The ubiquitin proteasome system (UPS) is the primary mechanism for targeted protein degradation in eukaryotic cells. Dysfunction of this system is a driver of human disease and a hallmark of aging and late-onset neurodegenerative disorders. Understanding the mechanisms that ensure robust protein turnover may provide new avenues for treatment in these contexts. E3 ubiquitin ligases play critical roles in supplying ubiquitinated substrates to the proteasome, with HUWE1 being an enormous, versatile, and highly conserved member of this family. Here, we show that the C. elegans HUWE1 ortholog EEL-1

INTRODUCTION AND IMPORTANCE: Gastrointestinal perforation is a very common surgical emergency with significant mortality and morbidity, which needs urgent intervention. Foreign body-related perforations are very rare, accounting for less than 1% of cases. However, Colonic perforation by Eel fish is an extremely rare phenomenon; only three cases have been reported in the existing literature. Here, we report a rare case of traumatic colonic perforation with septic shock by a rectally inserted Eel fish.
CASE PRESENTATION: A 25-year-old male presented to the emergency department with a 4-day hist

Influenza virus neuraminidase (NA) is an important target for antiviral development because it plays a crucial role in releasing newly assembled viruses. Two unique influenza-like virus genomes were recently reported in the Wuhan Asiatic toad and Wuhan spiny eel. Their NA genes appear to be highly divergent from all known influenza NAs, raising key questions as to whether the Asiatic toad influenza-like virus NA (tNA) and spiny eel NA (eNA) have canonical NA activities and structures and whether they show sensitivity to NA inhibitors (NAIs). Here, we found that both tNA and eNA have neuraminid

Lipids, particularly fatty acids (FAs), are major sources of energy and nutrients in aquatic ecosystems and play key roles during vertebrate development. The European eel Anguilla anguilla goes through major biochemical and physiological changes throughout its lifecycle as it inhabits sea- (SW), and/or brackish- (BW) and/or freshwater (FW) habitats. With the ultimate goal being to understand the reasons for eels adopting a certain life history strategy (FW or SW residency vs. 'habitat shifting'), we explored differences in lipid content and FA composition of muscle, liver and eyes from eels co

We prepared monoclonal antibodies (mAbs) against a recombinant tethered follicle-stimulating hormone (rec-FSH) from Japanese eel Anguilla japonica that was produced in Escherichia coli. Positive hybridomas (clones eFA-C5, eFA-C10, eFA-C11, eFA-C12, eFA-C13, and eFB-C14) were selected by using the eel FSH antigen in ELISA, and anti-eel FSH mAbs were purified from culture supernatants by performing affinity chromatography. Three of the 6mAbs were characterized and their isotypes were identified as IgG2b (eFA-C5 and eFA-C11) and IgG1 (eFB-C14). In western blotting assays, the mAbs recognized the

BACKGROUND: Homeostasis of ions and water is important for the maintenance of cellular functions. The regulation of the homeostasis is particularly important in euryhaline fish that migrate between freshwater (FW) and seawater (SW) environments. The fish gill, the major tissue that forms an interface separating the extracellular fluids and external water environment, has an effective transport system to maintain and regulate a constant body osmolality. In fish gills, the two major epithelial cells, pavement cells (PVCs) and mitochondria-rich cells (MRCs), are known to play key and complementar

The swamp eel, Monopterus albus, is an air-breathing teleost which typically lives in freshwater but can also be found in estuaries, where it has to deal with ambient salinity fluctuations. Unlike other teleosts, its gills are highly degenerate. Hence, it may have uncommon osmoregulatory adaptations, but no information is available on its osmoregulatory capacity and mechanisms at present. In this study M. albus was exposed to a 5 day progressive increase in salinity from freshwater (1 per thousand) to brackish water (25 per thousand) and subsequently kept in 25 per thousand water for a total o

The partition of O2 uptake between gills and skin was examined in the freshwater eel (Anguilla anguilla L.) at ambient PO2 ranging from hyperoxia (PO2 = 400 Torr) to severe hypoxia (PO2 = 10 Torr), using a technique of open-flow respirometry. All the expired water was collected, and the ventilatory flow and the mixed expired water PO2 were directly measured. The ventilatory water flow decreased moderately in hyperoxia, increased markedly between normoxia and 40 Torr, and below 40 Torr, hyperventilation was gradually reduced. Between PO2 400 and 70 Torr, the total O2 uptake was constant and the

Batrachotoxinin-A [(3)H]benzoate ([(3)H]BTX-B) binds specifically and with high affinity (K(D) 48 nM) to sites (B(max) 2.1 pmol/mg protein) associated with voltage-dependent sodium channels in rodent brain vesicular preparations. High affinity binding requires the presence of scorpion (Leiurus) venom and a membrane potential. Local anesthetics antagonize the binding. Nonspecific binding is defined in the presence of veratridine. In particulate preparations from electroplax of the eel Electrophorus electricus, [(3)H]BTX-B binds with a K(D) of about 140 nM and a B(max) of 2.5 pmol/mg protein in

We report the establishment of a platform for high-precision, meV-scale electron energy-loss spectroscopy (EELS) by the installation of the JEOL Atomic-Resolution Multi-Dimensional Transmission Electron Microscope (ARMD-TEM), which combines the CEOS Energy-Filtering and Imaging Device (CEFID) with a Wien-filter monochromator and aberration corrected optics, thus enabling ultra-stable, high-throughput low-loss spectroscopy with atomic-scale spatial resolution. Energy resolution and spectral stability are quantitatively assessed using zero-loss peak (ZLP) series of 10,000 spectra per dataset wit

