The use of a perfluorochemical emulsion as a vascular perfusate in drug absorption.

In-vitro simultaneous luminal and vascular perfusion using the perfluorochemical emulsion, FC-43 emulsion, as a vascular perfusate, was examined for drug absorption in rat jejunum. The intestinal membrane in this system was found to retain its normal barrier functions for drug transport, as evident from the following: (i) stable absorption clearance of tritiated water and salicylic acid at steady-state, (ii) agreement of this clearance with that by in-situ single-pass luminal perfusion, (iii) active transport of D-glucose and its inhibition by phloridzin and (iv) normal glutamate pyruvate tran

Changes in human cultured cells exposed to a perfluorocarbon emulsion.

A perfluorocarbon emulsion (Fluosol-DA, 20%) produced persistent cytotoxic changes and growth inhibition in fibroblast-like human cells. After 18 hours of exposure to culture medium containing 4 percent of this perfluorochemical emulsion, normal embryonic lung fibroblasts (IMR 90 cells) and their SV40 virus-transformed counterparts (AG 2804 cells) ceased proliferation and showed degenerative changes, even if Fluosol was washed off the cell monolayer and replaced with normal medium. The morphological manifestations of Fluosol cytotoxicity included cytoplasmic vacuolation of varying but frequent

Fat emulsion as a vehicle for diazepam. A study of 9492 patients.

Conventional preparations of diazepam for i.v. use contain solvents which cause pain on injection and thrombophlebitis in a high percentage of cases. However, diazepam can be dissolved with advantage in the oleaginous phase of an oil-in-water emulsion (Diazemuls). Diazemuls has been given to 9492 patients without serious side-effects. Following i.v. injection, 2435 patients were studied with respect to pain and clinical effect. Only 0.4% experienced pain. The intended clinical effect was recorded in 99% of the patients. I.m. injection of Diazemuls resulted in a significantly smaller frequency

Design and application of emulsion-filled hydrogels for functional foods.

Emulsion-filled hydrogels (EFHs) have emerged as transformative soft-matter systems that bridge the gap between lipid-based emulsions and polymeric hydrogels, enabling multifunctional applications in modern food design. Hydrogel networks act as a protective layer for emulsions by imparting tenable texture, enhanced stability, and responsiveness to environmental or physiological stimuli. This review provides a comprehensive overview of recent advances in the design, fabrication, and application of EFHs, emphasizing how formulation parameters and interfacial engineering govern their rheological,

Lipid emulsion infusion and its effect on anaesthetic drugs.

Lipid emulsion has several established clinical uses, including parenteral nutrition, treatment of local anaesthetic systemic toxicity and as an energy source in metabolic crises, such as maple syrup urine disease. In toxicological settings, it acts via a 'lipid sink' mechanism, sequestering lipophilic drugs and reducing their bioavailability. This raises the possibility that the pharmacokinetics of commonly used anaesthetic agents may be affected in patients with a high intravascular lipid load. We report the peri-operative management of a 14-year-old child with maple syrup urine disease rece

Nano-emulsion encapsulation for the efficient delivery of bacteriophage therapeutics.

Antibiotic resistance has become the major concern for global public health. Phage therapy is being considered as an alternative for antibiotics to treat the multidrug resistant bacterial infections. Bacteriophage therapeutic developments has faced many challenges, including the drug formulations for sustainable phage delivery. The nano-emulsion platform has been described as the best approach to retain phage efficacy, shelf life and stability. Encapsulated phage drugs ensure stable delivery of phages to the target site and integrate in the system. In this review, our main focus is on the nano

Fluorescent and Iodized Emulsion for Preoperative Localization of Pulmonary Nodules.

OBJECTIVE: This study was conducted to develop a fluorescent iodized emulsion comprising indocyanine green (ICG) solution and lipiodol (ethiodized oil) and evaluate its feasibility for use in a clinical setting. BACKGROUND: ICG use for the preoperative localization of pulmonary nodules is limited in terms of penetration depth and diffusion. METHODS: First, fluorescent microscopy was used to investigate the distribution of ICG-lipiodol emulsions prepared using different methods. The emulsions were injected in 15 lung lobes of 3 rabbits under computed tomography fluoroscopy guidance; evaluatio

Intravenous Lipid Emulsion for the Treatment of Perioperative Cocaine Intoxication.

Symptomatic cocaine intoxication in the preoperative period is a potentially life-threatening condition, especially before emergent surgery. The anesthesiologist is faced with a dilemma where the patient is deemed unsafe for induction of general anesthesia but also in need of immediate surgical intervention. Cocaine is a local anesthetic and, as such, has been proposed to respond to lipid emulsion treatment as other local anesthetics would. We present a case supporting this statement and review the relevant published literature on the topic.

