Development of Antisense Oligonucleotide Gapmers for the Treatment of Dyslipidemia and Lipodystrophy.

Although technological advances in molecular genetics over the last few decades have greatly expedited the identification of mutations in many genetic diseases, the translation of the genetic mechanisms into a clinical setting has been quite challenging, with a minimum number of effective treatments available. The advancements in antisense therapy have revolutionized the field of neuromuscular disorders as well as lipid-mediated diseases. With the approval of splice-switching antisense oligonucleotide (AO)-based therapies for the treatment of spinal muscular atrophy (SMA) and Duchenne muscular

Phytosterols and Cardiovascular Disease.

PURPOSE OF REVIEW: Coronary heart disease is the leading cause of mortality worldwide. Elevated blood cholesterol levels are not only the major but also the best modifiable cardiovascular risk factor. Lifestyle modifications which include a healthy diet are the cornerstone of lipid-lowering therapy. So-called functional foods supplemented with plant sterols lower blood cholesterol levels by about 10-15%. RECENT FINDINGS: In the recent revision of the ESC/EAS dyslipidemia guideline 2019, plant sterols are recommended for the first time as an adjunct to lifestyle modification to lower blood cho

Case Report: A family history of peanut allergy and hereditary alpha-tryptasemia.

CONTEXT: Hereditary alpha-tryptasemia (HαT) is associated with elevated basal serum tryptase (bST) and is associated with a higher risk of severe anaphylactic reactions in patients with clonal mast cell disorders or IgE-mediated Hymenoptera venom-induced anaphylaxis. The consequence of this genetic trait remains to be determined in other allergic diseases and food allergy in particular. OBJECTIVES: Here, we describe three cases of peanut allergy among siblings from a single family of four: two of them were associated with HαT, and the third one was associated with the tryptase wild-type genot

Diet in Treatment of Inflammatory Bowel Diseases.

There has been an alarming increase in the incidence of inflammatory bowel diseases (IBDs) worldwide over the past several decades. The pathogenesis of IBD involves genetic and environmental factors. Diet is a potentially modifiable environmental risk factor for IBD onset and severity. Diet can promote intestinal inflammation by dysregulating the immune system, altering intestinal permeability and the mucous layer, contributing to microbial dysbiosis, and other mechanisms. Dietary changes therefore might be incorporated into therapeutic strategies for IBD. Enteral nutrition is effective in the

Measuring Constipation in a Drosophila Model of Parkinson's Disease.

Non-motor symptoms in Parkinson's disease (PD) are common, difficult to treat, and significantly impair quality of life. One prevalent non-motor symptom is constipation, which can precede the diagnosis of PD by years or even decades. Constipation has been underexplored in animal models of PD and lacks specific therapies. This assay utilizes a Drosophila model of PD in which human alpha-synuclein is expressed under a pan-neuronal driver. Flies expressing alpha-synuclein develop the hallmark features of PD: the loss of dopaminergic neurons, motor impairment, and alpha-synuclein inclusions. This

Genetically Modified Microorganisms: Risks and Regulatory Considerations for Human and Environmental Health.

Advances in affordable genetic engineering have accelerated the creation and large-scale environmental release of genetically modified microorganisms (GMMs). While beneficial applications exist, GMMs may present unique, long-term risks to human and environmental health. Unlike static chemicals, GMMs are biologically active, self-replicating entities capable of rapid mutation and global dispersal. Current regulatory frameworks place responsibility on each country to regulate GMMs, without a clear, coordinated international policy. This review details critical risk scenarios, including horizonta

The Associations of Dietary Iron Intake and the Transferrin Receptor (TFRC) rs9846149 Polymorphism with the Risk of Gastric Cancer: A Case-Control Study Conducted in Korea.

BACKGROUND: A positive association between a high iron intake and colorectal cancer has been identified; however, the effect of dietary iron on gastric cancer (GC) remains unclear. Here, we investigate whether dietary iron is related to GC risk and whether the transferrin receptor (TFRC) rs9846149 polymorphism modifies this association. METHODS: A case-control study was designed to assess this association among 374 GC patients and 754 healthy controls. A self-administered questionnaire was used to collect information on demographics, medical history and lifestyle. Dietary iron intake was asse

Dietary preference, biological aging, and incident dementia: a prospective cohort study.

Dietary patterns are modifiable determinants of dementia risk, but the interactions between individual dietary preference profiles, biological aging, and dementia risk remain unclear. We aimed to investigate associations of dietary preference profiles with biological aging and incident dementia, as well as the potential modifying effect of genetic susceptibility. Dietary preference profiles were identified using latent profile analysis on food preferences questionnaire data. Cox models estimated the association between dietary preferences and dementia risk and their interaction with genetic ri

A Genetically Engineered Commercial Chicken Line Is Resistant to Highly Pathogenic Avian Leukosis Virus Subgroup J.

