LY294002-mediated inhibition of phosphatidylinositol 3-kinase activity triggers growth inhibition and apoptosis in CD40-triggered Ramos-Burkitt lymphoma B cells.

Cells of the Epstein-Barr virus genome-negative Ramos-Burkitt lymphoma (Ramos-BL) B cell line can be rescued from antigen receptor (AgR)-triggered growth inhibition and apoptosis by signals transduced through their surface CD40. This study investigates whether phosphatidylinositol 3-kinase (PI3-kinase), which has been reported to be intimately involved in the regulation of normal and neoplastic cell growth, plays a role in CD40-promoted Ramos-BL B cell survival and uses the selective and reversible PI3-kinase inhibitor, LY294002 (LY). LY-mediated inhibition of PI3-kinase activity triggers grow

Toxic effects of prenatal exposure to alcohol, tobacco and other drugs.

Tobacco, alcohol, cannabis and cocaine are the most consumed psychoactive drugs throughout the population. Prenatal exposure to these drugs could alter normal foetal development and could threaten future welfare. The main changes observed in prenatal exposure to tobacco are caused by nicotine and carbon monoxide, which can impede nutrient and oxygen exchange between mother and foetus, restricting foetal growth. Memory, learning processes, hearing and behaviour can also be affected. Alcohol may cause physical and cognitive alterations in prenatally exposed infants, fundamentally caused by alter

Adaptation to alkalosis induces cell cycle delay and apoptosis in cortical collecting duct cells: role of Aquaporin-2.

Collecting ducts (CD) not only constitute the final site for regulating urine concentration by increasing apical membrane Aquaporin-2 (AQP2) expression, but are also essential for the control of acid-base status. The aim of this work was to examine, in renal cells, the effects of chronic alkalosis on cell growth/death as well as to define whether AQP2 expression plays any role during this adaptation. Two CD cell lines were used: WT- (not expressing AQPs) and AQP2-RCCD(1) (expressing apical AQP2). Our results showed that AQP2 expression per se accelerates cell proliferation by an increase in ce

[Regulation of uterine cellular proliferation with estrogens and growth factors].

In this paper the role of estrogen and growth factors in the uterine cellular proliferation is analyzed. The evidences indicate that the estradiol-stimulate cell division is associated with the induction of expression of a variety of growth factors from the all major uterine cell types (epithelia, stroma and myometrium). These growth factors amplify the estrogen proliferation signal in autocrine and/or paracrin fashion. The best-studied growth factors in the uterine response to estradiol are epidermal growth factor (EGF) and insulin-like growth factor (IGF-1). Uterine cell proliferation is a c

Btn1 affects cytokinesis and cell-wall deposition by independent mechanisms, one of which is linked to dysregulation of vacuole pH.

btn1, the Schizosaccharomyces pombe orthologue of the human Batten-disease gene CLN3, is involved in vacuole pH homeostasis. We show that loss of btn1 also results in a defective cell wall marked by sensitivity to zymolyase, a beta-glucanase. The defect can be rescued by expression of Btn1p or CLN3, and the extent of the defect correlates with disease severity. The vacuole and cell-wall defects are linked by a common pH-dependent mechanism, because they are suppressed by growth in acidic pH and a similar glucan defect is also apparent in the V-type H(+) ATPase (v-ATPase) mutants vma1Delta and

Interplay between NETosis and the lncRNA-microRNA regulatory axis in the immunopathogenesis of cancer.

Neutrophil extracellular traps (NETs), web-like complex structures secreted by neutrophils, have emerged as key players in the modulation of immune responses and the immunopathogenesis of immune disorders. Initially described for their antimicrobial function, NETs now play a part in the fundamental processes of cancer biology, including cancer initiation, metastatic dissemination, and immune evasion strategies. NETs hijack anti-tumor immunity by entrapping circulating cancer cells, fostering the growth of tumors, and reorganizing the tumor microenvironment such that it is pro-malignancy. Emerg

Transforming growth factor β1 (TGFβ1) in physiology and pathology.

