[3H]-forskolin binding sites in rat submandibular salivary glands.

Specific [3H]-forskolin binding was linear as a function of the membrane protein concentration and reached steady-state by 30-40 min of incubation at 37 degrees C. Such binding was saturable and the bound radioligand could be readily dissociated by an excess of unlabelled forskolin. There was only one type of binding site, with a dissociation constant (Kd) of 0.16 microM. The concentration of the binding sites (Bmax) was 0.73 pmol/mg protein. There was no significant difference in the [3H]-forskolin binding characteristics in membranes of rats fed for 6 weeks on diets containing 10% corn oil,

Ca2+ not cyclic AMP mediates the fluid secretory response to isoproterenol in the rat mandibular salivary gland: whole-cell patch-clamp studies.

We have performed whole-cell patch-clamp studies on dispersed secretory cells of the rat mandibular gland to determine how beta-adrenergic stimulation causes fluid secretion. When the pipette contained a high K+ solution, the resting membrane potential averaged -33 mV +/- 1.1 (SEM, n = 34) and the clamped cell showed strong outward rectification. We monitored K+ and Cl- currents for periods of 15 min by recording the currents needed to clamp the cell potential at 0 and -80 mV, respectively. Isoproterenol (1-2 mumol/l) caused increases in the clamp current at 0 mV (the K+ current) and at -80 mV

Phorbol ester: effect on intraocular pressure, adenylate cyclase, and protein kinase in the rabbit eye.

Protein kinase C was identified as a major protein kinase enzyme activity in rabbit ciliary processes. Phorbol myristate acetate (4 beta-PMA) in the presence of Ca2+ activated protein kinase C but did not directly affect the cyclic AMP-dependent protein kinase enzyme isolated from ciliary processes. To elucidate possible roles of protein kinase C, PMA was injected intravitreally into rabbit eyes. Fifty pmoles of PMA produced approximately a 40% decrease of the intraocular pressure relative to the control eye lasting for more than 72 hr. A reduction of intraocular pressure was still elicited by

Potentiation of sympathetic neurosecretion by forskolin and cyclic AMP in the rabbit iris-ciliary body.

Forskolin has been reported to stimulate cAMP formation and reduce intraocular pressure in rabbit and primate eyes. In view of recent evidence for the involvement of cAMP in modulation of transmitter release at adrenergic synapses, we have investigated the presynaptic effects of forskolin and other cAMP activators on field-stimulated secretion of 3H-norepinephrine (3H-NE) in the isolated, perfused rabbit iris-ciliary body. Forskolin (10(-7)-10(-5) M) was found to markedly enhance stimulation-evoked 3H-NE release without affecting basal (spontaneous) release. The response to forskolin was poten

Simian virus 40 small-t antigen-induced theophylline resistance is not mediated by cyclic AMP.

Small-t antigen of simian virus 40 renders CV-1 cells resistant to growth arrest induced by theophylline and other methylxanthines. Elevated levels of cyclic AMP are not involved in growth arrest of CV-1 cells by methylxanthines, and small-t antigen does not alter cyclic AMP levels dramatically after infection.

cAMP-mediated regulation of chloride secretion by the opercular epithelium.

Catecholamine regulation of the Cl- secretion rate (short-circuit current, Isc) and adenosine 3',5'-cyclic monophosphate (cAMP) levels of the opercular epithelium was investigated by using 3-isobutyl-1-methylxanthine (IBMX), forskolin, and adrenergic agonists. In this tissue alpha-adrenergic agonists inhibit, and beta-adrenergic agonists stimulate, the Isc (J. Physiol. London 294: 483-495, 1979). IBMX and forskolin stimulated the Isc 125 and 85%, respectively, and simultaneously produced 2.5- and 70.0-fold elevations in the cAMP levels, respectively. These findings confirm previous observation

The adenylyl cyclase activator forskolin pretreatment reprograms CAR-T cells toward a stem-like state to enhance solid tumor therapy.

