Mammalian D-cysteine: A novel regulator of neural progenitor cell proliferation Mammalian D-cysteine: A novel regulator of neural progenitor cell proliferation Endogenous D-cysteine, the stereoisomer with rapid spontaneous in vitro racemization rate, has major neural roles.

D-amino acids are being recognized as functionally important molecules in mammals. We recently identified endogenous D-cysteine in mammalian brain. D-cysteine is present in neonatal brain in substantial amounts (mM) and decreases with postnatal development. D-cysteine binds to MARCKS and a host of proteins implicated in cell division and neurodevelopmental disorders. D-cysteine decreases phosphorylation of MARCKS in neural progenitor cells (NPCs) affecting its translocation. D-cysteine controls NPC proliferation by inhibiting AKT signaling. Exogenous D-cysteine inhibits AKT phosphorylation at

A far-red FRET fluorescent probe for ratiometric detection of l-cysteine based on carbon dots and N-acetyl-l-cysteine-capped gold nanoparticles.

A novel far-red fluorescence resonance energy transfer (FRET) fluorescent probe for ratiometric detection of l-cysteine (l-Cys) has been designed. The system was established a FRET assembly by positively charged carbon dots (CDs) and negatively charged N-acetyl-l-cysteine capped gold nanoparticles (NAC-AuNPs). The fluorescence of CDs at 539 nm could be effectively quenched in the presence of NAC-AuNPs owing to FRET process, while the emission of NAC-AuNPs at 630 nm was appeared. Subsequently, the interactions between l-Cys and NAC-AuNPs resulted in the decreased emission intensity of NAC-AuNPs

Cysteine protease activity and histamine release from the human mast cell line HMC-1 stimulated by recombinant streptococcal pyrogenic exotoxin B/streptococcal cysteine protease.

We constructed the expression vector pSK-SCP containing the streptococcal exotoxin B gene (spe b) which expressed protease activity. We showed that the recombinant streptococcal pyogenic exotoxin B/streptococcal cysteine protease (rSPE B/SCP) was secreted into the culture supernatant of the transformant and retained its SCP activity, which was equivalent to or greater than that of the naturally occurring molecule. The secreted rSPE B/SCP induced histamine release and degranulation of the human mast cell line HMC-1. This study may contribute to the understanding of the pathogenic role of SPE B/

Michael addition-elimination-cyclization based turn-on fluorescence (MADELCY TOF) probes for cellular cysteine imaging and estimation of blood serum cysteine and aminoacylase-1.

Oxidative stress is a hallmark of a range of diseases including cancer, HIV-AIDS and cardiovascular disorders. Blood plasma cysteine (Cys) is an established biomarker for oxidative stress, rendering the development of sensitive and technically straightforward approaches for its estimation extremely desirable. Herein, we achieve this goal by developing a series of Michael addition-elimination-cyclization based turn-on fluorescence (MADELCY TOF) probes that selectively detect Cys over other amino acids and biothiols such as homocysteine (Hcy) and glutathione (GSH). Cysteine detection by these co

Preparation, crystallization and X-ray diffraction analysis to 1.5 A resolution of rat cysteine dioxygenase, a mononuclear iron enzyme responsible for cysteine thiol oxidation.

Cysteine dioxygenase (CDO; EC 1.13.11.20) is an approximately 23 kDa non-heme iron metalloenzyme that is responsible for the oxidation of cysteine by O2, yielding cysteinesulfinate. CDO catalyzes the first step in the conversion of cysteine to taurine, as well as the first step in the catabolism of cysteine to pyruvate plus sulfate. Recombinant rat CDO was heterologously expressed, purified and crystallized. The protein was expressed as a fusion protein bearing a polyhistidine tag to facilitate purification, a thioredoxin tag to improve solubility and a factor Xa cleavage site to permit remova

Highlights in Cysteine Protease Inhibitors as Antiparasitic Agents Against Trypanosomatids.

