[Influences of hydroxylated recombinant human type III collagen on the functional characteristics of vascular smooth muscle cells].

This study was designed to investigate the effects of hydroxylated recombinant human type III collagen on the proliferation and migration of vascular smooth muscle cells (VSMCs). The viability of primarily cultured VSMCs was examined by the CCK-8 assay. A cell cycle assay kit was used to evaluate the effect of hydroxylated recombinant human type III collagen at different concentrations on cell proliferation. Cell migration was evaluated via random migration and wound healing assays. A protein-protein interaction (PPI) network was built to screen the key genes associated with functions of VSMCs

Microtubule involvement in NIH 3T3 Golgi and MTOC polarity establishment.

Scratch-wound assays are commonly used to study the ability of cells to polarize and migrate. In a previous study we showed that Golgi reorientation in response to a scratch wound is actin-dependent in NIH 3T3 cells but not in astrocytes. In this investigation, to study cell polarity and motility further, we used the polarization of the Golgi and microtubule organizing center (MTOC), as well as the ability of NIH 3T3 cells to migrate, in a scratch-wound assay. Unlike Golgi polarization, MTOC polarization was not dependent on actin, the Arp2/3 complex or Wiskott-Aldrich syndrome protein (WASP)-

Restoration of miR-1305 relieves the inhibitory effect of nicotine on periodontal ligament-derived stem cell proliferation, migration, and osteogenic differentiation.

BACKGROUND: Nicotine hinders the regenerative potentials of human periodontal ligament-derived stem cells (PDLSCs) and delays the healing process of periodontal diseases, but the underlying mechanism remains unclear. miR-1305 upregulation and its potential target RUNX2 downregulation exist in the PDLSCs exposed to nicotine. In this study, we aimed to investigate whether nicotine inhibits PDLSC proliferation, migration, and osteogenic differentiation by increasing miR-1305 level and decreasing RUNX2 level. METHODS: Quantitative real-time PCR (qRT-PCR) and Western blot assays were performed to

Inhibition of chondroitin sulfate proteoglycans enhances cell migration following enteric neural stem cell transplantation.

Enteric nervous system development relies on migration, proliferation and differentiation of enteric neural crest-derived cells (ENCDCs), processes that are regulated by mesenchymal morphogens and the extracellular matrix (ECM). Hirschsprung disease (HD) is a congenital neurocristopathy in which abnormal gut motility results from loss of ENCDC-derived enteric ganglia in the colorectum. While enteric neural stem cell transplantation is a proposed therapy, current efforts are limited by poor cell migration in HD models. To address this, we investigated ECM-derived inhibitory cues on enteric neur

Antitumor effects of curcumin in pediatric rhabdomyosarcoma in combination with chemotherapy and phototherapy in vitro.

Rhabdomyosarcoma (RMS), the most common pediatric soft tissue sarcoma, has an unfavorable outcome in advanced tumor stages with less than 30% failure‑free survival. Curcumin (CUR) is a promising drug in complementary oncology with few side effects but proven efficacy in various adult oncological entities. The present study analyzed the effects of CUR on pediatric (RMS) cell lines in vitro. RMS cell lines (RD and RH30), and skeletal muscle cells (SKMC) were treated with different doses of CUR (1.5‑30 µM) alone, with phototherapy (PDT, 488 nm) or in combination with vincristine (VCR) or dactinom

RSPO2-induced ferroptosis via PTBP1-mediated FSP1 mRNA decay suppresses breast cancer progression.

INTRODUCTION: Breast cancer is the most common malignant tumor worldwide, and its high heterogeneity limits the efficacy of current clinical therapies. R-spondin 2 (RSPO2) expression is closely associated with breast cancer subtypes, but the precise mechanisms by which it drives disease progression, particularly its relationship with ferroptosis, remain unclear. METHODS: RSPO2 expression was examined in primary breast cancer specimens and in MCF-7 and MDA-MB-231 cell lines. Functional assays, including proliferation, migration, and invasion tests, were performed following RSPO2 overexpression

Neurotensin drives theca cell migration during ovulation in primates†.

Theca cells are a critical steroidogenic cell type of the ovarian follicle and corpus luteum. The ovulatory luteinizing hormone (LH) surge (or human chorionic gonadotropin (hCG)) stimulates theca cell relocation from the stroma surrounding the dominant follicle to full integration into the developing corpus luteum. Luteinizing hormone/human chorionic gonadotropin also stimulates granulosa cells to produce local mediators of ovulation, including the peptide neurotensin (NTS). To determine if hCG-stimulated NTS regulates theca cell relocation within the ovulatory follicle, vehicle or an NTS rece

Combination of ionizing radiation and 2-thio-6-azauridine induces cell death in radioresistant triple negative breast cancer cells by downregulating CD151 expression.

BACKGROUND: Triple-negative breast cancer (TNBC) represents the most aggressive subtype of breast cancer and is frequently resistant to therapy, ultimately resulting in treatment failure. Clinical trials have demonstrated the potential of sensitizing radiation therapy (RT)-resistant TNBC through the combination of chemotherapy and RT. This study sought to explore the potential of CD151 as a therapy response marker in the co-treatment strategy involving ionizing radiation (IR) and the repurposed antiviral drug 2-Thio-6-azauridine (TAU) for sensitizing RT-resistant TNBC (TNBC/RR). LATS2 inhibits cell proliferation and metastasis through the Hippo signaling pathway in glioma.

As a core kinase in the Hippo pathway, large tumor suppressor kinase 2 (LATS2) regulates cell proliferation, migration and invasion through numerous signaling pathways. However, its functions on cell proliferation, migration and invasion in glioma have yet to be elucidated. The present study revealed that LATS2 was downregulated in glioma tissues and cells, as determined by reverse transcription‑quantitative polymerase chain reaction and immunohistochemistry. In addition, Cell Counting Kit‑8, scratch wound healing and Transwell assays revealed that overexpression of LATS2 in U‑372 MG cells inh

Overexpression of EGFL7 promotes angiogenesis and nerve regeneration in peripheral nerve injury.

Peripheral nerve injury (PNI) often leads to significant functional impairment. Here, we investigated the impact of epidermal growth factor-like domain-containing protein 7 (EGFL7) on angiogenesis and nerve regeneration following PNI. Using a sciatic nerve injury model, we assessed nerve function using the sciatic nerve function index. We analyzed the expression levels of EGFL7, forkhead box proteins A1 (FOXA1), nerve growth factor (NGF), brain-derived neurotrophic factors (BDNF), Neurofilament 200 (NF200), myelin protein zero (P0), cell adhesion molecule 1 (CD31), vascular endothelial growth
