Bioavailability of copper in analytical-grade and feed-grade inorganic copper sources when fed to provide copper at levels below the chick's requirement.

Young chicks were fed a casein-soy protein concentrate basal diet (.56 mg Cu/kg) containing graded levels of added Cu (0, .5, 1.0 mg/kg) from analytical-grade (AG) CuSO4.5H2O, Cu2O, CuO, CuCO3.Cu(OH)2, CuCl, and also from feed-grade (FG) CuSO4.5H2O and FG-CuO. Weight gain, hematocrit, hemoglobin, plasma Cu, liver Cu, gall bladder (bile) Cu, and tendon lysine were assessed to determine whether dose-dependent Cu responses would occur. Gall bladder Cu concentration as well as total Cu content of the gall bladder were found to be linear functions of supplemental Cu intake. Common-intercept multipl

Level and Source of Copper Affects Gene Expression of Copper-Regulatory Proteins and Soluble and Mucosal Copper Concentrations in the Small Intestine of Weanling Pigs.

A study was conducted to determine the effects of the level and source of copper (Cu) on Cu concentrations and the mRNA expression of Cu regulatory proteins in the small intestine of pigs. Thirty weanling castrated male pigs, approximately 21 days of age, were stratified by weight and assigned to dietary treatments consisting of control (6.7 mg Cu/kg from feed ingredients; no supplemental Cu) or 225 mg supplemental Cu from either Cu sulfate (CuSO4) or tribasic Cu chloride (TBCC). Pigs were harvested on days 35 or 36. The soluble Cu and mucosal Cu concentrations in the duodenum, proximal jejunu

Comparison of the effect of dietary copper nanoparticles and one copper (II) salt on the copper biodistribution and gastrointestinal and hepatic morphology and function in a rat model.

The aim of the study was to investigate the effect of two forms (CuCO3 (CuS); and Cu nanoparticles (CuNP)) and dosages (standard 6.5 mg/kg (H), half of the standard (L)) of additional dietary Cu administered to growing rats on gastrointestinal and hepatic function and morphology. Copper in the form of CuNP vs CuS caused lower Cu faecal/urinal excretion and increased Cu accumulation in the brain tissue. Hepatic high-grade hydropic degeneration and necrotic lesions were observed only in the CuNP-H animals. In the lower gut, the dietary application of CuNP stifled bacterial enzymatic activity of

Copper(I)-binding properties of de-coppering drugs for the treatment of Wilson disease. α-Lipoic acid as a potential anti-copper agent.

Wilson disease is an autosomal recessive genetic disorder caused by loss-of-function mutations in the P-type copper ATPase, ATP7B, which leads to toxic accumulation of copper mainly in the liver and brain. Wilson disease is treatable, primarily by copper-chelation therapy, which promotes copper excretion. Although several de-coppering drugs are currently available, their Cu(I)-binding affinities have not been quantitatively characterized. Here we determined the Cu(I)-binding affinities of five major de-coppering drugs - D-penicillamine, trientine, 2,3-dimercapto-1-propanol, meso-2,3-dimercapto

Nutritional copper deficiency in severely handicapped patients on a low copper enteral diet for a prolonged period: estimation of the required dose of dietary copper.

Six cases of nutritional copper deficiency were identified in a nursery institute for severely handicapped patients. All had been on prolonged enteral feeding of a copper-deficient diet and all had abnormalities related to the copper deficiency. Two of them had leukopenia, with or without macrocytic and normochromic anemia. After the oral administration of copper, the clinical and laboratory observations including neutrophil counts and serum copper and ceruloplasmin levels showed a complete recovery. The dietary copper requirement was estimated to be approximately 20 micrograms/kg/day for thes

Enzyme-bound copper of dopamine beta-monooxygenase. Activation of the holoenzyme by added copper and uncoupling of electron transfer from hydroxylation by copper salicylate.

preparations of dopamine beta-monooxygenase containing a full complement of copper (4.2 copper atoms per tetramer) show increased ascorbate-supported catalytic activities after addition of an excess of copper ions. The significance of this observation on the question of the number of copper atoms per active site is discussed. Low molecular weight copper complexes such as copper salicylate cause uncoupling of electron transport from hydroxylation. This uncoupling is probably the reason for the well-known inhibition of this enzyme observed at high copper concentration. The onset of inhibition by

Copper-marginal and copper-deficient diets decrease aortic prostacyclin production and copper-dependent superoxide dismutase activity, and increase aortic lipid peroxidation in rats.

