Haemonchus contortus (family Trichostrongylidae, Nematoda), a hematophagous gastrointestinal parasite found in small ruminants, has a great ability to develop resistance to anthelmintic drugs. We studied the biotransformation of the three benzimidazole anthelmintics: albendazole (ABZ), ricobendazole (albendazole S-oxide; RCB) and flubendazole (FLU) in females and males of H. contortus in both a susceptible ISE strain and resistant IRE strain. The ex vivo cultivation of living nematodes in culture medium with or without the anthelmintics was used. Ultrasensitive UHPLC/MS/MS analysis revealed 9,

Associations between intensity and frequency of Cryptosporidium and Giardia shedding with growth, carcase weight and dressing% were investigated using a longitudinal study of 1182 lambs on eight Australian farms. Live weight was recorded and faecal samples were collected on three sampling occasions; weaning (approximately 12 weeks of age), post-weaning (approximately 19 weeks) and pre-slaughter (approximately 29 weeks). Hot standard carcase weight (HSCW) and dressing% were measured at slaughter. Faecal samples were screened for presence and concentration of Cryptosporidium, Giardia and Haemonc

Whole gut homogenates of the blood-sucking nematode Haemonchus contortus induce protective immunity in goats, and some of these gut Ag are conserved among related parasitic nematode species. To identify gut Ag that induce protective immunity and have phylogenetically conserved epitopes, mAb were made to gut-surface Ag of H. contortus. Forty-nine mAb reacted with microvilli of the parasite gut. Two of these mAb (42/10.6.1 and 42/53.3.5) were analyzed here. Both of the mAb bound to the microvillar surface of freshly isolated gut, and each mAb recognized carbohydrate epitopes, based on sensitivit

Recent detailed studies of the physiologic changes occurring in cattle infected with some of the common gastrointestinal nematodes have shown that the weight loss and reduced carcass values resulting from these infections are a consequence of major alterations in host metabolism. Dramatic changes in protein metabolism were consistently found to involve decreased muscle synthesis and increased urinary nitrogen excretion. Because of the well-recognized loss of plasma proteins into the gut lumen and cellular damage at the mucosa occurring during nematode infections of the abomasum and duodenum, i

The objectives of the study were to determine the efficacy of the manufacturer's recommended dose (1 mg/kg body weight) and double dose (2 mg/kg body weight) of pour-on formulation of eprinomectin (EPN) on three sheep and six goat farms and to compare the EPN efficacy between both host species on two mixed farms. The efficacy of EPN against gastrointestinal nematodes (GINs) was evaluated by the faecal egg count reduction test. According to the WAAVP recommendations, the farms were evaluated as "resistant", "inconclusive", or "susceptible". The composition of the GINs was determined on the day

Anthelmintic resistance in livestock is a growing concern worldwide, with significant implications for animal health and agricultural productivity. This study explores the perceptions of veterinarians and farmers in Bosnia and Herzegovina regarding the factors contributing to anthelmintic resistance in Haemonchus contortus nematodes. Data were collected through structured questionnaires completed by 106 veterinarians and 188 farmers in 2022 and 2023. The analysis focused on self-reported therapeutic practices, farm management and environmental variables. Logistic regression, including Firth's

Small ruminants (sheep and goats) constantly suffer from endoparasitoses caused by gastrointestinal nematodes. Among these, the species Haemonchus contortus (Rudolphi, 1803) is considered to be the one of greatest importance within sheep farming. This nematode is difficult to control due to its resistance to most commercial anthelmintics. The aim of the present study was to assess the potential of macrochelid mites as macrobiological agents for controlling endoparasitoses of sheep caused by the nematode, H. contortus. For this, novel in vitro methodology was used, in which assessments were mad

Haemonchus contortus is one of the most pathogenic gastrointestinal parasites that infect small ruminants. The indiscriminate use of anthelmintics (i.e., benzimidazole class, BZ) to control infections has led to the reduction of drug efficacy in H. contortus populations worldwide. Resistance to BZ is associated with high frequencies of single nucleotide polymorphisms at F200Y, F167Y, and E198A positions of the β-tubulin isotype 1 gene. This study aimed to determine the frequency of single nucleotide polymorphisms associated with BZ resistance in H. contortus from 18 farms (545 sheep and 124 go

The purpose of the present study was to assay the in vitro and in vivo anthelmintic activity (AA) of Caesalpinia coriaria (Cc) mature fruits against the nematode Haemonchus contortus (Hc). The Hc infective larvae were used to assess the in vitro AA through larval mortality assay. The exposure of larvae to the different treatments was performed in 96-well microtitration plates. The treatments were as follows: hydroalcoholic extract (HA-E, at 25-100 mg/mL), aqueous fraction (Aq-F, at 12.5-50 mg/mL), organic fraction (EtOAc-F at 12.5-50 mg/mL), compounds (1, methyl gallate and 2, gallic acid at 1

