In vitro fertilization, intracytoplasmic sperm injection, in vitro maturation, and embryo culture.

Infertility is a highly prevalent disease affecting 1 in 6 couples worldwide. While there are numerous treatment options available to treat infertility, in vitro fertilization (IVF) is the most successful therapy available. IVF traditionally involves superovulation of the ovaries followed by ultrasound (US)-guided oocyte retrieval, fertilization of oocytes in vitro, and culture of resultant embryos. Embryo(s) are then either transferred into the uterus or cryopreserved. There have been tremendous advances in each step of this process over the past 40 years, however, much work still remains to

Preimplantation Genetic Testing for Aneuploidy (PGT-A) in In-Vitro Fertilisation (IVF) Treatment: Study Protocol for Pilot Phase of a Randomised Controlled Trial.

Introduction: Poor outcomes following IVF treatments are speculated to be due to the transfer of aneuploid embryos that cannot be identified based on morphological evaluation alone. This leads to patients requiring numerous embryo transfers and, consequently, a prolonged time interval before live birth. Embryo selection following preimplantation genetic testing for aneuploidy (PGT-A) with next-generation sequencing (NGS) has been suggested as an intervention to shorten time to pregnancy in women undergoing in vitro fertilisation (IVF). Past studies assessing the clinical efficacy of PGT-A in i

The use of autologous platelet-rich plasma in embryo culture to obtain high-quality embryos.

Transferring high-quality embryos is crucial for in vitro fertilization success, as they lead to better clinical pregnancy and live birth outcomes. Researchers have investigated adding supplements to culture media to enhance embryo quality because culture conditions greatly affect embryo quality and development. This study assessed whether adding autologous platelet-rich plasma containing growth factors and cytokines to the culture media could produce more usable and high-grade embryos. This retrospective study analyzed 175 in vitro fertilization cycles from 123 women with poor embryo developm

Sensitivity Variations in Developmental Toxicity of Imidacloprid to Zebrafish Embryos at Different Neurodevelopmental Stages.

Neonicotinoids are ubiquitous in global surface waters and pose a significant risk to aquatic organisms. However, information is lacking on the variations in sensitivity of organisms at different developmental stages to the neurotoxic neonicotinoids. We established a spectrum of toxicity to zebrafish embryos at four neurodevelopmental stages (1, 3, 6, and 8 h post fertilization [hpf]) and dechorionated embryos at 6 hpf based on external and internal exposure to imidacloprid as a representative neonicotinoid. Embryos at the gastrula stage (6 and 8 hpf) were more sensitive to imidacloprid than e

l-selenomethionine induces zebrafish embryo cardiovascular defects via down-regulating expression of lrp2b.

Selenium plays crucial roles in maintaining the growth and development of vertebrates including humans. However, excessive selenium in cells will lead to developmental defects and disease. Selenium has been reported to cause severe malformation in zebrafish embryos, but there are few studies on the mechanism of selenium excess-induced cardiovascular defects. In this study, the fertilized zebrafish embryos were treated with selenium for 96 h post fertilization (hpf). Under selenium stress, wild-type embryos showed pericardial edema, heart rate decrease, ectopic accumulation of hemoglobin; fli1-

Human stem cell-based embryo models: innovation, ethics, and policy.

The aim of this White Paper is to establish a foundational framework for research, technological development, and regulation in the emerging field of stem cell-based embryo models (SCBEMs). These models, generated from Pluripotent Stem Cells, are designed to recapitulate essential events in early stages of human development. They have the potential to illuminate the early stages of embryo development and implantation and hold promise as an avenue to address global health challenges, including infertility and pregnancy loss, congenital, neonatal and adult conditions, and the need for organ tran

Diabetic environment and genetic predisposition as causes of congenital malformations in NOD mouse embryos.

Congenital malformations such as neural tube defects and a kinky or waved vertebral column were observed at higher incidence in embryos from nonobese diabetic (NOD) female mice with overt diabetes (NOD-D; 40.3%, P less than 0.005) or without overt diabetes (NOD-N; 8.4%, P less than 0.05) than in control Institute of Cancer Research (ICR) mouse embryos (1%) at day 13 of gestation. In vivo and in vitro preimplantation development of NOD-N, NOD-D, and ICR embryos did not differ in rate of development, size, or morphology. Embryos cultured from one-cell to early blastocyst stage were mutually tran

A context-specific evaluation of polygenic embryo screening in best-prognosis preimplantation genetic testing cycles.