Agonist challenged aortic prostacyclin production was examined in copper-adequate, -marginal and -deficient rats fed AIN-based diets providing 6.7, 1.7 and 0.8 micrograms Cu/g, respectively. Aortic rings were incubated in Krebs-Henseleit salts, 10 mmol/L HEPES buffer, pH 7.4, 95%:5% O2:CO2, 37 degrees C, and equilibrated for 1 h. Equilibrated rings were challenged with buffer (basal), 273.0 nmol/L thrombin and angiotensin II at 84.6 pmol/L and 846.0 pmol/L. Prostacyclin production, determined at 10 minutes by RIA as 6-keto prostaglandin F1 alpha, in basal and 84.6 pmol/L angiotensin II ring in

Copper/Copper Oxide Nanoparticles: Biological Synthesis, Characterization and Potential Biomedical Applications: Advances and Perspectives.

The biosynthesis of copper and copper oxide nanoparticles (Cu/CuO-NPs) has attracted considerable interest due to its non-toxic, eco-friendly nature and wide-ranging applications, especially in nanomedicine and biomedical fields. Traditional nanoparticle production methods often involve toxic chemicals and generate harmful byproducts. In contrast, biological synthesis provides a cleaner, safer, more cost-effective, and sustainable alternative. Various biological sources, including plants, bacteria, fungi, and yeast, have been employed for the efficient and non-toxic production of Cu/CuO-NPs. T

Copper-Catalyzed Enantioselective Doyle-Kirmse Reaction of Azide-Ynamides via α-Imino Copper Carbenes.

[2,3]-Sigmatropic rearrangement reaction involving sulfonium ylide (Doyle-Kirmse reaction) generated from metal carbenes represents one of the powerful methods for the construction of C(sp3 )-S and C-C bonds. Although significant advances have been achieved, the asymmetric versions via the generation of sulfonium ylides from metal carbenes have been rarely reported to date, and they have so far been limited to diazo compounds as metal carbene precursors. Here, we describe a copper-catalyzed enantioselective Doyle-Kirmse reaction via azide-ynamide cyclization, leading to the practical and diver

Synthesis of Azoxy Compounds: from Copper Compounds to Mesoporous Silica-Encaged Ultrasmall Copper Catalysts.

Azoxy compounds have aroused extensive attention due to their unique biological activities, but the chemical synthesis of these compounds often suffers from limitations due to their requirement for stoichiometric oxidants, high costs, and restricted substrate range. Herein, a series of azoxy compounds were constructed via facile coupling reactions by using cost-effective N-methoxyformamide and nitroso compounds over Cu-based catalysts, affording high product yields with excellent tolerance of functional groups. Significantly, the mesoporous silica nanosphere-encapsulated ultrasmall Cu (Cu@MSN)

Copper/Carbon Hybrid Nanozyme: Tuning Catalytic Activity by the Copper State for Antibacterial Therapy.

Metal-carbon hybrid materials have shown promise as potential enzyme mimetics for antibacterial therapy; however, the effects of metal states and corresponding antibacterial mechanisms are largely unknown. Here, two kinds of copper/carbon nanozymes were designed, with tuned copper states from Cu0 to Cu2+. Results revealed that the copper/carbon nanozymes exhibited copper state-dependent peroxidase-, catalase-, and superoxide dismutase-like activities. Furthermore, the antibacterial activities were also primarily determined by the copper state. The different antibacterial mechanisms of these tw

Copper recovery and cyanide oxidation by electrowinning from a spent copper-cyanide electroplating electrolyte.

