Stimulation of cyclic AMP formation and nerve electrical activity by octopamine in the terminal abdominal ganglion of the female gypsy moth Lymantria dispar.

The biogenic amine octopamine is known to be present in the abdominal ganglia of some insects, but the expression of functional octopamine receptors in these neuronal structures has not yet been characterized. In the present study, we describe the presence in the female gypsy moth terminal abdominal ganglion (TAG), a key structure in the control of the insect reproductive behavior, of an octopamine receptor coupled to stimulation of adenylyl cyclase through the GTP-binding protein G(s). The rank order of potency of different antagonists, which discriminate between the different classes of octo

Stimulation of adenylate cyclase in relation to dopamine-induced long-term enhancement (LTE) of muscarinic depolarization in the rabbit superior cervical ganglion.

Dopamine (DA) induction of the long-term enhancement (LTE) of the slow muscarinic depolarizing response to methacholine (MCh), equivalent to the slow EPSP (S-EPSP), was previously found to be mimicked by exogenous cyclic AMP (cAMP) in the rabbit superior cervical ganglion (SCG). DA-induced LTE of the S-EPSP was shown to be depressed by some DA antagonists. We now show that DA (15 microM), its analog, 2-amino-6,7-dihydroxy-1,2,3,4-tetrahydronaphthalene (ADTN), and a D2 receptor antagonist, metoclopramide, each can induce both LTE of MCh depolarization and an increase in ganglionic cAMP. The effect of intrinsically photosensitive retinal ganglion cell (ipRGC) stimulation on axial length changes to imposed optical defocus in young adults.

PURPOSE: The intrinsically photosensitive retinal ganglion cells (ipRGCs) regulate pupil size and circadian rhythms. Stimulation of the ipRGCs using short-wavelength blue light causes a sustained pupil constriction known as the post-illumination pupil response (PIPR). Here we examined the effects of ipRGC stimulation on axial length changes to imposed optical defocus in young adults. MATERIALS AND METHODS: Nearly emmetropic young participants were given either myopic (+3 D, n = 16) or hyperopic (-3 D, n = 17) defocus in their right eye for 2 h. Before and after defocus, a series of axial leng

Dorsal Root Ganglion Stimulation for the Treatment of Non-Complex Regional Pain Syndrome Related Chronic Pain Syndromes: A Systematic Review.

BACKGROUND: While the majority of indications and approvals for dorsal root ganglion stimulation (DRGS) are for the refractory management of complex regional pain syndrome (CRPS), emerging evidence has suggested that DRGS may be favorably used for a plethora of other chronic pain phenomena. Consequently, we aimed to characterize the use and efficacy of DRGS for these non-CRPS-related chronic pain syndromes. MATERIALS AND METHODS: A systematic review of clinical studies demonstrating the use of DRGS for non-CRPS-related chronic pain syndromes. The literature search was performed using PubMed,

Dorsal root ganglion application of muscimol prevents hyperalgesia and stimulates myelin protein expression after sciatic nerve injury in rats.

BACKGROUND: Peripheral nerve injuries may result in debilitating pain that is poorly responsive to conventional treatment. Neuropathic pain induced by peripheral nerve injury is caused, in part, by ectopic discharges from the injury site or the dorsal root ganglia (DRG) resulting in enhanced central input and central hyperexcitability. A heterogeneous family of γ-aminobutyric acid (GABA)(A) channels is important in quieting neuronal excitability. We have recently reported that in vivo modulation of GABAergic neurons in DRG can alter the course of neuropathic pain development after peripheral n

Lens injury stimulates adult mouse retinal ganglion cell axon regeneration via both macrophage- and lens-derived factors.

In the present study the effects of lens injury on retinal ganglion cell axon/neurite re-growth were investigated in adult mice. In vivo, lens injury promoted successful regeneration of retinal ganglion cell axons past the optic nerve lesion site, concomitant with the invasion of macrophages into the eye and the presence of activated retinal astrocytes/Muller cells. In vitro, retinal ganglion cells from lens-lesioned mice grew significantly longer neurites than those from intact mice, which correlated with the presence of enhanced numbers of activated retinal astrocytes/Muller cells. Gasserian Ganglion and Supraorbital Nerve Stimulation for the Treatment of Herpetic-Related Neuralgia in the Ophthalmic Branch of the Trigeminal Nerve: A Single-Center Retrospective Study.

BACKGROUND: Herpes zoster infection involving the ophthalmic branch of the trigeminal nerve may lead to the development of ophthalmic herpetic-related neuralgia. Pharmacologic treatment of ophthalmic herpetic-related neuralgia often exhibits suboptimal clinical outcomes. However, limited reports suggest the use of Gasserian ganglion (GG) stimulation or supraorbital nerve stimulation (SON) through the foramen ovale to treat ophthalmic herpetic-related neuralgia. This study aims to investigate the efficacy and safety profile of peripheral nerve stimulation (PNS), including GG stimulation and SON

Comparative Effectiveness of Dorsal Root Ganglion and Spinal Cord Stimulation in Chemotherapy-Induced Peripheral Neuropathy: A Single-Center Retrospective Case Series.

