N6-methyladenosine (m6A) epitranscriptomic modifications play crucial roles in regulating both host and viral gene expression. Here, we revealed a novel mechanism by which paramyxoviruses exploit host m6A machinery to simultaneously enhance viral replication and suppress host immunity. Our results demonstrated that the viral matrix protein (M) of bovine parainfluenza virus type 3 (BPIV3) binds to the methyltransferase domain of METTL3 in the nucleus and facilitates its translocation to the cytoplasm through an exportin-1-dependent pathway. This mechanism is conserved across multiple paramyxovi

The growing and rapid development of high-throughput sequencing technologies have allowed a greater understanding of the mechanisms underlying gene expression regulation. Editing the epigenome and epitranscriptome directs the fate of the transcript influencing the functional outcome of each mRNA. In this context, non-coding RNAs play a decisive role in addressing the expression regulation at the gene and chromosomal levels. Long-noncoding RNAs, consisting of more than 200 nucleotides, have been shown to act as epigenetic regulators in several key molecular processes involving neurodegenerative

Small-Saunders et al. uncovered a new facet of artemisinin resistance in Plasmodium in which parasites use a previously underexplored arm of stress response mechanisms. Through altered epitranscriptomic modifications on tRNA, changed translation patterns adapt resistant cells to facilitate entry into a quiescent-like state which provides the parasite an escape from many drugs.

RNA N6-methyladenosine (m6A) is a key regulator of gene expression during early embryogenesis. Using SAC-seq (m6A-selective allyl chemical labeling and sequencing), an antibody-independent m6A profiling method, we generated the first single-nucleotide-resolution m6A map of bovine oocytes and preimplantation embryos. We observed both coordinated and uncoupled relationships between m6A modification and expression of protein-coding and noncoding genes. Integrative analysis of the transcriptome, m6A epitranscriptome, and translatome revealed dynamic m6A remodeling, particularly in ribosomal protei

Genome-wide approaches have unveiled multiple aspects of primary brain and spinal cord cancers regarding oncogenic processes, the tumor microenvironment, and mechanisms of resistance and sensitivity to therapy (among many others). In this review, we provide a brief overview of the possibilities that the current and highly diverse omics techniques offer for genomic landscape characterization, and how they can enhance our understanding of gliomas and glioneuronal tumors. More specifically, we discuss available methodologies for transcriptomics profiling (gene expression, alternative splicing, RN

Phytomelatonin has long been viewed as a pleiotropic stress signal that protects plants by scavenging reactive oxygen species (ROS), reinforcing antioxidant capacity, and coordinating hormonal responses. That view remains correct, but it is no longer sufficient. Recent work indicates that phytomelatonin also shapes the regulatory architecture that determines whether stress-responsive genes are accessible, whether transcripts are processed and translated efficiently, and whether stress-induced states can persist after the initial stimulus has disappeared. Here, we synthesize four emerging layer

Osteoporosis, characterized by imbalanced bone homeostasis, is driven by excessive osteoclast-mediated bone resorption, yet the epitranscriptomic regulation via m6A modification remains unclear. Here, we identify WTAP, a component of the m6A methyltransferase complex, as a critical negative regulator of osteoclastogenesis. Myeloid-specific Wtap knockout in mice exacerbates osteoclast formation and osteoporotic bone loss. Mechanistically, WTAP mediates m6A deposition on Csflr mRNA, promoting degradation via the key m6A reader YTHDF2 and downregulating CSF1R expression, thereby enhancing osteocl

BACKGROUND: RNA modifications, collectively known as the epitranscriptome, represent the third layer of gene regulation, influencing gene expression at transcriptional, post-transcriptional, and translational levels. RNA-modifying proteins (RMPs), including writers, erasers, and readers, are responsible for depositing, removing, and recognizing chemical modifications on RNA molecules. These modifications play a crucial role in linking molecular processes to cellular functions. Over the past few decades, a growing body of laboratory evidence, alongside advances in sequencing technologies, has u

Human ureaplasmas are minimal-genome bacteria and pathobionts of the urogenital tract. They must adapt to fluctuating pH conditions despite the absence of canonical transcriptional regulatory systems. However, the mechanisms underlying these responses remain unclear. This study aimed to construct a system-level model of pH adaptation in this minimal pathogen. We used an integrated multi-omics platform combining proteomics, metabolomics, and RNA modification profiling to construct a system-level model of pH adaptation. The results revealed a bifurcated strategy governed by the differential acti

