PC), a malignant neoplasm of the gastrointestinal tract, is linked to dismal prognoses and a restricted response to chemotherapy. Long noncoding RNAs (lncRNAs) play critical roles in regulating PC stemness. This study investigated the mechanism of cancer-associated fibroblasts (CAFs)-derived lncRNA nuclear paraspeckle assembly transcript 1 (NEAT1) promoting PC cell stemness via the microRNA-101-3p (miR-101-3p)/lysine demethylase 3A (KDM3A) axis. Human pancreatic CAFs were cultured in vitro and transfected with small interfering (si)-NEAT1. Extracellular vesicles (EVs) were i

Cancer-associated fibroblasts drive gemcitabine resistance in pancreatic cancer by promoting ferroptosis resistance via the PAX6/PAK3/SLC3A2 axis.

Gemcitabine serves as a cornerstone chemotherapeutic agent for pancreatic cancer; however, gemcitabine resistance remains a major obstacle to improving treatment efficacy. The aim of our study was to elucidate the underlying mechanisms of gemcitabine resistance in pancreatic cancer. Through transcriptomic sequencing of gemcitabine-resistant pancreatic cancer cells, we found that p21-activated kinase 3 (PAK3) was significantly upregulated in both gemcitabine-resistant cell lines and tissues. In gemcitabine-resistant pancreatic cancer cells, PAK3 phosphorylates serine 175 of SLC3A2, thereby enha

Cancer-associated fibroblasts promote osimertinib resistance in non-small cell lung cancer cells via METTL1-mediated NET1 m7G modification.

Osimertinib resistance remains a major challenge in the treatment of non-small cell lung cancer (NSCLC). Cancer-associated fibroblasts (CAFs) are the most abundant stromal cells in tumor microenvironment (TME), however, its role in osimertinib resistance in NSCLC is not fully understood. In this study, it was found that CAFs promoted osimertinib resistance in NSCLC cells via elevating RNA m7G modification. Methyltransferase 1 (METTL1) in NSCLC cells mediated CAFs' effect on m7G modification, and METTL1 was associated with NSCLC progression and poor prognosis. Further study demonstrates that CA

Inhibition of Gemcitabine-Induced Autophagy in Cancer-Associated Fibroblasts Reduces Collagen I and Enhances Chemotherapeutic Efficacy in Pancreatic Cancer.

In pancreatic cancer, increased collagen I impairs the efficacy of gemcitabine; however, the role of gemcitabine itself in collagen I accumulation remains unclear. This study aims to explore the mechanism of gemcitabine-induced fibrosis and provide new insights to enhance its therapeutic efficacy. We analyzed COL1A1 expression in pancreatic cancer patient tumor tissues and found that gemcitabine treatment upregulated COL1A1 expression. Subsequently, cancer-associated fibroblasts (CAFs) were modeled by inducing human adipose-derived mesenchymal stem cells with tumor-derived exosomes. Using the

Exosomes Derived from Non-small Cell Lung Cancer Cells Transfer LINC00467 to Induce the Proliferation of Cancer-associated Fibroblasts.

Cancer-associated fibroblasts (CAFs) significantly contribute to non-small cell lung cancer (NSCLC) progression by remodeling the extracellular matrix and mediating paracrine signaling. Although exosomal lncRNAs are key regulators of tumor-stroma interactions, the specific functions of individual lncRNAs remain poorly understood. LINC00467 and miR-133b were selected for further investigation in this study based on previous reports, prior evidence of lncRNA-miRNA interaction, and preliminary experimental observations in NSCLC-derived exosomes and fibroblast activation models. Exosomal CCT6A Secreted by Cancer-Associated Fibroblasts Interacts with β-Catenin to Enhance Chemoresistance and Tumorigenesis in Gastric Cancer.

Cancer-associated fibroblasts (CAFs) are pivotal components of the tumor microenvironment that drive gastric cancer (GC) progression and chemoresistance. Here, CCT6A is identified as a CAF-derived factor whose high expression is positively associated with GC progression and plays a key regulatory role in stemness, chemoresistance, and glucose metabolism. In a co-culture system of CAFs and cancer cells, it is discovered that CCT6A is transferred from CAFs to tumor cells mainly via exosomal transport, thereby enhancing stemness, chemoresistance, and glycolysis. Mechanistically, CCT6A interacts w

Fusobacterium nucleatum increases CTGF expression through TLR2-YAP signaling axis in cancer-associated fibroblasts, thereby promoting colorectal cancer progression.

Fusobacterium nucleatum (F. nucleatum) is increasingly recognized as a microbial driver of colorectal cancer (CRC) progression through immune modulation, inflammation dysregulation, and activation of oncogenic signaling cascades. Cancer-associated fibroblasts (CAFs), the predominant stromal constituents within the tumor microenvironment (TME), play pivotal roles in orchestrating tumorigenic processes. While F. nucleatum's interactions with epithelial and immune compartments have been extensively characterized, its immunological crosstalk with CAFs remains elusive. Through multimodal experiment

Cancer-associated fibroblasts promote cancer stemness by inducing expression of the chromatin-modifying protein CBX4 in squamous cell carcinoma.

The chromobox-containing protein CBX4 is an important regulator of epithelial cell proliferation and differentiation, and has been implicated in several cancer types. The cancer stem cell (CSC) population is a key driver of metastasis and recurrence. The undifferentiated, plastic state characteristic of CSCs relies on cues from the microenvironment. Cancer-associated fibroblasts (CAFs) are a major component of the microenvironment that can influence the CSC population through the secretion of extracellular matrix and a variety of growth factors. Here we show CBX4 is a critical regulator of the

In this study, we demonstrated that senescent cancer-associated fibroblasts (senCAF) drive lymphatic remodeling and LNM in early-stage PDAC. Mechanistically, senCAFs increased glucose metabolism and lactate production, which activated lactylation-mediated serine metabolism to protect lymphatic endothelial cells from oxidative stress. Moreover, we discovered CCR4+ regulatory T cells from the draining