Copper-cyanide bleed streams arise from contaminated baths from industrial electroplating processes due to the buildup of impurities during continuous operation. These streams present an elevated concentration of carbonate, cyanide and copper, constituting a heavy hazard, which has to be treated for cyanide destruction and heavy metals removal, according to the local environmental laws. In the Brazilian Mint, bleed streams are treated with sodium hypochlorite, to destroy cyanide and precipitate copper hydroxide, a solid hazardous waste that has to be disposed properly in a landfill or treated

Decreased brain copper due to copper deficiency has no effect on bovine prion proteins.

Copper (Cu) is believed to be integral in prion biology and the lack of Cu or replacement by other metal ions on prions may be involved in prion diseases. This theory has not been evaluated in the bovine. Thus, mature cows were used to determine the effects of Cu deficiency on brain Cu concentrations and prion functional characteristics. Two Cu states were induced, Cu-adequate (n=4) and Cu-deficient (n=4). Copper deficiency resulted in decreased (44%) brain Cu concentrations but had no effect on prion concentrations. Based on Western blot analysis, the molecular weights, glycoform distribution

Latent copper deficiency in patients receiving low-copper enteral nutrition for a prolonged period.

BACKGROUND: Copper deficiency has been reported in patients supported with long-term enteral nutrition. Occasionally, this leads to anemia and leukopenia. There is no detailed report relating to the onset time of copper deficiency and how the symptoms develop. This report describes the relation between copper deficiency symptoms and duration of enteral nutrition. METHODS: The study included 55 patients, with 82 measurements, at the neurologic ward of Nagoya Daini Red Cross Hospital. The mean age was 71 +/- 11 years. The daily average dosage of energy was 938 kcal/d. A commercial nutrient for

A copper-responsive transcription factor, CRF1, mediates copper and cadmium resistance in Yarrowia lipolytica.

The dimorphic yeast Yarrowia lipolytica is more resistant to high copper concentrations than Saccharomyces cerevisiae. This differential tolerance to copper ions has been observed in several strains arising from non-related isolates. To investigate the molecular basis of this resistance, we obtained several copper-sensitive mutants. By complementation of one of them, we isolated the YlCRF1 gene encoding for a copper-binding transcription factor of 411 amino acids homologous to ScAce1p, CgAmt1p, and ScMac1p. Naturally occurring copper-sensitive strains lack the CRF1 allele. The YlCRF1 transcrip

Regional specificity in alterations of rat brain copper and catecholamines following perinatal copper deficiency.

Perinatal copper deficiency was studied in 1-month-old female and male Sprague-Dawley rat offspring to investigate regional changes in brain copper and catecholamine levels. Offspring of dams given the low copper treatment beginning at day 7 of gestation exhibited signs characteristic of deficiency such as impaired growth and 10-fold lower liver copper levels compared with copper-adequate controls. Regional analysis of brain copper by graphite furnace atomic absorption spectroscopy revealed uniform and severe reduction of copper to levels 20 +/- 3% of controls in all regions, except the hypoth

Copper-64 metabolism in two patients with non-Wilsonian movement disorders and copper deficiency.

Copper-64 studies are presented of 2 patients with non-Wilsonian movement disorder and with abnormal copper handling. Both patients differed from the usual phenotype of non-Wilsonian low copper movement disorder as they had choreiform movement disorders with an onset in the first decade; one patient lacked significant intellectual impairment. Both patients had reduced serum total copper and marginal free copper and caeruloplasmin levels, and both patients were capable of incorporating 64Cu2+ into caeruloplasmin but the second case did so at markedly reduced level. Both showed slightly increase

Copper uptake and retention in liver parenchymal cells isolated from nutritionally copper-deficient rats.

Copper uptake and retention were studied in primary cultures of liver parenchymal cells isolated from copper-deficient rats. Male Sprague-Dawley rats were fed a copper-deficient diet (less than 1 mg Cu/kg) for 10 wk. Copper-deficient rats were characterized by low copper concentrations in plasma and liver, anemia, low plasma ceruloplasmin oxidase activity and increased 64Cu whole-body retention. Freshly isolated liver parenchymal cells from copper-deficient rats showed a higher 64Cu influx, which was associated with a higher apparent Vmax of 45 +/- 4 pmol Cu.mg protein-1.min-1 as compared with
