Investigations into the growth and formation of biofilm by Leptospira biflexa at temperatures encountered during infection.

The genus Leptospira comprises unique atypical spirochete bacteria that includes the etiological agent of leptospirosis, a globally important zoonosis. Biofilms are microecosystems composed of microorganisms embedded in a self-produced matrix that offers protection against hostile factors. Leptospires form biofilms in vitro, in situ in rice fields and unsanitary urban areas, and in vivo while colonizing rodent kidneys. The complex three-dimensional biofilm matrix includes secreted polymeric substances such as proteins, extracellular DNA (eDNA), and saccharides. The genus Leptospira comprises p

Phase-Transition-Induced Deformable OMV-Camouflaged Nanoparticles Enabling Deep Biofilm Penetration for Photothermal-Photodynamic Eradication of Bacterial Pneumonia.

Bacterial pneumonia remains refractory to conventional therapies due to the dense extracellular polymeric substance matrix of biofilms that severely restricts drug penetration. Here, we report a biomimetic, thermo-responsive nanoplatform (AIE/PCM@OMV) featuring phase-transition-induced deformability to overcome biofilm barriers and treat bacterial pneumonia. The nanoparticles comprise a phase-change material (PCM) core encapsulating an aggregation-induced emission (AIE) phototherapeutic agent and are camouflaged with bacterial outer membrane vesicles (OMVs). The OMV coating enables homologous

Surfactant-Engineered Niosomal Antibiotic Systems for Biofilm-Associated Infections: Design Principles and Translational Perspectives.

Persistent microbial biofilm infections remain a major obstacle to effective antimicrobial therapy due to restricted drug diffusion, metabolic heterogeneity, and the presence of tolerant bacterial subpopulations. In device-associated infections, biofilms substantially reduce antibiotic efficacy and contribute to chronic relapse despite adequate systemic exposure. Although nanocarrier-based delivery systems have been widely investigated, many formulations remain empirically developed with insufficient consideration of biofilm-specific physicochemical and biological barriers. This review examine

Nanocarrier-Integrated Topical Dosage Forms for Biofilm-Infected Wounds: Balancing Wound Retention, Biofilm Matrix Penetration, and Translational Feasibility.

Biofilm-infected wounds are difficult to treat because extracellular polymeric substance (EPS) matrices, wound exudate, polymicrobial spatial organization, impaired antimicrobial penetration, persistent inflammation, and antimicrobial tolerance limit the efficacy of conventional topical therapy. Nanocarriers can protect antimicrobial payloads, enable controlled and stimuli-responsive release, and improve interactions with biofilm matrices; however, their clinical performance also depends on the final topical dosage form that governs wound residence, local exposure, usability, and translation.

Environmental Adaptation in Nostoc sphaeroides: Spherical Multicellularity, Molecular Signalling and Implications for Sustainable Cultivation.

Nostoc sphaeroides is a diazotrophic cyanobacterium that forms macroscopic spherical colonies in seasonally flooded paddy fields. Studies of its environmental physiology, colony development and cultivation have largely proceeded independently. This review integrates strain-resolved genomic, transcriptomic, physiological and ecological evidence to examine spherical multicellularity as a possible adaptation to a variable habitat. Developmental transitions between motile hormogonia and vegetative, heterocyst-forming filaments link dispersal with establishment, while an extracellular polymeric sub

A Targeted Mitigation Strategy for Freshwater Harmful Algal Blooms through the Release of a Small-Molecule Polyphenolic Algaecide from Clay Nanotubes.

Harmful algal blooms (HABs) caused by Microcystis aeruginosa (MC) pose serious threats to freshwater ecosystems, public health, and water treatment infrastructure. Here, we present a targeted mitigation technology involving flocculation and settling of MC using a flocculant, polyaluminum chloride (PAC), and a nanotubular clay, halloysite, which serves both as a ballast to aid settling of the flocs and as a reservoir to deliver algaecides. The tubular morphology of halloysite allows loading of algaecide, thus constituting a targeted release of algaecide to MC cells in the floc. The specific alg

Star-shaped PCL nanoparticles combining quercetin release and α-amylase surface functionalization enable AFM-quantified removal of biofilms of clinical Staphylococcus aureus isolates.

