Discrimination between healthy participants and people with panic disorder based on polygenic scores for psychiatric disorders and for intermediate phenotypes using machine learning.

OBJECTIVE: Panic disorder is a modestly heritable condition. Currently, diagnosis is based only on clinical symptoms; identifying objective biomarkers and a more reliable diagnostic procedure is desirable. We investigated whether people with panic disorder can be reliably diagnosed utilizing combinations of multiple polygenic scores for psychiatric disorders and their intermediate phenotypes, compared with single polygenic score approaches, by applying specific machine learning techniques. METHODS: Polygenic scores for 48 psychiatric disorders and intermediate phenotypes based on large-scale

Discrimination of Benign and Malignant Thyroid Nodules through Comparative Analyses of Human Saliva Samples via Metabolomics and Deep-Learning-Guided Label-free SERS.

Thyroid nodules are a very common entity. The overall prevalence in the populace is estimated to be around 65-68%, among which a small portion (less than 5%) is malignant (cancerous). Therefore, it is important to discriminate benign thyroid nodules from malignant thyroid nodules. In this study, an equal number of participants with benign and malignant thyroid nodules (N = 10/group) were recruited. Saliva samples were collected from each participant, and SERS spectra were acquired, followed by validation using a metabolomics approach. An additional equal number of patients (N = 40/group) were

A preliminary study on early postmortem submersion interval (PMSI) estimation and cause-of-death discrimination based on nontargeted metabolomics and machine learning algorithms.

Postmortem submersion interval (PMSI) estimation and cause-of-death discrimination of corpses in water have long been challenges in forensic practice. Recently, many studies have linked postmortem metabolic changes with PMI extension, providing a potential strategy for estimating PMSI using the metabolome. Additionally, there is a lack of potential indicators with high sensitivity and specificity for drowning identification. In the present study, we profiled the untargeted metabolome of blood samples from drowning and postmortem submersion rats at different PMSIs within 24 h by liquid chromato

Using machine learning to discriminate non-classical 21-hydroxylase deficiency from polycystic ovary syndrome: an external validation study.

INTRODUCTION: Nonclassical 21-hydroxylase deficiency (NC21OHD) is a rare autosomal recessive disorder that is frequently misdiagnosed as polycystic ovary syndrome (PCOS) because of their overlapping clinical and biochemical presentations. The conventional diagnostic method, the cosyntropin stimulation test, is invasive, time consuming, and difficult to implement in routine clinical practice. To overcome these limitations, we previously developed a proof-of-concept machine learning model integrating basal serum steroid profiling by liquid chromatography with tandem mass spectrometry (LC-MS/MS)

Combining Clinical, Genetic and Protein Markers Using Machine Learning Models Discriminates Psoriatic Arthritis Patients From Those With Psoriasis.

BACKGROUND: Psoriatic Arthritis (PsA), an immune mediated inflammatory arthritis, affects a quarter of patients with cutaneous psoriasis, usually after psoriasis onset. Early diagnosis of PsA is challenging. A biomarker-based diagnostic test may facilitate early diagnosis. OBJECTIVES: We aimed to determine whether specific clinical features or genetic and protein markers, alone or in combination, can distinguish patients with PsA from those with psoriasis without PsA (PsC). METHODS: Patients with PsA and PsC were identified from a database of patients with psoriatic disease. Detailed demogra

Machine learning approach effectively discriminates between Parkinson's disease and progressive supranuclear palsy: Multi-level indices of rs-fMRI.

AIM: Parkinson's disease (PD) and progressive supranuclear palsy (PSP) present similar clinical symptoms, but their treatment options and clinical prognosis differ significantly. Therefore, we aimed to discriminate between PD and PSP based on multi-level indices of resting-state functional magnetic resonance imaging (rs-fMRI) via the machine learning approach. MATERIALS AND METHODS: A total of 58 PD and 52 PSP patients were prospectively enrolled in this study. Participants were randomly allocated to a training set and a validation set in a 7:3 ratio. Various rs-fMRI indices were extracted, f

Adversarial Erasing Enhanced Multiple Instance Learning (siMILe): Discriminative Identification of Oligomeric Protein Structures in Single Molecule Localization Microscopy.

Single-molecule localization microscopy (SMLM) achieves nanoscale imaging of complex protein structures in the cell. However, the ability to capture structural variability across cell conditions (cell lines, gene expression, treatment) from 3D point cloud SMLM data remains limited. We present siMILe, a weakly supervised multiple instance learning machine learning method to close this gap in interpretable subcellular discovery. siMILe identifies condition-specific changes in protein assemblies by leveraging their shape and network features, without requiring structure-level supervision. siMILe

Identification of discriminative neuroimaging markers for patients on hemodialysis with insomnia: a fractional amplitude of low frequency fluctuation-based machine learning analysis.

BACKGROUND AND OBJECTIVE: Insomnia is one of the common problems encountered in the hemodialysis (HD) population, but the mechanisms remain unclear. we aimed to (1) detect the spontaneous brain activity pattern in HD patients with insomnia (HDWI) by using fractional fractional amplitude of low frequency fluctuation (fALFF) method and (2) further identify brain regions showing altered fALFF as neural markers to discriminate HDWI patients from those on hemodialysis but without insomnia (HDWoI) and healthy controls (HCs). METHOD: We compared fALFF differences among HDWI subjects (28), HDWoI subj

Classification of Alzheimer's Disease, Mild Cognitive Impairment, and Cognitively Unimpaired Individuals Using Multi-feature Kernel Discriminant Dictionary Learning.

