Full thickness 3D in vitro conjunctiva model enables goblet cell differentiation.

In vitro culture and generation of highly specialized goblet cells is still a major challenge in conjunctival 3D in vitro equivalents. A model comprising all physiological factors, including mucus-secreting goblet cells has the potential to act as a new platform for studies on conjunctival diseases. We isolated primary conjunctival epithelial cells and fibroblasts from human biopsies. 3D models were generated from either epithelial layers or a combination of those with a connective tissue equivalent. Epithelial models were investigated for marker expression and barrier function. Full-thickness

Clinicopathological features of appendiceal goblet cell adenocarcinoma in Japan: a multicenter retrospective study.

PURPOSE: In the 5th edition of the World Health Organization classification, appendiceal goblet cell adenocarcinoma (GCA) is categorized separately from neuroendocrine tumors and other appendiceal adenocarcinomas. We clarified the clinicopathological characteristics of Japanese appendiceal GCA. METHODS: We designed a retrospective multicenter cohort study and retrieved the data of patients with appendiceal neoplasms and histologically diagnosed appendiceal goblet cell carcinoid (GCC) treated from January 2000 to December 2017 in Japan. The available GCC slides were reviewed and diagnosed with

Goblet Cell Carcinoma of the Appendix with Synchronous Adenocarcinoma of the Cecum: Distinct or Related Entities?

Goblet cell carcinoma (GCC) is a rare primary tumor of the appendix characterized by both epithelial and neuroendocrine components containing goblet cells. While in the past, the GCC has been associated with neuroendocrine tumors, recent studies consider that GCC is closer to adenocarcinoma than a neuroendocrine component. The association between gastro-intestinal (GI) carcinoids and second primary malignancies (SPMs) is widely described in the literature, but there is no reported case of GCC and synchronous adjacent adenocarcinoma of the colon. We describe the first case in the literature, to

Gastrointestinal Goblet Cell Adenocarcinomas Harbor Distinctive Clinicopathological, Immune, and Genomic Landscape.

Goblet cell adenocarcinoma (GCA) is a rare amphicrine tumor and difficult to diagnose. GCA is traditionally found in the appendix, but extra-appendiceal GCA may be underestimated. Intestinal adenocarcinoma with signet ring cell component is also very rare, and some signet ring cell carcinomas are well cohesive, having some similar morphological features to GCAs. It is necessary to differentiate GCA from intestinal adenocarcinomas with cohesive signet ring cell component (IACSRCC). The goal of this study is to find occurrence of extra-appendiceal GCA and characterize the histological, immunohis

Appendiceal goblet cell carcinomas have poor survival despite completion surgery.

PURPOSE: Appendiceal goblet cell carcinomas (aGCCs) are rare but aggressive tumours associated with significant mortality. We retrospectively reviewed the outcomes of aGCC patients treated at our tertiary referral centre. METHODS: We analysed aGCC patients, diagnosed between 1990-2016, assessing the impact of completion surgery and tumour factors on survival. Survival was assessed using Kaplan-Meier analysis. RESULTS: We identified 41 patients (23 F, 18 M); median age 61 (range 27-79) years. Mean tumour size was 10.5 (range 0.5-50) mm; most tumours were located in the appendiceal tip (n = 18

On the in vivo assessment of goblet cells of the human bulbar conjunctiva by confocal microscopy - A review.

BACKGROUND: In vivo confocal microscopy (IVCM) has been used for over 10 years to assess the goblet cell density (GCD) within the human conjunctiva, but the reported values have been variable with no obvious indications as to why. METHODS: From publications between 2008 and 2019, representative GCD values were extracted, as well as on the image sampling strategy used. RESULTS: Average GCD values for any particular group of individuals ranged from 7 to 979 goblet cells / sq. mm, and with one notable outlier removed, an overall group-mean value for GCD (+/- SD) from single site locations was 2

Advanced appendiceal goblet cell carcinoids with intestinal obstruction: two case reports.

A goblet cell carcinoid is quite rare and has features, wherein, a carcinoid-like image and an adenocarcinoma-like image coexist. We encountered two cases of rare goblet cell carcinoid originating in the appendix. Case 1 is that of a 48-year-old man with a chief complaint of abdominal distension and case 2 is that of a 64-year-old woman with a chief complaint of constipation. At the time of diagnosis, both cases had already metastasized to the peritoneum and other organs, and no radical surgical treatment could be administered in either case. Chemotherapies were performed according to the regi

Conjunctival epithelial and goblet cell function in chronic inflammation and ocular allergic inflammation.

PURPOSE OF REVIEW: Although conjunctival goblet cells are a major cell type in ocular mucosa, their responses during ocular allergy are largely unexplored. This review summarizes the recent findings that provide key insights into the mechanisms by which their function and survival are altered during chronic inflammatory responses, including ocular allergy. RECENT FINDINGS: Conjunctiva represents a major component of the ocular mucosa that harbors specialized lymphoid tissue. Exposure of mucin-secreting goblet cells to allergic and inflammatory mediators released by the local innate and adapti

The monoclonal antibody HCM31 specifically recognises the Sd(a) tetrasaccharide in goblet cell mucin.