Viral diseases remain a major concern for animal health and global food production in modern agriculture. In chickens, avian leukosis virus subgroup J (ALV-J) represents an important pathogen that causes severe economic loss. Until now, no vaccine or antiviral drugs are available against ALV-J and strategies to combat this pathogen in commercial flocks are desperately needed. CRISPR/Cas9 targeted genome editing recently facilitated the generation of genetically modified chickens with a mutation of the chicken ALV-J receptor Na+/H+ exchanger type 1 (chNHE1). In this study, we provide evidence t

Immune effector cell (IEC) therapies offer durable and sustained remissions in significant numbers of patients with hematological cancers. While these unique immunotherapies have improved outcomes for pediatric and adult patients in a number of disease states, as 'living drugs,' their toxicity profiles, including cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS), differ markedly from conventional cancer therapeutics. Benefits of genetically modified crops for the poor: household income, nutrition, and health.

The potential impacts of genetically modified (GM) crops on income, poverty and nutrition in developing countries continue to be the subject of public controversy. Here, a review of the evidence is given. As an example of a first-generation GM technology, the effects of insect-resistant Bt cotton are analysed. Bt cotton has already been adopted by millions of small-scale farmers, in India, China, and South Africa among others. On average, farmers benefit from insecticide savings, higher effective yields and sizeable income gains. Insights from India suggest that Bt cotton is employment generat

Genetic manipulation of cysH/cysJ in Citrobacter sp. XT1-2-2 enhanced cadmium immobilization by regulating metabolic pathways.

The Citrobacter sp. XT1-2-2 strain has emerged as a promising candidate for cadmium immobilization; however, the genetic basis underlying its sulfur-mediated bioremediation mechanisms remains inadequately understood. To address this gap, we concentrated on two pivotal genes, cysH and cysJ, within the sulfate assimilation pathway. We constructed Citrobacter sp. XT1-2-2-::APS and Citrobacter sp. XT1-2-2-::SiR strains with overexpression of cysH or cysJ for functional characterization. Transmission electron microscopy demonstrated a significant enhancement in the biosynthesis of CdS nanoparticles

Neurobehavioural genes exert indirect genetic effects on blood lipid profiles through behavioural traits.

Growing evidence reveals that indirect genetic effects (IGEs) contribute substantially to human complex traits. However, the genomic architecture and underlying mechanistic pathways of IGEs remain poorly understood. In this study, we employed a two-stage family study design to investigate IGEs and direct genetic effects (DGEs) on blood lipid profiles in the Fangshan Family-based Ischemic Stroke Study in China (FISSIC) cohort. We identified 16 IGE loci resulting in 14 candidate IGE genes, and 20 DGE loci resulting in 22 DGE genes, by an integrative functional mapping approach. The identified IG

Genetic variation in the vitamin D receptor (VDR) and the vitamin D-binding protein (GC) and risk for colorectal cancer: results from the Colon Cancer Family Registry.

Epidemiologic evidence supports a role for vitamin D in colorectal cancer (CRC) risk. Variants in vitamin D-related genes might modify the association between vitamin D levels and CRC risk. In this analysis, we did a comprehensive evaluation of common variants in the vitamin D receptor (VDR) and the vitamin D-binding protein (GC; group-specific component) genes using a population-based case-unaffected sibling control design that included 1,750 sibships recruited into the Colon Cancer Family Registry. We also evaluated whether any associations differed by calcium supplement use, family history

Response of the microbiome-gut-brain axis in Drosophila to amino acid deficit.

A balanced intake of macronutrients-protein, carbohydrate and fat-is essential for the well-being of organisms. An adequate calorific intake but with insufficient protein consumption can lead to several ailments, including kwashiorkor1. Taste receptors (T1R1-T1R3)2 can detect amino acids in the environment, and cellular sensors (Gcn2 and Tor)3 monitor the levels of amino acids in the cell. When deprived of dietary protein, animals select a food source that contains a greater proportion of protein or essential amino acids (EAAs)4. This suggests that food selection is geared towards achieving th

Improving the Dietary Vitamin A Content of Rural Communities in South Africa by Replacing Non-Biofortified white Maize and Sweet Potato with Biofortified Maize and Sweet Potato in Traditional Dishes.

Biofortification of staple crops has a potential for addressing micronutrient deficiencies, such as vitamin A deficiency (VAD), which are prevalent in South Africa. The poor acceptability of provitamin A (PVA)-biofortified foods could be improved by combining them with other food items to produce modified traditional dishes. The nutritional composition of the dishes could also be improved by the modification. The study aimed to investigate the effect of replacing white maize and cream-fleshed sweet potato (CFSP)] with PVA-biofortified maize and orange-fleshed sweet potato (OFSP) on the nutriti

The environment-pathogen-host axis in communicable and non-communicable diseases: recent advances in experimental and clinical research.

Allergies and autoimmune diseases are spreading worldwide. Control of infections, on the other hand, remains an issue, even in the post-antibiotic era. Chronic or poorly controlled infections occur in immune compromised individuals such as HIV patients, hospitalized patients exposed to multi-resistant bacteria, or patients on immunosuppressive treatment. They may become an even more emerging issue in an ageing population. At the same time, profound environmental changes such as global warming, urbanization, increasing environmental pollution and novel food engineering technologies may alter th
