Luteolin protects crucian carp against Aeromonas hydrophila by modulating inflammation-apoptosis axis.

Aeromonas hydrophila infection is a major bacterial disease that seriously threatens the healthy development of crucian carp aquaculture, highlighting the important practical value of developing safe and effective antibiotic alternatives. This study investigated the protective effects of the natural flavonoid luteolin against A. hydrophila infection in crucian carp and explored its underlying molecular mechanisms. The results showed that dietary supplementation with luteolin (20 μg/g diet per day for 4 weeks) significantly improved the survival rate of infected fish and alleviated histopatholo

Grass carp reovirus VP4 manipulates TOLLIP to degrade STING for inhibition of IFN production.

UNLABELLED: Although fish possess an effective interferon (IFN) system to defend against viral infection, grass carp reovirus (GCRV) still causes epidemic hemorrhagic disease and tremendous economic loss in grass carp. Therefore, it is necessary to investigate the immune escape strategies employed by GCRV. In this study, we show that the structural protein VP4 of GCRV (encoded by the S6 segment) significantly restricts IFN expression by degrading stimulator of IFN genes (STING) through the autophagy-lysosome-dependent pathway. First, overexpression of VP4 inhibited the expression of IFN induce

Mg2+ inhibits infection of spring viraemia of carp virus (SVCV) by enhancing IRF3 expression.

Magnesium ions (Mg2+), essential divalent cations, play a pivotal role in orchestrating a myriad of physiological and biochemical processes. However, the intricate relationship between Mg2+ homeostasis and antiviral immunity in fish remains largely unexplored. In this study, we found that Mg2+ enhances the interferon (IFN) response by increasing the expression of IFN regulatory factor 3 (IRF3) to inhibit spring viraemia of carp virus (SVCV) infection. In vivo studies showed Mg2+ increases the survival rate of SVCV-infected zebrafish while reducing viral transcription and load in liver tissues.

Exploring the dietary impacts and potential metabolic mechanisms of astaxanthin in common carp (Cyprinus carpio).

This study investigated the dietary effects and metabolic mechanisms of astaxanthin (Ast) in common carp (Cyprinus carpio). Supplementation with 100 mg/kg Ast resulted in a significant 19.83 % increase in growth. Data from the electronic tongue indicated that dietary Ast enhanced fillet sweetness while reducing bitterness and saltiness. Colorimetric and computer vision analyses showed a shift in fillet color toward yellow-red (amber) hue, with a* values rising by up to 59.93 % (redness) and b* values by 40.84 % (yellowness). Carotenoid quantification revealed that Ast was metabolized into viol

Transcriptomic Responses to Koi Herpesvirus in Isolated Blood Leukocytes from Infected Common Carp.

Koi herpesvirus (KHV, CyHV-3) causes severe economic losses in carp farms. Its eradication is challenging due to the establishment of latency in blood leukocytes and other tissues. To understand the molecular mechanisms leading to KHV infection in leukocytes, common carp were bath-exposed to KHV at 17 °C. After confirming the presence of viral transcripts in blood leukocytes at ten days post infection, RNA-Seq was performed on peripheral blood leukocytes on the Illumina NovaSeq. KHV infection triggered a robust immune response mediated by pattern recognition receptors, mainly toll-like recepto

Deltamethrin increased susceptibility to Aeromonas hydrophila in crucian carp through compromising gill barrier.

Pyrethroids serve as a significant method for managing and preventing parasitic diseases in fish. Among these, deltamethrin (DEL) is used extensively in aquatic environments. Our previous work has been confirmed that DEL exposure can induce oxidative stress and immunosuppression on the gill mucosal barrier of crucian carp (Carassius auratus). However, it is not clear whether DEL affects the susceptibility of farmed fish to bacterial infection. In this study, fish was pre-exposed to different DEL concentration (0, 0.3 and 0.6 μg L-1) and then challenged by immersion with Aeromonas hydrophila (1

Quercetin alleviates difenoconazole-induced growth inhibition in carp through intestinal-brain axis.

Difenoconazole (DIF) is frequently used for the management of fungal infections in fruit and vegetables and excessive residues in the aquatic environment can have adverse effects on fish such as growth inhibition. A treatment based on the dietary additive quercetin (QUE) is a promising approach to positively regulate the state of fish growth. This study focused on whether and how QUE alleviated DIF-induced growth inhibition in fish. In this study, carp were exposed to DIF (0.3906 mg/L) for consecutive 30 d, which showed growth inhibition. Disruption of the intestinal barrier led to elevated le

Preliminary study of BF/C2 on immune mechanism of grass carp against GCRV infection.

