Assessing Cardiac Amyloidosis Subtypes by Unsupervised Phenotype Clustering Analysis.

BACKGROUND: Cardiac amyloidosis (CA) is a set of amyloid diseases with usually predominant cardiac symptoms, including light-chain amyloidosis (AL), hereditary variant transthyretin amyloidosis (ATTRv), and wild-type transthyretin amyloidosis (ATTRwt). CA are characterized by high heterogeneity in phenotypes leading to diagnosis delay and worsened outcomes. OBJECTIVES: The authors used clustering analysis to identify typical clinical profiles in a large population of patients with suspected CA. METHODS: Data were collected from the French Referral Center for Cardiac Amyloidosis database (Hôp

QTAIM analysis dataset for non-covalent interactions in furan clusters.

Furan clusters are very important to understand the dynamics and properties of the furan solvent. They can be used combined with quantum cluster equilibrium theory to theoretically determine the thermodynamics properties of the furan solvent. To understand the structures of the furan clusters, one needs to understand the non-covalent interactions that hold the furan molecules together. In this paper, we have provided the data necessary to understand the non-covalent interactions in furan clusters. Firstly, the structures of the furan clusters have been generated using classical molecular dynam

Shunt failure clusters: an analysis of multiple, frequent shunt failures.

OBJECTIVE: Repeated failure of ventriculoperitoneal shunts (VPSs) is a problem familiar to pediatric neurosurgeons and patients. While there have been many studies to determine what factors are associated with the first shunt failure, studies of subsequent failures are much less common. The purpose of this study was to identify the prevalence and associated risk factors of clustered shunt failures (defined as 3 or more VPS operations within 3 months). METHODS: The authors reviewed prospectively collected records from all patients who underwent VPS surgery from 2008 to 2017 at their institutio

Analysis of muscle fiber clustering in the diaphragm muscle of sarcopenic mice.

INTRODUCTION: Sarcopenia likely comprises muscle fiber denervation and re-innervation, resulting in clustering of muscle fibers of the same type (classified by myosin heavy chain isoform composition). Development of methodology to quantitatively evaluate clustering of muscle fibers according to fiber type is necessary. METHODS: Fiber type specific immunofluorescence histology was used to quantify fiber clustering in murine diaphragm muscle (n = 15) at ages 6 and 24 months. RESULTS: With age, fiber type clustering is evidenced by fiber type specific changes in distances between fibers, specif

Cancer symptom clusters: an exploratory analysis of eight statistical techniques.

CONTEXT: Statistical methods to identify symptom clusters (SC) have varied between studies. The optimal statistical method to identify SC is unknown. OBJECTIVES: Our primary objective was to explore whether eight different statistical techniques applied to a single data set produced different SC. A secondary objective was to investigate whether SC identified by these techniques resembled those from our original study. METHODS: We reanalyzed a symptom data set of 1000 patients with advanced cancer. Eight separate cluster analyses were conducted on both prevalence and severity of 38 symptoms.

Analysis of FOXF1 and the FOX gene cluster in patients with VACTERL association.

VACTERL association, a relatively common condition with an incidence of approximately 1 in 20,000 -35,000 births, is a non-random association of birth defects that includes vertebral defects (V), anal atresia (A), cardiac defects (C), tracheo-esophageal fistula (TE), renal anomalies (R) and limb malformations (L). Although the etiology is unknown in the majority of patients, there is evidence that it is causally heterogeneous. Several studies have shown evidence for inheritance in VACTERL, implying a role for genetic loci. Recently, patients with component features of VACTERL and a lethal deve

A cluster analysis of not-in-treatment drug users at risk for HIV infection.

The purpose of the analysis described here was to classify not-in-treatment drug users participating in the National Institute on Drug Abuse (NIDA)-sponsored Cooperative Agreement study into several "homogeneous" HIV risk groups using cluster analysis. Data for this analysis (N=17,778) were collected at 19 study sites in the United States and Puerto Rico. Measures selected for the cluster analysis were limited to (a) current drug use and HIV risk behaviors, (b) mutually exclusive behaviors, (c) behaviors directly related to HIV risk, and (d) behaviors that were not statistically rare. Eight ho

Biochemical analysis of APOBEC3A-induced mutational signatures and clustered mutagenesis.

APOBEC3A is a single-stranded DNA cytidine deaminase that is involved in mutagenic processes in various cancers. Ample evidence indicates that APOBEC3A causes two major mutational signatures (SBS2 and SBS13) in cancer cells and cellular model systems, but the detailed molecular mechanisms that generate these two signatures from a single deaminase remain unclear. APOBEC3A-induced mutations are preferentially observed at DNA hairpin structures and are often observed as clusters in cancer genomes, but the molecular mechanisms behind the clustered mutations are unclear. Here, we developed a cell-f

Sputum Transcriptomic Analysis and Clustering Reveals Insight Into Asthma Heterogeneity.

