Distinctive pharmacological profile of a nonadrenergic inhibitory system in bullfrog lung.

1 Bullfrog hemilungs, pretreated with atropine, are markedly relaxed on addition of carbachol. Since the relaxant effect is inhibited by tetrodotoxin or hexamethonium, it is neurally mediated and involves stimulation of nicotinic receptors with release of an unknown inhibitory transmitter.2 Carbachol-induced relaxation is nonadrenergic since: (a) it considerably exceeds the maximal effects of isoprenaline or the effect of 10(-3)M adrenaline or noradrenaline; (b) it elicits marked further relaxation in preparations already relaxed by high concentrations of catecholamines; (c) it is not attenuat

Atropine-sensitive gastric excitation by stimulation of thoracic dorsal roots--antidromic activation of afferents?

In chloralosed cats, usually with ligated adrenals and paralysed with gallamine, the thoracic dorsal roots were electrically stimulated and gastric motor responses were recorded. Thoracic dorsal root stimulation regularly elicited gastric contractions after pretreatment with guanethidine and/or hexamethonium which were readily blocked by atropine. It was established that these gastric motor responses were due to peripheral stimulation of the dorsal root and not to spread to the ventral roots. Together with other results (Delbro & Lisander 1980) the findings suggest an antidromic activation of

Diabetes-induced alterations of autonomic nerve function in the cat.

The susceptibility to competitive ganglionic blocking agents such as hexamethonium (C6), tetraethylammonium bromide (TEAB), mecamylamine and d-tubocurarine (d-TC), of the superior cervical ganglion in cats with pancreatectomy and spontaneous diabetes or in animals treated with contrainsular drugs such as cortisone or dihydrochlorothiazide, was found to be decreased as compared to the reactivity of normal controls. The increased tolerance to ganglioplegics was not correlated with the elevation of the blood sugar level, and proved to be resistant to an acute administration of insulin. The result

Muscarinic involvement in vascular and adrenal medullary responses to splanchnic nerve stimulation in conscious calves.

Stimulation of the peripheral end of the right splanchnic nerve (4 Hz for 10 min) in the presence of hexamethonium caused a small but significant rise in mean aortic blood pressure which was subsequently abolished by atropine. There were also small but significant increases in the outputs of catecholamines, [Met5]-enkephalins and corticotrophin releasing factor (CRF) from the right adrenal gland. The catecholamine response was roughly halved after atropine while the outputs of enkephalins and CRF were unaffected. It is concluded that splanchnic sympathetic postganglionic neurones supplying the

VIP antisera inhibit the relaxatory motor responses of the feline sphincter of Oddi and gall-bladder induced by VIP or vagal nerve stimulation.

Regional administration of VIP elicited a dose-dependent relaxation of the feline sphincter of Oddi and gall-bladder. Relaxatory motor responses of these regions at efferent electrical stimulation of the vagal nerves were unmasked after atropine (resistant to propranolol but sensitive to hexamethonium). These findings in combination with the presence of a rich VIP-ergic innervation, including intrinsic VIP neurons, have made VIP a tentative post-ganglionic non-adrenergic, non-cholinergic neurotransmitter to these regions. The relaxatory motor responses elicited by VIP or vagal activation were

Mechanisms of methacholine-induced coronary vasospasm in an experimental model of variant angina in the anesthetized rat.

Methacholine (Mch), when injected near the ostia of the coronary arteries, induces an intense coronary vasospasm which can be measured by the degree of S wave elevation monitored from an electrocardiogram (ECG). The observed ECG changes resemble those occurring in patients with variant angina. The effects of Mch were blocked by atropine, but not by d-tubocurarine, hexamethonium, adrenergic receptor blockade, or prior reserpinization, indicating that Mch is acting directly on muscarinic receptors to produce a vasoconstriction of the coronary arteries. This model of Mch-induced coronary vasospas

Ganglion-blocking properties of atropine-like drugs.

The ganglion-blocking properties of atropine, atropine methyl nitrate, oxyphenonium, poldine, methantheline and propantheline have been examined and compared with those of hexamethonium, mecamylamine, pentolinium and tetraethylammonium. The ganglion-blocking activity was assessed by stimulating the preganglionic nerve to the superior cervical ganglion and recording directly the percentage depression in the postganglionic action potentials. In this way any ambiguity due to the peripheral actions of the drugs was removed. As a group the atropine-like compounds are not markedly less potent than t

Genomic and Pathologic Characterization of the First FAdV-C Serotype 4 Isolate from Black-Necked Crane.