Accurate classification of either patients with Alzheimer's disease (AD) or patients with mild cognitive impairment (MCI), the prodromal stage of AD, from cognitively unimpaired (CU) individuals is important for clinical diagnosis and adequate intervention. The current study focused on distinguishing AD or MCI from CU based on the multi-feature kernel supervised within-Class-similar discriminative dictionary learning algorithm (MKSCDDL), which we introduced in a previous study, demonstrating that MKSCDDL had superior performance in face recognition. Structural magnetic resonance imaging (sMRI)

Impaired discriminative fear-conditioning resulting from elevated fear responding to learned safety cues among individuals with panic disorder.

Classical fear-conditioning is central to many etiologic accounts of panic disorder (PD), but few lab-based conditioning studies in PD have been conducted. One conditioning perspective proposes associative-learning deficits characterized by deficient safety learning among PD patients. The current study of PD assesses acquisition and retention of discriminative aversive conditioning using a fear-potentiated startle paradigm. This paradigm was chosen for its specific capacity to independently assess safety- and danger learning in the service of characterizing putative anomalies in each type of l

Rhinal cortex removal produces amnesia for preoperatively learned discrimination problems but fails to disrupt postoperative acquisition and retention in rhesus monkeys.

To test whether the rhinal cortex (i.e., entorhinal and perirhinal cortex) plays a time-limited role in information storage, eight rhesus monkeys were trained to criterion on two sets of 60 object discrimination problems, one set at each of two different time periods separated by 15 weeks. After the monkeys had learned both sets, two groups balanced for preoperative acquisition rates were formed. One group received bilateral ablation of the rhinal cortex (n = 4), and the other was retained as an unoperated control group (n = 4). After a 2 week rest period, monkeys were assessed for retention o

Discrimination of the Lame Limb in Horses Using a Machine Learning Method (Support Vector Machine) Based on Asymmetry Indices Measured by the EQUISYM System.

Lameness detection in horses is a critical challenge in equine veterinary practice, particularly when symptoms are mild. This study aimed to develop a predictive system using a support vector machine (SVM) to identify the affected limb in horses trotting in a straight line. The system analyzed data from inertial measurement units (IMUs) placed on the horse's head, withers, and pelvis, using variables such as vertical displacement and retraction angles. A total of 287 horses were included, with 256 showing single-limb lameness and 31 classified as sound. The model achieved an overall accuracy o

Machine Learning-Driven Sensor Array Based on a DNA-Programmed Pt Nanozyme with Enhanced Dual-Enzyme-like Activities for Discrimination of Multiple Sulfur-Containing Species.

Nanozyme sensor arrays have garnered significant attention in the sensing field, whereas their practical application remains limited due to the issues of multiple independent sensing elements, concentration interference effects, and challenges in real-world implementation. Herein, we propose a machine-learning-empowered sensor array based on a single nanozyme with dual-enzyme-like activities for the simultaneous discrimination of multiple sulfur-containing species (S2-, S2O32-, S2O82-, SO32-, and SO42-). Hence, a DNA-modulating strategy to fabricate Pt nanozymes with improved peroxidase-like a

Comparison of pre-pulse inhibition, tactile discrimination learning and barrel cortical neural response in adult male rats following chronic exposure to morphine, methadone and buprenorphine.

Chronic exposure to opioids is the most common treatment plan to reduce the pain. In this study, the stereotyped behaviors and cognitive functions related to different types of tactile and auditory inputs were investigated in the rats following chronic exposure to the morphine, methadone, and buprenorphine. Here, three addicted groups received morphine, methadone, and buprenorphine while the control rats received saline for 21 days. Our results demonstrated that the opioid-treated groups showed stereotyped behaviors including grooming and rearing. In the behavioral level, prepulse inhibition a

Routine Laboratory-Based Machine Learning for Discriminating Multiple Myeloma from Clinical Mimickers: Development and Internal Validation of a Diagnostic Prediction Model.

Background: Multiple myeloma (MM) diagnosis requires specialised testing that is not universally available, contributing to diagnostic delays. The aim of this study was to develop and internally validate statistical and machine learning (ML) models that discriminate MM from clinically similar conditions using exclusively routine laboratory parameters. Methods: This retrospective diagnostic prediction model study included 422 consecutive patients referred to hematology for suspected MM at a tertiary care center (2017-2025). Fifteen routine demographic and laboratory variables were used to train

Visible microspectrophotometry coupled with machine learning to discriminate the erythrocytic life cycle stages of P. falciparum malaria parasites in functional single cells.

Malaria was regarded as the most devastating infectious disease of the 21st century until the COVID-19 pandemic. Asexual blood staged parasites (ABS) play a unique role in ensuring the parasite's survival and pathogenesis. Hitherto, there have been no spectroscopic reports discriminating the life cycle stages of the ABS parasite under physiological conditions. The identification and quantification of the stages in the erythrocytic life cycle is important in monitoring the progression and recovery from the disease. In this study, we explored visible microspectrophotometry coupled to machine lea

Synergistic pore size screening and dual time window kinetics: machine learning-based discrimination and concentration prediction of different types of preservatives in complex food matrices.

Distinguishing between different preservatives is of great significance in quality control. Therefore, developing effective methods to distinguish between them is of great importance. This study overcomes the limitation of traditional sensor arrays that identify only a single analyte. Through the synergistic integration of pore size screening, metal doping, and machine learning, this system achieves multi-category discrimination of different preservatives, with a minimum discrimination concentration of 150 nM. With the optimal pore size effect of the ZIF-8 nanozyme and a dual-window reaction a
