The Varicella-zoster virus ORF54 gene product encodes the capsid portal protein, pORF54.

The Varicella-zoster virus (VZV) ORF54 gene was characterized using a guinea pig antiserum prepared to a GST-pORF54 fusion protein. A protein of the predicted size, 87kDa, was detected in VZV-infected MeWo cells but not in mock-infected cells. Sucrose density gradient fractionation of pORF54 expressed in a recombinant baculovirus system resulted in samples containing enriched amounts of pORF54. Electron microscopic analysis suggested that the ORF54 gene encodes a protein that assembles into ring-like portal structures similar to those observed for numerous bacteriophages and other herpesviruse

Bacteriophage T4 capsid: a unique platform for efficient surface assembly of macromolecular complexes.

We report the first description of a macromolecular complex display system using bacteriophage T4. Decorated with two dispensable outer capsid proteins, Hoc (155 copies) and Soc (810 copies), the 120 nm x 86 nm T4 capsid particle offers a unique binding site-rich platform for surface assembly of hetero-oligomeric complexes. To display the 710 kDa anthrax toxin complex, two bipartite functional fusion proteins, LF-Hoc and LFn-Soc, were constructed. Using a defined in vitro binding system, sequential assembly was performed by first attaching LF-Hoc and/or LFn-Soc to hoc-soc- phage, saturating th

Arc capsid signaling between serotonergic and dopaminergic neurons sets sleep depth in Drosophila.

Arc genes evolved from Ty3 retrotransposons and encode proteins that self-assemble into virus-like capsids that package and transfer RNA between cells. This capsid-forming property may mediate a form of intercellular signaling distinct from classical synaptic transmission, but whether it operates within a defined neuronal system to control an ongoing behavior remains unclear. Here we show that Arc-dependent capsid signaling links serotonergic and PAM neurons to set sleep depth in Drosophila melanogaster . Loss of dArc genes deepened and consolidated sleep, increasing both sleep depth and the a

Purification of Disassembled Capsid Protein by Hydroxyapatite Chromatography Enhances Virus-Like Particle Recovery.

Virus-like particles (VLPs) are a promising modality with extensive applications in prophylactic and therapeutic vaccine design. Human Papillomavirus (HPV) vaccines are one such example where VLP vaccine applications have led to the successful reduction in HPV-associated diseases, such as cervical cancer. Current purification approaches utilize hydroxyapatite (HA) chromatography in bind-and-elute mode to purify HPV VLPs from host-cell impurities such as proteins and nucleic acids, though binding capacity for VLPs is low due to mass transfer limitations related to their large particle size. The

Recombinant Production and Immunological Evaluation of Tobamovirus fructirugosum (ToBRFV) Capsid and Movement Proteins.

Tomato brown rugose fruit virus (Tobamovirus fructirugosum, ToBRFV) is an emerging tobamovirus threatening tomato and pepper production worldwide. Accurate and scalable detection methods are essential to limit its agricultural impact. In this study, we report the recombinant production and characterization of ToBRFV capsid (CPToBRFV) and movement (MPToBRFV) proteins as candidate antigens for serological detection. Both coding sequences were amplified from infected tomato tissue and heterologously expressed in Escherichia coli BL21(DE3). CPToBRFV exhibited partial solubility under low-temperatu

Structural organization of HBV pgRNA genome driven by phase separation in capsid confinement.

Viruses rely on the precise packaging of their genomes within a capsid to execute essential life-cycle events, yet the principles governing genome structural organization in this confined environment remain elusive. Here, we reveal that hepatitis B virus (HBV) pregenomic RNA (pgRNA) exploits liquid-liquid phase separation (LLPS) inside the capsid to sculpt its architecture. Multiscale molecular dynamics (MD) simulations, supplemented by biochemical assays, show that pgRNA coalesces into a hollow, shell-like condensate along the inner capsid surface, with coexisting low- and high-density region

Alphaviral Capsid Proteins Inhibit Stress Granule Assembly via Competitive RNA Binding With G3BP1.

