Rapamycin Exerts Its Geroprotective Effects in the Ageing Human Immune System by Enhancing Resilience Against DNA Damage.

mTOR inhibitors such as rapamycin are among the most robust life-extending interventions known, yet the mechanisms underlying their geroprotective effects in humans remain incompletely understood. At non-immunosuppressive doses, these drugs are senomorphic, that is, they mitigate cellular senescence, but whether they protect genome stability itself has been unclear. Given that DNA damage is a major driver of immune ageing, and immune decline accelerates whole-organism ageing, we tested whether mTOR inhibition enhances genome stability. In human T cells exposed to acute genotoxic stress, we fou

Monitoring antioxidant defenses and free radical production in space-flight, aviation and railway engine operators, for the prevention and treatment of oxidative stress, immunological impairment, and pre-mature cell aging.

Degenerative diseases, immune impairment, and premature ageing commonly affect professional categories exposed to severe environmental and psychological stress. Among these, cosmonauts routinely experience extreme conditions due to microgravity, space radiation, altered oxygen supply, physical and mental fatigue during training, spaceflight, and post-flight. Long route aviation pilots display elevated oncogenic risk, connected with cosmic radiation overexposure, and high mortality rates for cardiovascular causes. Engine drivers, like pilots, are affected by health consequences of psycho-emotio

Predicting cancer rates in astronauts from animal carcinogenesis studies and cellular markers.

The radiation space environment includes particles such as protons and multiple species of heavy ions, with much of the exposure to these radiations occurring at extremely low average dose-rates. Limitations in databases needed to predict cancer hazards in human beings from such radiations are significant and currently do not provide confidence that such predictions are acceptably precise or accurate. In this article, we outline the need for animal carcinogenesis data based on a more sophisticated understanding of the dose-response relationship for induction of cancer and correlative cellular

Hibernation and Radioprotection: Gene Expression in the Liver and Testicle of Rats Irradiated under Synthetic Torpor.

Hibernation has been proposed as a tool for human space travel. In recent years, a procedure to induce a metabolic state known as "synthetic torpor" in non-hibernating mammals was successfully developed. Synthetic torpor may not only be an efficient method to spare resources and reduce psychological problems in long-term exploratory-class missions, but may also represent a countermeasure against cosmic rays. Here we show the preliminary results from an experiment in rats exposed to ionizing radiation in normothermic conditions or synthetic torpor. Animals were irradiated with 3 Gy X-rays and o

Mortality among US commercial pilots and navigators.

The airline industry may be an occupational setting with specific health risks. Two environmental agents to which flight crews are known to be exposed are cosmic radiation and magnetic fields generated by the aircraft's electrical system. Other factors to be considered are circadian disruption and conditions specific to air travel, such as noise, vibration, mild hypoxia, reduced atmospheric pressure, low humidity, and air quality. This study investigated mortality among US commercial pilots and navigators, using proportional mortality ratios for cancer and noncancer end points. Proportional ca

ibSLS as a biobank for democratizing access to multi-omics data and biospecimens from spaceflight research.

During spaceflight, astronauts are exposed to extreme conditions such as microgravity, cosmic radiation, and confinement, which can cause a wide range of health problems. To elucidate the molecular mechanisms underlying these issues and to develop intervention strategies for maintaining physiological homeostasis during space missions, space life science research using mouse models is actively conducted on the International Space Station (ISS). However, because of the high cost and technical complexity of spaceflight experiments, it is essential to maximize the scientific value of each mission

Celestial neuromorphics based on ferroelectric gallium oxide.

Efficient recognition of celestial activities demands hardware that can operate with high efficiency and robustness in radiation-rich space environments. Ultra-wide bandgap (UWBG) semiconductors are well-suited for such environments, but conventional UWBG transistors are not inherently compatible with advanced computing functions. To address this limitation, here, we report a κ-phase gallium oxide (κ-Ga2O3) based in-sensor reservoir computing system (κ-ISRC), which incorporates deep ultraviolet sensing, memory, and neuromorphic computation for celestial activity recognition. A ferroelectric hi

Beyond Earth's bounds: navigating the frontiers of Assisted Reproductive Technologies (ART) in space.

As interest in deep space travel grows exponentially, understanding human adaptation in becoming an interplanetary species is crucial. This includes the prospect of reproduction. This review summarizes recent updates and innovations in assisted reproductive technologies (ART) on Earth, while also discussing current challenges and areas for improvement in adapting ART studies to the space environment. We discuss the critical components of ART - gamete handling and preparation, fertilization, embryo culture, and cryopreservation - from the daily practice perspective of clinical embryologists and

Suppression of DNA-PK by RNAi has different quantitative effects on telomere dysfunction and mutagenesis in human lymphoblasts treated with gamma rays or HZE particles.

Basic to virtually all relevant biological effects of ionizing radiation is the underlying damage produced in DNA and the subsequent cellular processing of such damage. The damage can be qualitatively different for different kinds of radiations, and the genetics of the biological systems exposed can greatly affect damage processing and ultimate outcome--the biological effect of concern. The accurate repair of DNA double-strand breaks (DSBs) is critical for the maintenance of genomic integrity and function. Incorrect repair of such lesions results in chromosomal rearrangements and mutations tha

Some results of the effect of space flight factors on Drosophila melanogaster.

Two experiments with Drosophila melanogaster males were performed aboard the Salyut 6 orbital station. Mutagenic effects of a 8 day space flight on sex chromosome nondisjunction and intergene recombination in chromosome II were studied. The space flight factors (SFF) increased the frequency of chromosome nondisjunction and recombination. The model experiments showed that the combined effects of vibration and acceleration do not cover the whole spectrum of space flight mutagenic factors. These data suggest that heavy space ions are mainly responsible for the observed effect.