Fowl adenoviruses (FAdVs) are distributed worldwide in poultry and incriminated as the etiological agents for several health problems in fowls, and are capable of crossing species barriers between domestic and wild fowls. An FAdV strain was, for the first time, isolated from black-necked crane in this study, and was designated as serotype 4 Fowl aviadenovirus C (abbreviated as BNC2021) according to the phylogenetic analysis of its DNA polymerase and hexon gene. The viral genomic sequence analysis demonstrated that the isolate possessed the ORF deletions that are present in FAdV4 strains circul

Acute distal colitis impairs gastric emptying in rats via an extrinsic neuronal reflex pathway involving the pelvic nerve.

BACKGROUND AND AIMS: Patients with inflammatory bowel disease often present with abnormal gut motility away from the inflammatory site. We studied remote motility disturbances and their pathophysiology in a rat model of colitis. METHODS: Colitis was induced 72 h prior to experiments using trinitrobenzene sulphate (TNBS) instillation. Inflammation was verified using histology and myeloperoxidase (MPO) measurements. To assess gut motility, we determined gastric emptying, distal front and geometric centre (GC) of intestinal transit 30 min after intragastric administration of a semiliquid Evans b

The intercellular communication via nitric oxide and its regulation in coupling of cyclic GMP synthesis upon stimulation of muscarinic cholinergic receptors in rat superior cervical sympathetic ganglia.

Cyclic GMP (cGMP) production in rat superior cervical sympathetic ganglia (SCG) was markedly increased (ca. 7-9-fold) by the addition of either acetylcholine (ACh; 0.1 mM) or a muscarinic agonist, carbachol (Carb; 0.1 mM), in the presence of an inhibitor (3-isobutyl-1-methylxanthine) for cGMP hydrolytic enzyme during in vitro aerobic incubation at 37 degrees C for 5 min. The ACh-induced accumulation of cGMP in SCG was effectively blocked (-73%) by the further addition of atropine (10 microM), a muscarinic antagonist, whereas a nicotinic blocker, hexamethonium (10 microM) partially antagonized

Neurogenic and humoral factors maintaining arterial pressure in conscious dogs.

This study examined the contribution of each primary pressor system to resting arterial pressure and the ability of each to maintain pressure in the absence of the other systems in conscious dogs. In dogs with intact carotid sinus baroreflexes, ganglionic blockade (GB) reduced blood pressure by 13 +/- 3 mmHg, whereas blockade of the renin-angiotensin (RAS) and arginine vasopressin (AVP) systems did not significantly alter arterial pressure. After removal of the other two systems, the depressor response to blockade of the sympathetic nervous system (SNS) with hexamethonium and atropine was sign

Development of high sodium renal hypertension during chronic blockade of the vascular effects of vasopressin.

Studies in sodium-dependent models of hypertension have shown that arginine-vasopressin (AVP) plays an important role in the maintenance of blood pressure, predominantly through its vasoconstrictor action. In addition to AVP, the sympathetic nervous system (SNS) also acts to maintain blood pressure in high sodium one-kidney, figure-8 renal wrap hypertension. The purpose of this study was to determine if chronic blockade of vascular AVP (V1) receptors affected the induction of high sodium renal hypertension and the contribution of the SNS to the maintenance of blood pressure. Rats receiving chr

Neural control of electrical gastric activity in response to inflation of the antrum in the rabbit.

The respective roles of the extrinsic innervation (vagal and splanchnic) and of the myenteric plexus in the control of gastric motility were investigated in rabbits by quantifying the effect of cutting the extrinsic nerves on the electrical response of the antrum and pylorus to inflation of the antrum. In intact animals the frequency of bursts during distension was significantly increased above the frequency during the 30 min preceding inflations. The interval from the end of step inflation to the first burst recorded after inflation was significantly longer than the mean interval between burs

Biochemical characterization of the nicotinic cholinergic receptors in human brain: binding of (-)-[3H]nicotine.

(-)-[3H]Nicotine was found to bind specifically to membranes of human brains obtained at autopsy. The binding was stereospecific, (-)-nicotine being 40 times more potent than (+)-nicotine in displacing labeled (-)-nicotine. Saturation binding studies revealed the presence of two binding sites with dissociation constant (KD) values of 8.1 and 86 nM, and maximum binding capacity (Bmax) values of 36 and 90 fmol/mg protein, respectively. In competition studies, nicotinic agonists were 1,000 times more potent than ganglionic, neuromuscular, and muscarinic blocking drugs in displacing labeled (-)-ni

A study of the origin of altered pharmacological reactivity of synaptic structures caused by diabetes and pretreatment with contrainsular agents.

