Cognitive dysfunction in mice infected with Plasmodium berghei strain ANKA.

Cerebral malaria complicated by cognitive sequelae is a major cause of morbidity in humans infected with Plasmodium falciparum. To model cognitive function after malaria, we created a rodent model of cerebral malaria by infecting C57BL/6 mice with Plasmodium berghei strain ANKA. After 7 days, an object-recognition test of working memory revealed a significant impairment in the visual memory of infected mice. This impairment was observed in the absence of confounding effects of infection. The cognitive dysfunction correlated with hemorrhage and inflammation. Furthermore, microglial activity and

Potential mechanisms and markers of critical illness-associated cognitive dysfunction.

PURPOSE OF REVIEW: To review the current understanding of the potential mechanisms of critical illness-associated cognitive dysfunction and to provide insight into markers that could be used to evaluate the influence of specific mechanisms in individual patients. RECENT FINDINGS: Cognitive dysfunction is common in critically ill patients, not only during the acute illness but also long after its resolution. Several pathophysiologic mechanisms are thought to underlie critical illness-associated cognitive dysfunction, including neurotransmitter abnormalities and occult diffuse brain injury. Mar

Targeting Mitochondria for Postoperative Cognitive Dysfunction: From Mechanisms to Therapeutics.

Postoperative cognitive dysfunction (POCD) is a frequent neurological complication in older surgical patients, leading to substantial declines in quality of life and placing a considerable burden on society. This review provides a comprehensive overview of how mitochondrial dysfunction contributes to POCD, including disrupted energy metabolism, excessive reactive oxygen species production, calcium imbalance, and abnormalities in mitochondrial dynamics and quality control processes. These mitochondrial impairments further trigger neuroinflammation and activate multiple cell death pathways. In a

Linking subjective cognitive decline to glymphatic dysfunction in Parkinson's disease.

BACKGROUND: The role of glymphatic system in the subjective cognitive decline (SCD) stage of Parkinson's disease (PD) remains unexplored. This study investigated whether glymphatic dysfunction occurs in PD-SCD and its potential role in mediating the link between sleep disturbance and cognitive impairment. METHODS: We recruited 73 PD patients, categorized into three groups: PD with normal cognition (PD-NC, n = 24), PD-SCD (n = 22), and PD with mild cognitive impairment (PD-MCI, n = 27), alongside 27 healthy controls (HC). All participants underwent 3 T MRI to calculate the diffusion tensor ima

Features of cognitive dysfunction in late-onset temporal lobe epilepsy.

OBJECTIVE: A global increase in the aging population has resulted in more patients with late-onset epilepsy. Late-onset temporal lobe epilepsy (LOTLE) is one of the most common types of late-onset epilepsy and is frequently accompanied by memory impairments. Here, we investigated the relationships between memory function and clinical characteristics, including findings from long-term video-electroencephalograph (LVEEG) monitoring, in LOTLE. METHODS: We evaluated patients who had been diagnosed with LOTLE and Underwent LVEEG and neuropsychological tests (e.g., the Wechsler Memory Scale-Revised

The Relationship between Glymphatic Dysfunction and Post-stroke Cognitive Impairment.

BACKGROUND: Glymphatic dysfunction is proposed as a final common pathway to dementia. Cognitive impairment following ischemic stroke can gradually worsen, potentially leading to post-stroke dementia. This study aimed to examine the changes in glymphatic function in post-stroke patients and explore its relationship with cognition. METHODS: A total of thirty-two post-stroke patients and twenty-seven healthy controls (HCs) matched for age, sex, and educational level were enrolled in this study. All participants underwent neurological MRI scans and comprehensive cognitive assessments six months f

[Brain Pathology of Cognitive Dysfunction in Aged Non-human Animals].

Cognitive dysfunction, characterized by the deposition of amyloid β (Aβ), such as senile plaque (SP) and cerebral amyloid angiopathy (CAA), and phosphorylated tau are observed in several animal species, including primates, dogs, and cats, suggesting that these disorders are universal age-related phenomena in vertebrates. Additionally, argyrophilic neurofibrillary tangles positive for phosphorylated tau have been observed in primates, marine mammals, and felines. Recent findings regarding the pathology of cognitive dysfunction in aged non-human animals provide valuable information from a compar

Glymphatic system dysfunction in adult ADHD: Relationship to cognitive performance.

OBJECTIVES: While attention-deficit/hyperactivity disorder (ADHD) persists into adulthood, the relationship between glymphatic system function and cognitive performance in adult ADHD remains unclear. This study investigated the association between glymphatic system markers and cognitive outcomes in adults with ADHD. METHODS: This case-control study includes 41 adults with ADHD and 108 age-matched healthy controls (HCs). Glymphatic function was evaluated using choroid plexus volume (CPV), diffusion tensor imaging along the perivascular space (DTI-ALPS) index and coupling between blood‑oxygen-l

Optimized 5-HT2b inhibitors for neuropsychiatric syndromes with cognitive dysfunction.

