Trophic transfer of pollutants within two intertidal rocky shore ecosystems in different biogeographic regions of South Africa.

Tsitsikamma and Sheffield Beach are two relatively pristine sites along the South African east coast representing warm temperate and subtropical biogeographic rocky shore intertidal ecosystems, respectively. Stable isotopes (δ15N and δ13C), metals and organochlorine pesticides (OCPs) were measured in 38 intertidal components to study biomagnification or biodilution of metals and OCPs in these marine food webs. Comparison of the four species common to both sites revealed that the highest Al, Fe and OCP concentrations were measured in intertidal organisms from Sheffield Beach and was attributed

Quantitative microbial risk assessment for Escherichia coli O157:H7, Salmonella enterica, and Listeria monocytogenes in leafy green vegetables consumed at salad bars, based on modeling supply chain logistics.

Quantitative microbial risk assessments do not usually account for the planning and ordering mechanisms (logistics) of a food supply chain. These mechanisms and consumer demand determine the storage and delay times of products. The aim of this study was to quantitatively assess the difference between simulating supply chain logistics (MOD) and assuming fixed storage times (FIX) in microbial risk estimation for the supply chain of fresh-cut leafy green vegetables destined for working-canteen salad bars. The results of the FIX model were previously published (E. Franz, S. O. Tromp, H. Rijgersber

Methods of Controlling Microbial Contamination of Food.

The rapid growth of world population and increase in living standards have led to an increase in the demand for high-quality, safe food. The Food and Agriculture Organization of the United Nations (FAO) estimates that by 2050 the demand for food will increase by 60%, and production of animal protein will increase by 1.7% a year, with meat production to increase by nearly 70%, dairy products by 55%, and aquaculture by as much as 90%. Microbial contamination of food is a significant problem for the accessibility of safe food which does not pose a threat to the life and health of consumers. Campy

A robust nanobody-magnetic bead immunoassay for food safety monitoring: Sensitive detection of viral contamination in giant salamander.

The Chinese giant salamander is a valued food source of high-quality protein; however, the prevalence of Giant Salamander Iridovirus (GSIV) poses a significant threat to the safety and stability of the aquaculture food supply chain. Rapid and sensitive screening of viral pathogens in complex food matrices is therefore critical. We developed a bridge-free double-nanobody sandwich ELISA using magnetic beads (MBs) for detection of GSIV. AlphaFold 3 modeling suggested that covalent immobilization via conserved framework lysines may promote an optimal "upright" nanobody orientation, significantly e

Uptake and Accumulation of Nephrotoxic and Carcinogenic Aristolochic Acids in Food Crops Grown in Aristolochia clematitis-Contaminated Soil and Water.

Emerging evidence has suggested aristolochic acids (AAs) are linked to the development of Balkan endemic nephropathy (BEN), a chronic renal disease affecting numerous farmers living in the Balkan peninsula. However, the pathway by which AAs enter the human food chain and cause kidney disease remains poorly understood. Using our previously developed analytical method with high sensitivity and selectivity (Chan, W.; Lee, K. C.; Liu, N.; Cai, Z. J. Chromatogr. A 2007, 1164, 113-119), we quantified AAs in lettuce, tomato, and spring onion grown in AA-contaminated soil and culture medium. Our study

Concentration of mercury and other metals in an Arctic planktonic food web under a climate warming scenario.

Arctic marine ecosystems act as a global sink of mercury (Hg) and other metals, and high concentrations of these have been measured in higher trophic-level organisms. Nevertheless, the concentrations of metals at the basis of the marine food web in the Arctic is less known despite the likelihood of biomagnification from dietary sources. We investigated the concentrations of mercury (Hg) and other metals in different size fractions of plankton in West Greenland. All size fractions contained detectable levels of Hg (ranging from 4.8 to 241.3 ng g dw-1) at all stations, although with high geograp

Incorporating uncertainty reveals resilience and efficiency differences between three neighboring marine food webs.

