Based on studies on 610 cases of hereditary red cell membrane disorders, the characteristic features of the incidence of these disorders in the Japanese population are described. These patients were screened by a protocol on red cell morphology (scanning electron microscopy), on red cell membrane proteins (sodium dodecylsulfate polyacrylamide gel electrophoresis, and kinetics of membrane proteins), biophysical studies (ektacytometry, mechanical stability and fluorescence recovery after the photobleaching method), membrane transport (sodium influx and efflux, and anion transport), gene analysis

The anthrax toxin is a tripartite toxin, where the two enzymatic subunits require the third subunit, the protective antigen (PA), to interact with cells and be escorted to their cytoplasmic targets. PA binds to cells via one of two receptors, TEM8 and CMG2. Interestingly, the toxin times and triggers its own endocytosis, in particular through the heptamerization of PA. Here we show that PA triggers the ubiquitination of its receptors in a beta-arrestin-dependent manner and that this step is required for clathrin-mediated endocytosis. In addition, we find that endocytosis is dependent on the he

Human cytomegalovirus (HCMV) encodes an endoplasmic reticulum (ER)-resident glycoprotein, UL148, which activates the unfolded protein response (UPR) but is fully dispensable for viral replication in cultured cells. Hence, its previously ascribed roles in immune evasion and modulation of viral cell tropism are hypothesized to cause ER stress. Here, we show that UL148 is necessary and sufficient to drive the formation of prominent ER-derived structures that on average occupy 5% of the infected cell cytoplasm. The structures are sites where UL148 coalesces with cellular proteins involved in ER qu

Medullary cystic kidney disease/familial juvenile hyperuricemic nephropathy (MCKD/FJHN) are autosomal dominant renal disorders characterized by tubulo-interstitial fibrosis, hyperuricemia and medullary cysts. They are caused by mutations in the gene encoding uromodulin, the most abundant protein in urine. Uromodulin (or Tamm-Horsfall protein) is a glycoprotein that is exclusively expressed by epithelial tubular cells of the thick ascending limb of Henle's loop and distal convoluted tubule. To date, 37 different uromodulin mutations have been described in patients with MCKD/FJHN. Interestingly,

β-adrenergic receptor (β-AR) signaling in cardiac myocytes is central to cardiac function, but spatiotemporal activation within myocytes is unresolved. In rabbit ventricular myocytes, β-AR agonists or high extracellular [Ca] were applied locally at one end, to measure β-AR signal propagation as Ca-transient (CaT) amplitude and sarcoplasmic reticulum (SR) Ca uptake. High local [Ca]o, increased CaT amplitude under the pipette faster than did ISO, but was also more spatially restricted. Local isoproterenol (ISO) or norepinephrine (NE) increased CaT amplitude and SR Ca uptake, that spread along th

Polyethylene glycol (PEG) is widely recognized as a versatile model system for investigating protein-polymer interactions owing to its tunable physicochemical properties and hence has also been commonly used as macromolecular crowding agents. PEGs exist spanning a range of molecular weights, thereby exhibiting a diverse set of interactions with biomolecules. However, a systematic comparison between the different PEGs with regard to their properties as macromolecular crowders is hard to come by. The present work adopts a comprehensive approach in which both the behavior of a protein and the int

Patients with peritoneal metastasis (PM) of colorectal cancer (CRC) have poorer overall survival outcomes than those without PM. Cancer-associated fibroblasts (CAFs) are a major component of the tumor microenvironment and mediate CRC progression and PM. It is imperative to identify and develop novel therapeutic targets for PM-CRC driven by CAFs. Using lipidomics, we reveal that the abundance of phosphatidylcholine (PC) with unsaturated acyl chains was increased in clinical PM-CRC specimens. Additionally, we found that CAFs were present at a higher relative abundance in primary PM-CRC tumors an

BACKGROUND/AIM: WD40 repeats are found in many ciliary proteins. Although the WD40 repeat-containing WD repeat-containing protein 31 (WDR-31) is known to regulate ciliary protein trafficking and morphology, the specific contributions of its N-terminal, WD40, and C-terminal domains to protein localization and ciliary gate integrity remain unclear. Aim of this study is to dissect the functional roles of WDR-31 domains by investigating their contributions to ciliary localization, ciliary protein trafficking, and gate integrity.

BACKGROUND: Mitochondrial dysfunction and defective mitophagy are closely associated with cancer progression. Forkhead box O3 (FOXO3), a key transcription factor, promotes autophagy by activating autophagy-related genes. Polyphyllin VI (PPVI), a Paris polyphylla-derived steroidal saponin, displays potent antitumor activity.
PURPOSE: To investigate the antitumor effect of PPVI in breast cancer (BC) and elucidate its underlying mechanism.
METHODS: Differential gene expression and pathway enrichment were analyzed using RNA sequencing. Mitochondrial autophagic flux was assessed using the mKeima-

BACKGROUND: Liquid-liquid phase separation (LLPS) has emerged as a critical mechanism underlying oncogenic signaling and transcriptional regulation; however, its involvement in triple-negative breast cancer (TNBC) remains largely unknown. The pleckstrin homology-like domain family B member 2 (PHLDB2) has been implicated in cytoskeletal organization and cell migration, but its role in phase separation-mediated tumor progression has not been explored.
METHODS: We conducted an integrative multi-omics analysis combining transcriptomic profiling, bioinformatic modeling, and molecular biology exper