Trypanosoma cruzi, Trypanosoma brucei and Leishmania spp. are the causative agents of Chagas disease, human African trypanosomiasis, and leishmaniasis, respectively. These three neglected tropical diseases affect millions of individuals globally, and current therapeutic options remain severely limited. The major cysteine proteases (CPs) of these parasites play critical roles in their infectivity and survival, making them subjects of prolonged research. Therefore, cruzain, TbrCATL and CPB from Leishmania spp. have been recognized as the primary targets of most drugs under investigation. Among t

Non-specific cysteine modifications on proteins induced by trans-cyclooctene.

Tetrazine-trans-cyclooctene (Tz-TCO) click chemistry has been widely adopted for various applications due to its faster reaction kinetics and higher biocompatibility compared to conventional copper-catalyzed click chemistry. However, most click chemistry groups do not appear to be fully bioorthogonal, as several of them-including tetrazine-have been found to directly modify proteins. Here, we report the unexpected discovery of non-specific protein modifications by TCO during the development of a tetrazine-based proximity labeling method. TCO probes readily conjugate to cysteine residues on pro

Unique Cysteine-Directed Covalent Inhibition of PRMT1 Suppresses Breast Tumorigenesis.

Dysregulation of PRMT1, a key epigenetic enzyme, is strongly implicated in breast cancer pathogenesis. However, developing selective PRMT1 inhibitors has been challenging due to the high conservation of the catalytic domain across PRMT family members. We herein report 17zh (MS3-123), the first small-molecule, nanomolar covalent inhibitor targeting the unique Cys119 within the SAM-binding pocket of PRMT1 (IC50 = 11.4 nM) with over 17-96-fold selectivity against other type I PRMTs subfamily and >439-fold selectivity against type II PRMT5. 17zh selectively engages Cys119, inhibiting PRMT1 activit

α,β-Unsaturated carbonyl-based anticancer agents targeting cysteine residues.

The α,β-unsaturated carbonyl motif serves as an essential pharmacophore mediating the covalent modification of cysteine residues through Michael addition, thereby driving the biological activity of numerous natural products and targeted therapeutic agents. This review delineates the chemical rationale of the α,β-unsaturated carbonyl moiety as a soft electrophile and highlights its key applications in anticancer therapy in two areas: (1) as a warhead in covalent kinase inhibitors (e.g., targeting KRAS G12C, EGFR, and BTK) and (2) as electrophilic modulators of the Keap1-Nrf2 pathway via cystein

Cysteine-reactive mitigators of small vessel disease-related NOTCH3 mutants.

Pathogenic alterations in NOTCH3 cause CADASIL, an accelerated and currently untreatable form of cerebrovascular disease. CADASIL mutant NOTCH3, which frequently harbors abnormalities in EGF repeat cysteine number, adopt disulfide dependent abnormal conformations. To seek potential strategies to mitigate the impact of CADASIL mutations on NOTCH3, we investigated whether cysteine-targeting compounds may affect pathological NOTCH3. LSL-NOTCH3, a split luciferase assay that discriminates between benign and pathogenic NOTCH3 conformations, was used to quantify the capacity of 21 small molecule com

Cysteine import contributes to hypochlorous acid resistance in Staphylococcus aureus.

Hypochlorous acid (HOCl) is a potent antimicrobial oxidant produced by innate immune cells to eliminate invading bacterial pathogens, including Staphylococcus aureus. HOCl readily oxidizes thiols, amines, and macromolecules in S. aureus. Yet, some bacterial cells within an HOCl-challenged population withstand this oxidative stress and proceed to establish infection. The mechanisms that enable S. aureus to resist the effects of HOCl remain incompletely understood. Here, we show that cysteine uptake is a major determinant of HOCl resistance in S. aureus. Inactivation of the cysteine transporters

Crystallization of L-Cysteine in Heavy Water Induces Intrinsic Fluorescence.