Chimeric antigen receptor (CAR)-T cell therapy has achieved remarkable success in hematological malignancies; however, its efficacy against solid tumors remains limited, primarily due to T cell exhaustion. Enhancing memory-like properties and stemness of T cells has emerged as a promising strategy to overcome this barrier. In this study, we demonstrate that Forskolin, a stem cell-inducing compound, reprograms CAR-T cells toward a memory-like state characterized by elevated stemness. Forskolin-pretreated CAR-T cells exhibited enriched memory phenotypes, reduced exhaustion, and enhanced anti-tum

Functional selectivity of cannabinoids at CB1 receptors: Comprehensive analysis of G proteins, β-arrestin and cAMP signalling dynamics in HEK293T cells.

Therapeutic targeting of CB1 has been limited by adverse effects of orthosteric ligands. Allosteric modulation and biased signalling have been proposed to achieve pathway-selective control, but the receptor's complex coupling complicates interpretation of ligand actions. In this study, we systematically compared orthosteric and allosteric CB1 ligands in transfected HEK293T cells by measuring G-protein coupling and downstream cAMP regulation in real time using NanoBiT and GloSensor assays. Orthosteric agonists CP 55,940 and methanandamide activated both Gi and Gs with weak and transient β-arres

Forskolin Enhances Urokinase Plasminogen Activator Secretion and Angiogenic Activity of Xeno-Free Cultures of Human Adipose Tissue-Derived Stem Cells.

Adipose tissue-derived stem cells (ASCs) hold significant potential for treating various clinical conditions. To enhance their regenerative properties, ASCs can be chemically stimulated using various in vitro protocols. However, unsatisfactory results persist, partly due to the relatively costly long-term methods. Furthermore, current culturing techniques often rely on the use of xenogenic fetal bovine serum that can be immunogenic, limiting clinical translations. To facilitate clinical translation of ASCs-derived therapeutics, the effect of different stimulation protocols on human ASCs cultur

Valproic acid elevates HIF-1α-mediated CGB expression and suppresses glucose uptake in BeWo cells.

Placental dysfunction can disrupt pregnancy. However, few studies have assessed the effects of chemical-induced toxicity on placental function. Here, we examined the effects of valproic acid (VPA) as a model chemical on production of hormones and on glucose uptake in human choriocarcinoma cell line BeWo. Cells were treated with forskolin to differentiate into syncytiotrophoblasts, which were then treated with VPA for 72 hr. Real-time RT-PCR analysis showed that VPA significantly increased the mRNA expression of chorionic gonadotropin β (CGB), a hormone that is produced by the placenta in the f

Effect of cAMP on short-circuit current in isolated human ciliary body.

BACKGROUND: Cyclic adenosine monophosphate (cAMP) could activate chloride channels in bovine ciliary body and trigger an increase in the ionic current (short-circuit current, Isc) across the ciliary processes in pigs. The purpose of this study was to investigate how cAMP modulates Isc in isolated human ciliary processes and the possible involvement of chloride transport across the tissue in cAMP-induced Isc change. METHODS: In an Ussing-type chamber system, the Isc changes induced by the cAMP analogue 8-bromo-cAMP and an adenylyl cyclase activator forskolin in isolated human ciliary processes

Cyclic-AMP inhibits nitric oxide-induced apoptosis in human osteoblast: the regulation of caspase-3, -6, -9 and the release of cytochrome c in nitric oxide-induced apoptosis by cAMP.

Nitric oxide (NO) induces apoptotic cell death and cAMP has a significantly protective effect on NO-induced cytotoxicity in human osteoblasts, MG-63 cells. Treatment with S-nitroso-N-acetylpenicillamine (SNAP) (0.6 mM) resulted in genomic DNA fragmentation, characteristic of apoptosis. However, concomitant incubation of the cells with either DBcAMP or forskolin markedly inhibited SNAP-induced apoptosis in a dose-dependent manner. Furthermore, pretreatment of MG-63 cells with H-89 or KT5720, which is known to inhibit cAMP-dependent protein kinase (PKA), abolished the protective effect of DBcAMP

Inhibition of bronchoprotective effects of beta2-adrenoceptor agonists by peroxynitrite in guinea pig airways.

