Purification and spectroscopic characterization of a recombinant chloroplastic hemoglobin from the green unicellular alga Chlamydomonas eugametos.

Hemoglobins (Hb), which have the important task of delivering molecular oxygen by facilitating its reversible binding to the heme, are now thought to have evolved in all groups of organisms including prokaryotes, fungi, plants and animals. Our recent finding of a light-inducible chloroplastic Hb in the green unicellular alga Chlamydomonas eugametos has further extend this idea, while raising questions about the function that an Hb could play in a high oxygen environment such as in the chloroplast. In order to understand the role played by this new Hb, we have undertaken its biochemical charact

Developmental and light effects on the accumulation of FtsH protease in Arabidopsis chloroplasts--implications for thylakoid formation and photosystem II maintenance.

The chloroplast ATP-dependent metalloprotease FtsH is involved in the degradation of unassembled proteins, the repair of photosystem II (PSII) from photoinhibition, and, apparently, the formation of thylakoids. In Arabidopsis, it is encoded by a family of 12 genes. However, the products of only four of them, FtsH1, 2, 5 and 8, have been found in chloroplasts to date. Mutations in two of these, FtsH2 and 5, demonstrate a visible phenotype of variegated leaves, with the phenotype of the FtsH2 mutant being more pronounced. Moreover, the degree of variegation appears to be dependent on development

The role of the membrane-bound iron-sulphur centres A and B in the photosystem I reaction centre of spinach chloroplasts.

Photosystem I particles prepared from spinach chloroplast using Triton X-100 were frozen in the dark with the bound iron-sulphur Centre A reduced. Illumination at cryogenic temperatures of such samples demonstrated the photoreduction of the second bound iron-sulphur Centre B. Due to electron spin-electron spin interaction between these two bound iron-sulphur centres, it was not possible to quantify amounts of Centre B relative to the other components of the Photosystem I reaction centre by simulating the line-shape of its EPR spectrum. However, by deleting the free radical signal I from the EP

Exogenous melatonin enhances flooding tolerance in maize and is associated with coordinated redox, metabolic, and chloroplast responses.

Flooding stress (FS) at the maize seedling stage disrupts energy metabolism and redox homeostasis, impairing photosynthesis and causing cellular damage. This study evaluated whether exogenous melatonin (MT) alleviates FS injury in maize and characterized physiological and molecular responses associated with this protection. Maize seedlings were subjected to a 2 × 2 design (CK, CK-MT, FS, and FS-MT), while transcriptomic, metabolomic, and hormone profiling focused on CK, FS, and FS-MT. FS reduced net photosynthetic rate and stomatal conductance by 61.93% and 77.98%, respectively, increased H2O2

The Complete Chloroplast Genome of Clematis tangutica (Maxim.) Korsh. and an Adaptive Evolutionary Analysis of the ycf2 Gene.

BACKGROUND: Clematis tangutica (Maxim.) Korsh. is a Tibetan medicinal plant, but its chloroplast genome and plastid gene evolution remain unexplored. Here, we assembled the complete chloroplast genome of C. tangutica and investigated the adaptive evolution of the ycf2 gene. METHODS: Chloroplast DNA was extracted from fresh leaves and sequenced on the Illumina HiSeq PE150 platform. We then performed comprehensive genomic analyses, including genome structure characterization, repeat and SSR identification, comparative genomics, and positive selection analysis of the ycf2 gene. RESULTS: The gen

Chloroplastic protein prenylation: an internal or external affair? Reconciling biochemical data through a new hypothesis.

Protein prenylation facilitates membrane association, protein interactions, and trafficking by attaching isoprenoid groups to C-terminal cysteine residues in the cytosol, with subsequent proteolysis and carboxymethylation occurring in the endoplasmic reticulum. In contrast this classic pathway, an organelle-autonomous prenylation pathway in plastids has long been hypothesized, however, its biochemical mechanisms remain unconfirmed. This review synthesizes multi-omics and genetic evidence to propose that plastid prenylation likely involves nuclear-encoded proteins modified in the cytosol, with

The complete sequence of chloroplast genome of an important medicinal plant, Chamaemelum nobile Linaeus 1785 (Asteraceae).

Chamaemelum nobile L. (Asteraceae), commonly known as Roman chamomile, is a perennial herb with branched rhizomes and multiple leafy stems. The length of the complete cp genome is 149,827 bp, and it comprises 128 genes, including 83 protein-coding genes, 8 ribosomal RNA genes, and 37 transfer RNA genes. By conducting phylogenetic analyses based on chloroplast genomes, we found that C. nobile was more closely related to Matricaria chamomilla var. recutita within the chamomile species. This finding contributes to a better understanding of the phylogenetic relationship between C. nobile and other

Chloroplast Haplotype Analysis Reveals High Genetic Similarity Among Central Asian Prunus Species.

Genetic variation in four wild Prunus taxa (P. fruticosa, P. erythrocarpa, P. verrucosa and P. griffithii var. tianshanica) was investigated for the first time using six chloroplast DNA regions (matK, r rpl16, ycf1_1, ycf1_2, ndhF and trnH-psbA) analysed through CAPS-based SNP detection. The results revealed weak chloroplast differentiation among P. erythrocarpa, P. verrucosa and P. griffithii var. tianshanica. However, chloroplast variation exhibited a strong geographic signal across the studied populations. The observed chloroplast variation primarily reflected geographic structuring rather

The yellow-in-the-dark chIL Chlamydomonas mutant as a model for time-resolved chloroplast biogenesis and retrograde signaling.