Haemonchosis is a gastrointestinal parasitic disease of economic importance in ruminants especially sheep and goats. In view of the rising costs of conventional veterinary anthelmintics and the development of resistance by Haemonchus contortus, there is a need to develop alternative ethnoveterinary therapies for the treatment and management of Haemonchosis. This study investigated the anthelmintic activity of Dennettia tripetala G. Baker (Annonaceae) fruits against Haemonchus contortus in red Sokoto goats. The maximum tolerated dose was determined in adult albino rats administered, 3000, 4000,

Haemonchus contortus is a globally distributed and economically important gastrointestinal pathogen of small ruminants and has become a key nematode model for studying anthelmintic resistance and other parasite-specific traits among a wider group of parasites including major human pathogens. Here, we report using PacBio long-read and OpGen and 10X Genomics long-molecule methods to generate a highly contiguous 283.4 Mbp chromosome-scale genome assembly including a resolved sex chromosome for the MHco3(ISE).N1 isolate. We show a remarkable pattern of conservation of chromosome content with Caeno

BACKGROUND: Since pastoralists in South Darfur, Sudan, had complained about lack of albendazole (ABZ) efficacy to control nematodes in goats, the frequency of infection with gastrointestinal helminths was studied before in vivo faecal egg count reduction tests (FECRT) were conducted using ABZ orally either at the dose recommended for sheep, 5 mg/kg body weight (bw) or at 10 mg/kg bw. Experiments included goats naturally infected with gastrointestinal nematodes or experimentally infected with local Haemonchus contortus isolates. Three study areas (Nyala, Beleil and Kass) were visited in autumn

Anthelmintic resistance was explored by fecal egg count reduction test in a sheep flock from the French Pyrenees at the request of the veterinary practitioner after a poor response to anthelmintics was noted. The FECRT confirmed the suspicion with a mean percentage of reduction in egg excretions of 45% (CI 95%: - 40 to 78.5) and 0% (CI95%: - 162 to 49) within the ivermectin and the benzimidazole groups respectively. Haemonchus contortus was shown to be the IVM and BZ resistant species after morphological and molecular characterizations whereas Teladorsagia circumcincta was probably resistant t

Nicotinic acetylcholine receptors (nAChRs) play a key role in the normal physiology of nematodes and provide an established target site for anthelmintics. The free-living nematode, Caenorhabditis elegans, has a large number of nAChR subunit genes in its genome and so provides an experimental model for testing novel anthelmintics which act at these sites. However, many parasitic nematodes lack specific genes present in C. elegans, and so care is required in extrapolating from studies using C. elegans to the situation in other nematodes. In this review the properties of C. elegans nAChRs are rev

Recently, research for alternative methods to combat gastrointestinal parasites has increased, and the biological control activity of the fungus Duddingtonia flagrans stands out. In this study, the possible influence of temperature on the nematophagous activity of D. flagrans, after gastrointestinal passage, against Haemonchus contortus in sheep was analysed. Four female sheep, between 2 and 3 years of age and weighing between 40 and 50 kg, were used. Two sheep were parasitised with H. contortus, while two other sheep were dewormed. Before the collection of faeces, one of the dewormed animals

This article is the first of a series aimed at developing specific decision support software for on-farm optimisation of sustainable integrated management of haemonchosis. It contains a concept framework for such a system for use by farmers and/or their advisors but, as reported in the series, only the first steps have been taken on the road to achieve this goal. Anthelmintic resistance has reached such levels of prevalence and intensity that recently it evoked the comment that for small ruminants the final phase of resistance was being entered, without effective chemotherapeutic agents on som

Haemonchosis is a fatal disease of livestock caused by Haemonchus contortus (HC) - a blood-sucking parasite of the abomasum. This parasite is quite prevalent in sheep, causing mortality and production losses. The suppressor of cytokine signalling-2 (SOCS2) gene plays a vital role in sheep's immune response against gastrointestinal parasites. This study aimed to estimate the parasitic load of HC in three sheep breeds (Balkhi, Ghalji, and Michni) through faecal egg count and to identify SNPs in the SOCS2 gene associated with the susceptibility of sheep against HC. The results showed that the mea

This experiment aimed to assess the regulatory effects of treatment with Balanites aegyptiaca fruit ethanol extract (BA-EE) on oxidant/antioxidant status, anti-inflammatory cytokines, and cell apoptosis gene expression in the abomasum of Haemonchus contortus-infected goats. Twenty goat kids were assigned randomly to four equal groups: (G1) infected-untreated, (G2) uninfected-BA-EE-treated, (G3) infected-albendazole-treated, (G4) infected-BA-EE-treated. Each goat in (G1), (G3), and (G4) was orally infected with 10,000 infective third-stage larvae. In the fifth week postinfection, single doses o

Haemonchus contortus (H. contortus) ADP-ribosylation factor 1 (Hc-ARF1) and Hepatocellular carcinoma-associated antigen 59 (Hc-HCA59) are recognized to largely regulate the immune responses of host cells. However, studies about the protective efficacy of the two molecules are poorly unknown. In this research, combinations of recombinant Hc-HCA59 (rHc-HCA59) and Hc-ARF1 (rHc-ARF1) proteins were amalgamated with poly (lactic-co-glycolic acid) (PLGA) nanoparticles adjuvant in order to investigate their protection potential against H. contortus in goats. The results demonstrated that the levels of
