First case of a reversed Parabothus taiwanensis Amaoka & Shen, 1993 from Taiwan (Pleuronectiformes: Bothidae), with first evidence of situs inversus viscerum in bothid.

BACKGROUND: Reversed condition is rarely found in most flatfishes in natural environment, except for some certain species. The mechanism controlling the reversals in flatfishes has been studied in some cultivated species, whereas some have only few cases for the entire family and remain unclear. Here, we report the first record of a dextral (reversed) specimen of Parabothus taiwanensis Amaoka & Shen, 1993 collected off southwestern Taiwan recently. It represents the second reversed case ever recorded in Bothidae. We aim to provide a detailed description of this dextral specimen and compared to

Comparative Pharmacological Assessment of Amoxicillin in Five Cultured Fish Species: Implications for Off-Label Use in Aquaculture.

Background: Amoxicillin (AMOX) is widely used in aquaculture for bacterial infections due to its efficacy and safety. Despite official approval for select species, off-label use is common. This study evaluated the antibacterial efficacy and residue depletion of AMOX in five aquaculture species: olive flounder (Paralichthys olivaceus), rainbow trout (Oncorhynchus mykiss), Japanese eel (Anguilla japonica), black rockfish (Sebastes schlegelii), and Israeli carp (Cyprinus carpio). Methods: Fish were administered AMOX orally at 40 mg/kg and 80 mg/kg for seven days. Antibacterial efficacy was assess

Vaccine efficacy of Mycobacterium bovis BCG against Mycobacterium sp. infection in amberjack Seriola dumerili.

Mycobacteriosis, caused by the intracellular parasitism Mycobacterium sp., causes economic damages to aquaculture production in Japan, particularly in seriola fish production. Antibiotics are not effective against Mycobacterium sp. and so a potent vaccine is needed. We previously reported that BCG vaccine (Mycobacterium bovis BCG) induces adaptive immunity against Mycobacterium sp. in Japanese flounder, Paralichthys olivaceus. In a phylogenetic tree, the genes for a major antigen, the Ag85 complex, in Mycobacterium sp. TUMSAT-Msp001 are closely related to homologues in Mycobacterium ulcerans.

Molecular detection of myxosporean parasites in human fecal samples associated with foodborne illness.

In 2011, Kudoa septempunctata (Myxozoa: Myxosporea: Multivalvulida) was recognized as a causative agent of foodborne illness in Japan. Since then, studies have conducted surveillance for this parasite in flounder, a primary vehicle for foodborne outbreaks, and have reported the detection of K. septempunctata in fecal samples from patients. Although sporadic outbreaks suspected to be associated with the consumption of fish other than flounder have also been reported, information on examination of patient feces or leftover fish in such outbreaks remains limited. In this study, we investigated th

A portrait of 2 nematodes in liver of their paratenic fish hosts, illustrating different immunological approaches.

Comparative histopathological and ultrastructural investigations were performed on the livers of 2 fish species, namely, flounder (Platichthys flesus (L.)) naturally infected with the nematode Anisakis simplex (s.l.) (Rudolphi, 1809) larvae (L3) and tuvira (Gymnotus inaequilabiatus) (Valenciennes, 1839) harbouring the nematode Brevimulticaecum sp. (L3) (Shikhobalova and Mozgovoi, 1952). The intensity of infection by A. simplex (s.l.) larvae (L3) in flounders ranged from 3 to 10 parasites per organ. The worms were encapsulated by the peritoneal visceral serosa on the external surface of the liv

Will It Be Beneficial To Simulate the Antifreeze Proteins at Ice Freezing Condition or at Lower Temperature?

Antifreeze proteins (AFPs) enable the polar living species to survive subzero temperature conditions through effective lowering of the freezing point of body fluids. At the molecular level, AFPs directly interact with the growing seeds of ice crystals to inhibit their formation. To understand the structural and dynamic aspects of this interaction at the atomistic level, molecular dynamics (MD) simulations were carried out on several type I AFPs at multiple temperatures, including the physiologically relevant temperature of 273 K, a lower temperature of 227 K, and the conventional 300 K. A comp

Genome-wide DNA methylation mediates the resistance to vibriosis in Cynoglossus semilaevis.

Chinese tongue sole (Cynoglossus semilaevis) is an economically important marine fish in China. However, vibriosis has caused huge mortality and economic losses in its culturing industry. To reveal the effect of DNA methylation on the resistance to vibriosis in tongue sole, we conducted RNA sequencing and whole genome bisulfite sequencing (WGBS), and compared the gene expressions and DNA methylation patterns between the resistant and susceptible families. We identified a total of 741 significantly differentially expressed genes (DEGs) in kidney and 17460 differentially methylated genes (DMGs),

Exosomal miRNAs in the plasma of Cynoglossus semilaevis infected with Vibrio harveyi: Pleiotropic regulators and potential biomarkers involved in inflammatory and immune responses.

Exosomes are a class of extracellular vesicles with diameters ranging from ~50 to 150 nm. Incorporating diverse biological substances and being present extensively in biofluids, exosomes are involved in intracellular communication in various physiological and pathological processes and emerging as promising biomarkers for the prognosis and diagnosis of many diseases. Accumulating evidence shows that exosomes also play important roles in the inflammatory and immune responses to bacterial infection. However, the study of exosomes in teleost fish remains scarce. In the present study, we focused o

Removal of two pathogenic scuticociliates Miamiensis avidus and Miamiensis sp. using cells or culture filtrates of the dinoflagellate Alexandrium andersonii.

