Dynamics, function, and regulation of mammary stem cells in development and disease.

Understanding the heterogeneity of mammary epithelial cells and their contributions to postnatal development and regeneration is of great importance. Recent advances have defined the intrinsic complexity of the mammary epithelium, its stromal interactions, and the ontogeny of adult stem and progenitor cells. These insights have not only refined our understanding of normal mammary gland biology but also highlighted how specific epithelial subsets serve as cells of origin and targets of transformation in breast cancer. The close parallels between mammary stem cells and tumor-initiating cells, pa

Report on the 2nd World Congress on Fetal Origins of Adult Disease, Brighton, U.K., June 7-10, 2003.

In 1989, reports suggested that the fetal environment, as reflected in birth size, was related to the risk of noncommunicable diseases in adult life. This association was first described for coronary heart disease but rapidly extended to include type 2 diabetes, osteoporosis, and metabolic and endocrine homeostasis. This led to the development of the fetal origins of adult disease paradigm, which resulted in a refocusing of research effort over the next 10 y to consider the lifelong consequences of perinatal influences on chronic diseases. Previously, perinatal influences had largely been seen

Telomere shortening is associated with malformation in p53-deficient mice after irradiation during specific stages of development.

The natural ends of linear chromosomes, the telomeres, recruit specific proteins in the formation of protective caps that preserve the integrity of the genome. Unprotected chromosomes induce DNA damage checkpoint cascades and ultimately lead to senescence both in mouse and man in a p53 dependent manner and initial telomere length setting therefore determines the proliferative capacity of each cell. Yet, only little information is available on telomere biology during embryonic development. We have previously shown that the p53 gene plays a crucial role in the development of malformations (exenc

Male orangutan subadulthood: a new twist on the relationship between chronic stress and developmental arrest.

Both in the wild and in captivity, a marked and enduring arrest of secondary sexual developmental occurs in some male orangutans (Pongo pygmaeus) (Kingsley [1982] The Orang-Utan: Its Biology and Conservation, The Hague: Junk; Utami [2000]). Researchers have hypothesized that chronic stress, perhaps related to aggression from mature males, causes endocrine changes altering growth and maturation rates in these males (Maple [1980] Orangutan Behavior, New York: Van Nostrand Reinhold; Graham [1988] Orangutan Biology, Oxford: Oxford University Press). In this study, urine samples were collected over

Tooth number abnormality: from bench to bedside.

Tooth number abnormality is one of the most common dental developmental diseases, which includes both tooth agenesis and supernumerary teeth. Tooth development is regulated by numerous developmental signals, such as the well-known Wnt, BMP, FGF, Shh and Eda pathways, which mediate the ongoing complex interactions between epithelium and mesenchyme. Abnormal expression of these crutial signalling during this process may eventually lead to the development of anomalies in tooth number; however, the underlying mechanisms remain elusive. In this review, we summarized the major process of tooth devel

Human stem cell-based embryo models: from self-organization to recapitulating development.

Human stem cell-based embryo models (hereafter referred to as embryo models) are a rapidly evolving field at the intersection of developmental biology and bioengineering. In this review, we examine design principles, material systems, and engineering approaches that enable the construction of models arising from pluripotent stem cells. We consider how three-dimensional environments, synthetic hydrogels, and spatiotemporal modulation of morphogenetic signals guide self-organization. Together, these factors enable the recapitulation of stages from blastocyst formation to early organogenesis. We

Fetal mental programming: maternal psychological homeostasis as a placenta-mediated determinant of cognitive and emotional development - an integrative perspective.

BACKGROUND: Fetal programming has traditionally been interpreted through metabolic and endocrine paradigms, while maternal psychological regulation remains underdeveloped as a biological determinant of neurodevelopment. Emerging evidence from placental biology, epigenetics, and fetal neuroimaging suggests that maternal psychological states may act not only as contextual exposures but also as dynamic physiological signals during pregnancy. CONTENT: This article proposes an integrative perspective in which maternal psychological homeostasis is conceptualized as a multidimensional regulatory sta

The developmental lipidome of Nippostrongylus brasiliensis.

BACKGROUND: Nippostrongylus brasiliensis-a nematode of rodents-is commonly used as a model to study the immunobiology of parasitic nematodes. It is a member of the Strongylida-a large order of socioeconomically important parasitic nematodes of animals. Lipids are known to play essential roles in nematode biology, influencing cellular membranes, energy storage and/or signalling. METHODS: The present investigation provides a comprehensive, untargeted lipidomic analysis of four developmental stages/sexes (i.e. egg, L3, adult female and adult male stages) of N. brasiliensis utilising liquid chrom

Kidney organoids in translational research: disease modeling, drug discovery, and unresolved challenges.

Kidney organoids derived from human pluripotent stem cells (hPSCs) have emerged as powerful platforms for translational nephrology, enabling complex renal pathophysiology modeling in physiologically relevant three-dimensional contexts. This review synthesizes recent advances in kidney organoid applications for disease modeling and drug discovery, highlighting their translational potential beyond developmental biology. These organoids recapitulate key human kidney architectural features, including nephron-like structures with glomeruli and tubules, while exhibiting greater cellular heterogeneit

Clinical genetics and pathobiology of ciliary chondrodysplasias.