Developing noninvasive techniques that can probe how solvents modulate the nucleation pathways of bioorganic molecules in solution remains an active and open area of research. Herein, we investigate the crystallization of the amino acid L-Cysteine and show that both the structure of the crystal and its intrinsic fluorescence can be drastically altered by the solvent. Crystals formed in heavy water exhibit markedly different intermolecular packing as well as strikingly different monomer conformations compared to those in light water. Remarkably, these differences in the supramolecular packing r

Tunable Heteroaromatic Nitriles for Selective Bioorthogonal Click Reaction with Cysteine.

The binucleophilic properties of 1,2-aminothiol and its rare occurrence in nature make it a useful reporter for tracking molecules in living systems. The 1,2-aminothiol moiety is present in cysteine, which is a substrate for a biocompatible click reaction with heteroaromatic nitriles. Despite the wide range of applications for this reaction, the scope of nitrile substrates has been explored only to a limited extent. In this study, we expand the chemical space of heteroaromatic nitriles for bioconjugation under physiologically relevant conditions. We systematically assembled a library of 116 2-

A Bis(imidazole)-based cysteine labeling tool for metalloprotein assembly.

Precise metal-protein coordination by design remains a considerable challenge. Polydentate, high-metal-affinity protein modifications, both chemical and recombinant, can enable metal localization. However, these constructs are often bulky, conformationally and stereochemically ill-defined, or coordinately saturated. Here, we expand the biomolecular metal-coordination toolbox with the irreversible attachment to cysteine of bis(1-methylimidazol-2-yl)ethene ("BMIE"), which generates a compact imidazole-based metal-coordinating ligand. Conjugate additions of small-molecule thiols (thiocresol and N

S-Propargyl-Cysteine Ameliorates Peripheral Nerve Injury through Microvascular Reconstruction.

Microvascular reconstruction is essential for peripheral nerve repair. S-Propargyl-cysteine (SPRC), the endogenous hydrogen sulfide (H2S) donor, has been reported to promote angiogenesis. The aim of this study is to utilize the pro-angiogenic ability of SPRC to support peripheral nerve repair and to explore the potential mechanisms. The effects and mechanisms of SPRC on angiogenesis and peripheral nerve repair were examined under hypoxic condition by establishing a sciatic nerve crushed injury model in mice and rats, and a hypoxia model in human umbilical vascular endothelial cells (HUVECs) in

PbS biomineralization using cysteine: Bacillus cereus and the sulfur rush.

Bacillus sp. Abq, belonging to Bacillus cereus sensu lato, was isolated from an aquifer in New Mexico, USA and phylogenetically classified. The isolate possesses the unusual property of precipitating Pb(II) by using cysteine, which is degraded intracellularly to hydrogen sulfide (H2S). H2S is then exported to the extracellular environment to react with Pb(II), yielding PbS (galena). Biochemical and growth tests showed that other sulfur sources tested (sulfate, thiosulfate, and methionine) were not reduced to hydrogen sulfide. Using equimolar concentration of cysteine, 1 mM of soluble Pb(II) wa

Targeting a cysteine protease from a pathobiont alleviates experimental arthritis.

BACKGROUND: Several lines of evidence suggest that the pathobiont Porphyromonas gingivalis is involved in the development and/or progression of auto-inflammatory diseases. This bacterium produces cysteine proteases, such as gingipain RgpA, endowed with the potential to induce significant bone loss in model systems and in patients. OBJECTIVE: We sought to gain further insight into the role of this pathobiont in rheumatoid arthritis (RA) and to identify novel therapeutic targets for auto-inflammatory diseases. METHODS: We profiled the antibody response to RgPA-specific domains in patient sera.

Neuroprotective mechanisms of S-allyl-L-cysteine in neurological disease.

S-allyl-L-cysteine (SAC) is a sulfur-containing amino acid present in garlic and exhibits a wide range of biological activities such as antioxidant, anti-inflammatory, and anticancer agent. An earlier study demonstrated that SAC ameliorates oxidative damage in a model of experimental stroke. However, the antioxidant property of SAC does not suffice to explain its beneficial effects in terms of the underlying mechanisms. Endoplasmic reticulum (ER) stress and ER stress-induced cell death have been shown to be involved in various neurological diseases such as brain ischemia, Alzheimer's disease,
