Toward Predicting Pandemic Potential: A Comparative Analysis of Virus-Host Interactions Between Diverse Influenza A Viruses and the Human Innate Immune System.

Zoonotic infections caused by influenza A viruses (IAVs) of animal origin are a major public health concern, as they may represent the initial step toward the emergence of a pandemic strain. The host range of IAVs is shaped by species-specific virus-host interactions, and several host barriers have been identified that restrict the adaptation of avian viruses to humans. We used a comparative interaction screen by mammalian cell-based Nanoluciferase two-hybrid (mN2H) assay to investigate whether differences in the interaction networks between viral proteins from human, avian, and zoonotic IAVs

Antimicrobial Resistance Under a Changing Climate.

The landscape of antimicrobial resistance (AMR) is changing across the animal, human and environmental ecosphere, with both climate change and AMR recognised as leading threats to human health. In this viewpoint, we argue that climate change is likely to reshape the conditions under which AMR emerges, persists and spreads, with important implications for how risks are understood and managed in Aotearoa New Zealand (ANZ). These risks arise because climate-related exposures interact with existing vulnerabilities and adaptive capacity across human, animal and environmental systems. Addressing AMR

Ex vivo challenge models for infectious diseases.

Traditionally, molecular mechanisms of pathogenesis for infectious agents were studied in cell culture or animal models but have limitations on the extent to which the resulting data reflect natural infection in humans. The COVID-19 pandemic has highlighted the urgent need to rapidly develop laboratory models that enable the study of host-pathogen interactions, particularly the relative efficacy of preventive measures. Recently, human and animal ex vivo tissue challenge models have emerged as a promising avenue to study immune responses, screen potential therapies and triage vaccine candidates

Molecular detection and typing of pathogenic Leptospira species from livestock and small mammals in Uganda.

Leptospira are bacteria that cause leptospirosis in both humans and animals. Human Leptospira infections in Uganda are suspected to arise from animal-human interactions. From a nationwide survey to determine Leptospira prevalence and circulating sequence types in Uganda, we tested 2030 livestock kidney samples, and 117 small mammals (rodents and shrews) using real-time PCR targeting the lipL32 gene. Pathogenic Leptospira species were detected in 45 livestock samples but not in the small mammals. The prevalence was 6.12% in sheep, 4.25% in cattle, 2.08% in goats, and 0.46% in pigs. Sequence typ

Wild Boar: A Reservoir of Foodborne Zoonoses.

Wild boar populations around the world have increased dramatically over past decades. Climate change, generating milder winters with less snow, may affect their spread into northern regions. Wild boars can serve as reservoirs for a number of bacteria, viruses, and parasites, which are transmissible to humans and domestic animals through direct interaction with wild boars, through contaminated food or indirectly through contaminated environment. Disease transmission between wild boars, domestic animals, and humans is an increasing threat to human and animal health, especially in areas with high

Canine Rabies: A Looming Threat to Public Health.

Rabies is an acute, fatal viral disease that infects domestic and wild animals and is transmissible to humans. Worldwide, rabies kills over 55,000 people every year. The domestic dog plays a pivotal role in rabies transmission. Domestic dogs are not only part of our daily lives but also of our immediate surroundings, and this is reflected in the rise in pet dog ownership in developed and developing countries. This is important given that more frequent exposures and interactions at the animal-human interface increases the likelihood of contracting zoonotic diseases of companion animals. Despite

CILP1 interacting with YBX1 promotes hypertrophic scar formation by suppressing PPARs transcription.

Hypertrophic scar (HS) represents the most prevalent form of skin fibrosis, significantly impacting the quality of life. Despite this, the molecular mechanisms driving HS formation remain largely undefined, impeding the development of effective treatments. The study showed that Cartilage Intermediate Layer Protein 1 (CILP1) was predominantly expressed in myofibroblasts and was up-regulated in various forms of skin fibrosis, including human hypertrophic and keloid scars, and in animal models of HS. Notably, we detected elevated serum levels of CILP1 in fifty-two patients with HS compared to twe

The Opportunistic Pathogen Listeria monocytogenes: Pathogenicity and Interaction with the Mucosal Immune System.

