Chlorogenic acid suppresses monocrotaline-induced sinusoidal obstruction syndrome: The potential contribution of NFκB, Egr1, Nrf2, MAPKs and PI3K signals.

Hepatic sinusoidal obstruction syndrome (SOS) is a highly lethal liver disease. This study aims to observe the protection and its engaged mechanism of chlorogenic acid (CGA) against monocrotaline (MCT)-induced SOS. Results of detecting liver ascites, measuring serum transaminases, liver histological evaluation and scanning electron microscope observation all demonstrated that CGA prevented MCT-induced SOS in rats. CGA reduced MCT-induced increased liver myeloperoxidase (MPO) activity, tumor necrosis factor (TNF)α and interleukin (IL)-1β mRNA expression, toll-like receptor (TLR)-2,3,6,9 express

Nitrogen-doped SiQDs and their PVA fluorescent films for rapid fluorescence detection of chlorogenic acid in green coffee beans.

Nitrogen-doped silicon quantum dots (N-SiQDs) were prepared via a one-pot hydrothermal route, and subsequently utilized to establish a fluorescent sensing system for the rapid determination of chlorogenic acid (CGA). Mechanistic studies revealed a mixed quenching mechanism. The inner filter effect arises from the strong spectral overlap between CGA absorption and N-SiQD excitation. Meanwhile, CGA forms a non-fluorescent ground-state complex with N-SiQDs via hydrogen bonding and/or electrostatic interactions, as confirmed by a new absorption peak and a progressive emission red-shift. Such compl

Enhancement of antioxidant and antimicrobial properties of Whey protein isolate and chlorogenic acid complexes through enzymatic and alkaline techniques.

The interactions of whey protein isolate (WPI) with chlorogenic acid (CQA) using two techniques, alkaline (pH 9) and enzymatic (tyrosinase) were investigated. Complexes, formed between WPI and CQA by alkaline technique (AWPI-CQA) and enzymatic technique (EWPI-CQA), compared to control WPI (CWPI), were characterized in terms of their chemical, structural, emulsifying, antioxidant, and antibacterial properties. Compared to CWPI, both complexation methods significantly reduced free amino groups (CWPI: 588.00 nmol/mg; AWPI-CQA: 409.85 nmol/mg; EWPI-CQA: 412.50 nmol/mg), sulfhydryl groups (CWPI: 68

Synergistic Antioxidant Activity of Lycium barbarum Polysaccharide and Chlorogenic Acid and Its Effect on Inflammatory Response of NR8383 Cells.

It is inevitable for polyphenols and polysaccharides to interact during food preparation. Modifications in microstructure can lead to changes in the physical and chemical properties of food systems, which in turn may influence the nutritional characteristics and functional activities of the food. Recent studies have shown that, in addition to traditional Chinese medicine compounds, certain natural polysaccharides and polyphenols exhibit significant anti-inflammatory and antioxidant properties. These compounds are also associated with beneficial therapeutic effects for the prevention and treatm

Conceptual functional-by-design optimisation of the antioxidant capacity of trans-resveratrol, quercetin, and chlorogenic acid: Application in a functional tea.

Designing functional foods as delivery systemsmay become a tailored strategy to decrease the risk of noncommunicable diseases. Therefore, this work aims to optimise a combination of t-resveratrol (RES), chlorogenic acid (CHA), and quercetin (QUE) based on antioxidant assays and develop a functional tea formulation enriched with the optimal polyphenol combination (OPM). Experimental results showed that the antioxidant capacity of these compounds is assay- and compound-dependent. A mixture containing 73% RES and 27% QUE maximised the hydroxyl radical scavenging activity and FRAP. OPM upregulated

In Vitro and In Vivo Evaluation of Chlorogenic Acid-Encapsulated Lignin on Patulin Adsorption and Alleviation of Patulin-Induced Colonic Damage.

Patulin (PAT) is a water-soluble mycotoxin that causes digestive tract damage and liver and kidney function abnormalities. The current control approaches only reduce the amount of PAT in raw materials and food, which is difficult to remove once ingested in the body. In this study, lignin-based cross-linked particles loaded with chlorogenic acid were prepared, which intervened the liver and colon damage caused by PAT in mice. In the simulated digestion process in vitro, the accumulated adsorption capacity of the LB/CA-SH for PAT was 0.934 mg/g. LB/CA-SH intervention reversed the shortening of t

Temperature effect in the inhibition of PLA2 activity of Bothrops brazili venom by Rosmarinic and Chlorogenic acids, experimental and computational approaches.

Myotoxicity caused by snakebite envenoming emerges as one of the main problems of ophidic accidents as it is not well neutralized by the current serum therapy. A promising alternative is to search for efficient small molecule inhibitors that can act against multiple venom components. Phospholipase A2 (PLA2) is frequently found in snake venom and is usually associated with myotoxicity. Thus it represents an excellent target for the search of new treatments. This work reports the effect of temperature in the inhibition of catalytic properties of PLA2 from Bothrops brazili venom by Rosmarinic (RS

Cloning and characterization of Lonicera japonica p-coumaroyl ester 3-hydroxylase which is involved in the biosynthesis of chlorogenic acid.

Lonicera japonica is used in Chinese medicine as a source of antioxidants, primarily flavonoids, and a phenolic acid chlorogenic acid (CGA). Here we report the isolation and characterization of the full-length cDNA of LjC3H, a gene encoding p-coumaroyl ester 3-hydroxylase, an enzyme involved in CGA synthesis. Phylogenetic analysis indicated that is protein belongs to the CYP98A subfamily, and homology modeling revealed that its structure resembles that of other cytochrome P450 family proteins. Southern blot analysis indicated that more than one copy of sequences homologous to LjC3H is present

Synergistic CNTs@SnO2-MWW nanocomposite enables a high-performance miniaturized sensor for rapid detection of chlorogenic acid in food.

