Embryos derived from donor or patient oocytes are not different for in vitro fertilization outcomes when PGT allows euploid embryo selection: a retrospective study.

BACKGROUND: At our facilities, patients that received embryos using donor oocyte during in vitro fertilization (IVF), usually have had at least one failed attempt to produce at least one euploid embryo with their own oocytes; however, the current debate between using donor over patient oocytes remains inconclusive. We examined the aneuploidy rate and IVF clinical outcomes from embryos derived from either donor or patient oocytes. METHODS: Retrospectively, 973 cycles were examined of patients who underwent a standard IVF protocol. Chromosomal content was determined using Pre-implantation Genet

Reduced risk of transfusion-transmitted HIV in Kenya through centrally co-ordinated blood centres, stringent donor selection and effective p24 antigen-HIV antibody screening.

BACKGROUND: Following a 1994 study showing a high rate of transfusion-associated HIV, Kenya implemented WHO blood safety recommendations including: organizing the Kenya National Blood Transfusion Service (NBTS), stringent blood donor selection, and universal screening with fourth-generation p24 antigen and HIV antibody assays. Here, we estimate the risk of transfusion-associated HIV transmission in Kenya resulting from NBTS laboratory error and consider the potential safety benefit of instituting pooled nucleic acid testing (NAT) to reduce window period transmission. METHODS: From November to

The superior results of living-donor renal transplantation are not completely caused by selection or short cold ischemia time: a single-center, multivariate analysis.

BACKGROUND: The results of living-donor (LD) renal transplantations are better than those of postmortem-donor (PMD) transplantations. To investigate whether this can be explained by a more favorable patient selection procedure in the LD population, we performed a Cox proportional hazards analysis including variables with a known influence on graft survival. METHODS: All patients who underwent transplantations between January 1981 and July 2000 were included in the analysis (n=1,124, 2.6% missing values). There were 243 LD transplantations (including 30 unrelated) and 881 PMD transplantations.

Replacement of HLA class II serology by the HLA-DR microtitre plate oligotyping assay: a one-year experience in unrelated bone marrow donor selection.

Because the selection of unrelated bone marrow donors requires a more accurate HLA-typing technique than is used for sibling donors, we have replaced class II serology by a rapid and highly discriminative HLA-DR DNA oligotyping assay on microtitre plates. We present here our 1-year experience with class II typing for 112 potential donors identified for 33 consecutive leukaemic patients. Although the donors were selected to be HLA-A, -B, -DR serologically identical, the microtitre plate oligotyping assay detected DR incompatibilities in 52.7% of the patient/donor pairs. One third was due to inc

Human leukocyte antigen genotype modulates cytomegalovirus- and Epstein-Barr virus-specific T-cell response in healthy donors for optimized virus-specific T-cell donor selection.

INTRODUCTION: Reactivation of cytomegalovirus (CMV) and Epstein-Barr virus (EBV) is a major complication in immunocompromised patients, and understanding donor variability in antiviral immunity is essential for optimizing adoptive T-cell therapy. This study aimed to identify immunogenetic determinants of virus-specific T-cell responses in healthy donors to guide donor selection. METHODS: CMV- and EBV-specific T-cell responses were characterized using flow cytometry and IFN-g ELISpot after stimulation with peptide pools, followed by integrated analyses correlating the response magnitude with h

Insights into Inflammatory Priming of Adipose-Derived Mesenchymal Stem Cells: Validation of Extracellular Vesicles-Embedded miRNA Reference Genes as A Crucial Step for Donor Selection.

Mesenchymal stem cells (MSCs) are promising tools for cell-based therapies due to their homing to injury sites, where they secrete bioactive factors such as cytokines, lipids, and nucleic acids, either free or conveyed within extracellular vesicles (EVs). Depending on the local environment, MSCs' therapeutic value may be modulated, determining their fate and cell behavior. Inflammatory signals may induce critical changes on both the phenotype and secretory portfolio. Intriguingly, in animal models resembling joint diseases as osteoarthritis (OA), inflammatory priming enhanced the healing capac

Selective Angiography of Stimulant-Exposed Cardiac Donors Following Circulatory Death Does Not Impact Post-Transplant Outcomes.

Background/Objectives: Donation after circulatory death (DCD) has emerged to expand the heart-donor pool, but many DCD donors have risk factors such as cocaine or methamphetamine use. Stimulant use can cause coronary vasospasm and premature coronary artery disease, leading to routine donor coronary angiography (left heart catheterization, LHC) for coronary screening. However, performing LHC in DCD donors is challenging. We examined whether omitting LHC in stimulant-exposed DCD donors affects outcomes. Methods: A retrospective analysis was performed using the United Network for Organ Sharing (U

Selective Pathway toward Heteroleptic Spin-Crossover Iron(II) Complexes with Pyridine-Based N-Donor Ligands.

A new synthetic pathway is devised to selectively produce previously elusive heteroleptic iron(II) complexes of terpyridine and N,N'-disubstituted bis(pyrazol-3-yl)pyridines that stabilize the opposite spin states of the metal ion. Such a combination of the ligands in a series of the heteroleptic complexes induces the spin-crossover (SCO) not experienced by the homoleptic complexes of these ligands or shifts it to lower/higher temperatures respective to the SCO-active homoleptic complex. The midpoint temperatures of the resulting SCO span from ca. 200 K to the ambient temperature and beyond th

Vasopressor selection during critical care management of brain dead organ donors and the effects on kidney graft function.

BACKGROUND: Delayed graft function (DGF), the need for dialysis in the first week following kidney transplant, affects approximately one quarter of deceased-donor kidney transplant recipients. Donor demographics, donor serum creatinine, and graft cold ischemia time are associated with DGF. However, there is no consensus on the optimal management of hemodynamic instability in organ donors after brain death (DBDs). Our objective was to determine the relationship between vasopressor selection during donor management and the development of DGF. METHODS: Prospective observational data, including d

Selective omission of the donor cross-match before renal transplantation: efficacy, safety and effects on cold storage time.