Ciliary chondrodysplasias represent a heterogenous group of rare, nearly exclusively autosomal recessively inherited developmental conditions. While the skeletal phenotype, mainly affecting limbs, ribs and sometimes the craniofacial skeleton, is predominant, extraskeletal disease affecting the kidneys, liver, heart, eyes and other organs and tissues is observed inconsistently. Significant lethality, resulting from cardiorespiratory failure due to thoracic constriction as well as from renal and hepatic insufficiency or primary cardiac failure due to congenital heart disease, is observed with th

Global gene expression analysis of the zoonotic parasite Trichinella spiralis revealed novel genes in host parasite interaction.

BACKGROUND: Trichinellosis is a typical food-borne zoonotic disease which is epidemic worldwide and the nematode Trichinella spiralis is the main pathogen. The life cycle of T. spiralis contains three developmental stages, i.e. adult worms, new borne larva (new borne L1 larva) and muscular larva (infective L1 larva). Stage-specific gene expression in the parasites has been investigated with various immunological and cDNA cloning approaches, whereas the genome-wide transcriptome and expression features of the parasite have been largely unknown. The availability of the genome sequence informatio

The people behind the papers - Laura Lahti and Thomas Perlmann.

Midbrain dopamine (mDA) neuron degeneration is a major cause of Parkinson's disease, so understanding the mechanisms underpinning mDA neuron production could help inform future treatments. A new paper in Development identifies key transcription factors that regulate the timing of cell cycle exit in mDA progenitors. The study also identifies a population of ependymal cells that exhibit progenitor potential. To learn more about the story behind the paper, we caught up with first author Laura Lahti and corresponding author Thomas Perlmann, Professor in Molecular Developmental Biology at the Karol

Three dimensional HiLo-based structured illumination for a digital scanned laser sheet microscopy (DSLM) in thick tissue imaging.

Laser sheet based microscopy has become widely accepted as an effective active illumination method for real time three-dimensional (3D) imaging of biological tissue samples. The light sheet geometry, where the camera is oriented perpendicular to the sheet itself, provides an effective method of eliminating some of the scattered light and minimizing the sample exposure to radiation. However, residual background noise still remains, limiting the contrast and visibility of potentially interesting features in the samples. In this article, we investigate additional structuring of the illumination f

Integrin β1 signaling at the crossroads of cellular plasticity, the tumor microenvironment, and therapy resistance in medulloblastoma.

Medulloblastoma (MB), the most common malignant pediatric brain tumor, remains associated with relapse and long-term treatment-related toxicities. Tumor-microenvironment interactions increasingly appear to contribute to progression, stemness, and therapy resistance in brain cancers. Integrin β1, a major mediator of cell-extracellular matrix interactions, regulates survival signaling, mechanotransduction, and cancer stem cell maintenance in several tumor types, particularly glioblastoma. This mini-review discusses how developmental programs, extracellular matrix remodeling, hypoxia, and cancer

Molecular Genetic Analysis of Chlamydia Species.

Species of Chlamydia are the etiologic agent of endemic blinding trachoma, the leading cause of bacterial sexually transmitted diseases, significant respiratory pathogens, and a zoonotic threat. Their dependence on an intracellular growth niche and their peculiar developmental cycle are major challenges to elucidating their biology and virulence traits. The last decade has seen tremendous advances in our ability to perform a molecular genetic analysis of Chlamydia species. Major achievements include the generation of large collections of mutant strains, now available for forward- and reverse-g

MicroRNAs and atherosclerosis: new actors for an old movie.

To date, cardiovascular diseases (CVDs) are the leading causes of morbidity and mortality worldwide. MicroRNAs (miRNAs) are endogenous, short, non-coding RNA sequences able to regulate gene expression principally at the post-transcriptional level. Initially, they were thought to be involved only in developmental timing of worms. Their involvement in human biology was recently discovered and many studies have been performed to demonstrate the role of miRNA in human cancer. Since the first observation in 2005 of their implication in cardiac biology, many studies have demonstrated their role in t

Teaching Zebrafish Development in a STEM-Based Science Camp for Middle School Students.

Zebrafish have been used as an education tool for students of all ages and can be used in many learning environments to teach different fields of science. In this study, we focus on the biology of zebrafish. We describe an educational program within a weeklong science camp for students between 12 and 14 years old. The methodology described is based on running annual science camps over an 11-year period. In these camps, students learnt about the developmental stages of zebrafish, as well as general zebrafish biology, husbandry, ecology, behavior, and reproduction. This article describes how to

[Repair and formation of cartilage and bone: are they possible?].

Osteoporosis and osteoarthritis cause serious problems to the mobility, and therefore to the quality of life, of aged people. This is due to the fact that both diseases result from irreversible loss of quality or function of the tissues; i.e., bone and articular cartilage. Recent progress in molecular biology and developmental biology, however, have contributed a great deal to our understanding of the mechanisms regulating growth and differentiation of osteoblasts and chondrocytes, the cells responsible for producing bone and cartilage, respectively. In vivo studies using animals including hum

The people behind the papers - Denis Faerberg and Ilya Ruvinsky.

Male pheromones accelerate the development of hermaphrodite larvae in Caenorhabditis elegans, but the importance of this phenomenon is not well understood. A new paper in Development shows that pheromone exposure during larval stage 3 helps coordinate behaviour and development by modulating the timing of the transition to larval stage 4. To learn more about the story behind the paper, we caught up with first author Denis Faerberg who carried out the work in the lab of the corresponding author Ilya Ruvinsky at Northwestern University, USA.
