Freeze-induced chemical crosslinking of quaternized polysaccharides enables robust cryogels for continuous-flow removal of Congo red.

Continuous-flow adsorption utilizing macroporous cryogels is pivotal for the industrial-scale remediation of dye-containing wastewater. However, fabricating robust polysaccharide-based cryogels remains challenging due to the kinetic conflict between ice crystal growth and polymer crosslinking. Herein, we report a freeze-induced chemical crosslinking strategy for constructing robust quaternized polysaccharide-based cryogels. Using bis(vinylsulphonyl)methane (BVSM) as a crosslinker and microfibrillated cellulose (MFC) as a reinforcer, we successfully engineered a robust, macroporous quaternized

Photoreactive crosslinkers capture the ubiquitin-E2-E3 ligases intermediates.

E3 ligases catalyze the final step in transferring ubiquitin to protein substrates, but resulting intermediates are characterized by conformational flexibility. Recent work by Chandler and colleagues ( Biochem J. (2026) 483, 1115-1130) demonstrates that photocross-linking activity-based probes can trap transient conformations of the intermediates, providing a valuable tool to map interfaces during the ubiquitin transfer process.

Enzymatic double crosslinking strategy: remodeling of pea protein isolate hydrogels through transglutaminase and gelatin modification to redesign their functional properties.

Pea protein isolate (PPI) is rich in essential amino acids and exhibits low allergenicity; nevertheless, its poor gelation performance constrains its use in gel-based systems. To overcome this limitation, this study introduced gelatin (GEL) to modify pea protein isolate and constructed a double-crosslinked composite hydrogel based on a plant-animal protein combination via transglutaminase (TGase). The effects of different GEL/PPI ratios on the structure, mechanical properties, thermal stability, and swelling behavior of the hydrogels were investigated. The results showed that TGase effectively

A Chitosan-Containing Dual-Crosslinked Resilient Patch for Tissue Adhesive and Sealant.

Adhesive patches are optimum candidates for distinct biomedical applications, including tissue adhesion and leakage sealing in outpatient or surgical procedures. Unfortunately, most adhesive patches are significantly less effective at providing strong adhesion to the surface of wet tissue, which increases the risk and reduces the fault tolerance in surgery. Meanwhile, customization is difficult to achieve in terms of mechanical adaptability. To tackle this problem, we developed a chitosan-containing bioadhesive patch (ANC) comprising dual-crosslinked carboxymethyl chitosan methacrylate (CMCS-M

One-Step Enrichment and Quantitative Analysis of In Vivo Protein Complexes via Dimethylpiperidine Cross-Linker DPST.

In vivo cross-linking mass spectrometry (XL-MS) enables the proteome-wide characterization of protein complexes in living cells. However, most XL-MS methods face significant sample loss during enrichment, limiting their applications to limited-quantity samples, and suffer from poor reproducibility (20%-40%), hindering precise quantification. To overcome these challenges, we developed a novel membrane-permeable cross-linker, 2,6-dimethylpiperidine disuccinimidyl tridecanoate (DPST), in which the dimethylpiperidinyl group enables one-step enrichment of cross-linked peptides via tandem mass tags

A synergistic flexibility management strategy for self-healing carboxymethyl cellulose hydrogel with strain responsiveness based on p-phenylenediamine.

Ionic hydrogels, with adjustable physical and chemical properties, have been widely developed in the field of flexible sensing. The structural and functional integrity of ionic hydrogels under operational conditions is essential for their long-term stability and practical applicability. Herein, a rigid-flexible coupling crosslinking strategy was proposed, where p-phenylenediamine and urea served as complementary crosslinkers (rigid and flexible, respectively) within a carboxymethyl cellulose-based network. The rigid flexible coupling crosslinking network was constructed by a simple one-step me

Viscoelasticity of Fibrous Hydrogels Driven by Dynamic Intrafibrillar Imine Cross-Linking.

Viscoelasticity of biological fibrous networks impacts cell fates and may reflect pathological conditions in vivo. Imine-cross-linked fibrous hydrogels can serve as effective in vitro models for studying viscoelastic properties of biological tissues; however, the specific role of intrafibrillar dynamic covalent bonds in governing hydrogel elasticity and stress relaxation remains unexplored. Here, for fibrous hydrogels derived from cellulose nanocrystals and polyethylene glycol, we systematically varied the content of intrafibrillar imine cross-links to explore their impact on hydrogels' elasti

From micropores to mechanical strength: Fabrication and characterization of edible corn starch-sodium alginate double network hydrogels with Ca2+ cross-linking.

This study explores the fabrication and characterization of corn starch‑sodium alginate double network hydrogels using two distinct calcium ion cross-linking methods: the gluconolactone immersed method (GIM) and the calcium chloride immersed method (CCIM). We investigated the ionic cross-linking mechanism of these hydrogels and compared their microstructure and mechanical properties. Our results highlight significant differences between GIM and CCIM hydrogels, with the CCIM method producing a more uniform and compact network. At the same calcium ion concentration, CCIM hydrogel exhibited highe

Comparative study of polycarboxylic acids for sustainable crosslinking of silk fabrics: Evaluating flame retardancy and physical performances.

