Angiotensin-Converting Enzyme Inhibitors Used Concomitantly with Insulin Secretagogues and the Risk of Serious Hypoglycemia.

Serious hypoglycemia is a major adverse event associated with insulin secretagogues. Previous studies have suggested a potential relationship between angiotensin-converting enzyme inhibitors (ACEIs) used with sulfonylureas and serious hypoglycemia, and widely used drug compendia warn of this potential drug-drug interaction. We investigated the association between serious hypoglycemia and concomitant use of ACEIs in patients receiving insulin secretagogues, using the self-controlled case series design and Medicaid claims data from 5 US states linked to Medicare claims from 1999-2011. The exposu

Sexually Dimorphic Responses Reveal Multifaceted Benefits of Glibenclamide in Traumatic Brain Injury.

Sex disparities in traumatic brain injury (TBI) remain poorly understood. Previous data suggest that males are more susceptible to acute secondary injury processes and cell death, whereas females are more vulnerable chronically. Additional sex-based differences have been reported depending on injury model/severity and post-traumatic neurodegeneration. This gap in understanding limits therapy translation. We previously demonstrated sex-based differences in genetic modulation of a key pathway of secondary injury in TBI, sulfonylurea-receptor 1 (SUR1). Glibenclamide (GLI, SUR1-inhibitor) has show

Rapid Characterization of the Functional and Pharmacological Consequences of Cantú Syndrome KATP Channel Mutations in Intact Cells.

Gain-of-function of KATP channels, resulting from mutations in either KCNJ8 (encoding inward rectifier sub-family 6 [Kir6.1]) or ABCC9 (encoding sulphonylurea receptor [SUR2]), cause Cantú syndrome (CS), a channelopathy characterized by excess hair growth, coarse facial appearance, cardiomegaly, and lymphedema. Here, we established a pipeline for rapid analysis of CS mutation consequences in Landing pad HEK 293 cell lines stably expressing wild type (WT) and mutant human Kir6.1 and SUR2B. Cerebral blood flow during hemodilution and hypoxia in rats : role of ATP-sensitive potassium channels.

BACKGROUND AND PURPOSE: Hypoxia and hemodilution both reduce arterial oxygen content (CaO(2)) and increase cerebral blood flow (CBF), but the mechanisms by which hemodilution increases CBF are largely unknown. ATP-sensitive potassium (K(ATP)) channels are activated by intravascular hypoxia, and contribute to hypoxia-mediated cerebrovasodilatation. Although CaO(2) can be reduced to equal levels by hypoxia or hemodilution, intravascular PO(2) is reduced only during hypoxia. We therefore tested the hypothesis that K(ATP) channels would be unlikely to contribute to cerebrovasodilatation during hem

Uptake of relaxin in the uterus and cervix of rats in vivo: influence of ovarian steroids and tolerance.

The influence of oestradiol benzoate and progesterone treatment and of tolerance to relaxin on the uptake of 125I-labelled porcine relaxin by reproductive tissues was investigated in anaesthetized female rats. In ovary-intact rats, 125I-labelled relaxin uptake increased with time in reproductive tissues and in tissues concerned with metabolism and excretion. Administration of 50 micrograms unlabelled porcine relaxin before injection of 125I-labelled relaxin significantly reduced uptake of 125I-labelled relaxin into the uterus and cervix, but had no effect on uptake of 125I-labelled relaxin int

Clinical pharmacological studies of some possible interactions of lornoxicam with other drugs.

A series of studies in human subjects of potential interactions of lornoxicam with some other drugs is reviewed. No evidence of kinetic interaction was found with the antacids Maalox or Solugastrol, nor was there evidence of influence on antipyrine clearance. Lornoxicam reduced warfarin clearance and enhanced its hypoprothrombinaemic effect. It did not influence glibenclamide kinetics, but plasma insulin concentrations were significantly higher and plasma glucose concentrations significantly lower when lornoxicam and glibenclamide were co-administered. Lornoxicam produced a modest reduction in

Characterization of an important drug binding area on human serum albumin including the high-affinity binding sites of warfarin and azapropazone.

This paper reports a variety of experimental observations which strongly support the assumption that the warfarin binding site, or site I of human serum albumin, is better described as the warfarin-azapropazone binding area, consisting of the overlapping binding sites for warfarin and azapropazone. In general, drugs interacting with one of the two sites will also displace drugs bound to the other site, although their displacing potencies for both sites may vary considerably. This is most pronounced in the case of glibenclamide, which strongly inhibits the binding of drugs to the azapropazone s

Glibenclamide Serves as a Potent Vasopressor to Treat Vasoplegia After Cardiopulmonary Bypass and Reperfusion in a Porcine Model.