Scuticociliatosis, which is caused by parasitic protistan pathogens known as scuticociliates, is one of the most serious diseases in marine aquaculture worldwide. Thus, elimination of these ciliates is a primary concern for scientists and managers in the aquaculture industry. To date, formalin and other toxic chemicals have been used as anti-scuticociliate agents, but issues regarding their secondary effects often arise. Consequently, development of safer methods is necessary. To find out a safe method of controlling scuticociliate populations in aqua-tanks or small-scale natural environments,

Stable In Vitro Cultivation of the Myxozoan Enteromyxum leei for over 12 Months: Integration of a Physical Aggregation Technique for Long-Term Antiparasitic Interventions.

Emaciation disease, primarily caused by the myxosporean parasite Enteromyxum leei, poses a significant economic threat to the olive flounder (Paralichthys olivaceus) aquaculture industry. Research on E. leei has been severely restricted by its status as an unculturable obligate parasite and its extreme sensitivity to cryopreservation. In this study, we developed a standardized in vitro culture system that achieved stable maintenance of E. leei for over 12 months for the first time. To overcome the challenges of primary isolation, a physical aggregation technique was implemented, which effectiv

Anti-Protozoan Activities of Polar Fish-Derived Polyalanine Synthetic Peptides.

Chagas disease, sleeping sickness and malaria are infectious diseases caused by protozoan parasites that kill millions of people worldwide. Here, we performed in vitro assays of Pa-MAP, Pa-MAP1.9, and Pa-MAP2 synthetic polyalanine peptides derived from the polar fish Pleuronectes americanus toward Trypanosoma cruzi, T. brucei gambiense and Plasmodium falciparum activities. We demonstrated that the peptides Pa-MAP1.9 and Pa-MAP2 were effective to inhibit T. brucei growth. In addition, structural analyses using molecular dynamics (MD) studies showed that Pa-MAP2 penetrates deeper into the membra

Generation of VHSV replicon particles carrying transmembrane and C-terminal cytoplasmic region-deleted G gene (rVHSV-GΔTM) and comparison of vaccine efficacy with G gene-deleted VHSV (rVHSV-ΔG).

Vaccines based on viral replicon particles would be advantageous to induce immune responses compared to inactivated viruses in that they can infect host cells (only once) and can produce viral proteins in the infected cells like live viruses. Furthermore, as viral replicon particles are replication-defective, they are safer than live attenuated viruses. Previously, we had rescued viral hemorrhagic septicemia virus (VHSV) replicon particles lacking full ORF of G gene (rVHSV-ΔG). In the present study, to enhance the immunogenicity of VHSV replicon particles, we newly generated another form of VH

[Bacterial infections, what we can learn from each outbreak].

Studies on outbreaks or incidences of infectious diseases and food poisonings are the starting points in research. Analyses of the outbreaks will provide the mechanism by which the illnesses occur and the establishment of countermeasure. We report here some sensational outbreaks which recently occurred in Japan: 1) enterohemorrhagic Escherichia coli (EHEC) O157 outbreaks by pickled Chinese cabbage in Hokkaido, 2) EHEC O157/O111 outbreaks by raw beef in Toyama Prefecture, 3) parasitic food poisoning due to raw olive flounder consumption in the western Japan, and 4) botulism due to the consumpti

The corpuscles of Stannius, calcium-sensing receptor, and stanniocalcin: responses to calcimimetics and physiological challenges.

This study has examined whether the calcium-sensing receptor (CaSR) plays a role in control of stanniocalcin-1 (STC-1), the dominant calcium regulatory hormone of fish, comparable with that demonstrated for CaSR in the mediation of ionized calcium regulation of PTH secretion in mammals. In a previous study, we have cloned flounder STC-1 from the corpuscles of Stannius (CS). Here, we report the cloning and characterization of the CS CaSR, and the in vivo responses of this system to altered salinity, EGTA induced hypocalcemia, and calcimimetic administration. Aquatic models for the study of renal transport function and pollutant toxicity.

Studies of renal cell transport mechanisms and their impairment by xenobiotics are often limited by technical difficulties related to renal tubule complexity. Problems include the juxtaposition of multiple tubule segments with different transport functions and severely limited access to the tubular lumen. Some limitations can be overcome by the careful selection of an appropriate aquatic experimental system. Two aquatic models for the vertebrate proximal segment are discussed here. The first is the kidney from certain marine flounder, which offers the following advantages: long-term viability,

Disruption of diatom attachment on marine bioinspired antifouling materials based on Brill (Scophthalmus rhombus).

Bioinspired surfaces, due to their nano and micro topographical features, offer a promising approach for the development of novel antifouling solutions. The study of surface topography has gained popularity in recent years, demonstrating significant potential in mimicking natural structures that could be manufactured for application in the marine environment. This research focuses on investigating the antifouling (AF) performance of bio-inspired micro-textures inspired by Brill fish scales, Scophthalmus rhombus, under static laboratory conditions, using two common fouling diatom species, Ampho

Peritoneal exosomes mediate activation of CD4+ T cells and enhancement of vaccine efficacy in teleost.

BACKGROUND: Exosomes have emerged as promising vaccine candidates due to their role in intercellular communication and bioactive cargo-carrying capacity. However, their intrinsic properties in activating adaptive immunity have not yet been fully addressed. This study aims to elucidate how teleost peritoneal cell-derived exosomes (PDEs) elicit CD4+ T cell activation and contribute to vaccine-mediated adaptive immunity. METHODS: PDEs were isolated from flounder immunized with inactivated Vibrio anguillarum (ExoFKC) or non-immunized flounder (ExoCon) via differential centrifugation. Tissue and ce
