Fumonisin B1 (FB1) is a widespread mycotoxin produced by fungal Fusarium species-mainly in maize, one of the plants most commonly used for food and feed. Pigs and horses are the animal species most susceptible to this mycotoxin. FB1 exposure can cause highly diverse clinical symptoms, including hepatotoxicity, immunotoxicity, and intestinal barrier function disturbance. Inhibition of ceramide synthetase is a well-understood ubiquitous molecular mechanism of FB1 toxicity, but other more tissue-specific effects remain to be elucidated. To investigate the effects of FB1 in different exposed tissu

Hapten-carrier protein conjugates were made using five different small haptens (MW < 1000), two carrier proteins and two methods of conjugation. Nondenaturing agarose gel electrophoresis was used to demonstrate that when as few as two molecules of a small hapten are attached to the carrier, the conjugate band migrates differently from that of the carrier alone or of the coupling reagent-treated carrier. Furthermore, the direction of the change in migration of each conjugate correlates with the change in charge which occurs upon attachment of the hapten to the carrier.

Numerous recent, epidemiological studies reveal that Western populations are growing more and more deficient in daily Mg intake which have been linked to etiology of cardiovascular (CV) diseases. A growing body of evidence suggests that a major missing link to this dilemma may reside within the sphingolipid-ceramide pathways. For the past 25 years , our labs have been focusing on these pathways in Mg-deficient mammals. The objective of this paper is two-fold: 1) to test various hypotheses and 2) to review the current status of the field and how protein kinase C isoforms may be pivotal to solvin

Fusarium ear rot (FER), caused by Fusarium verticillioides, results in substantial yield losses and poses a significant threat to maize production worldwide. However, the genetic basis of FER resistance remains poorly understood. Utilizing QTL-seq and association analysis, we identified a gene encoding UDP-ᴅ-glucuronate 4-epimerase 1 (ZmGAE1). A 141-base pair insertion was revealed as the natural functional variation in the promoter of ZmGAE1, which decreases its expression and enhances resistance to FER. Functional validation confirmed that ZmGAE1 acts as a negative regulator of maize resista

Conditions were determined for rapid, convenient, and efficient determination of 16 common mycotoxins in tea samples. Mycotoxins in tea leaves and tea infusion samples were extracted using solid-liquid extraction/liquid-liquid extraction combined with ultrasonic-assisted extraction. The extraction solvent was 2-butanone/ethyl acetate (9/1 v/v) with 0.1 % formic acid. The established conditions enabled the analysis of these mycotoxins by ultra-high-performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) in 5.5 min. In addition, HPLC with a temperature-controlled fluorescence de

Fusarium species, which are widely distributed across agricultural ecosystems worldwide, are among the most economically harmful phytopathogens. They can colonize a broad range of agri-food crops, causing plant diseases and the accumulation of mycotoxins in edible plant tissues. Fusarium species produce a variety of secondary metabolites, posing toxicological risks to human and animal health. A total of 119 Fusarium strains isolated from agricultural soils in northwestern Iberian Peninsula were molecularly identified at the species level, and their capability to produce 17 mycotoxins was evalu

Fungal species produce various secondary metabolites with carcinogenic and genotoxic properties, including aflatoxins, ochratoxins, fumonisins, sterigmatocystin, and griseofulvin. Acute or chronic exposure to these mycotoxins through contaminated food and feed can cause DNA damage, chromosomal instability, and epigenetic changes, ultimately leading to cancer. This review focuses on IARC-classified carcinogenic mycotoxins and their contribution to cancer development by disrupting the DNA damage response (DDR), thereby affecting apoptosis, cell cycle regulation, and genomic stability. Current kn

Fungal contamination and mycotoxins remain important hazards in cereal, nut, oilseed, and processed-food chains. For grain handlers, processors, storage facilities, quality-assurance laboratories, and technology developers, the practical challenge is not only detection but determining when an AI-enabled optical output is sufficiently documented to support accepting, holding, sorting, or referring material for confirmatory analysis. This critical narrative review, supported by structured evidence appraisal, examines artificial intelligence (AI)-enabled hyperspectral imaging (HSI) for fungal and

Mycotoxins, toxic secondary metabolites of fungi are now recognised as major cause of food intoxications in Sub Saharan Africa (SSA). Aflatoxins, the most important of the group have been implicated in acute aflatoxicoses, carcinogenicity, growth retardation, neonatal jaundice and immunological suppression in SSA. The hot and humid tropical climate provides ideal condition for growth of toxigenicAspergillus spp, making food contamination to be widespread in SSA, with maize and groundnuts being the most contaminated. The available data suggests that cassava products (the most important African

Mycotoxins such as beauvericin (BEA), deoxynivalenol (DON), enniatin B (ENB), fumonisin B1 (FB1), T-2 toxin and zearalenone (ZEA) can co-occur in food commodities. This aim of this study was to assess the myelotoxicity of these mycotoxins in couple using in vitro human granulo-monocytic (Colony Forming Unit-Granulocyte and Macrophage, CFU-GM) hematopoietic progenitors. Clonogenic assays have been performed in the presence of the following couples of fusariotoxins: DON + BEA, DON + FB1, DON + T-2, DON + ZEA, T-2 + ZEA and BEA + ENB. Co-exposure of human CFU-GM to DON + BEA resulted in synergic

Trigonella foenum-graecum is a widely cultivated legume in Mediterranean regions, and it is used for human and animal consumption, as well as for medical purposes. High temperatures and abundant rainfall during the spring season in Sicily favor the formation of an environment suitable for the growth and proliferation of fungi with the production of mycotoxins. In this study, ochratoxin A, aflatoxin, deoxynivalenol, zearalenone, fumonisin, and T-2 toxin concentrations in Trigonella foenum-graecum were determined in feed administered to ruminants and also in blood samples from cattle and sheep i

Consumers are regularly exposed to well-known food contaminants (FCs), which are typically assessed for risk on an individual basis. However, there is limited knowledge about the overall levels and combinations of these compounds depending on dietary choices. The goal of this study was to estimate the real-life mixtures of FCs in different dietary models by integrating extensive data from the scientific literature concerning the reliable quantification of FCs in foods. A FAIR database detailing the occurrence of 73 FCs in 16 foods commonly consumed was built. The data were integrated into an o

Chronic exposure to mycotoxin-contaminated food and feed induces systemic toxicity in humans and animals, with emerging evidence identifying ferroptosis as a key mechanism driving this pathology. Key mycotoxins, including deoxynivalenol, aflatoxin B1, T-2 toxin, fumonisin B1, zearalenone, ochratoxin A, and patulin, trigger ferroptosis through interconnected pathways: iron metabolism disruption, lipid peroxides accumulation, Glutathione Peroxidase 4 (GPX4) antioxidant system imbalance, Ferroptosis Suppressor Protein 1 (FSP1) pathway modulation, and mitochondrial homeostasis disruption. In this work, the genome of Bacillus subtilis 34a, a strain isolated from healthy coffee plants, was sequenced and analyzed by genome mining to predict its antifungal potential. These predictions were subsequently evaluated by characterizing the volatile and diffusible chemical profiles associated with interactions between B. subtilis 34a and fungal pathogens (Fusarium spp. and Umbelopsis sp.). Dual co-cultivation assays revealed variable susceptibility among fungal strains to the antagonistic activity of Bacillus subtilis 34a, with radial growth inhibition ranging from 15.2% to 54.1%.
