Investigation of the association of growth rate in grower-finishing pigs with the quantification of Lawsonia intracellularis and porcine circovirus type 2.

As a part of a prospective cohort study in four herds, a nested case control study was carried out. Five slow growing pigs (cases) and five fast growing pigs (controls) out of 60 pigs were selected for euthanasia and laboratory examination at the end of the study in each herd. A total of 238 pigs, all approximately 12 weeks old, were included in the study during the first week in the grower-finisher barn. In each herd, approximately 60 pigs from four pens were individually ear tagged. The pigs were weighed at the beginning of the study and at the end of the 6-8 weeks observation period. Clinic

Distribution of porcine circovirus 2 cap antigen in the lymphoid tissue of pigs affected by postweaning multisystemic wasting syndrome.

The lymphatic organs of 50 pigs from a total of eight farms located at different sites in the epizootiological region of North Bačka County were studied to obtain data on the prevalence of circoviral infections in Serbia. All of the pigs examined had clinical signs suggestive of postweaning multisystemic wasting syndrome (PMWS). All pigs underwent necropsy and tissue samples were taken for histopathological, immunohistochemical (IHC) and PCR analysis. The presence of porcine circovirus 2 (PCV2) was established by PCR analysis in the organs of the pigs tested. The most frequent histopathologica

A genetically engineered chimeric vaccine against porcine circovirus type 2 (PCV2) is genetically stable in vitro and in vivo.

A vaccine against porcine circovirus type 2 (PCV2), designated PCV1-2 chimera, was recently developed by replacing the capsid gene of the non-pathogenic PCV1 with that of PCV2. The PCV1-2 chimera virus is attenuated in pigs but induces protective immunity against PCV2. In this study, the genetic stability of the PCV1-2 chimera was evaluated for its potential use as a live vaccine. The PCV1-2 chimera virus was serially passaged 11 times in PK-15 cells and 3 times in pigs. The PCV1-2 chimera virus used in this study contained a tracking marker mutation in the capsid gene (F to V at amino acid po

Assessment of a porcine circovirus type 2 vaccine prototype through anatomopathological analysis and its correlation with blood viral load.

Porcine circovirus type 2 (PCV2) is an endemic pathogen of global relevance, responsible for porcine circovirus-associated diseases (PCVAD). Consequently, vaccination against PCV2 is a standard practice in intensive swine production systems. This study aimed to evaluate the effect of a prototype vaccine on PCV2 viral load and associated lesions. Thirty-nine pigs from a high-health-status farm were randomly assigned to three experimental groups (n = 13 per group), corresponding to a placebo group, a commercial vaccine group and a prototype vaccine group. Viral load was assessed by qPCR from ser

A multiplex real-time PCR assay for identification porcine circovirus 2, atypical porcine pestivirus, and classical swine fever virus.

BACKGROUND: Porcine circovirus 2 (PCV2), atypical porcine pestivirus (APPV), and classical swine fever virus (CSFV) are significant pathogens causing congenital tremor in swine, resulting in substantial economic losses worldwide. METHODS: A TaqMan multiplex real-time PCR method was developed to detect PCV2, APPV, and CSFV, simultaneously. This method was then applied to investigate 476 clinical samples from newborn piglets with congenital tremor symptoms. RESULTS: This method exhibited high specificity with no cross-reactivity with other viruses. Furthermore, the assay demonstrated remarkable

A novel linear B cell epitope of the porcine circovirus type 3 capsid protein identified by phage display technology.

Porcine circovirus type 3 (PCV3) is endemic in swine worldwide and causes reproductive disorders, dermatitis and nephrotic syndrome, and multi-organ inflammation. PCV3 capsid protein (Cap) can self-assemble into virus-like particles (VLPs), and is an ideal candidate for vaccines and diagnostic reagents. In this study, the recombinant PCV3 Cap protein was successfully expressed in E. coli by deleting the nuclear localization sequence (NLS). The PCV3 VLPs were observed by transmission electron microscopy, and its immunogenicity was evaluated in six-week-old female BALB/c mice. A monoclonal antibo

Protective immunity induced by concurrent intradermal injection of porcine circovirus type 2 and Mycoplasma hyopneumoniae inactivated vaccines in pigs.

