Chemerin alleviates acute pancreatitis in the rat thorough modulation of NF-κB signal.

OBJECTIVE: Chemerin, an adipokine, works as the chemoattractant for the immune cells. The role of chemerin in the inflammatory reaction is controversial. Chemerin has been shown to aggravate the inflammatory response, but other studies demonstrated its anti-inflammatory influence. This study assessed the effects of chemerin on acute pancreatitis (AP) in vivo and in vitro. METHODS: For in vivo experiments male Wistar rats were used. For in vitro study rat pancreatic AR42J cells were employed. Chemerin (1, 5 or 10 μg/kg) was given to the rats prior to the induction of AP by subcutaneous caerule

FTY720 Attenuates Acute Pancreatitis in Hypertriglyceridemic Apolipoprotein CIII Transgenic Mice.

Hypertriglyceridemic pancreatitis (HTGP) is often encountered clinically as a common form of recurrent acute pancreatitis (AP). It is important to evaluate the management of severe hypertriglyceridemia (HTG) or anti-inflammation in the prophylaxis of HTGP in the clinic. FTY720 (2-amino-2[2-(4-octylphenyl) ethyl]-1, 3-propanediol) is a new anti-inflammatory agent with low toxicity and reported to ameliorate lung injury with pancreatitis in rat. We evaluated its protective affection on AP induced by seven hourly intraperitoneal injection of cerulein in apolipoprotein CIII transgenic mice with se

Gallbladder sensitivity to CCK in duodenal ulcer disease, highly selective and truncal vagotomy.

BACKGROUND/AIMS: Following truncal vagotomy, a heightened contractile response of the gallbladder to cholecystokinin (CCK) has been reported in patients. We investigated whether the gallbladder responsiveness to the CCK analog cerulein is also affected in patients with a highly selective vagotomy (HSV) and in duodenal ulcer patients, since most patients had truncal vagotomy for recurrent peptic ulcer disease. MATERIALS AND METHODS: Gallbladder emptying (cholescintigraphy) and plasma cholecystokinin like immunoreactivity (CCK-LI) levels were studied during infusion of graded doses of the CCK a

Hyperoncotic dextran and systemic aprotinin in necrotizing rodent pancreatitis.

BACKGROUND: Dextrans improve pancreatic microcirculation in acute experimental pancreatitis. They could therefore facilitate the transport of a protease inhibitor to ischemic areas of tissue injury and be of additional benefit. METHODS: To compare the effects of dextrans with and without aprotinin, necrotizing pancreatitis was induced in 33 male dextran-resistant Wistar rats by intraductal infusion of low-dose glycodeoxycholic acid (10 mmol/l) followed by intravenous cerulein (5 micrograms/kg/h) for 6 h. Three and four hours after the start of the cerulein infusion the animals received infusi

Evidence of local mechanism involvement in vasoactive intestinal polypeptide release from canine small intestine.

We studied the effect of intraileal administration of a 100-ml test meal, 100 ml of 5% glucose, amino acids, 10% Intralipos, or bile on canine plasma immunoreactive vasoactive intestinal polypeptide (VIP) levels in the mesenteric and femoral veins of dogs. The effects of intravenous administration of caerulein on plasma immunoreactive VIP levels were examined, with or without intraduodenal ballooning, to rule out the effect of bile and pancreatic and gastric juice. In addition, hexamethonium bromide or atropine sulfate was given intravenously before the administration of bile or intravenous ca

Evidence of cholecystokinin release by bombesin in the dog.

1 The intravenous infusion of bombesin elicited in the dog a contraction of the gall bladder with decreased opening pressure of the choledocho-duodenal junction and stimulation of pancreatic secretion.2 The pancreatic juice produced under the influence of bombesin was poor in bicarbonate and rich in protein. Threshold doses of the peptide were of the order of 0.25 mug kg(-1) h(-1) and maximum protein output was obtained with 1 mug kg(-1) h(-1). The pancreatic protein response to bombesin was very similar, in its onset and duration, to that elicited by intraduodenal infusion of L-tryptophan. In

Silencing of SPI1/OLAH Axis Inhibits Pancreatic Acinar Cell Oxidative Stress, Inflammation and Apoptosis to Alleviate Hyperlipidemic Acute Pancreatitis.