N6-methyladenosine (m⁶A), the most abundant internal RNA modification in eukaryotes, plays a critical role in post-transcriptional regulation. METTL14, a core component of the methyltransferase complex, emerges as a key epigenetic regulator with diverse functions in physiology and disease. Accumulating evidence reveals that METTL14 governs pyroptosis, a pro-inflammatory form of programmed cell death, via m⁶A-dependent mechanisms across various diseases, including neurological, cardiovascular, skeletal, and metabolic disorders. Importantly, m⁶A modification is implicated in multiple forms of re

The decidua, a dynamic and heterogeneous maternal tissue essential for pregnancy maintenance, has emerged as a key contributor to preeclampsia (PE) pathogenesis. Using methylated RNA immunoprecipitation sequencing (MeRIP-seq) and RNA sequencing (RNA-seq), we profiled N6-methyladenosine (m6A) methylation patterns and mRNA expression in the decidua of early-onset PE (EPE), late-onset PE (LPE), and normal pregnancy (NP) samples. Integrated analysis revealed that differentially methylated genes (DMGs) and differentially expressed genes (DEGs) were significantly enriched in pathways critical for de

Glaucoma is a leading cause of irreversible blindness, and for patients unresponsive to medical therapy, glaucoma filtration surgery (GFS) is the primary intervention to lower intraocular pressure (IOP). However, postoperative scarring often leads to surgical failure. Here, we identify RNA demethylase ALKBH5 as a key regulator of fibrotic remodeling after GFS via an epitranscriptomic mechanism involving N6-methyladenosine (m6A) modification. In both human patients and rabbit models, increased vascular perfusion and decreased bleb height, measured by AS-OCTA and UBM, were associated with elevat

2'-O-methylation (Nm) is a key epitranscriptomic mark across diverse RNAs, yet its role in antiviral defense remains largely unexplored. To systematically investigate the landscape and functional significance of internal Nm modifications on enterovirus RNAs, a sensitive sequencing method termed RACE-Nm was established, in combination with the machine learning-based predictor NanoNm, to map Nm sites on low-abundance RNAs at single-nucleotide resolution. A conserved Nm site was identified at nucleotide 307C within the viral internal ribosome entry site (IRES), which was catalyzed by the methyltr

Bromodomain-containing proteins (BRDs) are involved in many biological processes, most notably epigenetic regulation of transcription, and BRD dysfunction has been linked to many diseases, including tumorigenesis. However, the role of BRDs in the pathogenesis of uterine fibroids (UFs) is entirely unknown. The present study aimed to determine the expression pattern of BRD9 in UFs and matched myometrium and further assess the impact of a BRD9 inhibitor on UF phenotype and epigenetic/epitranscriptomic changes. Our studies demonstrated that the levels of BRD9 were significantly upregulated in UFs

Mutations in splicing factors are commonly observed in chronic lymphocytic leukemia (CLL); however, other mechanisms can also contribute to the dysregulation of alternative splicing. One example is the overexpression of the m6A RNA methyltransferase METTL3, that by depositing the epitranscriptomic mark in spliceosome transcripts leads to aberrant splicing, but at the same time creates vulnerability to METTL3 inhibitors. See related article by Wu et al., p. 228 (8) .

Radiation-induced pulmonary fibrosis, a devastating thoracic radiotherapy sequela, features aberrant ECM deposition and irreversible loss of functional lung architecture. However, the exact molecular mechanisms governing AECII dysfunction and fate transition during RIPF remain poorly understood. Here, we identify a novel metabolic-epigenetic-epitranscriptomic cascade that orchestrates the pro-fibrotic fate of AECIIs. We demonstrate that radiation exposure triggers a glycolytic shift in AECIIs, resulting in pathological intracellular lactate accumulation. This metabolic stress is directly coupl

Sevoflurane-induced developmental neurotoxicity is characterized by impaired mitophagy and excessive ferroptosis. Neuroglobin (NGB) exerts broad neuroprotective effects, yet its exact epitranscriptomic regulatory mechanism against sevoflurane injury remains elusive. Two complementary experimental systems with identical total sevoflurane exposure length were adopted for in vitro mechanistic exploration and in vivo physiological verification, respectively. HT22 hippocampal neurons underwent uninterrupted 6 h sevoflurane incubation, while neonatal rats received 2 h intermittent inhalation each da