Peroxynitrite plays an important role in the pathogenesis of inflammatory diseases, including those affecting the lung. In inflamed airways, simultaneous cellular production of superoxide anion (.O2-) and nitric oxide (NO) may occur, potentially resulting in continuous formation of peroxynitrite. However, because peroxynitrite has a short half-life, its in vivo physiologic effects in the airways may not be sufficiently evaluated with a single administration. Accordingly, this study was designed to use 3-morpholinosydnonimine (SIN-1), a compound that releases peroxynitrite, to determine whether

Invasion of cytotrophoblastic JEG-3 cells is stimulated by hCG in vitro.

Trophoblast invasion into the uterine wall is controlled by many factors. Previously, a human chorionic gonadotropin (hCG) receptor has been found to be expressed on invasive trophoblast as well as on choriocarcinoma cells implying a possible role for the hormone in trophoblast invasion. Therefore, this study examined the role of hCG in the invasion of trophoblastic (JEG-3) cells. Increasing hCG concentrations were applied in a trophoblast invasion model, JEG-3, through matrigel-coated filters. The proliferation was quantified by WST-1 cleavage assay. Cell migration was studied by examining th

No evidence for cell-to-cell coupling in rat colonic crypts: studies with Lucifer Yellow and with photobleaching.

Epithelial cells of exocrine glands (pancreas, lacrimal glands, salivary glands, sweat glands and gastric glands) are intimately linked together by gap junctions. Due to this close junctional coupling exocrine secretion occurs as the well concerted effort of a cell population. Colonic crypts have, on the one hand, anatomical and functional properties resembling those of exocrine glands (mostly crypt base cells) and, on the other hand, properties of absorbing cells (mostly surface cells). In the mid-crypt, depending on the functional status, absorption and secretion can occur. The present study

Effect of intracellular cAMP on K current in single gastric myocytes of guinea-pig.

In guinea-pig stomach, the effect of increased intracellular cAMP induced by forskolin, 8-Br-cAMP, or isobuthyl-methylxanthine (IBMX) on K current was studied in whole-cell currents. In enzymatically dispersed single cells, agents elevating [cAMP]in showed quite different effects on the IK depending on the recording conditions. Though using the same recording pipette containing 0.1 mM EGTA, agents elevating [cAMP]in reduced IK(Ca) recorded at various membrane potentials, while forskolin increased the spontaneous transient outward currents (STOCs) recorded at fixed holding potential, 0 mV. Attenuation of adenylate cyclase-induced increases in renal sodium excretion by the dopamine D-2 receptor agonist SK&F 89124.

1. Although the existence of D-2 receptor binding sites in kidney has been identified, their functional significance in terms of influencing renal sodium excretion is not clear. In the present study we have examined the renal effects of a selective D-2 receptor agonist, SK&F 89124, in anaesthetized rats. 2. Intravenous infusion of SK&F 89124 (0.3, 1 and 3 micrograms kg-1 min-1 respectively) produced dose-dependent decreases in mean arterial blood pressure, heart rate and renal blood flow without causing any significant changes in urine output, urinary sodium excretion, renal vascular resistanc

Oestrogen formation in genital and non-genital skin fibroblasts cultured from patients with hypospadias.

OBJECTIVE: Hypospadias is the most common birth defect in males. In most cases the aetiology is unknown. Since penile development is androgen dependent and oestrogen can modify androgen action, we compared the formation of oestrogen in penile tissue from patients with hypospadias to those with normal penile development. DESIGN AND PATIENTS: Oestrogen formation was assessed in fibroblast monolayers grown from biopsies of genital and non-genital skin from 11 males with normal genital development (controls) and 18 males with severe hypospadias utilizing the incorporation of tritium into H2O resu

Regulation of islet amyloid polypeptide in human pancreatic islets.

This study investigated the effect of glucose on islet amyloid polypeptide secretion, content, and mRNA synthesis of human pancreatic islets. The release of islet amyloid polypeptide from fresh isolated islets in response to glucose was parallel to that of insulin. The islet amyloid polypeptide-to-insulin molar ratios in response to 5.5 and 16.7 mM glucose were 1:16 and 1:15 respectively. Islets were cultured for 1 and 7 days at two different glucose concentrations (5.5 and 16.7 mM). The islet amyloid polypeptide response to the 1-day culture was similar to that of the fresh islets; however, a
