The effect of perfluorochemical artificial blood on lipoprotein metabolism in rats.

Experiments were performed to evaluate the utility of a perfluorochemical emulsion as an artificial blood substitute for studies of lipoprotein metabolism in rats. Perfusing the liver of fed rats with perfluorochemical emulsion FC-34 at the same rate as a 20% red blood cell (RBC) perfusate, there was comparable oxygen uptake; however, there was a greater release of glucose and production of lactate than in RBC perfused livers. Under the stimulation of a low level of free fatty acid, there was less free fatty acid uptake and less triglyceride secretion in emulsion perfused livers. The lipoprote

Novel PFAS Alternatives 6:2 Cl-PFESA and HFPO-TA as Plasma FXII Activators: Experimental Evidence and Computational Mechanistic Insights.

Per- and polyfluoroalkyl substances (PFAS) can persist in human blood and may perturb hematological homeostasis by interacting with coagulation factor XII (FXII), the initiator of the contact system. This study combined in vitro, ex vivo, and in vivo experiments with computational modeling to evaluate the effects of legacy PFAS (PFOS and PFOA) and their alternatives (6:2 Cl-PFESA, HFPO-DA, HFPO-TA, and HFPO-TeA) on FXII and downstream contact activation. In vitro and ex vivo screening showed that 6:2 Cl-PFESA had a greater capacity to activate FXII than PFOS, whereas HFPO-TA induced more prono

Multigeneration responses of the springtail Folsomia candida to short-chain per- and polyfluorinated substances.

Short-chain per- and polyfluorinated substances (PFAS) are widely distributed in the environment, but their chronic effects on soil organisms exposed over multiple generations remain largely unknown. The present study therefore aimed to investigate the toxicity of perfluorobutane sulfonic acid (PFBS) and its precursor perfluorobutane sulfonamide (FBSA) to the springtail Folsomia candida during five successive generations with the following endpoints: the median lethal concentration (LC50) for adult survival and the median effect concentration (EC50) for reproduction (EC50_repro) and population

Replacement per- and polyfluoroalkyl substance (PFAS)-free aqueous film-forming foams impact growth more than a PFAS-containing product in the hard clam, Mercenaria mercenaria.

Aqueous film-forming foams (AFFFs) are widely used fire suppression products that have been identified as a direct source of environmental per- and polyfluoroalkyl substances (PFAS). Per- and polyfluoroalkyl substance exposure has demonstrated chronic and sublethal effects on biota. Ongoing efforts aim to reduce and, ideally, eliminate PFAS use in AFFF products. However, there is little known about the potential toxic effects of the new PFAS-free AFFFs, specifically on benthic organisms. The objective of this study is to quantify the effects of seven AFFFs on growth in the hard clam, Mercenari

A novel constrained drop surfactometer demonstrates inhibition of lung surfactant function by PFAS aerosols in vitro.

Per- and polyfluoroalkyl substances (PFAS) are ubiquitous in both indoor and outdoor air, and there is an increasing need to effectively screen this diverse class of chemicals for inhalation toxicity potential. Because PFAS have strong surface-active properties, we hypothesized they may interfere with lung surfactant (LS) activity. We investigated the ability of 17 PFAS delivered as liquid aerosols to inhibit LS function in a newly developed constrained drop surfactometer. Using both fluorescent tracers and mass spectrometry techniques, deposition of PFAS aerosols onto exposed LS was determine

PFAS Across the Placenta and Its Potential Impact on Glucose Imbalance and Infant Growth.

Per- and polyfluoroalkyl substances (PFASs) are persistent pollutants that cross the placental barrier, yet their transfer dynamics and developmental impacts remain unclear. We quantified 16 PFASs in maternal serum, cord serum, and maternal/fetal placental surfaces from 102 mother-infant pairs, calculated transfer efficiencies, and evaluated associations with neonatal growth and 12-month development. Mediation analysis assessed the role of maternal glucose regulation in linking PFAS exposure to early life growth outcomes. All 16 PFASs were detected, with mean ∑16PFAS concentrations of 5.26 ng/

AOP-Based Analysis of Curated Transcriptomic Data Reveals a Context-Dependent Core Mechanism of PFAS-Induced Liver Steatosis.