INTRODUCTION: Neuropsychiatric syndromes such as anxiety and agitation are clinical presentations common to diverse neurodegenerative diseases and brain injury sequelae. They are a concern due to the impact on cognition, social interactions, and non-pharmacological treatments. Cognitive or behavioral disturbances occur at early disease stages and increase with disease progression. Coincident pathologies include the loss of serotonin (5-HT) neurons and appearance of neurofibrillary tangles in the raphe nucleus. Brain 5-HT2b receptor (5-HT2bR) levels are increased in Alzheimer's disease (AD), am

Alzheimer's disease and diabetes-associated cognitive dysfunction: the microglia link?

Alzheimer's disease (AD) is characterized by the accumulation of amyloid-β (Aβ) plaques and the aggregation of tau protein, resulting in intense memory loss and dementia. Diabetes-associated cognitive dysfunction (DACD) is a complication of diabetes mellitus, which is associated with decreased cognitive function and impaired memory. A growing body of literature emphasize the involvement of microglia in AD and DACD. Although AD and DACD share some common features related to symptomatology and pathophysiology, the characteristics and heterogeneity of microglia remain largely unknown in these two

Meningeal lymphatic dysfunction drives cognitive impairment after experimental subarachnoid hemorrhage.

More than half of subarachnoid hemorrhage (SAH) survivors develop delayed cognitive dysfunction, but the underlying mechanisms remain elusive. This study investigated the role of meningeal lymphatic vessels (mLVs) in this complication by examining their structural integrity, drainage capacity, and association with cognitive deficits post-SAH. In adult male C57BL/6J mice in which SAH was induced by intracisternal injection of autologous blood, spatial learning and memory, and hippocampal CA1 neuronal activity were impaired as early as 1 month post-surgery, with a marked exacerbation of these de

Microglia-Mediated Synaptic Dysfunction Contributes to Chemotherapy-Related Cognitive Impairment.

Chemotherapy-related cognitive impairment (CRCI) significantly impacts cancer survivors. Due to unclear mechanisms, effective treatments for cognitive deficits are lacking. Here, we examined if microglia-mediated deficits in synaptic plasticity drive CRCI. Adult male mice were treated with the chemotherapeutic drugs 5-fluorouracil and leucovorin (5-Fu/LV, intraperitoneal injection, I.P.) on Days 1, 8, and 15 at a dosage of 50 mg/kg for 5-Fu and 90 mg/kg for LV for 3 weeks. Cognitive function was assessed using a novel object recognition (NOR) test 4 weeks after completion of 5-Fu/LV treatment.

The cerebellum beyond motor control: cognitive dysfunction in spinocerebellar ataxias.

Spinocerebellar ataxias (SCAs) are a group of neurodegenerative disorders primarily characterized by progressive ataxia due to degeneration of the cerebellum and its afferent and efferent pathways. Although cerebellar motor symptoms are the clinical hallmark, considerable phenotypic variability exists across SCA subtypes and among individuals with the same genetic mutation. Depending on the subtype and the extent of cerebellar and extracerebellar involvement, various cognitive domains-including memory, attention, executive function, and language-may also be affected. Notably, the cognitive and

Postoperative cognitive dysfunction: Review of pathophysiology, diagnostics and preventive strategies.

Postoperative cognitive impairment is a common disorder after major surgery. Advances in medicine and treatment have resulted in an increasingly ageing population undergoing major surgical procedures. Since age is the most important risk factor for postoperative cognitive decline, it is not surprising that impairment of cognitive functions after surgery was recorded in almost a third of elderly patients. Postoperative cognitive dysfunction is part of the spectrum of postoperative cognitive impairment and researchers often confuse it with postoperative delirium and delayed neurocognitive recove

Cognitive Dysfunction and Exercise: From Epigenetic to Genetic Molecular Mechanisms.

Maintaining good health is crucial, and exercise plays a vital role in achieving this goal. It offers a range of positive benefits for cognitive function, regardless of age. However, as our population ages and life expectancy increases, cognitive impairment has become a prevalent issue, often coexisting with age-related neurodegenerative conditions. This can result in devastating consequences such as memory loss, difficulty speaking, and confusion, greatly hindering one's ability to lead an ordinary life. In addition, the decrease in mental capacity has a significant effect on an individual's

Endothelial Dysfunction and Pre-Existing Cognitive Disorders in Stroke Patients.

BACKGROUND: The origin of pre-existing cognitive impairment in stroke patients remains controversial, with a vascular or a degenerative hypothesis. OBJECTIVE: To determine whether endothelial dysfunction is associated with pre-existing cognitive problems, lesion load and biological anomalies in stroke patients. METHODS: Patients originated from the prospective STROKDEM study. The baseline cognitive state, assessed using the IQ-CODE, and risk factors for stroke were recorded at inclusion. Patients with an IQ-CODE score >64 were excluded. Endothelial function was determined 72 h after stroke s

Drop to Gate Nasal Drops Attenuates Sepsis-Induced Cognitive Dysfunction.

Nasal administration can bypass the blood-brain barrier and directly deliver drugs to the brain, providing a non-invasive route for central nervous system (CNS) diseases. Inspired by the appearance that a gate can block the outside world and the characteristics of the sol-gel transition can form a "gate" in the nasal cavity, a Drop to Gate nasal drop (DGND) is designed to set a gate in nose, which achieves protecting role from the influence of nasal environment. The DGND demonstrates the efficiency and application prospect of delivering drugs to the brain through the N-to-B. The effective conc
