Isocyanide terpene metabolites of Phyllidiella pustulosa, a nudibranch from the South China Sea.

A series of isocyanides, compounds 4-11, including diterpenes never before found in Phyllidiid nudibranchs, have been isolated from a Chinese population of the nudibranch Phyllidiella pustulosa. Three new sesquiterpenes (8, 10, and 11), with eudesmane, guaiane, and bisabolane skeletons, respectively, have been characterized by spectral methods and chemical comparison with known related molecules. The absolute stereochemistry of the major metabolite, the isocyanide 8, an enantiomer of the known sponge metabolite stylotelline (12), has also been determined.

Structure-dependent incorporation of terpenes into amphiphilic Poly(2-oxazoline) micelles.

Poly(2-oxazoline) (POx) amphiphilic block copolymers are highly effective carriers for poorly water-soluble small molecules, yet the structural features governing drug incorporation remain incompletely understood. Motivated by the unusually high loading of diterpene taxanes in POx micelles, we systematically examined whether shared terpene motifs support micellization with POx using natural compounds with established safety profiles. Representative mono-, di-, and triterpenes-α-pinene, carnosic acid (CA), carnosol (CARN), squalene, lupeol, betulin, and ursolic acid-were evaluated with a well-d

Oxidation and cyclization of casbene in the biosynthesis of Euphorbia factors from mature seeds of Euphorbia lathyris L.

The seed oil of Euphorbia lathyris L. contains a series of macrocyclic diterpenoids known as Euphorbia factors. They are the current industrial source of ingenol mebutate, which is approved for the treatment of actinic keratosis, a precancerous skin condition. Here, we report an alcohol dehydrogenase-mediated cyclization step in the biosynthetic pathway of Euphorbia factors, illustrating the origin of the intramolecular carbon-carbon bonds present in lathyrane and ingenane diterpenoids. This unconventional cyclization describes the ring closure of the macrocyclic diterpene casbene. Through tra

Kahweol Protects against Acetaminophen-Induced Hepatotoxicity in Mice through Inhibiting Oxidative Stress, Hepatocyte Death, and Inflammation.

Acetaminophen (APAP) can cause acute liver failure, but treatment options are still limited. Kahweol is the main diterpene compound of coffee and possesses antioxidant and anti-inflammatory properties. Emerging evidence suggests that this natural diterpene exerts favorable effects on several inflammatory diseases. However, the action of kahweol on APAP toxicity has not been addressed. The purpose of this study was to explore whether kahweol has a protective activity against APAP-induced hepatotoxicity and to investigate the mechanism. Administration of kahweol reduced serum levels of liver inj

Dietary cancer risk from conditional cancerogens in produce of livestock fed on species of spurge (Euphorbiaceae). II. Pathophysiological investigations in lactating goats fed on the skin irritant herb Euphorbia peplus and in their milk-raised kids.

Lactating goats were fed on aerial parts of the herb Euphorbia peplus L. admixed with their usual green fodder. During the experimental feeding period they showed symptoms of general poisoning. In necropsy the main toxic effects were seen in the heart, lung and liver. Histopathological examinations revealed that the primary toxic effects originated from degenerative changes in parenchymal and endothelial cells. Adverse symptoms in the liver and kidney were also reflected in an alteration of the levels of certain serum enzymes and of blood urea nitrogen. The milk of the goats fed on E. peplus,

Tremorgenic indole alkaloids potently inhibit smooth muscle high-conductance calcium-activated potassium channels.

Tremorgenic indole alkaloids produce neurological disorders (e.g., staggers syndromes) in ruminants. The mode of action of these fungal mycotoxins is not understood but may be related to their known effects on neurotransmitter release. To determine whether these effects could be due to inhibition of K+ channels, the interaction of various indole diterpenes with high-conductance Ca(2+)-activated K+ (maxi-K) channels was examined. Paspalitrem A, paspalitrem C, aflatrem, penitrem A, and paspalinine inhibit binding of [125I]charybdotoxin (ChTX) to maxi-K channels in bovine aortic smooth muscle sar