Understanding chloroplast biogenesis is key given the importance of photosynthesis and chloroplast metabolism. Chloroplast assembly has been extensively studied in angiosperms, where the main hallmarks have been established, including the need for nuclear and plastid genome coordination through anterograde and retrograde signals. However, recent works in other photosynthetic lineages have revealed both divergent and conserved regulators. These differences likely represent adaptive strategies that emerged during the evolution of the green lineage and highlight the need for evolutionary studies

Chloroplast Phylogenomics and Barcode Discovery in Medicinal Stachys Species Reveal Evolutionary Relationships and Adaptive Signatures.

The genus Stachys, a large and taxonomically intricate group within the Lamiaceae family, includes numerous species valued in traditional medicine, yet their evolutionary relationships are often obscured by morphological complexity. To resolve these persistent difficulties, we sequenced and assembled the complete chloroplast genomes of two medicinally important Iranian species, Stachys persica and Stachys germanica, and compared them against seven congeneric genomes. Despite an overall picture of strong structural conservation across the genus, we discovered that evolutionary variation concent

The chloroplast CLPD chaperone: consequences of under- and overexpression, interaction with the CLP protease core, and candidate substrates.

Expression of the chloroplast AAA+ chaperone CLPD gene increases during senescence and drought, but its function in chloroplast proteostasis is poorly understood. This study provides a comprehensive analysis of Arabidopsis CLPD protein accumulation across development from early seedlings to senescence, and compares results to its homologs CLPC1,2, as well as CLPB3 and cpHSP90. The developmental consequences of complete loss of CLPD expression (clpd-1), as well as overexpression of functional CLPD or CLPD impaired in ATP hydrolysis (CLPD-TRAP), were determined in Arabidopsis. clpd-1 has acceler

Molecular identification and phylogenetic analysis of Polygonatum Kingianum with different floral colors on the basis of chloroplast genomes.

Polygonatum kingianum, a plant with both medicinal and edible properties, demonstrates considerable variation in perianth coloration, making its differentiation from related species through morphological characteristics particularly challenging. This variation has led to frequent substitution of P. kingianum with other Polygonatum species in commercial markets, thereby compromising product quality and posing risks to the safety and efficacy of clinical applications. This research reports the sequencing and assembly of chloroplast genomes from three P. kingianum phenotypes with distinct floral

Thermal protection of the plastid-encoded RNA polymerase complex by PAP9 ensures chloroplast function in Arabidopsis.

The Plastid-encoded RNA polymerase (PEP) complex is a multi-subunit machinery essential for chloroplast biogenesis and photosynthetic gene transcription. Its structural integrity and enzymatic activity rely critically on a suite of PEP-associated proteins (PAPs). Mutations in most PAP-encoding genes typically lead to severe albinism and developmental arrest in Arabidopsis thaliana. However, the pap9 mutant exhibits phenotype closely resembling the wild type under standard growth conditions. In this study, we demonstrate that pap9 mutants display impaired chloroplast development, reduced photos

Chloroplast transformation for bioencapsulation and oral delivery using the immunoglobulin G fragment crystallizable (Fc) domain.

Proinsulin Like Growth Factor (prolGF1) and myostatin (Mstn) regulate muscle regeneration when intravenously delivered. We set out to test if chloroplast bioencapsulated forms of these proteins may serve as a non-invasive means of drug delivery through the digestive system. We created tobacco (Nicotiana tabacum) plants carrying GFP-Fc1, proIGF-I-Fc1, and Mstn-Fc1 fusion genes, in which fusion with the immunoglobulin G Fc domain improved both protein stability and absorption in the small intestine. No transplastomic plants were obtained with the Mstn-Fc1 gene, suggesting that the protein is tox

The whole chloroplast genome of Secale strictum subsp. kuprijanovii, a wild and perennial kinship to cultivated rye.

Secale strictum subsp. kuprijanovii is a perennial, hermaphrodite wild rye species and a progenitor of the modern cultivated rye, Secale cereale. With high adaptive capacity in stress conditions, it is valuable for enriching the germplasm resources of rye. Therefore, to elucidate its genetic and phylogenetic relationship is of great importance. We hereby sequenced, assembled and presented for the first time the complete chloroplast genome of this less studied species. The whole genome is 137,079 bp in size, including a large single copy region of 81,099 bp, a small single copy region of 12,820

Proteome analysis of chloroplast proteins in stage albinism line of winter wheat (triticum aestivum) FA85.

The "stage albinism line of winter wheat" FA85 was a specific natural mutant strain on leaf color. This physiological mutation was controlled by cytogene. In order to reveal the genetic and biochemical mechanism of albinism, 2-DE was used to investigate the difference of chloroplast protein expression pattern between FA85 and its parent wheat Aibian 1. From the results of 2-DE gels analysis, approximately 683 spots were detected on each gel, and 57 spots were expressed differently at least two-fold. Using MALDI-TOF/TOF MS, 14 of 57 spots were identified, which could be categorized into four cl

New advances in the production of edible plant vaccines: chloroplast expression of a tetanus vaccine antigen, TetC.

Vaccines are a proven method of controlling disease. However there are issues with the delivery and administration of vaccines. A particular problem is that the majority of vaccines currently used are injected, which can be unsafe if needles are reused in areas where blood-borne diseases are prevalent. Vaccines targeting the mucosal immune system avoid many of the problems associated with injections. One potential form of mucosal vaccine is based on the expression of vaccine antigens in plants. Current research in this area has focused on the expression of immunogens from the plant's nuclear g