BACKGROUND: Oleoyl-ACP hydrolase (OLAH), a fatty acid metabolism-related gene, is abnormally expressed in many diseases. Using the GEO database, we found that OLAH was highly expressed in acute pancreatitis (AP) patients. However, whether it mediates hyperlipidemic AP (HAP) progression remains unclear. MATERIALS AND METHODS: Mouse pancreatic acinar cells (MPC-83) were treated with palmitic acid (PA) and cerulein (CER) to mimic HAP cell models, and HAP mice models were constructed by injecting with P-407 and CER. The mRNA and protein levels of OLAH and Salmonella pathogenicity island 1 (SPI1)

IGF-1 protects against acute pancreatitis by suppressing NF-κB activation through the β-arrestin1/STAT3 pathway.

Acute pancreatitis (AP) is a prevalent inflammatory condition with an upward trend in incidence in recent years. Recent studies have demonstrated the important role of IGF-1 and β-arrestin-1 in inflammation, but their role in AP remains unexplored. Our study explored the role of IGF-1 in acute pancreatitis (AP) using caerulein-induced AP models in wild-type (WT) and β-arrestin1-knockout (KO) mice treated with IGF-1, picropodophyllotoxin (PPP, an IGF-1R inhibitor), Bay11708 (an NF-κB inhibitor), and the STAT3 inhibitor VI. Caerulein injection induced AP, characterized by elevated serum amylase

Dachaihu Decoction alleviates chronic pancreatitis by regulating MAPK signaling pathway: Insights from network pharmacology and experimental validation.

ETHNOPHARMACOLOGICAL RELEVANCE: Chronic pancreatitis (CP), a syndrome characterized by inflammatory fibrosis, can impair both the internal and external secretory functions of the pancreas. The global incidence of this disease is gradually increasing. However, the exact pathogenesis remains unclear, resulting in a lack of targeted clinical therapies. According to the principles of traditional Chinese medicine, CP can be attributed to Shaoyang and Yangming syndromes, which manifest as abdominal pain and hypochondriac distension. Dachaihu Decoction (DCHD) is a classic formula from the "Treatise o

Functional disturbance of biliary indocyanine green excretion in rat cerulein pancreatitis followed by endotoxemia: role of the prime and the second attack.

CONTEXT: Hepatic injury is considered one of the critical complications associated with acute pancreatitis. It was proposed that initial insults to the liver in the early phase of the attack have an important priming effect, and the subsequent infectious attack (e.g. infectious pancreatic necrosis, bacterial translocation episode) constitutes a second attack on the liver. OBJECTIVE: To evaluate the role of priming by induction of cerulein pancreatitis and a following second attack by endotoxemia. MAIN OUTCOME MEASURES: Plasma clearance and biliary excretion of indocyanine green (a hepatophil

Defect in pancreatic exocrine and endocrine response to CCK in genetically diabetic OLETF rats.

Clinical as well as experimental studies in insulinopenic diabetes mellitus have demonstrated abnormal pancreatic exocrine responses to cholecystokinin (CCK). In the present study, we examined pancreatic exocrine and endocrine function in the recently developed genetically diabetic Otsuka Long-Evans Tokushima fatty (OLETF) rats and compared them with those in the control Long-Evans Tokushima Otsuka (LETO) rats of the same age. Stepwise increasing doses of CCK octapeptide (CCK-8; 0.027-7.0 nmol.kg-1.h-1) evoked a characteristic biphasic dose-response curve for pancreatic juice and protein outpu

Effect of cerulein hyperstimulation on the paracellular barrier of rat exocrine pancreas.

BACKGROUND/AIMS: Cerulein-induced pancreatitis causes a rapid increase in pancreatic enzyme levels in serum and decreases in pancreatic duct secretion and interstitial edema. One mechanism to explain these early events is disruption of the actin tight junction paracellular seal of acinar and intralobular pancreatic duct cells. METHODS: To examine the paracellular barrier of the proximal exocrine pancreas, rats were hyperstimulated with 5.0 micrograms.kg-1.h-1 of cerulein. Actin was visualized with rhodamine phalloidin and by electron microscopy and tight junctions were visualized with antibod

Anticholecystokinin activities of loxiglumide.