INTRODUCTION: Asthma is a heterogenous disease shaped by different inflammatory pathways. The aim is to investigate transcriptomic profiles in asthmatic patients and associate these with inflammation, airway damage and lung function. METHODS: Adult asthma patients attending the outpatient pneumology clinic in our tertiary center, underwent diagnostic sputum induction and upon consent remaining sputum RNA was used for bulk RNA-sequencing (n = 56) coupled with unsupervised clustering. A retrospective analysis of comorbidities was performed. Sputum cytokine mRNA levels were determined via qPCR.

RaCE: A rank-clustering estimation method for network meta-analysis.

Ranking multiple interventions is a crucial task in network meta-analysis (NMA) to guide clinical and policy decisions. However, conventional ranking methods often oversimplify treatment distinctions, potentially yielding misleading conclusions due to inherent uncertainty in relative intervention effects. To address these limitations, we propose a novel Bayesian rank-clustering estimation approach, termed rank-clustering estimation (RaCE), specifically developed for NMA. Rather than identifying a single "best" intervention, RaCE enables the probabilistic clustering of interventions with simila

Rhinoconjunctivitis symptoms in children and adolescents with asthma: Longitudinal clustering analysis.

BACKGROUND: Rhinoconjunctivitis phenotypes are conventionally described on the basis of symptom severity, duration and seasonality, and aeroallergen sensitization. It is not known whether these phenotypes fully reflect the patterns of symptoms seen at a population level. OBJECTIVE: We sought to identify phenotypes of rhinoconjunctivitis on the basis of symptom intensity and seasonality using an unbiased approach and to compare their characteristics. METHODS: A cohort of children with asthma in low-income urban environments was prospectively followed with a rhinoconjunctivitis activity questi

Factor analysis and clustering of motor and psychiatric dimensions in idiopathic blepharospasm.

INTRODUCTION: Idiopathic blepharospasm is a clinically heterogeneous form of focal dystonia, also associated with psychiatric symptoms. The identification of the most relevant sets of motor and psychiatric manifestations may help better understand the specific phenomenology of the condition and delineate blepharospasm subtypes more accurately. METHODS: Patients with idiopathic blepharospasm were from the Dystonia Coalition project. Factor analysis of several motor and psychiatric scales was performed to identify the relevant determinants of blepharospasm severity. The selected items were then

Maternal mortality in Brazil: an analysis of temporal trends and spatial clustering.

This article aims to analyze spatial and temporal patterns of maternal mortality in Brazil during the period 2010-2020 and identify related socioeconomic indicators. We conducted an ecological study of the maternal mortality ratio (MMR) in Brazil's municipalities using secondary data. Temporal analysis was performed using the joinpoint method. Bayesian statistics, spatial autocorrelation, the Getis Ord Gi* technique and the scan statistic were used to identify spatial clusters, and multiple non-spatial and spatial regression models were used to assess the association between factors and the MM

Computational Comparative Analysis of Small Atomically Precise Copper Clusters.

Atomically precise copper clusters (APC) have attracted attention for their promise in sensing, water remediation, and electrochemical technologies. However, smaller-sized APCs and the evolution of their properties as a function of size and composition are not clearly understood. Here, we have performed an investigation into the electronic structure, geometry, and optical properties of small atomically precise copper clusters using density functional theory (DFT) and time-dependent DFT. Through comparative analysis, we show that the electronic structures of the experimentally characterized clu

Analysis of neural clusters due to deep brain stimulation pulses.

Deep brain stimulation (DBS) is an established method for treating pathological conditions such as Parkinson's disease, dystonia, Tourette syndrome, and essential tremor. While the precise mechanisms which underly the effectiveness of DBS are not fully understood, several theoretical studies of populations of neural oscillators stimulated by periodic pulses have suggested that this may be related to clustering, in which subpopulations of the neurons are synchronized, but the subpopulations are desynchronized with respect to each other. The details of the clustering behavior depend on the frequ

Global pathway view analysis of microRNA clusters in myasthenia gravis.

The significant roles of microRNAs (miRNAs) in the pathogenesis of myasthenia gravis (MG) have been observed in numerous previous studies. The impact of miRNA clusters on immunity has been demonstrated in previous years; however, the regulation of miRNA clusters in MG remains to be elucidated. In the present study, 245 MG risk genes were collected and 99 MG risk pathways enriched by these genes were identified. A catalog of 126 MG risk miRNAs was then created; the MG risk miRNAs were located on each chromosome and a miRNA cluster was defined as a number of miRNAs with a relative distance of <6