Rat small intestinal goblet cell mucins reacting with monoclonal antibody HCM31 increase significantly during regeneration from experimental mucosal damage and at the period of expulsion of parasitic nematode, Nippostrongylus brasiliensis (N.b). The reduction in reactivity of HCM31 with mucin upon neuraminidase treatment, suggested that HCM31 recognizes sialylated oligosaccharide on mucin. HCM31-reactive sialomucins are therefore considered to play an important role in the physiological and pathological changes in the gastrointestinal mucosa. To determine the epitope for HCM31, oligosaccharide

Role of histamine and its receptor subtypes in stimulation of conjunctival goblet cell secretion.

PURPOSE: The purpose of this study was to determine the effect of histamine and its receptors on goblet cell secretion. METHODS: Cultured rat and human goblet cells were grown in RPMI 1640. Goblet cell secretion of high molecular weight glycoconjugate was measured by an enzyme-linked lectin assay. Cultured rat goblet cells were homogenized and either RNA was isolated for RT-PCR or proteins were isolated for Western blot analysis for presence of histamine receptors subtypes H₁ through H₄. The localization of these receptors was determined in rat and human goblet cells by immunofluorescence mic

Goblet cell carcinoids at extraappendiceal locations of gastrointestinal tract: an underrecognized diagnostic pitfall.

BACKGROUND: Goblet cell carcinoid (GCC) is a clinicopathologically distinctive tumor that typically arises in appendix and metastasizes frequently. Although rare cases of ostensibly primary extraappendiceal GCC (EGCC) have been reported, the distinction from extraappendiceal metastasis of occult appendiceal primary may be problematic and has not been dealt with systematically in literature. METHODS: We reviewed our combined experience with EGCC at four North American hospitals and reevaluated all EGCC cases published in literature. RESULTS: We encountered 16 cases that were initially reporte

Application of light microscopical and ultrastructural immunohistochemistry in the study of goblet cell carcinoid in the appendix.

BACKGROUND: Goblet cell carcinoids appear less frequently in the appendix than do other carcinoids. In the presented work a case with a goblet cell carcinoid of the appendix is described. METHODS: Routine histological and histochemical methods were employed, with a combination of histochemistry and immunohistochemistry on one section and light and electron microscopical immunohistochemisty on paraffin-embedded material, were applied to identify the type of the carcinoid and to reveal the fine structure of cell types in the tumour nests of the appendix. RESULTS: During the biopsy of a patient

Adenocarcinoid tumor, goblet cell type, arising in a ureteroileal conduit: a case report.

The adenocarcinoid tumor is a potentially malignant neoplasm usually arising in the appendix. We describe a 55-year-old woman with a ureteroileal conduit in place for 12 years who suffered obstruction of the ileal segment of the conduit, with recurrent pyelonephritis and abdominal pain. Pathological examination of a segmentally resected portion of the conduit revealed a submucosal proliferation of nests of argentaffin/argyrophil cells and large, mucin-distended goblet cells. The patient showed no evidence of metastasis and has done well to date following excision. A discussion of neoplasms ari

The fine structural localization of peroxidase activity in goblet cells of the conjunctival epithelium of rats.

Cytochemical investigation of peroxidase was made on the conjunctival goblet cells, using the diaminobenzidine method. Many goblet cells in the conjunctiva demonstrated this reaction. The reaction was present in secretory granules as well as in the perinuclear cisterna, the rough endoplasmic reticulum, several Golgi saccules and adjacent small smooth-surfaced vesicles, and condensing vacuoles. Each secretory granule within a goblet cell showed a different degree of reaction, hence it is concluded that some goblet cells probably produce both mucous substance and peroxidase in one cell. The mode

Acomparison of changes in the mucous glands and goblet cells of nasal, sinus, and bronchial mucosa.

A comparison of mucous gland size in the bronchial and nasal mucosa was performed on material from 24 random necropsies. The Reid index and counts of mucous gland acini were used as measures of hypertrophy. In 14 of these cases the relationship between goblet cell hyperplasia in the sinus mucosa and the presence of chronic bronchitis was investigated by comparing the proportion of goblet cells with the corresponding Reid index. No significant correlation was found between nasal and bronchial mucous gland size, and no association was demonstrated between the proportion of goblet cells in the si

Autophagy is required during high MUC2 mucin biosynthesis in colonic goblet cells to contend metabolic stress.

Goblet cells are specialized for the production and secretion of MUC2 glycoproteins that forms a thick layer covering the mucosal epithelium as a protective barrier against noxious substances and invading microbes. High MUC2 mucin biosynthesis induces endoplasmic reticulum (ER) stress and apoptosis in goblet cells during inflammatory and infectious diseases. Autophagy is an intracellular degradation process required for maintenance of intestinal homeostasis. In this study, we hypothesized that autophagy was triggered during high MUC2 mucin biosynthesis from colonic goblet cells to cope with me

Moxifloxacin based axially swept wide-field fluorescence microscopy for high-speed imaging of conjunctival goblet cells.

Goblet cells (GCs) in the conjunctiva are specialized epithelial cells producing mucins on the ocular surface. GCs play important roles in maintaining homeostasis of the ocular surface, and GC dysfunction is associated with various complications including dry eye diseases. Current GC examination methods, which are conjunctival impression cytology and confocal reflection microscopy, have limitations for routine examination. Fluorescence microscopy using moxifloxacin was recently introduced as a non-invasive and high-contrast imaging method, but further development is needed to be used for GC ex