The anticholecystokinin activities of loxiglumide, (D,L-4-(3,4-dichloro-benzoylamino)-5-(N-3-methoxypropyl-pentylamino++ +)-5-oxo- pentanoic acid, CR 1505) are described. Loxiglumide antagonizes in vivo the contractions of the gall bladder of guinea pig induced or mediated by cholecystokinin-8 (CCK-8) (i.v. ED50 = 0.24 mumol/kg), the emptying of the gall bladder of the mouse induced by CCK-8 (i.v. Effects of rociverine and other spasmolytic agents on caerulein-induced delay in gastric emptying in the conscious rat.

Caerulein (C)-induced delay in gastric emptying (GE), due to pylorospasm, was used as an experimental model for a quantitative test of the activity of some spasmolytic drugs in conscious rats. Rociverine at doses of 10 and 15 mg kg-1 i.p., though not at higher doses, considerably reduced this delay. Of the other drugs tested only papaverine had this effect and to a lesser degree. Atropine, N-butylscopolammonium bromide and dicyclomine proved to be inactive or further reduced GE. The effect of spasmolytics on C pylorospasm seems to be the outcome of two actions possessed by the drugs in varying

Effects of caerulein and trypsin inhibitors on the endocrine mouse pancreas.

Oral administration to mice with soybean trypsin inhibitor (SBTI) (27-30 mg/mouse/day) or aprotinin (5500-6000 KIU/mouse/day) for six weeks increased the total pancreatic insulin (IRI). The pancreatic IRI was also increased after sc injections of synthetic caerulein (0.05 microgram/mouse/day divided into 3 daily doses), being 82% above the control levels when expressed per g pancreas. Aprotinin (6000 KIU/mouse/day divided into 3 daily doses) injected sc had no effect on the insulin content. The total glucagon did not change significantly in any of the groups, but the molar ratio of insulin to

Evidence for a role in stimulus--secretion coupling of prostaglandins derived from release of arachidonoyl residues as a result of phosphatidylinositol breakdown.

That stimulation of secretion in exocrine and endocrine glands is associated with increased turnover of phosphatidylinositol and phosphatidic acid has been known for many years. In the present work, mouse pancreases were prelabeled with [14C]arachidonic acid in the presence of the secretogogue carbamoylcholine. They were then incubated in media containing atropine and 1% albumin. The atropine causes the tissue to revert to the resting state, and the albumin binds free [14C]arachidonic acid. The tissues were finally incubated in media containing no stimulant or the stimulant caerulein, which is

Genetic and pharmacologic modulations elucidate NRF2 protective role in acute pancreatitis.

Acute pancreatitis (AP) is a potentially fatal disease of the exocrine pancreas. The disease pathogenesis remains obscure, and no effective treatment is available. Uncontrolled, deregulated inflammation is a major cause of systemic complications in AP; however, approaches to reduce inflammation in human disease by inhibiting inflammatory mediators have encountered multiple challenges and have not been successful. Oxidative stress drives inflammation in many diseases but its role in AP remains poorly understood. This study evaluates the role of nuclear factor erythroid 2-related factor 2 (NRF2)

Intraarterial Infusion of Lidocaine is Superior to the Subcutaneous Injection of Low Molecular Weight Heparin for Improving the Course of Cerulein-Induced Acute Pancreatitis in Rats.

This study aimed to determine the efficacy of low molecular weight heparin (LMWH) and lidocaine combined with LMWH for improving the course of acute pancreatitis (AP). A total of 30 rats were divided into three groups: the NaCl group, which received an intraarterial infusion of 0.9% sodium chloride; the Heparin group, which received a subcutaneous injection of LMWH; and the Lidocaine-Heparin group, which received an intraarterial infusion of 1% lidocaine, with subsequent subcutaneous injection of LMWH. LRG1 inhibition promotes acute pancreatitis recovery by inducing cholecystokinin Type 1 receptor expression via Akt.

Rationale: Acute pancreatitis (AP) is a common gastrointestinal disease affecting nearly 3 million people annually worldwide. Although AP is typically self-limiting, up to 20% of patients may develop life-threatening complications. Individuals who suffer from AP also have an increased likelihood of developing other exocrine and endocrine pancreatic disorders. However, to date, there are no specific, targeted treatment modalities that can effectively improve the clinical outcomes of AP.