Viral infection is one of the conditions that induce stress granule (SG) formation, a cellular defense mechanism that exerts antiviral effects. To counteract this host response, viruses have evolved a broad spectrum of strategies to inhibit SG formation. However, the molecular mechanisms underlying SG inhibition remain poorly understood. The nucleocapsid proteins play a critical role in virus replication and host interaction. Here, using Semliki Forest Virus (SFV) as a model, we uncover the function of the alphavirus nucleocapsid in SG inhibition. This inhibitory function depends on oligomeriz

Identification of a capsid-derived Zika virus epitope with high IgG discriminatory performance.

The identification of specific serological targets for Orthoflavivirus zikaense (ZIKV) remains challenging due to the high degree of structural similarity and antibody cross-reactivity among co-circulating arboviruses, particularly dengue virus (DENV). In this context, immunoinformatics-based approaches may contribute to the rational selection of candidate epitopes with potentially improved analytical specificity. In the present study, five conserved ZIKV peptides previously identified through an immunoinformatic pipeline were experimentally evaluated as candidate serological targets. Peptide

Profiling Human Papillomavirus Lineage-Specific Capsid Antigenicity With Geographically Diverse Natural Infection Antibodies.

Human papillomavirus variants are classified into lineages based on their whole genome sequence, but the impact of lineage variation on the structure or function of encoded proteins is unclear. We used a global panel of lineage-specific natural infection sera to assess antibody-binding specificity for lineage-specific L1L2 antigens, and we used these data to create relational antigenic maps. Merging these data with neutralizing antibody data demonstrated similar spatial geometry and revealed dependency on a limited number of amino residues on the capsid surface. These data inform the degree of

Membrane Expression Enhances Folding, Multimeric Structure Formation, and Immunogenicity of Viral Capsid Proteins.

Viral capsid proteins are widely explored for subunit vaccine development but are often hampered by their complexity of production and low immunogenicity. Here, we report a simple approach to overcoming these challenges by combining mRNA vaccine technology with protein engineering. Using African swine fever virus (ASFV) capsid proteins P72 and penton as models, we engineered them into membrane-bound and secreted forms and compared their immunogenicity to that of the native intracellular form in mice and pigs through mRNA vaccination. The membrane-bound and secreted P72 and penton folded into t

Exploiting the chikungunya virus capsid protein: a focused target for antiviral therapeutic development.

Chikungunya disease is spread by the bite of infected Aedes mosquitoes. It is considered a neglected tropical disease that has the potential to cause sporadic epidemics in naive populations. Despite the substantial investment in research, there are no approved antiviral treatments for chikungunya. Several screening approaches have been used to identify potential antiviral molecules that target the whole virus, viral proteins, and viral-host interactions, often in conjunction with computational studies. The genome of chikungunya virus (CHIKV) encodes four nonstructural and five structural prote

The role of chikungunya virus capsid-viral RNA interactions in programmed ribosomal frameshifting.

UNLABELLED: Chikungunya virus (CHIKV) is a globally significant arthropod-borne virus that causes outbreaks in tropical and subtropical regions and is rapidly expanding in geographical range. Infection can lead to acute febrile illness with debilitating joint pain that may persist as chronic arthritis. Efficient CHIKV replication and virion assembly rely on coordinated interactions between viral RNA and the capsid protein, a structural component essential for particle formation. In addition to its well-established structural role, the alphavirus capsid protein has been shown in Sindbis virus t

Adeno-associated virus serotype 2 capsid variants for improved liver-directed gene therapy.

