In contrast to 15 or more validated and/or proposed tick-borne spotted fever group species, only three named medically important rickettsial species are associated with insects. These insect-borne rickettsiae are comprised of two highly pathogenic species, Rickettsia prowazekii (the agent of epidemic typhus) and R. typhi (the agent of murine typhus), as well as R. felis, a species with unconfirmed pathogenicity. Rickettsial association with obligate hematophagous insects such as the human body louse (R. prowazekii transmitted by Pediculus h. humanus) and several flea species (R. typhi and R. f

Tomato spotted wilt virus (TSWV) is a harmful pathogen that causes severe disease in tomato, pepper, and other horticultural and agronomic crops. Its genome comprises three linear single-stranded RNA molecules (segments L, M, and S), which are unequally packed in nucleocapsids. Although its genome structure and molecular mechanism of infection are well understood, the evolutionary dynamics of this virus require further analysis to determine the most probable viral ancestor, the date of divergence, the ancestral geographical source of dispersal, and the demographic history of TSWV. For this, we

Around 150 million years ago, eusocial termites evolved from within the cockroaches, 50 million years before eusocial Hymenoptera, such as bees and ants, appeared. Here, we report the 2-Gb genome of the German cockroach, Blattella germanica, and the 1.3-Gb genome of the drywood termite Cryptotermes secundus. We show evolutionary signatures of termite eusociality by comparing the genomes and transcriptomes of three termites and the cockroach against the background of 16 other eusocial and non-eusocial insects. Dramatic adaptive changes in genes underlying the production and perception of pherom

A wide diversity of pathogenic mosquito-borne viruses circulate in the Brazilian Amazon, and the intense deforestation can contribute to the spread of these viruses. In this context, this study aimed to investigate the viral diversity in mosquitoes of the genera Aedes, Culex, Haemagogus, and Sabethes from a transition area between the Amazon, Cerrado, and Caatinga biomes in Brazil. Metagenomic high-throughput sequencing was used to characterize the virome of 20 mosquito pools. A total of 15 virus-like genomes were identified, comprising species genomically close to insect-specific viruses of t

We present a draft sequence of the genome of Aedes aegypti, the primary vector for yellow fever and dengue fever, which at approximately 1376 million base pairs is about 5 times the size of the genome of the malaria vector Anopheles gambiae. Nearly 50% of the Ae. aegypti genome consists of transposable elements. These contribute to a factor of approximately 4 to 6 increase in average gene length and in sizes of intergenic regions relative to An. gambiae and Drosophila melanogaster. Nonetheless, chromosomal synteny is generally maintained among all three insects, although conservation of orthol

BACKGROUND: Fleas (Insecta: Siphonaptera) are small flightless parasites of birds and mammals; their blood-feeding can transmit many serious pathogens (i.e., the etiological agents of bubonic plague, endemic and murine typhus). The lack of flea genome assemblies has hindered research, especially comparisons to other disease vectors. Accordingly, we sequenced the genome of the cat flea, Ctenocephalides felis, an insect with substantial human health and veterinary importance across the globe.
RESULTS: By combining Illumina and PacBio sequencing of DNA derived from multiple inbred female fleas w

Schmallenberg virus (SBV) belongs to the Simbu serogroup within the family Peribunyaviridae, genus Orthobunyavirus and is transmitted by Culicoides biting midges. Infection of naïve ruminants in a critical phase of gestation may lead to severe congenital malformations. Sequence analysis from viremic animals revealed a very high genome stability. In contrast, sequence variations are frequently described for SBV from malformed fetuses. In addition to S segment mutations, especially within the M segment encoding the major immunogen Gc, point mutations or genomic deletions are also observed. Analy

Adjusting reproduction timing to environmental cues is essential for lifetime fitness. In many insects, reproductive diapause shows clinal variation along environmental gradients such as photoperiod and temperature. How such continuous trait variation may be encoded at the molecular level and maintained in the presence of gene flow remains largely elusive. The fruit fly Drosophila triauraria is distributed across a wide latitudinal range of the Japanese archipelago. Northern strains exhibit a strong photoperiodic reproductive diapause in females, whereas southern strains fail to arrest ovarian

Olfaction is critical to the host preference selection behavior of many disease-transmitting insects, including the mosquito Anopheles gambiae sensu stricto (hereafter A. gambiae), one of the major vectors for human malaria. In order to more fully understand the molecular biology of olfaction in this insect, we have previously identified several members member of a family of candidate odorant receptor proteins from A. gambiae (AgORs). Here we report the cloning and characterization of an additional AgOR gene, denoted as AgOr5, which shows significant similarity to putative odorant receptors in

Bacillus thuringiensis (Bt) is a well-known entomopathogenic bacterium widely used in biopesticide formulations due to its diverse arsenal of insecticidal toxins and environmental adaptability. However, the genetic diversity and virulence potential of native Bt strains from South Asia remain underexplored. In this study, we performed whole-genome sequencing and comparative genomic analysis of B. thuringiensis strain JSd1, isolated from Bangladeshi soil. The high-quality draft genome, assembled at 75x coverage, comprises 5.39 Mb in 64 contigs with a GC content of 35.28% and 99.26% completeness.