OBJECTIVES: Chemotherapy-induced peripheral neuropathy (CIPN) is a debilitating consequence of many necessary oncologic treatments. Spinal cord stimulation (SCS) is the more common option for refractory cases, but growing evidence suggests dorsal root ganglion stimulation (DRG-S) is another viable option. This study aims to retrospectively compare the use of DRG-S and SCS to determine the modality chosen for permanent implantation for treatment of lower extremity CIPN. MATERIALS AND METHODS: This is a retrospective, single-center case series in which five patients with lower extremity CIPN un

Blue Light Induces Retinal Ganglion Cell Damage by Stimulating Drp1-Dependent Mitochondrial Fission and Activating NF-κB/NOX4 Axis.

PURPOSE: This study aimed to investigate the mechanisms of blue light-induced neurotoxicity in retinal ganglion cells (RGCs), focusing on the roles of mitochondrial dynamics and oxidative stress. METHODS: The impact of blue light exposure was assessed in vitro and in vivo. Key molecular changes were analyzed, and the effects of pharmacological inhibition of Drp1 and/or NOX4 were evaluated on mitochondrial function and RGC apoptosis. RESULTS: Blue light triggered mitochondrial fission by upregulating Drp1 and downregulating MFN2. This disruption promoted the nuclear translocation and phosphor

Long-Term Effectiveness and Safety of Dorsal Root Ganglion Stimulation for Persistent Spinal Pain Syndrome: Results From an Expanded Prospective Registry.

INTRODUCTION: Persistent spinal pain syndrome (PSPS) with predominant low-back pain (LBP) is difficult to manage when conservative and surgical treatments fail. Dorsal root ganglion stimulation (DRG-S) offers segmental, focal neuromodulation that may be particularly suited for managing axial LBP, yet long-term evidence from real-world practice remains limited. This study evaluated the effectiveness, safety, and impact on medication use of bilateral T12 DRG-S in patients with intractable LBP, using prospectively collected registry data. METHODS: Of 33 consecutive patients with chronic LBP asse

Repetitive magnetic stimulation prevents dorsal root ganglion neuron death and enhances nerve regeneration in a sciatic nerve injury rat model.

Peripheral nerve injury (PNI) often leads to retrograde cell death in the spinal cord and dorsal root ganglia (DRG), hindering nerve regeneration and functional recovery. Repetitive magnetic stimulation (rMS) promotes nerve regeneration following PNI. Therefore, this study aimed to investigate the effects of rMS on post-injury neuronal death and nerve regeneration. Seventy-two rats underwent autologous sciatic nerve grafting and were divided into two groups: the rMS group, which received rMS and the control (CON) group, which received no treatment. Motor neuron, DRG neuron, and caspase-3 posit

Dorsal root ganglion stimulation provides functional improvement from debilitating abdominal pain in Crohn's disease: A 12-month follow-up.

BACKGROUND: Crohn's disease (CD) is a chronic relapsing-remitting, immunological, inflammatory bowel disease involving any part of the gastrointestinal tract, most commonly, the terminal ileum. Abdominal pain is a prominent debilitating symptom of CD due to continuous intestinal inflammation, associated with disease severity and complications. However, abdominal pain has shown to occur even with disease remission. CASE PRESENTATION: A female college student with a history of Crohn's Disease was referred for severe, chronic abdominal pain, with frequent flare-ups and hospitalizations. Due to h

Successful Treatment of Refractory Neuropathic Pain Following Axillary Dissection and Lipoma Resection Using Cervical and Thoracic Dorsal Root Ganglion Stimulation.

BACKGROUND: Neuropathy is a pain condition that is caused by an insult to the somatosensory system. Providing adequate management of neuropathic pain (NP) conditions can be challenging as the dis-ease may become progressively nonresponsive to conventional therapies. If conventional treatment options prove ineffective, cervical dorsal root ganglion (DRG) stimulation is a relatively effective, off-label-use therapy for NP, which requires expert cervical placement to be completed safely and effectively. The Neuromodulation Appropriateness Consensus Committee guidelines highlight safety features a

Case Report: Novel Anchoring Technique and Surgical Nuances for Trigeminal Ganglion Stimulation in the Treatment of Post-Herpetic Trigeminal Neuropathic Facial Pain.

INTRODUCTION: Trigeminal ganglion stimulation is a neuromodulatory surgical procedure utilized to treat trigeminal neuropathic pain. This technique involves the placement of a stimulating electrode adjacent to the trigeminal ganglion and can be trialed before permanent implantation. Wider adoption by surgical practitioners is currently limited by complications such as lead migration from the trigeminal ganglion, which can result in loss of therapy and cannot be rectified without repeat surgery. We describe a novel surgical modification that successfully anchors the trigeminal ganglion electrod