Vaccines against porcine circovirus type 2 (PCV2) and Mycoplasma hyopneumoniae (Mhp) are routinely used by intramuscular injection. However, since intramuscular vaccination causes stress and increases the risk of cross-contamination among pigs, research on intradermal vaccination is currently being actively conducted. This study was designed to evaluate the efficacy of intradermally administered inactivated vaccines against PCV2 and Mhp in pigs. Three-week-old specific pathogen-free pigs were divided into three groups (5 pigs per group). Pigs in the two groups were intradermally vaccinated wit

High Co-infection Status of Novel Porcine Parvovirus 7 With Porcine Circovirus 3 in Sows That Experienced Reproductive Failure.

Porcine parvoviruses (PPVs) and porcine circoviruses (PCVs) infect pigs worldwide, with PPV1-7 and PCV2 infections common in pigs. Although PPV7 was only identified in 2016, co-infection of PPV7 and PCV2 is already common, and PPV7 may stimulate PCV2 replication. PCV3, a novel type of circovirus, is prevalent in pig populations worldwide and considered to cause reproductive disorders and dermatitis nephrotic syndrome. In recent studies, pigs were commonly infected with both PCV3 and PPV7. Our objective was to investigate the co-infections between PPV7 and PCV3 in samples from swine on farms in

Viral metagenomics in Brazilian Pekin ducks identifies two gyrovirus, including a new species, and the potentially pathogenic duck circovirus.

Viral metagenomics coupled to high-throughput sequencing has provided a powerful tool for large-scale detection of known and unknown viruses associated to distinct hosts and environments. Using this approach, known and novel viruses have been characterized from sylvatic and commercial avian hosts, increasing our understanding of the viral diversity in these species. In the present work we applied an exploratory viral metagenomics on organs (spleen, liver and bursa of Fabricious) of Pekin ducks from Southern Brazil. The virome contained sequences related to a known duck pathogen (duck circoviru

Clearance of porcine circovirus and porcine parvovirus from porcine-derived pepsin by low pH inactivation and cation exchange chromatography.

The contamination of oral rotavirus vaccines by porcine circovirus (PCV) raised questions about potential PCV contamination of other biological products when porcine trypsin or pepsin is used in production process. Several methods can be potentially implemented as a safety barrier when animal derived trypsin or pepsin is used. Removal of PCV is difficult by the commonly used viral filters with the pore size cutoff of approximately 20 nm because of the smaller size of PCV particles that are around 17 nm. It was speculated that operating the chromatography step at a pH higher than pepsin's low p

Full-genome sequences of porcine circovirus 3 (PCV3) and high prevalence in mummified fetuses from commercial farms in Brazil.

The clinical implications of recently discovered porcine circovirus 3 (PCV3) infections are still unknown. The potential role of this emerging virus in reproductive loss in swine has been described. Herein, we report a high prevalence of PCV3 in mummified fetuses from sows maintained in modern farms in Rio Grande do Sul, Santa Catarina, Paraná, Goiás, and Mato Grosso do Sul states, Brazil. For this analysis, 276 mummified fetuses from 11 commercial swine farms were included. The presence of PCV3 DNA was confirmed using PCR, and the complete sequence of five different viral strains was obtained

A Porcine circovirus type 2b (PCV2b)-based experimental vaccine is effective in the PCV2b-Mycoplasma hyopneumoniae coinfection pig model.

Porcine circovirus type 2 (PCV2) is one of the major swine pathogens causing high economic losses due to PCV2-associated disease (PCVAD). PCV2 infection is not only immunosuppressive by damaging lymphoid tissues but is also exacerbated by co-infections with other pathogens including Mycoplasma hyopneumoniae. While PCV2 can be divided into several genotypes, currently only PCV2a, PCV2b and PCV2d are globally prevalent and considered of major importance. Most commercial PCV2 vaccines are based on PCV2a isolates; however, the high prevalence of PCV2b and PCV2d in the global pig population is rais

Immunity Elicited by an Experimental Vaccine Based on Recombinant Flagellin-Porcine Circovirus Type 2 Cap Fusion Protein in Piglets.