Healthcare-associated infections (HAIs) driven by multi-drug-resistant biofilms remain a critical challenge in clinical settings. The emergence of antibiotic-resistant strains has intensified the need for innovative antimicrobial strategies, particularly for effective surface treatments in clinical environments. This study presents a novel bifunctional nanoplatform designed to eradicate Staphylococcus aureus biofilms through a dual-action mechanism. We synthesized a custom star-shaped polycaprolactone (PCL-COOH) architecture to serve as a robust nanocarrier, enabling high quercetin loading (PC

Elucidating the anti-biofilm and anti-quorum sensing potential of selenocystine against respiratory tract infections causing bacteria: in vitro and in silico studies.

Bacteria are increasingly relying on biofilms to develop resistance to antibiotics thereby resulting in their failure in treating many infections. In spite of continuous research on many synthetic and natural compounds, ideal anti-biofilm molecule is still not found thereby warranting search for new class of molecules. The current study focuses on exploring anti-biofilm potential of selenocystine against respiratory tract infection (RTI)-causing bacteria. Anti-bacterial and anti-biofilm assays demonstrated that selenocystine inhibits the growth of bacteria in their planktonic state, and format

Infrared spectroscopy with multivariate analysis to interrogate the interaction of whole cells and secreted soluble exopolimeric substances of Pseudomonas veronii 2E with Cd(II), Cu(II) and Zn(II).

Extracellular polymeric substances (EPS) are bacterial products associated to cell wall or secreted to the liquid media that form the framework of microbial mats. These EPS contain functional groups as carboxyl, amino, hydroxyl, phosphate and sulfhydryl, able to interact with cations. Thus, EPS may be considered natural detoxifying compounds of metal polluted waters and wastewaters. In this work Attenuated Total Reflectance-Fourier Transform Infrared spectroscopy (ATR-FTIR) in combination with multivariate analysis (Principal Component Analysis-PCA-) were used to study the interaction of Cd(II

Mechanisms of sludge reduction in Nostocoides-driven enhanced biological phosphorus removal process.

Nostocoides, a key phosphate-accumulating organism (PAOs), is abundant in wastewater treatment plants (WWTPs); however, the mechanism of sludge reduction in biological phosphorus removal systems under its dominance remains largely unknown. This study employed a lab-scale, exploratory multiparameter approach to examine extracellular polymeric substance (EPS) profiles, intracellular metabolites, microbial community, membrane integrity, and physiological activity to elucidate how Nostocoides-enriched enhanced biological phosphorus removal (EBPR) configurations accomplished sludge reduction. Waste

Low Concentrations of Chlorhexidine Inhibit the Formation and Structural Integrity of Enzyme-Treated Multispecies Oral Biofilms.

The self-produced matrix of biofilms, consisting of extracellular polymeric substances, plays an important role in biofilm adhesion to surfaces and the structural integrity of biofilms. In dentistry, biofilms cause multiple diseases such as caries, periodontitis, and pulpitis. Disruption of these biofilms adhering to dental hard tissues may pose a major challenge since biofilms show higher tolerance to antimicrobials and antibiotics than planktonic cells. In this study, the effect of low concentrations of chlorhexidine (CHX) on enzyme-treated multispecies oral biofilm was investigated in an in

c-Type cytochrome-dependent formation of U(IV) nanoparticles by Shewanella oneidensis.

Modern approaches for bioremediation of radionuclide contaminated environments are based on the ability of microorganisms to effectively catalyze changes in the oxidation states of metals that in turn influence their solubility. Although microbial metal reduction has been identified as an effective means for immobilizing highly-soluble uranium(VI) complexes in situ, the biomolecular mechanisms of U(VI) reduction are not well understood. Here, we show that c-type cytochromes of a dissimilatory metal-reducing bacterium, Shewanella oneidensis MR-1, are essential for the reduction of U(VI) and for

Pseudomonas aeruginosa Biofilms in Cystic Fibrosis: Interactions, Methods, and Therapeutic Strategies.