The effect of membrane-stabilizing ganglionic blocking agents was found to be decreased in the ganglia of diabetic cats. Similarly, the sensitivity to membrane-stabilizing muscle relaxants of motor endplates in diabetic rats was also diminished, while the effectivity of depolarizing agents was augmented. Analysing the mechanism of action of these phenomena, it was shown that in diabetes developing as a result of pancreas removal or contrainsular treatment, a factor appeared in the blood of the experimental animals which was heat labile and facilitated the transmission process in the cholinergi

The ethanol withdrawal syndrome in the rat: effects of drug treatment on adrenal gland and urinary catecholamines.

It has been found that a short period of severe intoxication with ethanol produces a marked reduction of adrenal catecholamines. Animals so treated demonstrate moderate to severe withdrawal signs. Accordingly, adrenal and urinary catecholamines were determined in rats undergoing withdrawal after treatment with several drugs known to modify sympathetic function. The depressant diazepam obtained withdrawal severity without altering peripheral catecholamines, propranolol reduced urinary catecholamines and transiently ameliorated withdrawal, whereas hexamethonium elevated urinary catecholamines an

Forgotten Circulation: Reduced Mesenteric Venous Capacitance in Hypertensive Rats Is Improved by Decreasing Sympathetic Activity.

BACKGROUND: The mesenteric venous reservoir plays a vital role in mediating blood volume and pressure changes and is richly innervated by sympathetic nerves; however, the precise nature of venous sympathetic regulation and its role during hypertension remains unclear. We hypothesized that sympathetic drive to mesenteric veins in spontaneously hypertensive (SH) rats is raised, increasing mean circulatory filling pressure (MCFP), and impairing mesenteric capacitance. METHODS: Arterial pressure, central venous pressure, mesenteric arterial, and venous blood flow were measured simultaneously in c

Distinct patterns of myogenic motor activity identified in isolated human distal colon with high-resolution manometry.

BACKGROUND: Colonic high-resolution manometry (HRM) has been used to reveal discrete, propagating colonic motor patterns. To help determine mechanisms underlying these patterns, we used HRM to record contractile activity in human distal colon ex vivo. METHODS: Surgically excised segments of descending (n = 30) or sigmoid colon (n = 4) were immersed in oxygenated Krebs solution at 36°C (n = 34; 16 female; 67.6 ± 12.4 years; length: 24.7 ± 3.5 cm). Contractility was recorded by HRM catheters. After 30 minutes of baseline recording, 0.3 mM lidocaine and/or 1 mM hexamethonium were applied. Ascend

M-currents (Kv7.2-7.3/KCNQ2-KCNQ3) Are Responsible for Dysfunctional Autonomic Control in Hypertensive Rats.

UNLABELLED: Autonomic dysfunctions play important roles in hypertension, heart failure and arrhythmia, often with a detrimental and fatal effect. The present study analyzed if these dysfunctions involved M-channels (members of the Kv7/KNCQ family) in spontaneously hypertensive rats (SHR). Cardiac output and heart rate (HR) were recorded by a flow probe on the ascending aorta in anesthetized SHR and normotensive rats (WKY), and blood pressure (BP) by a femoral artery catheter. Total peripheral vascular resistance (TPR) was calculated. XE-991 (Kv7.1-7.4-inhibitor) reduced resting HR in WKY but o

Interleukin-1 (IL-1) stimulates arginine vasopressin (AVP) release from superfused rat hypothalamo-neurohypophyseal complexes independently of cholinergic mechanism.

We studied whether interleukin-1 (IL-1) affects the release of arginine vasopressin (AVP) from the superfused hypothalamo-neurohypophyseal complex (HNC) of rats. Involvement of the cholinergic system in the mediation of IL-1 on AVP release from HNC was also examined. Both human recombinant IL-1 alpha and -1 beta elicited a rapid increase of AVP from HNC in a dose-dependent manner at concentrations ranging from 0.1 to 10 nM. However, neither IL-1 alpha nor -1 beta at concentrations of 100 nM increased AVP, and even suppressed the stimulatory effect of 10 nM IL-1 alpha and -1 beta added later. A