In a recent study, we reported that a recombinant protein from fusion expression of flagellin to porcine circovirus type 2 (PCV2) Cap induced robust humoral and cell-mediated immunity that afforded full protection for PCV2 infection using BALB/c mice. Here, we further evaluated the immunogenicity and protection of the recombinant protein using specific pathogen free (SPF) pigs. Twenty-five 3-week-old piglets without passively acquired immunity were divided into 5 groups. All piglets except negative controls were challenged with a virulent PCV2 at 21 days after booster vaccination and necropsie

Evaluation of natural porcine circovirus type 2 (PCV2) subclinical infection and seroconversion dynamics in piglets vaccinated at different ages.

This study aimed to determine the porcine circovirus type 2 (PCV2) serological and virological dynamics in piglets vaccinated at different ages in a PCV2 subclinical infection (PCV2-SI) scenario. Six hundred and forty-four 2 week-old healthy piglets were selected and distributed into four treatment groups: vaccination at 3, 6 or 10 weeks of age (3W-VAC, 6W-VAC and 10W-VAC groups, respectively) and unvaccinated pigs (NON-VAC group). Blood (n = 112 pigs) and oral fluid (OF) (n = 40 pens) samples were taken throughout the study to assess PCV2 load, humoral immunity and viral genotyping. Percentag

Construction and immunogenicity of a recombinant pseudorabies virus co-expressing porcine circovirus type 2 capsid protein and interleukin 18.

A novel recombinant pseudorabies virus (PRV) expressing porcine circovirus type 2 (PCV2) capsid protein and IL-18 was constructed. The PCV2 open reading frame 2 (ORF2) and porcine IL-18 genes were amplified by PCR and then inserted into the PRV transfer vector (pG) to produce a recombinant plasmid (pGO18). Plasmid pGO18 was transfected into porcine kidney cell (PK15) pre-infected with PRV HB98 vaccine strain to generate a recombinant virus. The recombinant virus PRV-ORF2-IL18 was purified by green fluorescent plaque purification and the inserts were confirmed by PCR, enzyme digestion, sequenci

Assessment of neutralizing and non-neutralizing antibody responses against Porcine circovirus 2 in vaccinated and non-vaccinated farmed pigs.

Vaccination is the most efficacious procedure to curtail Porcine circovirus 2 (PCV2)-associated diseases (PCVAD). Experimental studies indicate that PCV2 vaccine-induced virus-neutralizing antibodies play a major role in protection from PCVAD. However, the immune response to PCV2 vaccination of pigs on farms is less clear. Analysing groups of age-matched vaccinated and non-vaccinated farmed pigs, we found significantly increased levels of virus-neutralizing antibodies only in vaccinated pigs belonging to the age group with the highest risk for developing PCVAD. Serum levels of PCV2 genomes wer

Porcine circovirus type 2 decreases the infection and replication of attenuated classical swine fever virus in porcine alveolar macrophages.

Recently, it has been noted that porcine circovirus type 2 (PCV2) infection adversely affects the protective efficacy of Lapinized Philippines Coronel (LPC) vaccine, an attenuated strain of classical swine fever virus (CSFV), in pigs. In order to investigate the possible mechanisms of the PCV2-derived interference, an in vitro model was established to study the interaction of LPC virus (LPCV) and PCV2 in porcine alveolar macrophages (AMs). The results showed that PCV2 reduced the LPCV infection in AMs and the levels of PCV2-derived interference were dose-dependent. The PCV2-derived interferenc

Testicular processing fluid as a useful matrix for the detection of porcine circovirus type 2 DNA and virus-specific antibodies.

INTRODUCTION: Testicular processing fluid (PF) obtained during boar castration may serve as a diagnostic matrix for monitoring porcine circovirus type 2 (PCV2) presence. METHODS: Anti- PCV2 antibodies in PF were detected using an indirect ELISA, and PCV2 DNA was detected by real-time PCR (qPCR), and the effects of sample pooling were evaluated. Paired sera and PF from boars and sera from gilts were tested with commercial ELISA and qPCR kits. PF-specific cut-offs were set by ROC (ELISA OD; qPCR Ct).
