Phytophthora infestans, the pathogen responsible for the devastating potato disease known as late blight, poses a significant threat to plant health. A major determinant of its pathogenic success is the deployment of cytoplasmic effector proteins, particularly the RXLR class of effectors, which accumulate within the host cells and manipulate plant immune responses. These effectors function as key virulence factors that interfere with multiple layers of plant defense, including pattern-triggered immunity (PTI) and effector-triggered immunity (ETI). Recent studies have revealed that RXLR effecto

Plastid-based production of recombinant proteins has emerged as a transformative platform for global health applications, offering high-yield, low-cost, and scalable alternatives to conventional microbial and mammalian systems. Chloroplasts facilitate exceptional transgene expression (often exceeding 40% of total soluble protein (TSP)), due to leveraging high genome copy numbers per cell, precise multi-gene stacking, minimal position effects, and maternal inheritance for biocontainment. Critically, recent advances have progressed beyond proof-of-concept. Edible crops, such as lettuce, have bee

Soil salinization and alkalization severely threaten global agricultural sustainability. Although N-acetylcysteine (NAC) is well known as an antioxidant in animal research, its effects on plant salt-alkali tolerance and the underlying mechanisms remain unclear. This study investigated the regulatory effects of foliar-sprayed NAC on two soybean (Glycine max) cultivars-salt-tolerant HF50 and salt-sensitive HN95-under saline-alkali stress. Exogenous NAC alleviated seedling growth inhibition by supplying cysteine, which elevated glutamate-cysteine ligase (GCL) activity and promoted homoglutathione

Expression of OXIDATIVE SIGNAL-INDUCIBLE1 (OXI1) is induced by a number of stress conditions and regulates the interaction of plants with pathogenic and beneficial microbes. In this work, we generated Arabidopsis OXI1 knockout and genomic OXI1 overexpression lines and show by transcriptome, proteome, and metabolome analysis that OXI1 triggers ALD1, SARD4, and FMO1 expressions to promote the biosynthesis of pipecolic acid (Pip) and N-hydroxypipecolic acid (NHP). OXI1 contributes to enhanced immunity by induced SA biosynthesis via CBP60g-induced expression of SID2 and camalexin accumulation via

Genetic and genomic studies indicate that copper deficiency triggers changes in the expression of genes encoding key enzymes in various chloroplast-localized lipid/pigment biosynthetic pathways. Among these are CGL78 involved in chlorophyll biosynthesis and HPPD1, encoding 4-hydroxyphenylpyruvate dioxygenase catalyzing the committed step of plastoquinone and tocopherol biosyntheses. Copper deficiency in wild-type cells does not change the chlorophyll content, but a survey of chlorophyll protein accumulation in this situation revealed increased accumulation of LHCSR3, which is blocked at the le

Plant mitochondria and chloroplasts are sources of reactive oxygen species (ROS) that trigger oxidative DNA lesions. Among these, 8-oxo-7,8-dihydroguanine (8-oxodG) is particularly mutagenic because it exhibits ambiguous base pairing, as DNA polymerases efficiently incorporate adenine opposite 8-oxodG, leading to G·C→T·A transversions. Despite exposure to oxidative stress, plant organellar genomes exhibit some of the lowest nucleotide substitution rates in nature, implying that their DNA replication machinery may have evolved mechanisms to cope with oxidative lesions such as 8-oxodG. In family

Chloroplast development is an integral part of plant survival and growth, and occurs in parallel with chlorophyll biosynthesis. However, little is known about the mechanisms underlying chloroplast development in hexaploid wheat. Here, we obtained a spaceflight-induced wheat albino mutant mta. Chloroplast ultra-structural observation showed that chloroplasts of mta exhibit abnormal morphology and distribution compared to wild type. Photosynthetic pigments content was also significantly decreased in mta. Transcriptome and chloroplast proteome profiling of mta and wild type were done to identify

The ALAD gene encodes an enzyme that is essential for chlorophyll biosynthesis and is involved in many other physiological processes in plants. In this study, the CaALAD gene was cloned from pepper and sequenced. Multiple sequence alignment and phylogenetic analysis of ALAD proteins from nine plant species showed that ALAD is highly conserved, and that CaALAD shows the highest homology with the ALAD protein from eggplant. Subcellular localization indicated that the CaALAD protein is mainly localized to the chloroplasts. After transferring CaALAD into the Arabidopsis thaliana genome, cold toler

Phytoplasmas are unculturable, phloem-restricted bacterial pathogens responsible for devastating diseases in crops and ornamentals worldwide. Their mechanism for nutrient acquisition from host plants remains largely unknown. This study demonstrated that infection with potato purple top phytoplasma induced extensive remodeling of lipid metabolism in tomato plants, closely linked to autophagy activation. Western blot and confocal analyses revealed increased ATG8 lipidation and autophagosome formation at endoplasmic reticulum stress sites, alongside the redistribution of lipid droplets toward phy

The potent neurotoxin domoic acid is produced by some species of diatoms in the genus Pseudo-nitzschia, which can lead to amnesic shellfish poisoning (ASP) events in marine food webs. Of the toxin-producing Pseudo-nitzschia, P. australis has caused the most toxic and widespread events, making it a species of concern. Toxic and non-toxic Pseudo-nitzschia species cannot be distinguished easily by light microscopy, necessitating molecular testing. We designed a real-time quantitative PCR assay for rapid and accurate detection of P. australis in environmental DNA (eDNA) samples, targeting the inte