While general principles of ecosystem-based approaches to fisheries management have been articulated for over a century, their large-scale implementation remains a challenge. Operationalizing ecosystem-based management demands the ongoing development of indicators and ecological reference points, analogous to biological reference points of single species fisheries management. For such ecosystem-level indicators to be useful in applied contexts, we must appropriately account for uncertainty. Here, we focused on advancing the utility of one specific indicator, relative ascendancy, by incorporati

Effect of web inclination on debris-induced damage in Leucauge venusta webs (Araneae: Tetragnathidae).

Spiders are among the most conspicuous trap builders in nature, and the structure and orientation of spider webs are presumably under strong selection pressure. Most ecribellate orb-weavers (Araneoidea) build vertically-orientated webs (0°; ie the plane of the web is orthogonal to the ground). However, Leucauge venusta (Tetragnathidae; Walckenaer, 1842) often build horizontally-orientated webs (90°; ie parallel to the ground), despite evidence that prey capture and retention rates are higher in vertical webs. Falling debris is a common hazard for webs in the forest understory, and basic geomet

Rapid diversification of a free-living protist is driven by adaptation to climate and habitat.

Microbial eukaryotes (protists) have major functional roles in aquatic ecosystems, including the biogeochemical cycling of elements as well as occupying various roles in the food web. Despite their importance for ecosystem function, the factors that drive diversification in protists are not known. Here, we aimed to identify the factors that drive differentiation and, subsequently, speciation in a free-living protist, Synura petersenii (Chrysophyceae). We sampled five different geographic areas and utilized population genomics and quantitative trait analyses. Habitat and climate were the major

Controlling Foodborne Antimicrobial-Resistant Bacteria: The Gen(i)e is Out of the Bottle.

As a conduit between the environment and human and animal populations, food sits at the nexus of One Health and is a major vehicle for the transmission of pathogenic and nonpathogenic antibiotic-resistant bacteria. To alleviate the associated public health burden, it is therefore critical to reduce human exposure to antibiotic-resistant bacteria via food consumption. The objective of this narrative literature review was to demonstrate that while reduction of antibiotic use in agriculture is necessary, it is not sufficient to protect humans from exposure to antibiotic-resistant foodborne infect

A State-of-Art Review on Multi-Drug Resistant Pathogens in Foods of Animal Origin: Risk Factors and Mitigation Strategies.

Most of the foodborne microbial diseases are linked to foods of animal origin such as milk, meat, and poultry. Nowadays, the presence of multi-drug resistant (MDR) pathogens in foods is becoming an increasingly public health concern worldwide due to the overuse of antimicrobial drugs in animal feed. MDR pathogens can enter the food chain by posing a significant risk to both animals and consumers. MDR pathogens causing infections are untreatable due to their resistance to various antibiotics, primarily cephalosporin and carbapenems and to their extended-spectrum beta-lactamase (ESBL)-producing

Sample Preparation for qPCR Detection of Listeria from Food.

Quantitative PCR, if performed properly, is a highly sensitive and robust tool. Nevertheless, its application to the particular case of pathogen detection from foodstuffs necessitates special requirements for reliable results. Firstly, a robust analytical chain, involving sample preparation and DNA isolation with purification, is necessary to ensure optimal performance. Secondly, for reliable quantification of Listeria monocytogenes from food, reproducible controls for all steps of the analytical chain are needed, which can give quantitative information about the performance of each individual

Role of peptide YY(3-36) in the satiety produced by gastric delivery of macronutrients in rats.

Peptide YY(3-36) [PYY(3-36)] is postulated to act as a hormonal signal from gut to brain to inhibit food intake. PYY(3-36) potently reduces food intake when administered systemically or into the brain. If action of endogenous PYY(3-36) is necessary for normal satiation to occur, then pharmacological blockade of its receptors should increase food intake. Here, we determined the effects of iv infusion of Y1, Y2, and Y5 receptor antagonists (BIBP 3226, BIIE 0246, CGP 71683) during the first 3 h of the dark period on food intake in non-food-deprived rats. Our results showed that 1) Y2 receptor blo

Differential PFAS transfer through the terrestrial and aquatic food web to predatory spiders.