Health conditions in aircrew - Association with neurotoxic substances and other occupational factors.

Aircrew are exposed to in-cabin engine oil and hydraulic fumes during work. A wide spectrum of symptoms, referred to as Aerotoxic Syndrome (AS), has been described in case series and health surveys. In addition, epidemiological studies have consistently shown elevated risks for breast cancer in females and for melanoma in both male and female aircrew members. The concentrations of neurotoxic, endocrine-disrupting, and carcinogenic components of fumes have been too low to explain chronic health effects. Instead, ionizing radiation, elevated ozone levels, and disruption of circadian rhythms have

Lichens survive in space: results from the 2005 LICHENS experiment.

This experiment was aimed at establishing, for the first time, the survival capability of lichens exposed to space conditions. In particular, the damaging effect of various wavelengths of extraterrestrial solar UV radiation was studied. The lichens used were the bipolar species Rhizocarpon geographicum and Xanthoria elegans, which were collected above 2000 m in the mountains of central Spain and as endolithic communities inhabiting granites in the Antarctic Dry Valleys. Lichens were exposed to space in the BIOPAN-5 facility of the European Space Agency; BIOPAN-5 is located on the outer shell o

Reconstruction of the radiological component of the exposome in the CONSTANCES cohort.

As an innovative approach to contribute to the study of the health effects of ionizing radiation (IR) exposure, we developed a method to characterize the radiological component of the exposome as exhaustively as possible. We estimated annual equivalent IR doses in millisieverts (mSv) received by six target organs (brain, breast, lung, colon, prostate and thyroid) from the largest possible set of sources, for 21,720 adult participants in the French general population cohort CONSTANCES from birth (age 0 year) until the age they reached in 2020. We considered the following environmental sources o

GeneLab Database Analyses Suggest Long-Term Impact of Space Radiation on the Cardiovascular System by the Activation of FYN Through Reactive Oxygen Species.

Space radiation has recently been considered a risk factor for astronauts' cardiac health. As an example, for the case of how to query and identify datasets within NASA's GeneLab database and demonstrate the database utility, we used an unbiased systems biology method for identifying key genes/drivers for the contribution of space radiation on the cardiovascular system. This knowledge can contribute to designing appropriate experiments targeting these specific pathways. Microarray data from cardiomyocytes of male C57BL/6 mice followed-up for 28 days after exposure to 900 mGy of 1 GeV proton or

36Cl concentrations from polar ice cores set new constraints on the Carrington event.

The Carrington event of 1859 CE is considered one of the largest geomagnetic storms of the observational era and often used as a benchmark for a worst-case scenario. Yet, there exists no robust evidence of an associated solar energetic particle (SEP) event of a significant magnitude, based on measurements of cosmogenic radionuclides 10Be and 14C from ice cores and tree rings, respectively. In this study, we present two 36Cl records from Greenland with 2- and 4-year resolution from the EGRIP and NGRIP ice core sites, together with semi-annual 10Be data from EGRIP, as well as annual 10Be and 36C

Somatic mutation profiles in aged military nuclear test veterans: A comparative whole-genome sequence study.

Veterans of the British nuclear testing programme represent an aged group of ex-military personnel who may have been exposed to ionising radiation through their participation at nuclear testing sites. This study aimed to compare the somatic mutational landscape of a cohort of 30 nuclear test veterans with that of an age-matched cohort of 30 control veterans. Variants were identified from publicly available whole-genome sequencing data using a bioinformatics pipeline developed in accordance with the gold standard approaches as defined by the Broad Institute. The resulting set of raw SNV and IND

Ecological Evidence for Lower Risk of Lymphoma with Greater Exposure to Sunlight and Higher Altitude.

Introduction: Sunlight exposure increases vitamin D-related immune modulation and motility of T lymphocytes. Blue light exposure from the sun can stimulate immune function and help promote healthy circadian rhythm. Hence, greater sunlight exposure may lower the risk of Hodgkin lymphoma and non-Hodgkin lymphoma (NHL). Altitude may also lower the risk of these cancers through an oxygen-related mechanism, and because cosmic radiation has less shield from the atmosphere at higher levels, it allows for radiation hormesis. Methods: An ecological study design was used, with county-level lymphoma, sun

Ultrahigh-energy gamma-ray emission associated with black hole-jet systems.

Black holes (BHs), one of the most intriguing objects in the universe, can manifest themselves through electromagnetic radiation initiated by the accretion flow. Some stellar-mass BHs drive relativistic jets when accreting matter from their companion stars, forming microquasars. Non-thermal emission from the radio to teraelectronvolt gamma-ray band has been observed from microquasars, indicating the acceleration of relativistic particles. Here we report detection of four microquasars (SS 433, V4641 Sgr, GRS 1915+105, MAXI J1820+070) of spectra extending to the ultrahigh-energy (UHE; photon ene

3D biofabrication and space: A 'far-fetched dream' or a 'forthcoming reality'?

The long duration space missions across the Low Earth Orbit (LEO) often expose the voyagers to an abrupt zero gravity influence. The severe extraterrestrial cosmic radiation directly causes a plethora of moderate to chronic healthcare crises. The only feasible solution to manage critical injuries on board is surgical interventions or immediate return to Earth. This led the group of space medicine practitioners to adopt principles from tissue engineering and develop human tissue equivalents as an immediate regenerative therapy on board. The current review explicitly demonstrates the constructiv