BACKGROUND AND AIMS: Current liver-directed gene therapies look for adeno-associated virus (AAV) vectors with improved efficacy. With this background, capsid engineering is explored. Whereas shuffled capsid library screenings have resulted in potent liver targeting variants with one first vector in human clinical trials, modifying natural serotypes by peptide insertion has so far been less successful. Here, we now report on two capsid variants, MLIV.K and MLIV.A, both derived from a high-throughput in vivo AAV peptide display selection screen in mice. APPROACH AND RESULTS: The variants transd

A single nanobody neutralizes multiple epochally evolving human noroviruses by modulating capsid plasticity.

Acute gastroenteritis caused by human noroviruses (HuNoVs) is a significant global health and economic burden and is without licensed vaccines or antiviral drugs. The GII.4 HuNoV causes most epidemics worldwide. This virus undergoes epochal evolution with periodic emergence of variants with new antigenic profiles and altered specificity for histo-blood group antigens (HBGA), the determinants of cell attachment and susceptibility, hampering the development of immunotherapeutics. Here, we show that a llama-derived nanobody M4 neutralizes multiple GII.4 variants with high potency in human intesti

Chemical modification of AAV9 capsid with N-ethyl maleimide alters vector tissue tropism.

Although more adeno-associated virus AAV-based drugs enter the clinic, vector tissue tropism remains an unresolved challenge that limits its full potential despite that the tissue tropism of naturally occurring AAV serotypes can be altered by genetic engineering capsid vie DNA shuffling, or molecular evolution. To further expand the tropism and thus potential applications of AAV vectors, we utilized an alternative approach that employs chemical modifications to covalently link small molecules to reactive exposed Lysine residues of AAV capsids. We demonstrated that AAV9 capsid modified with N-e

Antiviral compounds modulate elasticity, strength and material fatigue of a virus capsid framework.

This study investigates whether the biochemical and antiviral effects of organic compounds that bind different sites in the mature human immunodeficiency virus capsid may be related to the modulation of different mechanical properties of the protein lattice from which the capsid is built. Mechanical force was used as a probe to quantify, in atomic force microscopy experiments at physiological pH and ionic strength, ligand-mediated changes in capsid lattice elasticity, breathing, strength against local dislocation by mechanical stress, and resistance to material fatigue. The results indicate th

Integrative structural biology of HIV-1 capsid protein assemblies: combining experiment and computation.

HIV-1 is the causative agent of acquired immunodeficiency syndrome (AIDS), a global pandemic that has claimed 32.7 million lives since 1981. Despite decades of research, there is no cure for the disease, with 38 million people currently infected with HIV. Attractive therapeutic targets for drug development are mature HIV-1 capsids, immature Gag polyprotein assemblies, and Gag maturation intermediates, although their complex architectures, pleomorphism, and dynamics render these assemblies challenging for structural biology. The recent development of integrative approaches, combining experiment

Design, synthesis, and antiviral activity of phenylalanine derivatives as HIV-1 capsid inhibitors.

The HIV-1 Capsid (CA) is considered as a promising target for the development of potent antiviral drugs, due to its multiple roles during the viral life cycle. Herein, we report the design, synthesis, and antiviral activity evaluation of series of novel phenylalanine derivatives as HIV-1 CA protein inhibitors. Among them, 4-methoxy-N-methylaniline substituted phenylalanine (II-13c) and indolin-5-amine substituted phenylalanine (V-25i) displayed exceptional anti-HIV-1 activity with the EC50 value of 5.14 and 2.57 μM respectively, which is slightly weaker than that of lead compound PF-74 (EC50 =

Genetic and Chemical Capsid Modifications of Adenovirus Vectors to Modulate Vector-Host Interactions.

Adenovirus-based vectors are playing an important role as efficacious genetic vaccines to fight the current COVID-19 pandemic. Furthermore, they have an enormous potential as oncolytic vectors for virotherapy and as vectors for classic gene therapy. However, numerous vector-host interactions on a cellular and noncellular level, including specific components of the immune system, must be modulated in order to generate safe and efficacious vectors for virotherapy or classic gene therapy. Importantly, the current widespread use of Ad vectors as vaccines against COVID-19 will induce antivector imm
