Cannabinoid modulation of prefrontal-limbic activation during fear extinction learning and recall in humans.

Pre-extinction administration of Δ9-tetrahydrocannibinol (THC) facilitates recall of extinction in healthy humans, and evidence from animal studies suggest that this likely occurs via enhancement of the cannabinoid system within the ventromedial prefrontal cortex (vmPFC) and hippocampus (HIPP), brain structures critical to fear extinction. However, the effect of cannabinoids on the underlying neural circuitry of extinction memory recall in humans has not been demonstrated. We conducted a functional magnetic resonance imaging (fMRI) study using a randomized, double-blind, placebo-controlled, be

Synthetic Pathway of Nonpsychoactive Cannabidiol from Psychoactive Tetrahydrocannabinol: A DFT Guide.

In this study, a reaction pathway for converting the psychoactive compound Δ9-THC into the nonpsychoactive cannabidiol (CBD) was proposed. Related cannabinoids, including Δ8-THC and cannabinol (CBN), were also examined. The transformation proceeds via two key steps: (i) hydrogen elimination and cyclic ether ring cleavage, and (ii) hydrolysis to form hydroxyl groups, facilitated by boron trichloride (BCl3) as a Lewis acid and 2,6-di-tert-butyl-4-methylpyridine as a selective base. Energy calculations indicate that TS4-5 is the rate-determining transition state for Δ9-THC, Δ8-THC, and CBN, with

The subjective effects of △9-tetrahydrocannabinol: A systematic review and dose-response meta-regression.

Global shifts towards the liberalisation of cannabis policies have increased the potency of cannabis products, alongside their accessibility, diversity, and the number of inexperienced cannabis consumers. There is a lack of systematic evidence on the subjective effects of cannabis' main psychoactive compound, Δ9-tetrahydrocannabinol (THC). We systematically reviewed and meta-analysed data from 41 randomised, double-blind, and placebo-controlled experimental studies on the acute subjective effects of THC in 1020 healthy individuals with infrequent cannabis use (<2 weekly; <100 lifetime). Pairwi

Single trichome phytocannabinomics of two different cannabis varieties.

The present study investigates the phytocannabinoid profiles of Cannabis sativa L. by analyzing individual glandular trichomes from both a medicinal cannabis variety (FM2) and a high-CBD (cannabidiol) hemp variety. The analysis was performed using an untargeted metabolomics approach (phytocannabinomics) that enabled the identification of nearly 70 phytocannabinoids in single trichomes, a significant improvement over previous studies, which typically analyzed pooled or enriched trichome fractions. For comparison, the phytocannabinoid profiles of whole inflorescences were also examined. Results

Kynurenine amplifies tetrahydrocannabinol-induced sensorimotor impairment and classic "tetrad" effects in mice.

BACKGROUND: L-kynurenine (KYN), a kynurenine pathway (KP) metabolite, is the main precursor for the neuroactive metabolite kynurenic acid (KYNA). Several studies suggest a patho-physiologically relevant association between increased brain KYNA levels and cognitive dysfunctions in individuals with schizophrenia. Δ9-tetrahydrocannabinol (Δ9-THC; i.e. the main psychoactive compound of cannabis) can worse schizophrenia-related psychosis, often leads to the development of cannabis use disorder in individuals with schizophrenia, and increases the risk of earlier onset of schizophrenia symptoms in th

High-Potency Cannabis Use and Health: A Systematic Review of Observational and Experimental Studies.

OBJECTIVE: Amid continuously rising concentrations of delta-9-tetrahydrocannabinol (THC) in cannabis (i.e., potency), high-potency cannabis is a major topic in contemporary cannabis policy discussions, yet its impact on health is not well understood. The authors conducted a systematic review of observational and experimental studies examining the relationship between high-potency cannabis use and a range of health outcomes. METHODS: Records were obtained from a systematic search of five biomedical research databases. The authors developed ecologically relevant potency (percent THC) exposure-c

Recently, semi-synthetic cannabinoids have entered the illegal market and are produced to mimic the psychoactive effects of tetrahydrocannabinol (THC). This is a case report of a 19-year-old man, who was hospitalized due to severe sedation, hypotension and tachy- and bradycardia after ingestion of the semi-synthetic cannabinoids hexahydrocannabinol (HHC) and hexahydrocannabiphorol (HHC-P) mixed in food. HHC-P, HHC and metabolites were identified in blood samples. The amount of semi-synthetic cannabinoids in illegal products can be high, which can explain the described prolonged clinical effect

The Relationship Between Cannabis, Cognition, and Schizophrenia: It's Complicated.

The consequences of cannabis use, especially in the context of schizophrenia, have gained increased importance with the legalization of cannabis in North America and across the globe. Cannabis use has multifaceted impacts on cognition in schizophrenia patients and healthy subjects. Healthy subjects, particularly those who initiated cannabis use at earlier ages and used high-potency cannabis for longer durations, exhibited poorer cognition mainly in working memory and attention. Cannabis use in schizophrenia has been associated with symptom exacerbation, longer and more frequent psychotic episo

An Individuality of Response to Cannabinoids: Challenges in Safety and Efficacy of Cannabis Products.