Emulsion Effect on the Measurement of Standard Laboratory Blood Parameters.

BACKGROUND: A number of pharmaceutical agents have limited water solubility and are therefore often prepared in a lipid emulsion. Emulsion renders plasma opaque and this could interfere with the accuracy of some standardized laboratory measurements, especially for optical or mechanical based assays. We determined the interference on some laboratory diagnostic values of blood specimens after propofol addition in vitro as well as in vivo when infused into swine. METHODS: In vitro, laboratory parameters were measured immediately after mixing swine blood diluted with increasing amounts of propofo

'Emulsion locks' for the containment of hydrocarbons during surfactant flushing.

Reversible double water in oil in water (W/O/W) emulsions were developed to contain subsurface hydrocarbon spills during their remediation using surfactant flushing. Double emulsions were prepared by emulsifying CaCl2 solutions in canola oil, and subsequently by emulsifying the W/O emulsions in aqueous sodium alginate solutions. The formation of double emulsions was confirmed with confocal and optical microscopy. The double emulsions reversed and gelled when mixed with the surfactants sodium dodecyl sulfate (SDS) and cocamidopropyl betaine (CPB). Gels can act as 'emulsion locks' to prevent spr

The Role of Intralipid Emulsion in the Rat Model of Digoxin Intoxication.

Although the mechanism of action is not well known, intravenous lipid emulsion (ILE) has been shown to be effective in the treatment of lipophilic drug intoxications. It is thought that, ILE probably separates the lipophilic drugs from target tissue by creating a lipid-rich compartment in the plasma. The second theory is that ILE provides energy to myocardium with high-dose free fatty acids activating the voltage-gated calcium channels in the myocytes. In this study, effects of ILE treatment on digoxin overdose were searched in an animal model in terms of cardiac side effects and survival. For

Successful intravenous lipid emulsion therapy: Olanzapine intoxication.

Olanzapine is an antipsychotic drug used in psychiatric diseases. At high doses it exhibits cardiovascular and neurological sideeffects in particular. Lipid emulsion therapy for the removal of medication from plasma in high-dose lipophilic drug use has recently become very widespread. In the light of current literature, this report discusses the successful treatment of a patient within 4 hrs of olanzapine overdose as an attempted suicide, who presented with agitation and clouded consciousness.

Emulsion Hydrogel Soft Motor Actuated by Thermal Stimulation.

An emulsion hydrogel motor (E-H motor), constituted by low-boiling-point oil fuel and a hydrogel matrix, is prepared through a simple yet versatile oil-in-water (O/W) emulsion template method. The E-H motor can be efficiently propelled by the bubbles generated under a thermal stimulus. As thermally induced explosion occurs inside the E-H motor (diameter ∼4.0 mm and length ∼6.0 mm), the gas bubbles resulting from thermotropic phase transition are violently ejected from one side, leading to a fast speed of 14.78 ± 4.82 mm s-1 in a 60 °C aqueous solution. Additionally, multiple water-insoluble or

Intravenous Lipid Emulsion Therapy for Bromethalin Toxicity in a Dog.

Bromethalin is a central nervous system toxin currently incorporated into several different rodenticides. In 2008, the EPA requested that manufacturers phase out second-generation anticoagulant rodenticides. In response, manufacturers began to increase production of bromethalin-based rodenticides. It is likely that pet exposure to bromethalin will increase in the future. Bromethalin has no known antidote and tends to deposit in fat. Intravenous lipid emulsions (ILEs) are being used with increasing frequency in both human and veterinary medicine to treat numerous acute systemic toxicities. A 4

Design and development of oral nanoparticulated insulin in multiple emulsion.

The present research aimed at developing an injection-free nanoparticulated formulation in multiple emulsion form, for oral delivery of insulin, which otherwise undergoes degradation in the gastric environment if administered orally. Insulin-polymeric nanoparticles were prepared using layer by layer (LbL) adsorption method and incorporated into an emulsion to form a nanoparticulated multiple emulsion. Using 0.6 M sodium chloride, the insulin nanoaggregates of 300-400 nm size were obtained about a yield of 94%. The characteristics of a representative nanoparticle were as follows: particle size

Investigation of different emulsion systems for dermal delivery of nicotinamide.

Nicotinamide (NA) has been shown to have beneficial effects on several skin diseases such as tumor, acne vulgaris, photodamage, cellulite and atopic dermatitis. The purpose of this study was to develop a multiple emulsion and a microemulsion formulation as delivery systems for NA. A two-step process was used to prepare the W/O/W multiple emulsion. Optimum microemulsion formulation was selected by using construction of pseudo-ternary phase diagram. The physicochemical properties such as droplet size and viscosity measurements, stability studies were also evaluated. Ex-vivo permeation studies we