Nitric oxide (NO) is a key factor in inflammation. Endothelial nitric oxide synthase (eNOS), whose activity increases after stimulation with proinflammatory cytokines, produces NO in endothelium. NO activates two pathways: 1) soluble guanylate cyclase-protein kinase G and 2) S-nitrosylation (NO-induced modification of free-thiol cysteines in proteins). S-nitrosylation affects phosphorylation, localization, and protein interactions. NO is classically described as a negative regulator of leukocyte adhesion to endothelial cells. However, agonists activating NO production induce a fast leukocyte a

Recently, mitophagy-mediated bone mineralization of mesenchymal stem cells has emerged as another bone formation pattern, but whether mitophagy-mediated bone mineralization shapes craniofacial development remains unknown. Here, we demonstrate that loss of OPTN, a keystone macroautophagy/autophagy receptor, impairs mitophagy and acidic calcium phosphate (ACP) transport in orofacial bone mesenchymal stem cells (OMSCs), leading to craniofacial bone mineralization defects. We substantiate that OPTN undergoes LLPS both in vitro and in vivo, driven by S173 phosphorylation within its intrinsically di

AIM: Pulpitis is a common and painful disease characterised by complex and dynamic immune responses and may lead to irreversible tissue damage or tooth loss. Macrophages act as key sentinels that exhibit differential metabolic reprogramming, tightly orchestrating the immune microenvironment. However, the specific metabolic mechanisms governing macrophage-driven inflammatory activation in pulpitis remain unclear. Here, we reveal a specific lipid metabolism-driven immune pathway within macrophages in pulpitis.
METHODOLOGY: An integrative analysis of single-cell RNA sequencing (scRNA-seq), lipid

LIM kinases (LIMKs) are mainly in the cytoplasm and regulate actin dynamics through cofilin phosphorylation. Recently, it has been reported that nuclear localization of LIMKs can mediate suppression of cyclin D1 expression. Using immunofluorescence monitoring of enhanced green fluorescent protein-tagged LIMK2 in combination with photobleaching techniques and leptomycin B treatment, we demonstrate that LIMK2 shuttles between the cytoplasm and the nucleus in endothelial cells. Sequence analysis predicted two PKC phosphorylation sites in LIMK2 but not in LIMK1. One site at Ser-283 is present betw

Most epithelial cells secrete a glycoprotein-rich apical extracellular matrix that can have diverse but still poorly understood roles in development and physiology. Zona Pellucida (ZP) domain glycoproteins are common constituents of these matrices, and their loss in humans is associated with a number of diseases. Understanding of the functions, organization and regulation of apical matrices has been hampered by difficulties in imaging them both in vivo and ex vivo. We identified the PAN-Apple, mucin and ZP domain glycoprotein LET-653 as an early and transient apical matrix component that shape

Lipid droplets (LDs) are cytoplasmic organelles that store neutral lipids for use as an energy supply in times of nutrient deprivation and for membrane assembly. Misregulation of LD function leads to many human diseases, including lipodystrophy, obesity and neutral lipid storage disorders. A number of proteins have been shown to localize to the surface of lipid droplets, including lipases such as adipose triglyceride lipase (ATGL) and the PAT-domain proteins ADRP (adipophilin) and TIP47, but the mechanism by which they are targeted to LDs is not known. Here we demonstrate that ATGL and ADRP, b

BACKGROUND: The perivascular conduct pathway that connects the cerebrospinal fluid spaces with the interstitial fluid in the parenchyma are of importance for solute clearance from the brain. In this pathway, the relatively wide perivascular space (PVS) surrounding the pial arteries provides a low-resistant passage while around the perforating arteries, the solute movement is along the basement membrane (BM), that prevents the free exchange of interstitial fluids and solutes. We hypothesize that this selectivity involves specific components of the vascular BM, which is mainly composed of type I

The paranodal junctions flank mature nodes of Ranvier and provide a barrier between ion channels at the nodes and juxtaparanodes. These junctions also promote node assembly and maintenance by mechanisms that are poorly understood. Here, we examine their role in the accumulation of NF186, a key adhesion molecule of PNS and CNS nodes. We previously showed that NF186 is initially targeted/accumulates via its ectodomain to forming PNS (hemi)nodes by diffusion trapping, whereas it is later targeted to mature nodes by a transport-dependent mechanism mediated by its cytoplasmic segment. To address th

The characterization of diffusion through biological tissues has played an important role in fundamental medical research and product development. Understanding the diffusion phenomena allows for the identification of new concepts in fundamental science, evolving medical knowledge and improving future standards and protocols. To illustrate, the structure of cortical bone changes upon the onset of osteoporosis, altering the limited porous compartment through which nutrients and essential signaling molecules travel to bone cells. Estrogen hormone replacement therapy (HRT) is one of the gold stan

Equid alphaherpesvirus 1 (EHV1) is a DNA virus that causes severe disease in equids. Some strains are neurotropic and cause disease in the central nervous system, whereas others are non-neurotropic and can cause negative reproductive outcomes. Mechanisms governing EHV1 pathotype are not understood. However, EHV1 pathotypes have different infection efficiencies and replication kinetics in various cell types. They are also differentially susceptible to epigenetic inhibitors. Aside from this observation little is known about EHV1 chromatin or how its regulation influences infection. To build know