The hemodynamic stabilization of patients after complex cardiac surgery is a daily challenge. The use of high doses of catecholamines is common but has potential adverse effects. Glibenclamide, a KATP blocker, seems to attenuate vasoplegia in different animal models of septic shock. Therefore, the aim of this study was to investigate the impact of Glibenclamide on the vasoplegic syndrome after cardiopulmonary bypass in a porcine model. In this experimental study, 20 landrace pigs were randomized into two groups and examined: In the control group, standard medical therapy, including norepinephr

The effect of nitrazepam on depression and curiosity in behavioral tests in mice: The role of potassium channels.

Evidence show that gamma-aminobutyric acid (GABA) receptors are involved in depression, so the aim of this study was to investigate the effect of nitrazepam as agonist of GABAA receptors on depression and curiosity in male mice and the role of potassium channel in antidepressant-like response. For this purpose, we studied the antidepressant-like properties of fluoxetine, nitrazepam, glibenclamide, and cromakalim by both forced swimming test (FST) and tail suspension test (TST). Animals were injected by various doses of nitrazepam (0.05, 0.1, and 0.5mg/kg). Nitrazepam at dose of 0.5mg/kg signif

Histopathology of the endocrine pancreas of a fresh-water fish, Clarias batrachus (L.) IX: Effect of the new antidiabetic drug, glibenclamide (HB 419).

Oral administration of glibenclamide, the new antidiabetic drug, caused hypoglycemia in Clarias batrachus. The effect was maximal at 15 hours, when some fish had practically no sugar in their plasma. The hypoglycemic effect was insignificant in partially-pancreatectomized fish, indicating that the drug potentiated the effect of insulin in such animals. In fasting normal fish, the drug provoked an increase in the liver glycogen contents while in the diabetic animals, a reverse condition occurred. Significant beta-cell degranulation and vacuolisation was noted in the Langerhans' islets, the effe

Pharmacological Basis for the Antidiarrheal and Antispasmodic Effects of Cuminaldehyde in Experimental Animals: In Silico, Ex Vivo and In Vivo Studies.

BACKGROUND: Medicinal herbs are frequently used for the management of gastrointestinal disorders because they contain various compounds that can potentially amplify the intended therapeutic effects. Cuminaldehyde is a plant-based constituent found in oils derived from botanicals such as cumin, eucalyptus, myrrh, and cassia and is responsible for its health benefits. Despite the utilization of cuminaldehyde for several medicinal properties, there is currently insufficient scientific evidence to support its effectiveness in treating diarrhea. Hence, the present investigation was carried out to e

Vascular contraction of umbilical arteries of pregnant women with preeclampsia.

OBJECTIVE: Potassium channels have an important role in the vascular adaptation during pregnancy and a reduction in the expression of adenosine triphosphate-sensitive potassium channels (Katp) has been linked to preeclampsia. Activation of Katp induces vasodilation; however, no previous study has been conducted to evaluate the effects of the inhibition of these channels in the contractility of preeclamptic arteries. Glibenclamide is an oral antihyperglycemic agent that inhibits Katp and has been widely used in vascular studies. METHODS: To investigate the effects of the inhibition of Katp, um

Integrated control of coronary blood flow in exercising swine by adenosine, nitric oxide, and KATP channels.

The interplay of mechanisms regulating coronary blood flow (CBF) remains incompletely understood. Previous studies in dogs indicated that CBF regulation by KATP channels, adenosine, and nitric oxide (NO) follows a nonlinear redundancy design and fully accounted for exercise-induced coronary vasodilation. Conversely, in swine, these mechanisms appear to regulate CBF in a linear additive fashion with considerable exercise-induced vasodilation remaining when all three mechanisms are inhibited. A direct comparison between these studies is hampered by the different doses and administration routes (

Comparison of the effectiveness and safety of insulin and oral hypoglycemic drugs in the treatment of gestational diabetes mellitus: a meta-analysis of 26 randomized controlled trials.

OBJECTIVE: Oral hypoglycemic drugs for the treatment of gestational diabetes mellitus (GDM) are still controversial because they can pass through the placenta. The purpose of this meta-analysis is to evaluate the safety and effectiveness of oral hypoglycemic drugs. METHODS: PubMed, Ovid Embase, Web of Science, and Cochrane databases were systematically searched (inception to 20 April 2021). Rev Man 5.0 was used to analyze the data. A random-effects model was used to compute the summary risk estimates. RESULTS: There were 26 randomized controlled trials (RCTs) involving 4921 GDM patients whic

Expanded Physiologically-Based Pharmacokinetic Model of Rifampicin for Predicting Interactions With Drugs and an Endogenous Biomarker via Complex Mechanisms Including Organic Anion Transporting Polypeptide 1B Induction.

As rifampicin can cause the induction and inhibition of multiple metabolizing enzymes and transporters, it has been challenging to accurately predict the complex drug-drug interactions (DDIs). We previously constructed a physiologically-based pharmacokinetic (PBPK) model of rifampicin accounting for the components for the induction of cytochrome P450 (CYP) 3A/CYP2C9 and the inhibition of organic anion transporting polypeptide 1B (OATP1B). This study aimed to expand and verify the PBPK model for rifampicin by incorporating additional components for the induction of OATP1B and CYP2C8 and the inh
