Integrating virtual screening and molecular dynamics simulations to identify emodin as a PYCR1 inhibitor modulating docetaxel sensitivity in prostate cancer.

Docetaxel (DTX) resistance is the main cause of treatment failure in castration-resistant prostate cancer (CRPC). Pyrroline-5-carboxylic acid reductase 1 (PYCR1) is an enzyme involved in proline metabolism. It is highly expressed in various cancers and promotes malignant progression, yet its role in DTX resistance in prostate cancer remains unclear. In this study, bioinformatics analyses and in vitro/vivo experiments demonstrated that interfering with PYCR1 expression modulates the sensitivity of prostate cancer cells to DTX. Subsequently, via structure-based virtual screening, molecular dynam

Emodin-Loaded Thermoresponsive Hydrogel as a Potential Drug Delivery System for Periodontal Disease in a Rat Model of Ligature-Induced Periodontitis.

Periodontitis, a chronic inflammatory disease, causes alveolar bone loss. Current treatments show limitations in achieving dual antimicrobial and anti-inflammatory effects. We evaluated an emodin-loaded thermoresponsive hydrogel as a local drug delivery system for periodontitis treatment. Emodin itself demonstrated antibacterial activity against Porphyromonas gingivalis, with minimal inhibitory and minimal bactericidal concentrations of 50 μM. It also suppressed mRNA expression of proinflammatory cytokines [tumor necrosis factor alpha, interleukin (IL)-1β, and IL-6] in lipopolysaccharide-stimu

De novo biosynthesis of the plant-derived aloe-emodin in Aspergillus nidulans via artificial pathway assembly and temporal flux control.

Traditional botanical extraction of medicinal anthraquinones like aloe-emodin is limited by low yields, long growth cycles, and environmental dependence. Microbial biosynthesis provides a sustainable alternative, but de novo reconstruction of aloe-emodin remains difficult due to unresolved pathway steps and poor metabolic flux distribution in engineered hosts. In this study, we engineered Aspergillus nidulans for the de novo biosynthesis of aloe-emodin via modular assembly of an artificial pathway. We first established an emodin-producing chassis by reconstructing a minimal mdp-derived upstrea

Emodin Alleviates Acute Pancreatitis-Associated Acute Lung Injury by Inhibiting Serum Exosomal miRNA-21-3p-Induced M1 Alveolar Macrophage Polarisation.

Severe acute pancreatitis (SAP) is a common abdominal emergency in clinical practice. Approximately 20%-40% of patients with SAP will be associated with acute lung injury (SAP-ALI), which is a major cause of death. Emodin (EMO) is a naturally occurring anthraquinone derivative with various pharmacological properties. EMO has a therapeutic effect on SAP-ALI; however, the underlying mechanism remains unclear. Exosomes mediate intercellular communication in disease progression. Therefore, this study aimed to explore the role of serum exosomes in SAP-ALI and the potential mechanisms by which EMO r

Emodin reduces surgical wounding-accelerated tumor growth and metastasis via macrophage suppression in a murine triple-negative breast cancer model.

It has been suspected that tumor resection surgery itself may accelerate breast cancer (BC) lung metastasis in some patients. Emodin, a natural anthraquinone found in the roots and rhizomes of various plants, exhibits anticancer activity. We examined the perioperative use of emodin in our established surgery wounding murine BC model. Emodin reduced primary BC tumor growth and metastasis in the lungs in both sham and surgical wounded mice, consistent with a reduction in proliferation and enhanced apoptosis (primary tumor and lungs). Further, emodin reduced systemic inflammation, most notably th

Emodin Successfully Inhibited Invasion of Brucella abortus Via Modulting Adherence, Microtubule Dynamics and ERK Signaling Pathway in RAW 264.7 Cells.

The aim of this work is to investigate the protective efficacy of emodin, an active, naturally-occurring anthraquinone derivative of several traditional Chinese herbs, against Brucella abortus infection in macrophages. Brucella were incubated with different concentrations of emodin and showed that bacterial survival rates were markedly reduced in a dose-dependent manner at increasing incubation time points. Through bacterial infection assay, the highest non-cytotoxic concentration of emodin demonstrated attenuated invasion of Brucella into macrophages, however it did not inhibit the growth of

Aloe-emodin ameliorates non-alcoholic fatty liver disease by regulating the ZNRF3/Wnt/β-catenin axis and suppressing hepatic lipogenesis.

Non-alcoholic fatty liver disease (NAFLD) is a highly prevalent metabolic disorder with limited approved pharmacological treatment options, creating an urgent need for novel therapeutic agents. This study investigated the efficacy and underlying molecular mechanism of aloe-emodin, a natural anthraquinone from Rhei Radix et Rhizoma, in treating NAFLD. In a high-fat diet (HFD)-induced mouse model, aloe-emodin administration significantly reduced body and liver weight, ameliorated hepatic steatosis, lowered serum ALT and hepatic lipid levels, and suppressed the expression of key lipogenic genes.

Carboxymethyl chitosan-modified pH/redox dual-responsive hollow mesoporous manganese dioxide nanoparticles for targeted delivery of emodin in breast cancer.

Emodin is a natural anthraquinone derivative with a variety of pharmacological effects such as anti-inflammatory and anti-cancer. However, its clinical application is hindered by poor water solubility and low bioavailability. To address these limitations, we developed carboxymethyl chitosan and poly(acrylic acid)/poly(allylamine hydrochloride)-modified hollow mesoporous manganese dioxide nanoparticles (CPPMNP) for emodin loading (ECPPMNP). The zeta potential of ECPPMNP was -26.4 ± 0.5 mV, the average particle size was (67.2 ± 4.7) nm, and the drug loading and encapsulation efficiency were (23.

Polygonum cuspidatum Extract (Pc-Ex) Containing Emodin Suppresses Lung Cancer-Induced Cachexia by Suppressing TCF4/TWIST1 Complex-Induced PTHrP Expression.

Cachexia, which is characterised by the wasting of fat and skeletal muscles, is the most common risk factor for increased mortality rates among patients with advanced lung cancer. PTHLH (parathyroid hormone-like hormone) is reported to be involved in the pathogenesis of cancer cachexia. However, the molecular mechanisms underlying the regulation of PTHLH expression and the inhibitors of PTHLH have not yet been identified. The PTHLH mRNA levels were measured using quantitative real-time polymerase chain reaction, while the PTHrP (parathyroid hormone-related protein) expression levels were measu

Emodin ameliorates glucose-induced morphologic abnormalities and synthesis of transforming growth factor beta1 and fibronectin by human peritoneal mesothelial cells.

OBJECTIVE: Excessive synthesis and deposition of matrix proteins by peritoneal mesothelial cells can lead to structural and functional changes in the peritoneal membrane, jeopardizing the long-term efficacy of peritoneal dialysis (PD). Prolonged exposure to high glucose concentrations in PD fluid has been implicated as a major stimulus to matrix accumulation, through the induction of transforming growth factor beta1 (TGFbeta1). This study investigated the effect of emodin (3-methyl-1,6,8-trihydroxyanthraquinone) on TGFbeta1 and fibronectin (FN) synthesis in human peritoneal mesothelial cells (