This review explores the role of Pseudomonas aeruginosa biofilms in cystic fibrosis (CF) pathogenesis. Biofilms, the main bacterial lifestyle in CF lungs, are key in therapy failure, immune evasion, and chronic infection persistence. This review examines biofilm structure, emphasizing extracellular polymeric substances (Psl, Pel, alginate, eDNA) and their roles in structural stability, resistance to antibiotics, and immune modulation. Regulatory mechanisms, including c-di-GMP signaling and quorum-sensing systems, are detailed as key drivers of biofilm formation and maintenance. The review also

Effect of sonication on staphylococcal and gram-negative biofilms relevant to prosthetic joint infections: an in vitro study.

BACKGROUND: Periprosthetic joint infections (PJIs) are severe complications of arthroplasty in which biofilm formation on implant surfaces compromises microbiological diagnosis and antimicrobial efficacy. Although staphylococci remain the predominant pathogens, Gram-negative bacilli have increasingly been associated with diagnostic failure and unfavorable clinical outcomes. This study aimed to evaluate the formation, maturation, and structural organization of Gram-positive and Gram-negative bacterial biofilms and to investigate the effects of a standardized sonication protocol on biofilm disru

Enhanced Clearing of Wound-Related Pathogenic Bacterial Biofilms Using Protease-Functionalized Antibiotic Nanocarriers.

Biofilms are prevalent in chronic wounds and once formed are very hard to remove, which is associated with poor outcomes and high mortality rates. Biofilms are comprised of surface-attached bacteria embedded in an extracellular polymeric substance (EPS) matrix, which confers increased antibiotic resistance and host immune evasion. Therefore, disruption of this matrix is essential to tackle the biofilm-embedded bacteria. Here, we propose a novel nanotechnology to do this, based on protease-functionalized nanogel carriers of antibiotics. Such active antibiotic nanocarriers, surface coated with t

Disrupting Mycobacterium smegmatis biofilm using enzyme-immobilized rifampicin loaded silk fibroin nanoparticles for dual anti-bacterial and anti-biofilm action.

Biofilm formation is a major challenge in the treatment of tuberculosis, leading to poor treatment outcomes and latent infections. The complex and dense extracellular polymeric substances (EPS) of the biofilm provides safe harbour for bacterium enabling persistence against anti-TB antibiotics. In this study, we demonstrated that rifampicin-encapsulated silk fibroin nanoparticles immobilized with antibiofilm enzymes can disrupt the Mycobacterium smegmatis biofilm and facilitate the anti-bacterial action of Rifampicin (RIF). The EPS of M.smegmatis biofilm predominantly comprised of lipids (48.8

Catalase Activity is Critical for Proteus mirabilis Biofilm Development, Extracellular Polymeric Substance Composition, and Dissemination during Catheter-Associated Urinary Tract Infection.

Proteus mirabilis is a leading uropathogen of catheter-associated urinary tract infections (CAUTIs), which are among the most common health care-associated infections worldwide. A key factor that contributes to P. mirabilis pathogenesis and persistence during CAUTI is the formation of catheter biofilms, which provide increased resistance to antibiotic treatment and host defense mechanisms. Another factor that is important for bacterial persistence during CAUTI is the ability to resist reactive oxygen species (ROS), such as through the action of the catalase enzyme. Potent catalase activity is

Role of exopolysaccharide secreted by Chlorella vulgaris SSAU 8 and Desertifilum salkalinema SSAU 7 in fertility enhancement of saline soil.

Soil salinization has emerged as a major constraint to global agricultural productivity, severely disrupting soil structure, nutrient cycling, and plant establishment. This study evaluates the functional potential of the cyanobacterium Desertifilum salkalinema SSAU-7 and the microalga Chlorella vulgaris SSAU-8 as bio-ameliorants for saline soils. Both isolates demonstrated high salinity tolerance and exhibited key plant growth-promoting traits, including indole-3-acetic acid (IAA) and hydrogen cyanide (HCN) production. Under escalating salt concentrations, D. salkalinema SSAU-7 and the mixed c