For protein fibers, polycarboxylic acids represent a green strategy to enhance durability without using formaldehyde. This study evaluated the physical and flame retardant properties of silk fabrics treated with three formaldehyde-free crosslinkers: citric acid (CA), 1,2,3,4-butanetetracarboxylic acid (BTCA), and 2-phosphonobutane-1,2,4-tricarboxylic acid (PBTCA). Results showed that these acids bond with silk protein through esterification and amidation, improving washing durability. Particularly, PBTCA integrates phosphorus into silk, boosting flame retardancy. While BTCA led to the highest

Cell fixation improves performance of in situ crosslinking mass spectrometry while preserving cellular ultrastructure.

Crosslinking mass spectrometry (XL-MS) has the potential to map the interactome of the cell with high resolution and depth of coverage. However, current in vivo XL-MS methods are hampered by crosslinkers that demonstrate low cell permeability and require long reaction times. Consequently, interactome sampling is not high and long incubation times can distort the cell, bringing into question the validity any protein interactions identified by the method. We address these issues with a fast formaldehyde-based fixation method applied prior to the introduction of secondary crosslinkers. Using huma

Engineering Hibiscus-Like Riboflavin/ZIF-8 Microsphere Composites to Enhance Transepithelial Corneal Cross-Linking.

Riboflavin-5-phosphate (RF) is the most commonly used photosensitizer in corneal cross-linking (CXL), but its hydrophilicity and negative charge limit its penetration through the corneal epithelium into the stroma. To enhance the corneal permeability of RF and promote its efficacy in the treatment of keratoconus, novel hibiscus-like RF@ZIF-8 microsphere composites [6RF@ZIF-8 NF (nanoflake)] are prepared using ZIF-8 nanomaterials as carriers, which are characterized by their hydrophobicity, positive potential, biocompatibility, high loading capacities, and large surface areas. Both hematoxylin

Complications of corneal collagen cross-linking.

Corneal collagen cross-linking (CXL) is an effective treatment for arresting progression in patients with keratoconus. CXL was approved by United States Food and Drug Administration for the treatment of progressive keratoconus in 2016. It is a relatively safe procedure with a low complication rate. As this approach becomes more popular, it is paramount to be familiar with the potential complications associated with the procedure and its management. This article aims to report and review the complications of CXL for the treatment of keratoconus and post-LASIK ectasia.

Accurate and Automated High-Coverage Identification of Chemically Cross-Linked Peptides with MaxLynx.

Cross-linking combined with mass spectrometry (XL-MS) provides a wealth of information about the three-dimensional (3D) structure of proteins and their interactions. We introduce MaxLynx, a novel computational proteomics workflow for XL-MS integrated into the MaxQuant environment. It is applicable to noncleavable and MS-cleavable cross-linkers. For both, we have generalized the Andromeda peptide database search engine to efficiently identify cross-linked peptides. For noncleavable peptides, we implemented a novel dipeptide Andromeda score, which is the basis for a computationally efficient N-s

Infectious keratitis after corneal crosslinking: systematic review.

Corneal crosslinking is a U.S. Food and Drug Administration--approved therapy to stiffen the cornea and prevent progression of corneal ectasia in patients with keratoconus. The standard procedure involves removal of the corneal epithelium (epithelial-off) prior to treatment. Variations to the standard procedure include accelerated crosslinking and transepithelial procedures. This study reviewed what is known regarding the risk for infection after epithelial-off crosslinking, the spectrum of pathogens, and clinical outcomes. 26 publications were identified. All eyes were fit with a bandage cont

Nonlinear optical crosslinking (NLO CXL) for correcting refractive errors.

Ultraviolet A (UVA) light-based photoactivation of riboflavin (Rf) to induce corneal crosslinking (CXL) and mechanical stiffening is now a well-known treatment for corneal ectasia and Keratoconus that is being used in a topographically guided photorefractive intrastromal CXL (PiXL) procedure to treat low degrees of refractive errors. Alternative approaches for non-invasive treatment of refractive errors have also been proposed that use femtosecond lasers (FS) that provide much faster, more precise, and safer results than UVA CXL. One such treatment, nonlinear optical crosslinking (NLO CXL), ha

Tunable bioactivity and mechanics of collagen-based tissue engineering constructs: A comparison of EDC-NHS, genipin and TG2 crosslinkers.

Due to its ubiquity and versatility in the human body, collagen is an ideal base material for tissue-engineering constructs. Chemical crosslinking treatments allow precise control of the biochemical and mechanical properties through macromolecular modifications to the structure of collagen. In this work, three key facets regarding the collagen crosslinking process are explored. Firstly, a comparison is drawn between the carbodiimide-succinimide (EDC-NHS) system and two emerging crosslinkers utilising alternate chemistries: genipin and tissue transglutaminase (TG2). By characterising the chemic

Chromatin Immunoprecipitation.

Chromatin immunoprecipitation, commonly referred to as ChIP, is a powerful technique for the evaluation of in vivo interactions of proteins with specific regions of genomic DNA. Formaldehyde is used in this technique to cross-link proteins to DNA in vivo, followed by the extraction of chromatin from cross-linked cells and tissues. Harvested chromatin is sheared and subsequently used in an immunoprecipitation incorporating antibodies specific to protein(s) of interest and thus coprecipitating and enriching the cross-linked, protein-associated DNA. The cross-linking process can be reversed, and