This review describes precisely the consequence of TGFβ1 prevalence in the organism, and its significant influence on physiological and pathophysiological processes. Organ and tissue distinctiveness hinder unambiguous characterisation of the cytokine. However, there are constant functions of TGFβ1 inducing no controversy: it participates in foetal development, control of cell growth and differentiation, induces fibrosis and scar formation (the process of 'wound healing'), causes the suppression of immune response, is involved in angiogenesis, the development of tumours, and inflammatory proces

Elucidating the roles of essential genes in autotrophic metabolism and cell morphology of Clostridium ljungdahlii by CRISPRi.

Understanding the function of essential genes in Clostridium ljungdahlii is critical for unraveling its autotrophic metabolism and optimizing its potential as a platform for syngas fermentation. However, study on essential genes of this species remains insufficient. Here, we employed an inducible CRISPR interference (CRISPRi) system to investigate the roles of key metabolic and cell division genes in C. ljungdahlii. Targeted repression of genes encoding pyruvate:ferredoxin oxidoreductase (PFOR1, PFOR2), acetaldehyde:ferredoxin oxidoreductase (AOR1, AOR2), and glyceraldehyde phosphate hydrogena

TUC338 Overexpression Leads to Enhanced Proliferation and Reduced Apoptosis in Tongue Squamous Cell Carcinoma Cells In Vitro.

PURPOSE: Long noncoding RNAs are closely related to the development of tumors. In this study, we explored the contribution of the long noncoding RNA TUC338 to cellular processes in tongue squamous cell carcinoma (TSCC). MATERIALS AND METHODS: First, we detected TUC338 expression using quantitative reverse transcription-polymerase chain reaction in 25 patients. Then, we transfected a short hairpin RNA to silence TUC338 expression in the CAL-27 and SCC-9 cell lines. Tumor cell growth was determined by the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) assay, and apoptos

Salicylic Acid Steers the Growth-Immunity Tradeoff.

Plants possess an effective immune system to combat most microbial attackers. The activation of immune responses to biotrophic pathogens requires the hormone salicylic acid (SA). Accumulation of SA triggers a plethora of immune responses (like massive transcriptional reprogramming, cell wall strengthening, and production of secondary metabolites and antimicrobial proteins). A tradeoff of strong immune responses is the active suppression of plant growth and development. The tradeoff also works the opposite way, where active growth and developmental processes suppress SA production and immune re

Astacin Metalloproteinase PpMC5 of Mesenchyme Cells Regulates Epithelial Cell Proliferation and Survival to Promote Starfish Larval Development.

The simple, triploblastic, epithelial body structure of the Patiria pectinifera starfish larva contains one mesenchyme cell type distributed across the blastocoel. In this study, we elucidated the role of a novel astacin metalloproteinase, PpMC5, tethered to the membrane of mesenchyme cells. PpMC5 knockdown by injection of morpholino oligo (MO) into the oocyte inhibited normal growth throughout the bipinnaria stage and caused a dwarfed phenotype consisting of reduced cell number and size, developmental delay, and cell debris in the blastocoel. When transforming growth factor β, P. pectinifera-

Interplay between growth factor and nutrient signaling: lessons from Drosophila TOR.

During normal development, cellular and organismal growth is coordinately regulated. Each cell and each individual organ integrates information about nutrient availability, hormonal signals, and intrinsic growth programs. Describing the signaling pathways involved in these processes and how they are integrated is important to understand how growth is controlled during development and may also permit the development of means to curb uncontrolled growth in disease. In recent years, the biochemical analysis of cellular growth in cultured cells and the genetic dissection of growth control in model

Development, Epigenetics and Metabolic Programming.

It is now widely recognized that the environment in early life can have important effects on human growth and development, including the 'programming' of far-reaching effects on the risk of developing common metabolic and other noncommunicable diseases in later life. We have shown that greater childhood adiposity is associated with higher maternal adiposity, low maternal vitamin D status, excessive gestational weight gain and short duration of breast-feeding; maternal dietary patterns in pregnancy and vitamin D status have been linked with childhood bone mineral content and muscle function. Effects of diadenosine tetraphosphate on FGF9-induced chloride flux changes in achondroplastic chondrocytes.