Toxicology faces the need to shift from generalized hazard evaluation toward precision approaches that account for the impact of the exposed biological systems. This need is particularly evident for per- and polyfluoroalkyl substances (PFAS), a highly persistent and diverse chemical class whose multiorgan apical toxicities are well documented, yet whose mechanistic understanding remains fragmented. To address this gap, a comprehensive toxicogenomic collection covering multiple PFAS and biological systems is curated, harmonized, and standardized. Through systematic integration of these data wit

Environmental Chemical-Induced Cardiometabolic Disorders: Combined Epidemiological and Experimental Evidence.

Screening environmental pollutants that are harmful to the cardiometabolic status and understanding their key toxic pathways are crucial for effective clinical intervention. Based on exposure data of 46 chemicals in a nationally representative 13,286 people, logistic regression and mixture modeling were used to preliminarily identify environmental pollutants with significant impacts on 12 indicators for cardiometabolic disorders. A total of 15 chemicals were found to be associated with the integrated latent class, among which four chemicals (perfluorononanoic acid [PFNA], perfluorooctanoic aci

Decontamination and Surface Analysis of PFAS-Contaminated Fire Suppression System Pipes: Effects of Cleaning Agents and Temperature.

Per- and polyfluoroalkyl substances (PFAS)-containing firefighting foam have been used in stationary fire suppression systems for several decades. However, there is a lack of research on how to decontaminate PFAS-contaminated infrastructure and evaluate treatment efficiency. This study assessed the removal of PFAS from stainless steel pipe surfaces using different cleaning agents (tap water, methanol, and aqueous solutions containing 10 and 20 wt % of butyl carbitol (BC)) at different temperatures (20 °C, 40 °C, and 70 °C). The content of the remaining fluorine (F)-containing compounds on the

A systematic review for non-targeted analysis of per- and polyfluoroalkyl substances (PFAS).

This review follows the PRISMA guidelines to provide a systematic review of 115 peer reviewed articles that used non-targeted analysis (NTA) methods to detect per- and polyfluoroalkylated substances (PFAS). This literature highlights the significant positive impact of NTA in understanding PFAS in the environment. Within the literature a geographical bias exists, with most NTA studies (∼60 %) conducted in the United States and China. Future studies in other regions (such as South America and Africa) are needed to gain a more global understanding. More research is required in marine environments

Evaluation of PFAS extraction and analysis methods for biosolids.

Per- and polyfluoroalkyl substances (PFAS) in the environment is a growing concern leading to a focus on PFAS occurrence in biosolids, a byproduct of wastewater treatment processes, often applied to improve soil health. This led to the need for analytical method development for assessing PFAS in biosolids. This study compares three methods for PFAS quantitation, evaluating solvent extraction, clean-up techniques, and final injection solvents. Three biosolids examined included not stabilized, anaerobically digested, and activated sludge with long-term lagoon-stabilized solids, resulting in diff

Removal of perfluorooctanoic acid (PFOA) in the liquid culture of Phanerochaete chrysosporium.

Perfluorooctanoic acid (PFOA) is becoming a concern due to its persistence, bioaccumulation, and potential harmful effects on humans and the environment. In this study, the fungus Phanerochaete chrysosporium (P. chrysosporium) was used to remove the PFOA in liquid culture system. The results showed that the average activities of laccase (Lac), lignin peroxidase (LiP), and manganese peroxidase (MnP) enzymes secreted by P. chrysosporium were 0.0003 U/mL, 0.013 U/mL, and 0.0059 U/mL, respectively, during the incubation times of 0-75 days. The pH of 3 and incubation time of 45-55 days were the opt

Confirming sulfluramid (EtFOSA) application as a precursor of perfluorooctanesulfonic acid (PFOS) in Brazilian agricultural soils.

