Pharmacological targeting of the KIT growth factor receptor: a therapeutic consideration for mast cell disorders.

KIT is a member of the tyrosine kinase family of growth factor receptors which is expressed on a variety of haematopoietic cells including mast cells. Stem cell factor (SCF)-dependent activation of KIT is critical for mast cell homeostasis and function. However, when KIT is inappropriately activated, accumulation of mast cells in tissues results in mastocytosis. Such dysregulated KIT activation is a manifestation of specific activating point mutations within KIT, with the human D816V mutation considered as a hallmark of human systemic mastocytosis. A number of other activating mutations in KIT

Zebrafish gon4la mutants recapitulate human GON4L-related growth disorders and reveal novel metabolic organs abnormalities.

Biallelic mutations in human GON4L have been linked to short stature, craniofacial abnormalities, and microcephaly, yet the underlying mechanisms remain unclear. GON4L is a nuclear protein composed of a YY1AP1-like region, two PAH domains, and one SANT domain, all implicated in transcriptional or chromatin regulation. Here, we define the function of Gon4la (a zebrafish ortholog of human GON4L) in post-embryonic growth and tissue homeostasis. We generated three gon4la mutant lines using CRISPR-Cas9, each predicted to express truncated proteins lacking the C-terminal PAH/SANT domains and differi

Application of artificial intelligence in diagnosis and management of fetal growth disorders: a comprehensive review.

Fetal growth disorders, including both fetal growth restriction and macrosomia, remain major contributors to perinatal morbidity and long-term health risks in adulthood. While ultrasound is the most frequently employed technique for the diagnosis of intrauterine growth abnormalities, its efficacy is constrained by the operator's experience and variable accuracy. This review explores the role of artificial intelligence (AI) in advancing the detection and management of fetal growth disorders. We conducted a comprehensive literature search of major databases to identify original and review articl

The ElonginB/C-Cullin5-SOCS-Box-Complex Is a Potential Biomarker for Growth Hormone Disorders.

Insulin-like growth factor 1 (IGF-1) is the standard biochemical marker for the diagnosis and treatment control of acromegaly and growth hormone deficiency (GHD). However, its limitations necessitate the screening for new specific and sensitive biomarkers. The elonginB/C-cullin5-SOCS-box-complex (ECS-complex) (an intracellular five-protein complex) is stimulated by circulating growth hormone (GH) and regulates GH receptor levels through a negative feedback loop. It mediates the cells' sensitivity for GH and therefore, represents a potent new biomarker for those diseases. In this study, individ

Connective Tissue Disorders and Cardiovascular Complications: The Indomitable Role of Transforming Growth Factor-β Signaling.

Marfan Syndrome (MFS) and Loeys-Dietz Syndrome (LDS) represent heritable connective tissue disorders that segregate with a similar pattern of cardiovascular defects (thoracic aortic aneurysm, mitral valve prolapse/regurgitation, and aortic dilatation with regurgitation). This pattern of cardiovascular defects appears to be expressed along a spectrum of severity in many heritable connective tissue disorders and raises suspicion of a relationship between the normal development of connective tissues and the cardiovascular system. With overwhelming evidence of the involvement of aberrant Transform

Impact of Pituitary Autoimmunity and Genetic Disorders on Growth Hormone Deficiency in Children and Adults.

Growth hormone (GH), mostly through its peripheral mediator, the insulin-like growth factor 1(IGF1), in addition to carrying out its fundamental action to promote linear bone growth, plays an important role throughout life in the regulation of intermediate metabolism, trophism and function of various organs, especially the cardiovascular, muscular and skeletal systems. Therefore, if a prepubertal GH secretory deficiency (GHD) is responsible for short stature, then a deficiency in adulthood identifies a nosographic picture classified as adult GHD syndrome, which is characterized by heart, muscl

Epidemiology of male and female reproductive disorders and impact on fertility regulation and population growth.

The National Institute of Child Health and Human Development has conducted a series of vision workshops to set an ambitious but tenable broad agenda, transcending traditional organizational and disciplinary boundaries. This presentation sets forth a vision regarding how the aspects of a woman's reproductive lifespan are interrelated, likely starting from childhood all the way to the later reproductive years. As a woman propagates, this circular interaction is then transmitted to the next generation. We need an understanding of the clinical processes, the time course, the natural history, and d

[Growth disorders of the upper end of the femur following treatment of congenital hip luxation].

From approximately 2,500 cases of treated congenital dislocation of the hip, the authors have selected 82 cases with 100 hips showing disturbance of the superior epiphysis and metaphysis of the femur. These have been observed after several types of treatment - napkins in abduction, abduction harnesses, abduction splintage, closed reduction by Lorenz technique, progressive reduction by Somerville technique or open reduction. The risk factors have been analysed in 22 hips treated by progressive reduction. It is concluded that changes in the metaphysis and in the growth plate are the main cause o

From challenge to growth: A qualitative study of parental adaptation to Autism Spectrum Disorder.

Raising a child with Autism Spectrum Disorder (ASD) profoundly affects family dynamics and parental well-being. While research has often focused on stress and negative outcomes, less is known about the processes and resources that foster adaptation and growth in parents. This qualitative study explored the adaptation process among parents of children with ASD, examining both challenges and transformative experiences, and identifying key personal and contextual factors that support or hinder adjustment. Thirty-six parents (19 mothers, 17 fathers) of children aged 5-11 years with ASD (severity l