Sand flies are the natural vectors for the Leishmania species that produce a spectrum of diseases in their mammalian hosts, including humans. Studies of sand fly/Leishmania interactions have been limited by the absence of genome editing techniques applicable to these insects. In this report, we adapted CRISPR (clustered regularly interspaced palindromic repeat)/Cas9 (CRISPR-associated protein 9) technology to the Phlebotomus papatasi sand fly, a natural vector for Leishmania major, targeting the sand fly immune deficiency (IMD) pathway in order to decipher its contribution to vector competence

Vibrio cholerae is one of the deadliest pathogens in the history of humankind. It is the causative agent of cholera, a disease characterized by a profuse and watery diarrhoea that still today causes 95.000 deaths worldwide every year. V. cholerae is a free living marine organism that interacts with and infects a variety of organisms, from amoeba to humans, including insects and crustaceans. The complexity of the lifestyle and ecology of V. cholerae suggests a high genetic and phenotypic plasticity. In this review, we will focus on two peculiar genomic features that enhance genetic plasticity i

The insecticidal molecules of spiders persistently evolve to ensure rapid paralysis of their prey, and the best molecules are transmitted to their progeny. Here, we cloned two insecticidal peptides, Bs2 and Bs3, from the venom glands of the theraphosid Brachypelma smithi. Bs2 and Bs3 are 90.2% identical, but they exhibit interesting structural differences at their C-termini, including a connecting disulfide bond (residues Cys15-Cys36 for Bs2 and Cys15-Cys30 for Bs3). The genomic origin of Bs2 and Bs3 may be a cause for gene duplication events. Moreover, Bs2 differs in two residues from Tal1 (9

Olfaction contributes to many crucial behaviors in insects, such as foraging, locating hosts, mating, and avoiding predators. In the first step of the olfaction process in insects, odorant-binding proteins (OBPs) bind with the odorants and transport hydrophobic odorants. OBPs are also believed to accelerate the termination of the odorant response. The oriental fruit fly, Bactrocera dorsalis, is one of the most destructive fruit-eating pests, causing enormous economic losses to the fruit and vegetable industry worldwide. However, information relating to the number, diversity, and expression pat

Wolbachia pipientis is known to infect only arthropods and nematodes (mainly filarial worms). A unique feature shared by the two Phyla is the ability to replace the exoskeleton, a process known as ecdysis. This shared characteristic is thought to reflect a common ancestry. Arthropod moulting is induced by the steroid hormone 20-hydroxyecdysone (20E) and a role for ecdysteroids in nematode ecdysis has also been suggested. Removing Wolbachia from filarial worms impairs the host's development. From analyses of the genome of Wolbachia harbored by the filarial nematode Brugia malayi and that of its

BACKGROUND: The small hive beetle, Aethina tumida, is a rapidly emerging global pest of honey bee colonies. Small hive beetle infestation can be extremely destructive, which may cause honey bees to abscond and render colony infrastructure unusable. Due to the impacts small hive beetles have on honey bees, a wide variety of physical, cultural, and chemical control measures have been implemented to manage small hive beetle infestations. The use of insecticides to control small hive beetle populations is an emerging management tactic. Currently, very little genomic information exists on insectici

P-element-induced wimpy testis (PIWI) -interacting RNAs (piRNAs) are short non-coding RNAs crucial for silencing transposable elements (TEs) and maintaining genome stability, particularly in germ cells. While most research on piRNAs and PIWI proteins has traditionally focused on model organisms such as Drosophila, emerging studies are now extending to other insect orders. However, little is known about piRNAs in insects of the Hemiptera order. Here, we investigated the role of piRNAs in the brown planthopper (BPH), Nilaparvata lugens, a significant pest of rice that belongs to the Hemiptera or

BACKGROUND: Neuropeptides play important roles in insects, but in many cases their functions remain obscure. Comparative neuropeptidome analyses may provide clues to what these functions might be. Praying mantises are predators and close relatives of cockroaches that are scavengers. Cockroach neuropeptidomes are well established, but little is known about mantis neuropeptides. The recently published genome assembly of the praying mantis Tenodera sinensis makes it Possible to change that.
METHODS: The genome assembly from T. sinensis was analyzed for the presence of genes coding neuropeptides.

BACKGROUND: Transcription initiation regulation is mediated by sequence-specific interactions between DNA-binding proteins (transcription factors) and cis-elements, where BRE, TATA, INR, DPE and MTE motifs constitute canonical core motifs for basal transcription initiation of genes. Accurate identification of transcription start site (TSS) and their corresponding promoter regions is critical for delineation of these motifs. To this end, the genome scale analysis of core promoter architecture in insects has been confined to Drosophila. The recently sequenced Tsetse fly genome provides a unique
