[Pharmacokinetics of ceftazidime in children and newborn infants. Study in 14 patients and review of the literature].

A pharmacokinetic study of ceftazidime was conducted in 1983, together with a clinical study, on 7 neonates (3 of them premature), 4 infants and 3 children. The antibiotic was administered by slow intravenous injection in mean doses of 30 mg/kg twice in 24 h in neonates and thrice in 24 h in older children. Blood samples were collected by micropuncture, and assays were performed by the microbiological method on agar plates. Curves of plasma concentrations over time showed two slopes compatible with a two-compartment model. The alpha half-life was the same in neonates and infants; the beta half

Pseudomonas peritonitis in neutropenic rats treated with amikacin, ceftazidime and ticarcillin, alone and in combination.

Peritonitis in neutropenic rats, caused by a strain of Pseudomonas aeruginosa resistant to amikacin and ticarcillin alone but susceptible to ceftazidime and combinations of amikacin with ticarcillin, amikacin with ceftazidime and ticarcillin with ceftazidime, was investigated. Four hours after a bacterial challenge with a LD70 intraperitoneal dose of P. aeruginosa (1.5 X 10(8) cfu/ml), either amikacin, ticarcillin, ceftazidime, amikacin-ticarcillin, amikacin-ceftazidime, or ticarcillin-ceftazidime was administered at dosing intervals that mimicked the serum concentrations of the drugs found in

Evaluating the efficacy of ceftazidime/avibactam plus amikacin combination therapy as a carbapenem-sparing agent against ceftazidime/avibactam-resistant Escherichia coli using a hollow fiber infection model: a proof-of-concept study.

Carbapenems, often regarded as the last line of defense in treatment of urinary tract infections and urosepsis caused by Escherichia coli, are under pressure from emerging resistance. Thus, it is necessary to investigate carbapenem-sparing agents to mitigate the dissemination of carbapenem resistance. In this study, we aimed to evaluate the efficacy of ceftazidime/avibactam plus amikacin combination therapy against two ceftazidime/avibactam-resistant E. coli clinical isolates harboring IMP-4 carbapenemase (CTAP #226 and CTAP #233). To achieve this, we used a hollow fiber infection model (HFIM)

Ceftazidime-avibactam or best available therapy in patients with ceftazidime-resistant Enterobacteriaceae and Pseudomonas aeruginosa complicated urinary tract infections or complicated intra-abdominal infections (REPRISE): a randomised, pathogen-directed, phase 3 study.

BACKGROUND: Carbapenems are frequently the last line of defence in serious infections due to multidrug-resistant Gram-negative bacteria, but their use is threatened by the growing prevalence of carbapenemase-producing pathogens. Ceftazidime-avibactam is a potential new agent for use in such infections. We aimed to assess the efficacy, safety, and tolerability of ceftazidime-avibactam compared with best available therapy in patients with complicated urinary tract infection or complicated intra-abdominal infection due to ceftazidime-resistant Gram-negative pathogens. METHODS: REPRISE was a path

Ceftazidime-Avibactam Effectiveness in Carbapenem-Resistant Enterobacterales Bacteremia: A Retrospective Cohort Study in Korea.

BACKGROUND: Carbapenem-resistant Enterobacterales (CRE) infections pose a global health concern owing to their increasing incidence and limited treatment options. Ceftazidime-avibactam (CAZ-AVI) has emerged as a promising therapeutic option, with some studies reporting improved outcomes in CRE bacteremia. However, evidence regarding its survival benefit remains unclear, particularly in Korea. In this retrospective cohort study, we compared the effectiveness of CAZ-AVI with the best available therapy (BAT) in patients with CRE bacteremia. METHODS: We included adult patients with CRE bacteremia

Comparative pharmacokinetics of ceftazidime in Siamese crocodiles after intramuscular administration between forelimb and hindlimb.

The pharmacokinetics of ceftazidime (CAZ) were evaluated in Siamese crocodiles (Crocodylus siamensis) following a single 20 mg/kg intramuscular administration into either the forelimb or hindlimb. The pharmacokinetic profiles were substantially equivalent between the injection sites, with no significant differences (p > 0.05) in key parameters (Cmax, Tmax, AUC, t1/2λz, Cl, and bioavailability). CAZ was absorbed rapidly at both injection sites, with a median Tmax value of 0.5 h and high bioavailability (90.35% for the forelimb, 86.12% for the hindlimb), indicating efficient systemic exposure, r

Complex evolutionary trajectories in vivo of two novel KPC variants conferring ceftazidime-avibactam resistance.

More and more ceftazidime-avibactam-resistant KPC-producing Klebsiella pneumoniae have been reported with its widespread use, and the detection rate of KPC variants has increased dramatically. However, the evolutionary mechanism and fitness effects during KPC mutation remained unknown. Here, we report the complex in vivo evolutionary trajectories of two novel KPC variants, KPC-155 (L169P/GT242A) and KPC-185 (D179Y/GT242A), from K. pneumoniae in the same patient. The novel variants were shown to confer ceftazidime-avibactam resistance but restore carbapenem susceptibility based on the results o

First report of KPC variants conferring ceftazidime-avibactam resistance in Colombia: introducing KPC-197.

UNLABELLED: Resistance to ceftazidime-avibactam (CZA) due to Klebsiella pneumoniae carbapenemase (KPC) variants is increasing worldwide. We characterized two CZA-resistant clinical Klebsiella pneumoniae strains by antimicrobial susceptibility test, conjugation assays, and WGS. Isolates belonged to ST258 and ST45, and produced a KPC-31 and a novel variant KPC-197, respectively. The novel KPC variant presents a deletion of two amino acids on the Ω-loop (del_168-169_EL) and an insertion of two amino acids in position 274 (Ins_274_DS). Continued surveillance of KPC variants conferring CZA resistan

DNase I and chitosan enhance efficacy of ceftazidime to eradicate Burkholderia pseudomallei biofilm cells.

Biofilm-associated Burkholderia pseudomallei infection contributes to antibiotic resistance and relapse of melioidosis. Burkholderia pseudomallei biofilm matrix contains extracellular DNA (eDNA) that is crucial for biofilm establishment. However, the contribution of eDNA to antibiotic resistance by B. pseudomallei remains unclear. In this study, we first demonstrated in vitro that DNase I with the administration of ceftazidime (CAZ) at 24 h considerably inhibited the 2-day biofilm formation and reduced the number of viable biofilm cells of clinical B. pseudomallei isolates compared to biofilm