Achondroplasia, the most common type of dwarfism, is characterized by a mutation in the fibroblast growth factor receptor 3 (FGFR3). Achondroplasia is an orphan pathology with no pharmacological treatment so far. However, the possibility of using the dinucleotide diadenosine tetraphosphate (Ap(4)A) with therapeutic purposes in achondroplasia has been previously suggested. The pathogenesis involves the constitutive activation of FGFR3, resulting in altered biochemical and physiological processes in chondrocytes. Some of these altered processes can be influenced by changes in cell volume and ion

PRMT3 drives glioblastoma progression by enhancing HIF1A and glycolytic metabolism.

Glioblastoma (GBM) is the most common and aggressive primary brain tumor, but the mechanisms underlying tumor growth and progression remain unclear. The protein arginine methyltransferases (PRMTs) regulate a variety of biological processes, however, their roles in GBM growth and progression are not fully understood. In this study, our functional analysis of gene expression networks revealed that among the PRMT family expression of PRMT3 was most significantly enriched in both GBM and low-grade gliomas. Higher PRMT3 expression predicted poorer overall survival rate in patients with gliomas. Kno

[Scanning electron microscopic examination of 50 cases of non-Hodgkin's lymphoma (NHL)].

Fifty cases of NHL were studied by scanning electron microscopy to study the possible differences in the surface morphology of tumor cells between T cell and B cell NHL. Acid naphthyl acetate esterase (ANAE) activity test, the stain of antiserum samples of gamma, mu, alpha, kappa, lambda chains with the peroxidase antiperoxidase immune complex method (PAP method) and classification under light microscope were done for all the cases. It was found that the surface morphology of T cell lymphoma cells differed from that of B cell lymphoma. There were a lot of granular protrusions with a few pseudo

Metabolic control of cell division in α-proteobacteria by a NAD-dependent glutamate dehydrogenase.

Prior to initiate energy-consuming processes, such as DNA replication or cell division, cells need to evaluate their metabolic status. We have recently identified and characterized a new connection between metabolism and cell division in the α-proteobacterium Caulobacter crescentus. We showed that an NAD-dependent glutamate dehydrogenase (GdhZ) coordinates growth with cell division according to its enzymatic activity. Here we report the conserved role of GdhZ in controlling cell division in another α-proteobacterium, the facultative intracellular pathogen Brucella abortus. We also discuss the

Thermo-sensitive PLGA-PEG-PLGA hydrogel for sustained release of EGF to inhibit cervical cancer recurrence.

Overexpression of epidermal growth factor receptor (EGFR) in cancer is a key cause of recurrence of cervical cancer (CC). Although the EGF-EGFR pathway has been studied for decades, preventing tumor growth and recurrence caused by peripheral EGF remains a great challenge. In this work, a strategy is proposed to reduce the stimulation of high concentration EGF on tumor growth by using a thermo-sensitive hydrogel. The hydrogel is a triblock copolymer composed of polyethylene glycol (PEG) and poly (lactide glycolide) (PLGA). Based on the excellent temperature sensitivity, carrier capacity, swelli

Gambogic acid exhibits promising anticancer activity by inhibiting the pentose phosphate pathway in lung cancer mouse model.

BACKGROUND: The pentose phosphate pathway (PPP) plays a crucial role in the material and energy metabolism in cancer cells. Targeting 6-phosphogluconate dehydrogenase (6PGD), the rate-limiting enzyme in the PPP metabolic process, to inhibit cellular metabolism is an effective anticancer strategy. In our previous study, we have preliminarily demonstrated that gambogic acid (GA) induced cancer cell death by inhibiting 6PGD and suppressing PPP at the cellular level. However, it is unclear whether GA could suppress cancer cell growth by inhibiting PPP pathway in mouse model. PURPOSE: This study a