Chlorogenic acid (CGA) is a bioactive polyphenol found in tea and fruit, which exhibits anti-inflammatory, antibacterial and anticancer properties that hold significant potential in food and health applications. However, traditional sensing materials suffer from drawbacks such as slow surface kinetics and poor selectivity in complex matrices, making the development of highly functional sensing materials challenging. To address this issue, this study synthesizes a layered nano MWW zeolite (MWW framework topology) with high specific surface area and diffusion properties. Via an in-situ growth st

Chlorogenic acid promotes macrophage phagocytosis through the METTL3-SIRPA axis in an m6A-dependent manner for colon cancer therapy.

Epigenetic modifications, particularly N6-methyladenosine (m6A) modifications, alter the phenotype of macrophages. Among the m6A-related proteins, methyltransferase 3 (METTL3), as the catalytic core methyltransferase subunit, plays an essential role in regulating the function of macrophages. This study aimed to explore novel m6A inhibitors from natural products that target METTL3 to modulate the phagocytic function of macrophages in the treatment of colon cancer. In this work, first, a high-throughput screening of biological m6A by colorimetric assay was conducted to rapidly identify candidate

Carboxyl Amidation of Chlorogenic Acid Improves Anti-Inflammatory Activity and Biosafety via Potent AKR1B1 Inhibition in LPS-Induced Macrophages.

Persistent inflammation driven by excessive AKR1B1 activation contributes to the progression of multiple chronic inflammatory diseases. Natural chlorogenic acid (CGA) exhibits anti-inflammatory activity but suffers from poor membrane permeability, while its laurate ester derivative (CGL) shows clear dose-dependent cytotoxicity in macrophages. Here, we synthesized a novel chlorogenic acid amide derivative (CGAA) by selective amidation of the aliphatic carboxyl group on the quinic acid moiety of CGA, fully preserving the chiral polyphenol scaffold. Structural identity was confirmed by HSQC, HMBC

A Novel Mechanism of Chlorogenic Acid in Cardioprotection: Blocking NLRP3 Inflammasome via Ca2+/CaMKIIα Signaling in Sepsis-Induced Cardiomyopathy.

Sepsis-induced cardiomyopathy (SICM) is a severe complication of sepsis, characterized by myocardial inflammation, oxidative stress, and cardiac dysfunction. Chlorogenic acid (CGA), a natural polyphenol with known anti-inflammatory and antioxidant properties, is abundant in many traditional medicinal plants used for cardiovascular and inflammatory disorders. However, its cardioprotective effects in SICM and the underlying mechanisms remain unclear. An in vivo cecal ligation and puncture (CLP) model was used to induce SICM in rats, followed by CGA treatment. Cardiac function and myocardial inju

Chlorogenic acid counteracts mycophenolate mofetil-induced neurotoxicity in zebrafish and PC12 cells: A potential bioremediation strategy for aquatic pharmaceuticals.

Pharmaceutical pollution, particularly from immunosuppressants like mycophenolate mofetil (MMF), has emerged as a significant environmental contaminant in aquatic ecosystems, frequently detected in wastewater at concentrations of 0.1-2.5 μg/L with an occurrence rate exceeding 80 %. This study demonstrates that chlorogenic acid (CGA), a plant-derived polyphenol found in coffee, fruits, and medicinal plants, effectively counters MMF-induced neurotoxicity in both zebrafish and neuronal cells. Specifically, CGA significantly protected PC12 cells from H2O2-induced damage by promoting cell prolifera

Chlorogenic acid alleviates thioacetamide-induced toxicity and promotes liver development in zebrafish (Danio rerio) through the Wnt signaling pathway.

Chlorogenic acid (CGA) is a phenylpropanoid compound that is well known to improve the antioxidant capacity and other biological activities. However, the roles of CGA in the liver development of organisms are unclear. In the present study, we aimed to investigate the function of CGA in the hepatic development in thioacetamide (TAA)-induced zebrafish embryos. We found that CGA exerted certain beneficial effects on zebrafish larvae from TAA-exposed zebrafish embryos, such as increasing the liver size, body length, heart rate, acetylcholinesterase activity, and motor ability. In addition, CGA dis

Decaffeination and Neuraminidase Inhibitory Activity of Arabica Green Coffee (Coffea arabica) Beans: Chlorogenic Acid as a Potential Bioactive Compound.

Coffee has been studied for its health benefits, including prevention of several chronic diseases, such as type 2 diabetes mellitus, cancer, Parkinson's, and liver diseases. Chlorogenic acid (CGA), an important component in coffee beans, was shown to possess antiviral activity against viruses. However, the presence of caffeine in coffee beans may also cause insomnia and stomach irritation, and increase heart rate and respiration rate. These unwanted effects may be reduced by decaffeination of green bean Arabica coffee (GBAC) by treatment with dichloromethane, followed by solid-phase extraction

Multi-omics analyses reveal distinct and complementary effects of chlorogenic acid and bile acids on hepatic lipid metabolism and redox homeostasis in juvenile rainbow trout.

Long-term feeding with a commercial basal diet may induce moderate hepatic lipid accumulation and sub-optimal redox status in juvenile rainbow trout (Oncorhynchus mykiss), forming a mutually reinforcing hepatic sub-health cycle. Chlorogenic acid (CGA) and bile acids (BAs) are promising additives for improving hepatic lipid metabolism and redox balance, but their individual and combined effects remain unclear. Here, juvenile rainbow trout were fed four diets for 90 days: basal diet (C), 600 mg/kg CGA (TC), 1000 mg/kg BAs (TB), or both additives (TCB). Unlike previous single-additive studies, th
