Neurotoxic and cardiotoxic effects of cocaine and ethanol.

INTRODUCTION: Concurrent abuse of alcohol and cocaine results in the formation of cocaethylene, a powerful cocaine metabolite. Cocaethylene potentiates the direct cardiotoxic and indirect neurotoxic effects of cocaine or alcohol alone. CASE REPORT: A 44-year-old female with history of cocaine and alcohol abuse presented with massive stroke in the emergency department. CT scan revealed extensive left internal carotid artery dissection extending into the left middle and anterior cerebral arteries resulting in a massive left hemispheric infarct, requiring urgent decompressive craniectomy. The pa

Perceiving the effects of ethanol intoxication on voice.

Many conditions operate to degrade the quality of the human voice. Alcohol intoxication is one of them. In this project, the objectives were to examine the ability of human listeners to accurately estimate both the presence and severity of intoxication from two types of speech samples. A review of available data suggests that, although listeners can often identify individuals who are intoxicated simply by hearing samples of their voice, they are less efficient at accurately determining the severity of this condition. A number of aural-perceptual studies were carried out to test these relations

Anthocyanins: are they beneficial in treating ethanol neurotoxicity?

Heavy alcohol exposure produces profound damage to the developing central nervous system (CNS) as well as the adult brain. Children with fetal alcohol spectrum disorders (FASD) have a variety of cognitive, behavioral, and neurological impairments. FASD currently represents the leading cause of mental retardation. Excessive alcohol consumption is associated with Wernicke-Korsakoff syndrome (WKS) and neurodegeneration in the adult brain. Although the cellular/molecular mechanism underlying ethanol's neurotoxicity has not been fully understood, it is generally believed that oxidative stress plays

A fatal case of pure ethanol ingestion.

An adult male was found dead in a car with two empty bottles (500 ml x 2) labeled dehydrated ethanol (>99.5%, v/v). At autopsy, extensive pancreatic necrosis with severe hemorrhage was observed. High concentrations of ethanol were detected in blood (8.14 mg/ml), urine (8.12 mg/ml) and tissue specimens. The cause of death was determined to be an acute alcohol intoxication caused by ingesting approximately 1l dehydrated ethanol.

Mecamylamine and ethanol preference in healthy volunteers.

BACKGROUND: Recent evidence suggests that some of the behavioral effects of alcohol may be mediated through actions on nicotinic acetylcholine receptors. Mecamylamine, a nicotinic acetylcholine receptor antagonist, reduces alcohol preference and consumption in alcohol-preferring rats, and in humans, mecamylamine dampens some of the subjective, or mood-altering, effects of alcohol. This experiment was designed to investigate the effects of mecamylamine on consumption of alcohol in healthy social drinkers. METHODS: Healthy volunteers (12 men, 12 women) participated in a choice procedure in whic

An unusual form of fatal ethanol intoxication.

Forensic pathologists are very familiar with deaths due to ethanol intoxication. The overwhelming majority of these deaths are a result of the oral ingestion of ethanol. We report an unusual case of an individual who expired in his secured residence after self administration of a wine enema. Toxicology showed an ethanol concentration of 0.40 g/dL in the blood and 0.41 g/dL in the vitreous fluid. Scene investigation was of paramount importance in determining the unusual method by which the decedent absorbed the alcoholic beverage.

A novel ethanol-hypersensitive mutant of Arabidopsis.

A novel ethanol-hypersensitive mutant, geko1 (gek1), was isolated from Arabidopsis thaliana. The gek1 mutant displays an enhanced sensitivity (10-100 times greater than the wild type) to ethanol in growth medium, while it grows normally in the absence of ethanol, and responds normally to other alcohols and to environmental stresses such as heat shock and high salinity. The ethanol-hypersensitive phenotype of gek1 requires alcohol dehydrogenase activity, indicating that gek1 is sensitive not to ethanol itself but to the metabolites of ethanol. Consistent with this, gek1 shows enhanced sensitivi

Effects of ethanol on cocaine discrimination in rats.

Ethanol and cocaine are frequently abused in combination, but little is known about how the subjective effects of the two drugs interact. The ability of ethanol and other GABA(A)-active compounds to alter the discriminative stimulus effects of cocaine was tested. Male Sprague-Dawley rats were trained to discriminate cocaine (10 mg/kg ip) from saline using either single- or cumulative-dosing methods. In single-dose testing, ethanol (0.1-0.5 g/kg) dose-dependently decreased cocaine-appropriate responding following the training dose of cocaine. Ethanol (0.5 g/kg) produced a rightward shift in the

Developmental differences in the acquisition of tolerance to ethanol.

This study was designed to compare the development of tolerance to ethanol in adolescent and adult rats. Rats were pretreated with ethanol (4 g/kg) twice daily by intragastric gavage for 3 or 7 days, and then challenged with a single IP dose of ethanol (5 g/kg). Throughout the pretreatment period body temperature was measured before and after the morning dosage. During the IP challenge test we measured body temperature, duration of the loss of righting reflex, and blood ethanol level upon regaining the righting reflex. The adolescent rats that were pretreated for 7 days developed greater toler

Individual differences in the biphasic effects of ethanol.