The low energy region (< 50 eV) of the electron energy loss spectrum (EELS) can contain a great deal of spectral detail associated with excitations of the valence electrons. Calculation of the spectra from first principles can assist with interpretation and the most widely used method is the random phase approximation (RPA), usually neglecting local field effects (LFE). For KBr this approach is insufficient due to the importance of quasiparticle and excitonic effects. Calculations including these multi-electron effects are shown to give much improved agreement with the experimental spectra, an

Breast cancer is the most common cancer type amongst women throughout the world. Mammography is a successful imaging modality for the earliest diagnosis of breast cancer in its beginning stages. Owing to less visibility and bad contrast in mammogram (MG) images, an earlier identification of breast cancer is a critical step for efficacious treatment of this disease. Here, the Eel and Grouper Lyrebird Optimizer-based Fractal Deep Spiking Residual Network (EGLO_FDSRN) is introduced for the detection of breast cancer using MG images. Initially, the MG image from mammographic image datasets is cons

To compare the fatty acid composition in different tissues of Anguilla japonica before and after sea entry, we analyzed the muscle, liver, and ovary of eels from the Yangtze Estuary and offshore waters, and describe the distribution patterns of various fatty acids in different tissues. Comparing different tissues, in eels from the estuary, the muscle had the highest content of EPA, the liver had the highest content of DHA, and the ovary had the highest contents of ARA. In offshore eels, the muscle had the highest contents of C16:1 and C18:1n9c; the liver had the highest content of C16:0; and t

Ricefield eel is an important economic fish in China. However, large-scale intensive breeding has increased the incidence of diseases in eels. In this study, we conducted an 8-week feeding trial to investigate the effects of β-glucan on the growth performance, intestinal health, and Aeromonas veronii resistance of Monopterus albus (M. albus). Three hundred healthy fish (initial body weight: 12.38 ± 0.50 g) were randomly divided into five groups: A1 (basal diet) was considered the control group, whereas A2, A3, A4, and A5 were the experimental groups. The fish in the experimental groups were fe

BACKGROUND: An increasing number of long noncoding RNAs (lncRNAs) have been found to play important roles in sex differentiation and gonad development by regulating gene expression at the epigenetic, transcriptional and posttranscriptional levels. The ricefield eel, Monopterus albus, is a protogynous hermaphroditic fish that undergoes a sequential sex change from female to male. However, the roles of lncRNA in the sex change is unclear.
RESULTS: Herein, we performed RNA sequencing to analyse lncRNA expression patterns in five different stages of M. albus development to investigate the roles o

Invasive parasites have been implicated in the declines of several freshwater species. The swim bladder nematode Anguillicola crassus was introduced into Europe in the 1980s and is considered a threat to the European eel (Anguilla anguilla). Infection affects stress resistance and swimming behaviour. European eels produce an immune response against the parasite during the late stages of infection and after repeated infections. We used RNA-seq to examine the molecular response to infection during the poorly understood early stage and identify expression of genes and associated processes that ar

Complete sexual maturation of European eels (Anguilla anguilla) in captivity can only be achieved via injections with gonadotropins. For female eels this procedure takes 4-6months and the response ranges from "unresponsive" to final maturation and ovulation. Reproductive success could be significantly increased via early selection of responders based on predictive markers and minimally invasive sampling methods. To get a better understanding of the genetic background of ovarian maturation of the European eel we performed a pilot deep-sequencing transcriptome analysis of ovarian tissue derived

A total of 80 specimens of the European eel Anguilla anguilla were collected during the period from February 2013 to March 2014 at the coast of the Gulf of Suez (Red Sea, Egypt). A new species of parasitic nematodes was recovered and described as Cucullanus egyptae. It was found in the eel's intestine with a prevalence of 68.7%. The morphology of the recovered parasite was studied by light and scanning electron microscopy. The adult worms had a wide cephalic extremity with a slit-like oral aperture being surrounded by a cuticular ring and delimited internally by a row of small teeth in additio

Luteinizing hormone (Lh) and follicle-stimulating hormone (Fsh) control many aspects of gonadal development and function in teleosts. In the present paper, the specific antisera against ricefield eel Lhb (Lh beta subunit), Fshb (Fsh beta subunit), and Cga (the common pituitary glycoprotein hormone alpha subunit) were generated, and the cellular localization, initial appearance, and subsequent development of gonadotrophs in relation to early ovarian differentiation and development in the ricefield eel, a protogynous sex-changing teleost, were examined with immunochemistry. Lhb- and Fshb-immunor

Anguillid herpesvirus (AngHV) is the causative agent of 'mucus sloughing and hemorrhagic septicemia disease' in eels. Egg yolk immunoglobulin (IgY) has been reported to serve as oral administration of antibodies against bacterial and viral pathogens. In this study, an inactivated vaccine of AngHV was developed to immunise the laying hens, thereby facilitating the production of anti-AngHV IgY. The yolks were subsequently collected and processed into a dry yolk powder, and IgY was extracted from the yolk powder. The anti-AngHV efficacy of the IgY was assessed through both in vitro and in vivo as