STUDY QUESTION: In best-prognosis preimplantation genetic testing (PGT) cycles-defined by the availability of multiple transferable embryos-can polygenic embryo screening (PES) provide meaningful, context-dependent stratification that may influence embryo prioritization? SUMMARY ANSWER: In a sister-pair breast cancer validation cohort, the polygenic risk score (PRS) showed modest predictive performance, and when applied retrospectively to embryos, differences in modelled susceptibility were context-dependent, primarily influenced by the number of available embryos, parental polygenic risk pro

Incubator type affects human blastocyst formation and embryo metabolism: a randomized controlled trial.

STUDY QUESTION: Does the type of incubator used to culture human preimplantation embryos affect development to the blastocyst stage and alter amino acid utilization of embryos in assisted reproduction? SUMMARY ANSWER: Culturing embryos in a time lapse system (TLS) was associated with a higher Day 5 blastocyst formation rate and altered amino acid utilization when measured from Day 3 to Day 5 compared to the standard benchtop incubator. WHAT IS KNOWN ALREADY: Culture environment is known to be important for the developing preimplantation embryo. TLSs provide a stable milieu allowing embryos t

Predicting pregnancy outcomes in IVF cycles: a systematic review and diagnostic meta- analysis of artificial intelligence in embryo assessment.

INTRODUCTION: Embryo selection remains a key challenge in in vitro fertilization (IVF), as many morphologically "normal" embryos fail to implant. Artificial intelligence (AI) offers a promising tool for improving embryo assessment by providing more objective and accurate predictions of pregnancy outcomes. This study aims to systematically review and conduct a diagnostic meta-analysis to evaluate the effectiveness of AI-based tools in embryo selection for predicting pregnancy outcomes in IVF. METHODS: We conducted a systematic review following PRISMA guidelines, searching Web of Science, Scopu

Emerging in vitro platforms and omics technologies for studying the endometrium and early embryo-maternal interface in humans.

Human reproductive success relies on the intricate interplay between the developing embryo and the maternal endometrium. These highly-coordinated interactions facilitate implantation, setting in motion a series of developmental programs to establish a sustained fetal-maternal interface. Understanding endometrial function and the early human embryo-maternal dialogue is thus an important prerequisite for refining clinical approaches to alleviate implantation failure, early pregnancy loss and other obstetric complications. Yet, many mediators of implantation remain elusive. Driven by endocrine fa

Optimal utilization of modern reproductive technologies to maximize the gross margin of milk production.

In this study, a linear programming model was developed to maximize the gross margin of milk production by determining the optimal use of different reproductive technologies in a dairy herd. The model has the potential to vary the use of conventional artificial insemination, insemination with X-sorted sperm, and the use of unselected or sex-selected embryo recovery and transfer. Data from Finnish dairy herd recording systems were used to parameterize the model. This paper presents the results of 6 scenarios for a herd size of 60 dairy cows. In the basic scenario, the optimum economic combinati

Targeted management of adenomyosis: frozen embryo transfer outcomes with GnRH-agonist and letrozole therapy: a case series.

BACKGROUND: Adenomyosis negatively impacts fertility treatment outcomes by lowering clinical pregnancy rates, reducing live birth rates, and increasing miscarriage rates following in vitro fertilization. Hormonal suppression therapy has emerged as a preferred treatment for patients with adenomyosis, particularly before endometrial preparation for frozen-thawed embryo transfer; however, the combination therapy for individualized patients can improve the outcomes. Here we present three cases in wherein we applied a combined protocol using GnRH agonists and letrozole for endometrial preparation i

Embryo transfer in competition horses: Managing mares and expectations.

Embryo transfer (ET) is an accepted and successful technique for obtaining foals from mares without interrupting their competition careers. Recent research, however, suggests that the potential of factors including heat, exercise, repeated embryo flushing and repeated manipulation of the reproductive cycle using exogenous hormones to have a negative impact on fertility may have been underestimated. This paper reviews the evidence base for involvement of these factors in repeated failures to recover embryos from nongeriatric competition mares without obvious clinical or pathological indications

Does assisted hatching improve implantation rates after in vitro fertilization or intracytoplasmic sperm injection in all patients? A prospective randomized study.

PURPOSE: Preliminary data from some research centers indicate that assisted hatching might be of value to increase embryo implantation rate in the human, at least in selected cases. It is not clear, however, whether this technique would be of benefit for all patients undergoing an embryo transfer. We therefore performed a prospective randomized study to evaluate the effect of assisted hatching on the implantation rate in our in vitro fertilization (IVF)/intracytoplasmic sperm injection (ICSI) program. METHODS: In total, 120 couples undergoing an embryo transfer were randomized between two gro