BACKGROUND: A donor lymphocyte cross-match (XM) test performed before renal transplantation is considered mandatory but may delay the transplant and increase the cold storage time. With careful documentation of sensitizing events and with knowledge of previous antibody screening results, it is often possible to predict the XM result for a given donor HLA mismatch. In this study, a policy was adopted of omitting the pretransplant XM in patients in whom a negative result was predicted with absolute confidence. METHODS: Recipients were selected for cadaveric donor kidney transplantation using a

Allogeneic peripheral stem cell transplantation using positively selected CD34+ cells from HLA-mismatched donors.

We examined five children who underwent allogeneic peripheral stem cell transplantation (PSCT) using positively selected CD34+ cells from three or two loci-mismatched donors. CD34+ cells mobilized from peripheral blood were separated by immunomagnetic beads. CD34+ cells at 2.2-6.2 x 10(6)/kg were transplanted into three patients with refractory leukemia, a patient with relapsed medulloblastoma and a patient with Fanconi's anemia following a conditioning regimen which included irradiation, alkylating agents and antithymocyte globulin treatment. [Analysis of HLA class I molecules using isoelectric focusing. Its application in selection of bone marrow donors].

BACKGROUND: Differences between HLA proteins class I are assessed by serological typing using HLA allo-antisera. This method suffices for the assessment of HLA signs in allogenic transplantations of bone marrow in genotypically identical siblings. However, it does not suffice in the selection of donors from the wider family of a related or not related donor from the register of voluntary bone marrow donors. In order to assess the polymorphism of the HLA-class I not detected by serological typing other more sensitive techniques are introduced. In the authors department one-dimensional isoelectr

Twin-twin transfusion syndrome: rapid development of severe hydrops of the donor following selective feticide of the hydropic recipient.

Twin-twin transfusion syndrome is a serious complication of monozygotic twin pregnancies and is associated with high rates of perinatal morbidity and mortality. Unfortunately, therapies to disrupt the arteriovenous anastomoses are limited, and the success rate of these therapeutic modalities are poor. We present a case of severe twin-twin transfusion syndrome at 23 weeks' gestation in which selective feticide of the recipient hydropic twin by intracardiac injection of potassium chloride was followed by rapid development of hydrops in the donor twin.

Nickel(II) driven activity enhancement of a donor-acceptor porous organic polymer for selective photoreduction of carbon dioxide.

We report the design and synthesis of porous organic polymers (Py-DPEN) as a photocatalyst for the photoreduction of CO2 to carbon monoxide (CO). Incorporation of Ni(II) drastically enhanced the photocatalytic activity under visible light irradiation (70 W m-2, 470 nm), resulting in a significantly higher CO yield (38 µmol) with excellent selectivity. This pronounced improvement demonstrates the crucial role of Ni(II) in driving CO2 conversion using TEOA as a sacrificial electron donor in an acetonitrile-water medium. Comprehensive characterization, photocatalytic performance evaluation, and p

Multidimensional analysis of selected bioelements in rat's brain subjected to stroke procedure and treatment with H2S donor AP-39.

BACKGROUND: A stroke is characterized by a sudden disruption in blood flow to the brain. According to WHO statistics, stroke is the second most common cause of death. Its pathophysiology involves complex mechanisms: oxidative stress, inflammation, cytotoxicity and neuronal cell death. Middle cerebral artery occlusion (MCAO) in rats is commonly used to study the pathophysiology of stroke, as well as the efficacy of therapeutic strategies e.g. application of H2S donors. OBJECTIVES: The aim of this study was to determine the concentrations of minerals (Mg, Na, K), and trace elements (Fe, Cu, and

Simultaneous en bloc kidney and pancreas transplantation from pediatric donors: Selection, surgical strategy, management, and outcomes.

Pediatric donors are underutilized for simultaneous pancreas-kidney transplantation due to concerns about technical complications and inadequate islet and/or renal mass. We analyzed our experience with simultaneous en bloc kidney and pancreas transplantation using pediatric donors on 8 consecutive adult patients from 1997-2018. En bloc kidney transplants were implanted intraperitoneally and contralaterally to right-sided pancreas grafts. All patients became insulin-independent immediately; with 1 case of delayed kidney function and 1 case of insulin resistance; there were no graft thromboses.

Yield stability of selected rice breeding lines and donors across conditions of mild to moderately severe drought stress.

Although mild to moderately severe drought stress may have less of an effect on rice grain yield than severe drought stress during reproductive stage, its prevalence across rice farmers' fields at the global level may be more economically significant. In this study, field experiments were conducted on selected genotypes with known tolerance to severe reproductive-stage drought in order to identify those that would produce high and stable grain yield across seasons and soil moisture conditions varying from well-watered to mild and moderately severe drought stress. Mild stress generally occurred

Selective Transfer Hydrogenation of Furfural into Furfuryl Alcohol on Zr-Containing Catalysts Using Lower Alcohols as Hydrogen Donors.

A series of zirconium-based catalysts were prepared for the selective transfer hydrogenation of biomass-derived furfural (FFR) into furfuryl alcohol with lower alcohols as hydrogen sources. The sample structures were clearly characterized using various methods, such as X-ray powder diffraction, thermogravimetric analysis, scanning electron microscope, NH3-temperature-programmed desorption (TPD), CO2-TPD, and nitrogen physisorption. Excellent furfuryl alcohol yield of 98.9 mol % was achieved over Zr(OH)4 using 2-propanol as a hydrogen donor at 447 K. The poisoning experiments indicated that bas
