Metallo-beta-lactamases in Gram-negative bacteria: introducing the era of pan-resistance?

Metallo-beta-lactamases (MBLs) are being reported with increasing frequency and from several countries worldwide and are becoming the prevalent and most clinically significant determinants of carbapenem resistance. Furthermore, MBL-producing strains that exhibit a pan-resistant phenotype are increasingly detected. Initially MBLs were detected in Pseudomonas aeruginosa, however nowadays they are frequently found in Acinetobacter baumannii, Klebsiella pneumoniae and other Enterobacteriaceae. MBLs spread easily on plasmids and cause nosocomial infections and outbreaks with excess mortality. Such

[Clinically important beta-lactamases of gram-negative bacteria: extended-spectrum beta-lactamases (ESBL)].

Beta-lactamases are the commonest cause of a resistance of gram-negative bacteria to beta-lactam antimicrobial agents. The introduction of the third- and fourth-generation cephalosporins into clinical practise is the reason of an evolution of new beta-lactamases being able to hydrolyze these antibiotics. Extended-spectrum beta-lactamases (ESBLs) are the major group of these enzymes. Most of the ESBLs are mainly structural mutants of penicillinases TEM-1, TEM-2 and SHV-1. For several reasons, ESBL-producing isolates should, by definition, be reported as resistant to all penicillins, cephalospor

Multidrug resistance in Gram-negative bacteria that produce extended-spectrum beta-lactamases (ESBLs).

In 1983, just two years after the introduction of the oxymino-beta-lactams to the market , the first extended-spectrum beta-lactamases were isolated in Germany from Klebsiella pneumoniae strains. Since then several outbreaks have been reported in many European countries and the USA, and nowadays in several places worldwide the problem seems to reach endemic dimensions, with rates exceeding 50% in some countries, such as Portugal and Turkey. On the other hand not only K. pneumoniae but also Escherichia coli strains, with Enterobacter aerogenes predominating among the other enterobacteriaceal sp

Bacteremias in liver transplant recipients: shift toward gram-negative bacteria as predominant pathogens.

During the 1990s, gram-positive bacteria emerged as major pathogens after liver transplantation. We sought to determine whether the pathogens associated with bacteremias in liver transplant recipients have changed. Patients included 233 liver transplant recipients transplanted between 1989 and 2003. The proportion of all infections due to bacteremias increased significantly over time (P <.0001). Of other major infections, a trend toward a decrease in fungal infections (P =.089) and a significant decrease in cytomegalovirus (CMV) disease (P =.0004) were documented. Whereas the proportion of bac

Gram-negative bacteria can induce contact lens related acute red eye (CLARE) responses.

PURPOSE: Twelve volunteers participated in a study designed to measure the overnight corneal edema response with a variety of hydrogel contact lenses. During the study four subjects (5 eyes) experienced a contact lens related acute red eye (CLARE) reaction, which manifested as severe ocular pain, photophobia, corneal infiltration, and conjunctival hyperemia. An additional five subjects (7 eyes) developed corneal infiltrates only. Twelve eyes (of 9 subjects) showed no response. METHODS AND RESULTS: Upon microbiological examination of the contact lenses and storage solutions, gram-negative bact

Prevalence of multidrug resistant gram-negative bacteria and associated factors among gram-negative blood culture isolates at Tikur Anbessa Specialized Hospital: a retrospective study.

BACKGROUND: The rising incidence of multidrug-resistant (MDR) gram-negative bacteria causing bloodstream infections (BSI) is a significant concern, particularly in Sub-Saharan Africa, where inadequate control over antibiotic access, poor antimicrobial stewardship, and weak infection prevention measures worsen the issue. This study aims to assess prevalence, bacteriologic profile, and associated risk factors of MDR-gram negative isolates and carbapenem resistant gram-negative BSI admitted patients of Tikur Anbessa Specialized Comprehensive Hospital (TASH) a tertiary hospital in Adiss Ababa that

Insights into membrane disruption and oxidative stress in Gram-positive and Gram-negative bacteria: role of sodium benzoate and phenoxyethanol.

This study investigates the antibacterial effects of two widely used preservatives in foods, pharmaceuticals, and personal care products, sodium benzoate and phenoxyethanol, on Gram-positive bacteria Staphylococcus aureus (ATCC 6538) and Gram-negative bacteria Pseudomonas aeruginosa (MTCC 1688). Despite their widespread usage, the exact antibacterial mechanisms of these preservatives are not yet fully understood. Our study demonstrated that the presence of SB and POE lead to extensive cellular rupture, membrane damage, and intracellular content leakage in bacteria. The results were validated t

Reducing the effective dose of doxycycline using chitosan silver nanocomposite as a carriers on gram positive and gram-negative bacteria.

Doxycycline (Doxy) is a tetracycline antibiotic with a potent antibacterial activity against a broad range of bacteria. Using nanotechnology is one feasible way to increase the antibiotics' ability to penetrate the body and increase their antibacterial effectiveness. In this work, we report the formation of a stable green synthesized silver nanoparticles (AgNPs) by chitosan with Doxy nanocomposite for the first time. The obtained nanoparticles were characterized by transmission electron microscopy (TEM), zeta-potential, UV-Visible spectroscopy and four transform infrared spectroscopy (FTIRs).

Research note: Antimicrobial properties of commercial black soldier fly (Hermetia illucens) meal against gram-positive and gram-negative bacteria.

Black soldier fly larvae meal (BSFM) is increasingly recognised as a sustainable and functional protein source in the livestock feed industry, owing to its high crude protein and rich fatty acid content. Beyond its nutritional value, BSFM has demonstrated antibacterial properties, enhancing resistance in animals to pathogens such as Salmonella species. These findings are critical as antimicrobial resistance continues to rise in the livestock sector. The black soldier fly (BSF) genome encodes antimicrobial peptides (AMPs) that can be classified under families such as cecropins, defensins, and a

Differentiating Live Versus Dead Gram-Positive and Gram-Negative Bacteria With and Without Oxidative Stress Using Buoyant Mass Measurements.

Expeditious and accurate determination of pathogenic bacteria cell viability is of great importance to public health for numerous areas including medical diagnostics, food safety, and environmental monitoring. In this work a cell buoyant mass classifier approach is presented to assess bacteria cell viability in real time. Buoyant mass measurements for live and dead Gram-positive and Gram-negative bacteria populations were acquired with a commercial suspended microchannel resonator, Archimedes, to generate receiver operating characteristic (ROC) curves. To quantitatively assess the difference i

Filamentous forms of Gram-negative bacteria on microscopy: Report of a case and review of literature.

Gram-negative bacteria (GNB) are known to undergo morphological changes like elongation, abrupt bulge formation at the site of potential cell division and bulge stagnation following exposure to β-lactam antibiotics. Positive blood cultures of a 78-year-old man on Gram staining showed a long filamentous GNB with spherical enlargement along some bacterial filaments. Cultures on solid media grew pan sensitive K. pneumoniae of normal morphology. A total of 16 cases of GNB showing filamentous or spheroplast forms have been reported in literature. Clinicians and microbiologists should be aware of po