BF/C2 is a crucial molecule in the coagulation complement cascade pathway and plays a significant role in the immune response of grass carp through the classical, alternative, and lectin pathways during GCRV infection. In vivo experiments demonstrated that the mRNA expression levels of BF/C2 (A, B) in grass carp positively correlated with GCRV viral replication at various stages of infection. Excessive inflammation leading to death coincided with peak levels of BF/C2 (A, B) mRNA expression and GCRV viral replication. Correspondingly, BF/C2 (A, B) recombinant protein, CIK cells and GCRV co-incu

Silybin mitigates chronic Avermectin exposure-induced intestinal damage and growth inhibition in carp.

Avermectins, as a new type of environmental pollutant, have received significant attention in recent years. Previous research has shown that acute exposure to avermectins can induce oxidative stress and inflammation in non-target fish species, such as carp. Flavonoid lignans, particularly Silybin, have demonstrated promising biological activities, including regulation of non-alcoholic fatty liver and cerebral ischemia-reperfusion injury. This study aims to investigate the impact of dietary supplementation with Silybin on the intestinal damage in carp caused by chronic exposure to avermectins a

Enrichment and distribution of 3,6-dichlorocarbazole in red crucian carp (Carassius auratus) and its hepatotoxicity.

Polyhalogenated carbazoles (PHCZs) are a class of organohalogen compounds where the hydrogen atom on the carbazole ring is replaced by a halogen atom. In recent years, PHCZs have drawn increasing concern due to their persistence, dioxin-like toxicity, bioaccumulation, potential ecological hazards and widespread occurrence in the environment. Current research on the enrichment and depuration of PHCZs in biological tissues and organs is insufficient, and the liver toxicity is unclear. Herein, to understand the enrichment and elimination of 3,6-DCCZ in fish tissues and organs as well as the hepat

Evaluation of metabolic stress status in common carp (Cyprinus carpio) exposed to the fungicide folpet.

Among pesticides, fungicides are the most extensively used ones in agriculture and their effects on fish health and indirectly human health are needed to evaluate. Folpet has been used for over 50 years as a fungicide across the world. The mechanism of action of folpet on non-target aquatic organisms is so poorly understood and there is no available information about potential acute toxicity of folpet and its mechanism of action in non-target aquatic organisms. With this motivation, two successive experiments were set up: first, 96 h-LC50 value of folpet for common carp (Cyprinus carpio) was d

IRF2 Cooperates with Phosphoprotein of Spring Viremia of Carp Virus to Suppress Antiviral Response in Zebrafish.

IFN regulatory factor (IRF) 2 belongs to the IRF1 subfamily, and its functions are not yet fully understood. In this study, we showed that IRF2a was a negative regulator of the interferon (IFN) response induced by spring viremia of carp virus (SVCV). Irf2a-/- knockout zebrafish were less susceptible to SVCV than wild-type fish. Transcriptomic analysis reveals that differentially expressed genes (DEGs) in the irf2a-/- and irf2a+/+ cells derived caudal fins were mainly involved in cytokine-cytokine receptor interaction, mitogen-activated protein kinase (MAPK) signaling pathway, and transforming

Proteomics-based molecular and functional characteristic profiling of muscle tissue in Triploid crucian carp.

Triploid crucian carp (TCC) is a kind of artificially bred fish with huge economic value to China. It has several excellent characteristics, such as fast growth, strong disease resistance and delicious taste. However, as a regionally specific fish, the underlying molecular mechanisms of these characteristics are largely unknown. In this study, we performed quantitative proteomics on the muscle tissues of TCC and its parents, allotetraploid (♂), red crucian carp (♀) and common carp. Combined with multiple bioinformatic analysis, we found that the taste of TCC can be mainly attributed to umami a

Cyanobacteria Microcystis aeruginosa Contributes to the Severity of Fish Diseases: A Study on Spring Viraemia of Carp.

Fish are exposed to numerous stressors in the environment including pollution, bacterial and viral agents, and toxic substances. Our study with common carps leveraged an integrated approach (i.e., histology, biochemical and hematological measurements, and analytical chemistry) to understand how cyanobacteria interfere with the impact of a model viral agent, Carp sprivivirus (SVCV), on fish. In addition to the specific effects of a single stressor (SVCV or cyanobacteria), the combination of both stressors worsens markers related to the immune system and liver health. Solely combined exposure re