The environmental persistence of Per- and polyfluoroalkyl substances (PFAS) results in prolonged exposure of organisms, which in turn leads to bioaccumulation and trophic transfer. However, if trophic transfer is studied at all, different ecosystems are generally examined in isolation. The aim of this study was therefore to unravel the PFAS transfer through a contaminated terrestrial and aquatic food web, with spiders as top predators. To this end, organisms from the aquatic and terrestrial compartments of a PFAS-contaminated ecosystem were collected and screened for 44 PFAS. The trophic posit

Gellan Gum Modification and Structural Regulation for Food Applications: Delivery Systems and Digestive Behavior.

Gellan gum (GG) is an anionic microbial polysaccharide widely used in foods because of its efficient gelation, thermal stability, and ion-responsive structuring. However, the contrasting limitations of high- and low-acyl GG, together with their sensitivity to molecular weight, ions, pH, thermal history, and food-matrix composition, restrict their performance in complex products. This review distinguishes four intervention levels of GG modification and structural regulation: covalent modification, chain-architecture regulation, supramolecular regulation, and food-matrix composite structuring. Next-Day Salmonella spp. Detection Method Based on Real-Time PCR for Foods.

Reliable detection of Salmonella spp. remains essential for food safety, yet conventional culture-based methods are limited by their long turnaround times. Advances in molecular diagnostics, particularly real-time PCR (qPCR), have transformed pathogen detection by enabling faster, more sensitive, and more flexible analytical approaches. qPCR has gained prominence due to its ability to detect low levels of contamination and its robustness across diverse food matrices. In this chapter, we describe a qPCR-based strategy for the rapid detection of Salmonella spp. that adheres to the principles of

Tropical wood resistance to the West Indian drywood termite Cryptotermes brevis: If termites can't chew….

BACKGROUND: The importance and impact of invasive species are usually considered based on their economic implications, particularly the direct damage that they cause. The West Indian drywood termite Cryptotermes brevis (Walker) is an example and is a concern in structural lumber, furniture, and other wood products. Despite its importance, its tropical wood preferences and the wood physical characteristics contributing to resistance have not been investigated to date. Here, we developed wood testing units to allow the X-ray recording of termite colonization and then subsequently tested tropical

Establishment of a simple and rapid identification method for Listeria spp. by using high-resolution melting analysis, and its application in food industry.

Listeria monocytogenes is the causative bacteria of listeriosis, which has a higher mortality rate than that of other causes of food poisoning. Listeria spp., of which L. monocytogenes is a member, have been isolated from food and manufacturing environments. Several methods have been published for identifying Listeria spp.; however, many of the methods cannot identify newly categorized Listeria spp. Additionally, they are often not suitable for the food industry, owing to their complexity, cost, or time consumption. Recently, high-resolution melting analysis (HRMA), which exploits DNA-sequence

Survival of salmonellae on and in tomato plants from the time of inoculation at flowering and early stages of fruit development through fruit ripening.

The fate of salmonellae applied to tomato plants was investigated. Five Salmonella serotypes were used to inoculate tomato plants before and after fruits set, either by injecting stems with inoculum or brushing flowers with it. Ripe tomato fruits were subjected to microbiological analysis. Peptone wash water, homogenates of stem scar tissues, and homogenates of fruit pulp were serially diluted and plated on bismuth sulfite agar before and after enrichment. Presumptive Salmonella colonies were confirmed by serological tests, PCR assay using HILA2 primers, and enterobacterial repetitive intergen

Innovations and technology disruptions in the food sector within the COVID-19 pandemic and post-lockdown era.

BACKGROUND: COVID-19 pandemic has caused a global lockdown that has abruptly shut down core businesses and caused a worldwide recession. The forecast for a smooth transition for the agri-food and drink industry is, at best, alarming. Given that COVID-19 shutdown multiple core services (such as aviation, food services, supply chains, and export and import markets), there is an enormous deficiency in critical information to inform priority decision making for companies where this uncertainly is likely to impact negatively upon recovery. SCOPE AND APPROACH: The current article investigates poten