Ethanol exerts both stimulant-like and sedative-like subjective and behavioral effects in humans depending on the dose, the time after ingestion and, we will argue, also on the individual taking the drug. This study assessed stimulant-like and sedative-like subjective and behavioral effects of ethanol during the ascending and descending limbs of the blood alcohol curve across a range of doses in nonproblem social drinkers. Forty-nine healthy men and women, 21 to 35 years old, consumed a beverage containing placebo or ethanol (0.2, 0.4, or 0.8 g/kg) on four separate laboratory sessions, in rand

Very unusual ethanol distribution in a fatality.

A 48-year-old man with an extensive history of alcoholism was found dead at home. He was lying face down on a carpet. There was evidence of gastric aspiration at autopsy and histologic examination. The distribution of ethanol was very unusual (concentrations in mg/100 mL or mg/100 g): femoral blood, 257 and 273 (two samples); heart blood, 643; vitreous humor, 763; urine, 84; bile, 616; liver, 250; and gastric, 4660 (2470 mg/53 g). In addition, this man ingested isopropanol, and, according to the history, may also have ingested acetone in the form of nail polish remover. The distribution of bot

Interaction of ethanol and the organophosphorus insecticide parathion.

Phosphorothioate insecticides such as parathion (O,O-diethyl-O-p-nitrophenyl phosphorothioate) undergo P450-dependent oxidative desulfuration, leading to both activation and detoxification of these compounds. Consequently, alterations in P450-dependent oxidative desulfuration may affect the acute toxicities of these insecticides. In the present study, pretreatment of mice with 15% ethanol in the drinking water for 6 days antagonized the acute toxicity of parathion, but not its toxic metabolite paraoxon (O,O-diethyl-O-p-nitrophenyl phosphate), suggesting that ethanol affected the oxidative desu

Death caused by an allergic reaction to ethanol.

Death as a result of anaphylaxis remains rare and is only occasionally related to ingestion of common substances. This article reports a death due to an allergic reaction to ethanol, as well as a review of the reported deaths in the literature attributed to ethanol. This report emphasizes the importance of awareness of such obscure allergies by forensic pathologists, the significance of antemortem testing, and the value of gathering accurate medical information, both antemortem and postmortem.

Influence of thiamine on the behavioral sensitivity to ethanol.

Changes in sensitivity to ethanol's rate-decreasing effects on operant performance were examined in control rats and cohorts that received diet-induced or diet+pyrithiamine-induced thiamine deficiency. Seven groups of male Sprague-Dawley rats (12 rats/group) were trained in a 5-cycle lever-press operant task under a fixed-ratio 30 schedule of food reinforcement. Once trained to maintain consistent operant performance across all 5 cycles, each rat was tested with various doses of ethanol injected at the beginning of each time-out cycle. Each group of rats demonstrated equivalent saline baseline

Effects of ethanol on cultured fetal astroglia.

This study investigated the effects of a 4-day ethanol exposure on cultured rhombencephalic astroglia. The contents of astroglial protein and DNA, and astroglial uptake of serotonin (5-HT) were determined. Fetal rhombencephalic astroglia were examined because of this laboratory's evidence that in utero ethanol exposure markedly impairs the development of serotonergic neurons, which are located in this fetal brain area, and because of the recently demonstrated importance of local support glia in neuronal development. The results of these experiments demonstrated that protein was significantly r

The pharmacodynamics of ethanol: effects on performance and judgment.

The objective and subjective effects produced by increasing and decreasing ethanol concentrations were studied in healthy volunteers on three separate occasions. A randomized, double-blind, placebo-controlled, four-way crossover study was used to determine whether there is any disparity between the time course of blood ethanol concentration and its effects on either objective test performance or self rated impairment. On each study day the subjects received one of four treatments consisting of either placebo or sufficient alcohol to achieve peak estimated blood alcohol concentration (Est.BAC)

Interactions of cocaine with barbital, pentobarbital and ethanol.

This study deals with the interactions of cocaine with barbital, pentobarbital and ethanol in nontolerant and tolerant male Sprague-Dawley rats. Cocaine hydrochloride (50 mg) pellets implanted s.c. in rats prior to the i.p. injections of sodium barbital (150 mg/kg dose once daily for 4 days) potentiated the hypothermic response 2 hr after the barbital injection, when maximum hypothermia occurred. The s.c. implantation of the same type of pellets prior to the i.p. injections of sodium pentobarbital (75 mg/kg dose once daily for 5 days) potentiated the pentobarbital hypnosis as measured by the d

Effects of ethanol on drug and metabolite pharmacokinetics.

Pharmacokinetic interactions of ethanol with other drugs, including its effects upon drug metabolite disposition, are reviewed in terms of clearance concepts. This approach is particularly useful in understanding the mechanisms of ethanol-drug interactions, i.e. in separating the effects of ethanol upon drug clearance, volume of distribution and plasma protein binding. The application of clearance concepts provides the basis for understanding the qualitative differences in ethanol interactions with low and high hepatic extraction ratio drugs. The effects of short and long term ethanol consumpt

Interactions of paraldehyde with ethanol and chloral hydrate.

The interaction of the depressant and toxic actions of paraldehyde with those of ethanol and chloral hydrate were investigated in two strains of male mice by determination of the ED50 for anesthetic action (righting reflex loss) and the 24 hr LD50, respectively. In all instances less than simple additive synergism was observed. Onset and duration times for the anesthetic actions of paraldehyde and ethanol were observed to be almost identical. Investigation of the time of maximum toxic effect revealed paraldehyde itself and mixtures containing 40% or more paraldehyde to be more acutely toxic th