Malaria parasites have evolved unusual metabolic adaptations that specialize them for growth within heme-rich human erythrocytes. During blood-stage infection, Plasmodium falciparum parasites internalize and digest abundant host hemoglobin within the digestive vacuole. This massive catabolic process generates copious free heme, most of which is biomineralized into inert hemozoin. Parasites also express a divergent heme oxygenase (HO)-like protein (PfHO) that lacks key active-site residues and has lost canonical HO activity. The cellular role of this unusual protein that underpins its retention

Opium poppy, a member of the Papaveraceae family, is an ancient herbaceous plant and well-known medical resource in the pharmaceutical industry. However, opium poppies are grown worldwide for producing illicit drugs, significantly increasing the incidence of narcotic drug abuse. Since the narcotic poppy has not yet been genetically investigated, we characterized a novel variable number tandem repeat (VNTR) marker of forensically important poppy species based on the genetic analysis of 164 samples collected from two locations spanning the Jeolla province and Jeju island of South Korea. Respiratory burst oxidase homologs (RBOHs), also known as NADPH oxidases, contribute significantly to the production of ROS in plants, alongside other major sources such as photosynthesis and electron transport in chloroplasts. It has been shown that plant RBOHs play an active role in plant adversity response and electron transport. However, the phylogenetic analysis and characterization of the SlRBOH gene family in tomatoes have not been systematically studied. This study identified 11 SlRBOH genes in the tomato genome using a genome-wide search approach. The physicochemical properties, chrom

Microbial metabolites regulate plant growth and stress responses, yet their roles in crassulacean acid metabolism (CAM) plants remain insufficiently characterized. Here, we investigated the effects of metabolites from Bacillus licheniformis strain BlCP6, identified by 16S rDNA sequencing and whole-genome analysis, on growth and stress responses in Phalaenopsis orchids. Genome sequencing and metabolite profiling revealed that siderophores represent major bioactive compounds with direct plant growth-promoting potential, whereas osmoprotectants and extracellular lytic enzymes may contribute indir

The cuticle represents the outer most protective barrier against biotic and abiotic stresses. It is composed of cutin and waxes and protects plants from desiccation, UV, cold, mechanical stresses, and pathogens. GWAS/BSAseq combined with SeqSNP analyses in an association panel of 34 potato cultivars had revealed that the acyl-ACP thioesterase FATA (Soltu.DM.06G033680.1) is significantly associated with drought tolerance in potato. Apart from three FATB genes, only one FATA gene is present in potato that has the highest homology to FATA2 in Arabidopsis. FATA is responsible for the export of C18

The extraction of high-quality microbial DNA from environmental samples is critical for many downstream applications, including short- and long-read metagenomic sequencing. However, environmental DNA is prone to low recovery, degradation, and contamination by enzymatic inhibitors, with the extent of these issues largely dependent on the DNA purification method. The embedding of bacterial cells in a mucoid matrix within biofilms further complicates the process, making the study of algal symbionts particularly challenging. This study benchmarked five methods to recover microbial cells from biofi

The family Lauraceae, a hyper-diverse magnoliid family comprising approximately 63 genera and over 3,000 species, plays a key ecological role in tropical and subtropical forests. Yet deep relationships among its nine tribes remain unresolved, likely due to limited sampling and complex evolutionary processes such as incomplete lineage sorting (ILS) and gene flow. To address these challenges, we generated datasets of 255 single-copy nuclear genes and chloroplast genomes using a newly designed Lauraceae-specific probe set, achieving the most comprehensive genus-level sampling (84%) to date. Pentatricopeptide repeat (PPR) proteins bind RNA and are present in mitochondria and chloroplasts of Eukaryota. In fungi, they are responsible for controlling mitochondrial genome expression, mainly on the posttranscriptional level. Candida albicans is a human opportunistic pathogen with a facultative anaerobic metabolism which, unlike the model yeast Saccharomyces cerevisiae, possesses mitochondrially encoded respiratory Complex I (CI) subunits and does not tolerate loss of mtDNA. We characterized the function of 4 PPR proteins of C. albicans that lack orthologs in S. cerevisiae and found tha

A population encounters a variety of environmental stresses, so the full source of its resilience can only be captured by collecting all the signatures of adaptation to the selection of the local environment in its population history. Based on the multiomic data of Arabidopsis thaliana, we constructed a database of phenotypic adaptations (p-adaptations) and gene expression (e-adaptations) adaptations in the population. Through the enrichment analysis of the identified adaptations, we inferred a likely scenario of adaptation that is consistent with the biological evidence from experimental work

Jatropha mollissima is endemic to Brazil and is used for traditional medicinal purposes, including the treatment of snakebite. In this study, latex obtained from this plant was fractioned using reversed-phase chromatography, and the fractions were then screened for peptides. A 755 g/mol peptide was obtained, and MS/MS analyses indicated it had a cyclic sequence (Pro-Leu-Gly-Val-Leu-Leu-Tyr). This peptide sequence was present in the Jatropha genome database, and an identity value of 90.71%, an E-value of 0.0, and a score of 883 with NO-associated protein 1/chloroplastic/mitochondria of Jatropha