Perfluorooctanesulfonic acid (PFOS) is a manmade chemical with several industrial applications and also a potential byproduct of many other per- and polyfluorinated substances (PFAS) in the environment. Due to the gathered evidence on its environmental persistence, long-range transport, toxicity, and bioaccumulative and biomagnifying properties, PFOS, its salts and perfluorooctane sulfonyl fluoride (PFOSF), were listed for global restriction under the Stockholm Convention on Persistent Organic Pollutants in 2009. Nevertheless, Brazil has granted an acceptable purpose exemption for using PFOSF

Ecotoxicity Evaluation of Fire-Extinguishing Water from Large-Scale Battery and Battery Electric Vehicle Fire Tests.

Electrified transport has multiple benefits but has also raised some concerns, for example, the flammable formulations used in lithium-ion batteries. Fires in traction batteries can be difficult to extinguish because the battery cells are well protected and hard to reach. To control the fire, firefighters must prolong the application of extinguishing media. In this work, extinguishing water from three vehicles and one battery pack fire test were analyzed for inorganic and organic pollutants, including particle-bound polycyclic aromatic hydrocarbons and soot content. Additionally, the acute tox

Dual effects of PFOA or PFOS on reductive dechlorination of trichloroethylene (TCE).

PFASs and chlorinated solvents are the common co-contaminants in soil and groundwater at firefighter training areas (FTAs). Although PFASs mixtures could have adverse impacts on bioremediation of trichloroethylene (TCE) by inhibiting Dehalococcoides (Dhc), little is known about the effect and contribution of PFOA or PFOS on dechlorination of TCE by non-Dhc organohalide-respiring bacteria (OHRB). To study this, PFOA and PFOS were amended to the growth medium of a non-Dhc OHRB-containing enrichment culture to determine the impact on dechlorination. This study demonstrated that high levels of PFO

Hepatotoxic Effects of Hexafluoropropylene Oxide Trimer Acid (HFPO-TA), A Novel Perfluorooctanoic Acid (PFOA) Alternative, on Mice.

As an alternative to perfluorooctanoic acid (PFOA), hexafluoropropylene oxide trimer acid (HFPO-TA) has been increasingly used for fluoropolymer manufacture in recent years. Its growing detection in environmental matrices and wildlife raises considerable concern about its potential health risks. Here we investigated the effects of HFPO-TA on mouse liver following 28 days of exposure to 0.02, 0.1, or 0.5 mg/kg/d of HFPO-TA via oral gavage. Results showed that HFPO-TA concentrations increased to 1.14, 4.48, and 30.8 μg/mL in serum and 12.0, 32.2, and 100 μg/g in liver, respectively. Liver injury

Efficient treatment of perfluorohexanoic acid by nanofiltration followed by electrochemical degradation of the NF concentrate.

The present study was aimed at the development of a strategy for removing and degrading perfluorohexanoic acid (PFHxA) from industrial process waters at concentrations in the range 60-200 mg L-1. The treatment train consisted of nanofiltration (NF) separation followed by electrochemical degradation of the NF concentrate. Using a laboratory-scale system and working in the total recirculation mode, the DowFilm NF270 membrane provided PFHxA rejections that varied in the range 96.6-99.4% as the operating pressure was increased from 2.5 to 20 bar. The NF operation in concentration mode enabled a vo

Ultrasound contrast agents: an overview.

With the introduction of microbubble contrast agents, diagnostic ultrasound has entered a new era that allows the dynamic detection of tissue flow of both the macro and microvasculature. Underpinning this development is the fact that gases are compressible, and thus the microbubbles expand and contract in the alternating pressure waves of the ultrasound beam, while tissue is almost incompressible. Special software using multiple pulse sequences separates these signals from those of tissue and displays them as an overlay or on a split screen. This can be done at low acoustic pressures (MI<0.3)