Since legalization, cannabis/marijuana has been gaining considerable attention as a functional ingredient in food. ∆-9 tetrahydrocannabinol (THC), cannabidiol (CBD), and other cannabinoids are key bioactive compounds with health benefits. The oral consumption of cannabis transports much less hazardous chemicals than smoking. Nevertheless, the response to cannabis is biphasically dose-dependent (hormesis; a low-dose stimulation and a high-dose inhibition) with wide individuality in responses. Thus, the exact same dose and preparation of cannabis may be beneficial for some but toxic to others. T

Cannabis as a Source of Approved Drugs: A New Look at an Old Problem.

Cannabis plants have been used in medicine since ancient times. They are well known for their anti-diabetic, anti-inflammatory, neuroprotective, anti-cancer, anti-oxidative, anti-microbial, anti-viral, and anti-fungal activities. A growing body of evidence indicates that targeting the endocannabinoid system and various other receptors with cannabinoid compounds holds great promise for addressing multiple medical conditions. There are two distinct avenues in the development of cannabinoid-based drugs. The first involves creating treatments directly based on the components of the cannabis plant.

The Utility of THC Cutoff Levels in Blood and Saliva for Detection of Impaired Driving.

Background: Δ9-Tetrahydrocannabinol (THC) is the psychoactive component in cannabis and a relationship of THC to driving impairment is expected. Despite this, there are discrepant findings with respect to the relationship of blood THC to driving. This study investigated the relationship of blood, urine, and saliva THC/THC-COOH levels to "weaving," as measured by a driving simulator. Methods: Participants smoked cannabis alone or with alcohol. THC/THC-COOH levels in blood, urine, and saliva were correlated with standard deviation of lateral position (SDLP), measuring "weaving." In addition, SDL

Time- and temperature-dependent postmortem ∆9-tetrahydrocannabinol concentration changes in rabbits following controlled inhaled cannabis administration.

ostmortem redistribution (PMR), a well-known phenomenon in forensic toxicology, can result in substantial changes in drug concentrations after death, depending on the chemical characteristics of the drug, blood collection site, storage conditions of the body and postmortem interval (PMI). Limited PMR data are available for ∆9-tetrahydrocannabinol (THC), the primary psychoactive component in Cannabis sativa. PMR was evaluated after controlled cannabis inhalation via a smoking machine and exposure chamber in New Zealand white rabbits. Necropsies were performed on five control rabbits immediately

Tetrahydrocannabinol and Cannabidiol in Tourette Syndrome.

BACKGROUND: Tourette syndrome is characterized by chronic motor and vocal tics. There is preliminary evidence of benefit from cannabis products containing Δ9-tetrahydrocannabinol (THC) and that coadministration of cannabidiol (CBD) improves the side-effect profile and safety. METHODS: In this double-blind, crossover trial, participants with severe Tourette syndrome were randomly assigned to a 6-week treatment period with escalating doses of an oral oil containing 5 mg/ml of THC and 5 mg/ml of CBD, followed by a 6-week course of placebo, or vice versa, separated by a 4-week washout period. Review of delta-8-tetrahydrocannabinol (Δ8 -THC): Comparative pharmacology with Δ9 -THC.

The use of the intoxicating cannabinoid delta-8-tetrahydrocannabinol (Δ8 -THC) has grown rapidly over the last several years. There have been dozens of Δ8 -THC studies dating back over many decades, yet no review articles have comprehensively covered these findings. In this review, we summarize the pharmacological studies of Δ8 -THC, including receptor binding, cell signalling, in vivo cannabimimetic activity, clinical activity and pharmacokinetics. We give special focus to studies that directly compared Δ8 -THC to its more commonly studied isomer, Δ9 -THC. Overall, the pharmacokinetics and ph

Effects of psychotropic drugs on ocular parameters relevant to traffic safety: A systematic review.

Driving is a complex neurobehavioural task necessitating the rapid selection, uptake, and processing of visual information. Eye movements that are critical for the execution of visually guided behaviour such as driving are also sensitive to the effects of psychotropic substances. The Embase (via Ovid), EBSCOHost, Psynet, Pubmed, Scopus and Web of Science databases were examined from January 01st, 2000 to December 31st, 2021. Study selection, data extraction and Cochrane Risk of Bias (RoB2) assessments were conducted according to PRISMA guidelines. The review was prospectively registered (CRD42

Potential effects of nicotine content in cigarettes on use of other substances.

A national nicotine reduction policy has the potential to reduce cigarette smoking and associated adverse health impacts among vulnerable populations. However, possible unanticipated adverse effects of reducing nicotine content in cigarettes, such as increasing the use of alcohol or other abused substances, must be examined. The purpose of this study was to evaluate the effects of exposure to varying doses of nicotine in cigarettes on use of other substances. This was a secondary analysis (n = 753) of three simultaneous, multisite, double-blind, randomized-controlled trials examining 12 weeks

Determination of Cross-Reactivity of Contemporary Cannabinoids with THC Direct Immunoassay (ELISA) in Whole Blood.

Immunoassay procedures, such as enzyme-linked immunosorbent assay (ELISA), are widely used for screening samples in both driving under the influence of drugs (DUID) and postmortem (PM) investigations. While these are sensitive and widely used techniques, they lack specificity compared to more novel instrumental screening platforms. In this study, the cross-reactivities of several cannabinoid isomers and related compounds were evaluated in whole blood using the Cannabinoids Direct ELISA kit from Immunalysis. The compounds of interest were supplemented individually at three different concentrati