Listeria monocytogenes is an opportunistic foodborne pathogen causing listeriosis, an often fatal infection leading to meningitis, sepsis, or infection of the fetus and abortion in susceptible individuals. It was recently found that the bacterium can also cause acute, self-limiting febrile gastroenteritis in healthy individuals. In the intestinal tract, L. monocytogenes penetrates the mucosa directly via enterocytes, or indirectly via invasion of Peyer's patches. Animal models for L. monocytogenes infection have provided many insights into the mechanisms of pathogenesis, and the development of

Behavioural Welfare Implications of Human Handling Practices in Zoo Snakes.

BACKGROUND: Handling reptiles, particularly snakes, is common in ambassador-animal and animal-visitor interaction programmes. However, relatively little is known about the behavioural and welfare impacts of these interactions on snakes. OBJECTIVES: This study examined whether staff-led handling during visitor interactions impacted snake behaviour by comparing before and after handling and between handling and non-handling days. METHODS: Observations were conducted at Cleland Wildlife Park (South Australia, Australia) on three individual snakes: one woma python (Aspidites ramsayi) and two Chi

Compendium of measures to prevent disease associated with animals in public settings, 2007: National Association of State Public Health Veterinarians, Inc. (NASPHV).

Certain venues encourage or permit the public to contact animals, resulting in millions of human-animal interactions each year. These settings include county or state fairs, petting zoos, animal swap meets, pet stores, zoologic institutions, circuses, carnivals, farm tours, livestock-birthing exhibits, educational exhibits at schools, and wildlife photo opportunities. Although multiple benefits of human-animal contact exist, infectious diseases, rabies exposures, injuries, and other human health problems associated with these settings are possible. Infectious disease outbreaks reported during

Assessing systemic and non-systemic transmission risk of tick-borne encephalitis virus in Hungary.

Estimating the tick-borne encephalitis (TBE) infection risk under substantial uncertainties of the vector abundance, environmental condition and human-tick interaction is important for evidence-informed public health intervention strategies. Estimating this risk is computationally challenging since the data we observe, i.e., the human incidence of TBE, is only the final outcome of the tick-host transmission and tick-human contact processes. The challenge also increases since the complex TBE virus (TBEV) transmission cycle involves the non-systemic route of transmission between co-feeding ticks

Biology, host relations, and epidemiology of Sarcoptes scabiei.

Scabies continues to be an important parasitic disease of humans, and other mammals. Suprisingly for a disease that has afflicted humans since antiquity, little is directly known about the basic biology of the parasite, the host-parasite interactions, the host immune response, and host susceptibility. Much more research in these areas is needed if we are to understand fully the occurrence, transmission, and epidemiology of both human and animal scabies.

Animal models for dissecting Vibrio cholerae intestinal pathogenesis and immunity.

The human diarrheal disease cholera is caused by the bacterium Vibrio cholerae. Efforts to develop animal models that closely mimic cholera to study the pathogenesis of this disease began >125 years ago. Here, we review currently used non-surgical, oral inoculation-based animal models for investigation of V. cholerae intestinal colonization and disease and highlight recent discoveries that have illuminated mechanisms of cholera pathogenesis and immunity, particularly in the area of how V. cholerae interacts with the gut microbiome to influence infection. The emergence of high-throughput tools

Leptospira and Leptospirosis: A Review of Species Classifications, Genomes, Morphological Structures, Antimicrobial Resistances, Transmissions, and Clinical Manifestations.

Leptospirosis, also known as "rat-urine disease", is a neglected zoonotic and waterborne disease that is caused by Leptospira spp. This disease is transmitted by direct and indirect exposure to the urine and stool of infected animals. The current estimate has highlighted that leptospirosis has caused at least one million cases and 60,000 deaths, with high endemicity in tropical regions. With climate change, urbanisation, and increasing human-animal interaction, the threat of leptospirosis and other zoonotic diseases will continue to emerge. Investing in multidisciplinary research, technology,

Toxic shock syndrome.

In the past 10 years, we have learned much about TSS and S. aureus and its toxins. A number of important biologic principles have been reemphasized in this first decade of TSS research: S. aureus is a very complex organism, one not likely to yield quick answers; in vitro observations must always be confirmed in the patient; animal models may not always be reliable replicates of human disease; and epidemiologic associations cannot be equated with causation. Toxic shock is an intricate phenomenon with many interesting scientific facets. Unraveling its mysteries will undoubtedly teach us more abo

Public health threat of novel zoonotic diseases: literature review.

Zoonoses, diseases transmitted from animals to humans, continue to challenge public health despite advancements in controlling infectious diseases. The intricate link between human, animal, and environmental health is emphasised by the fact that zoonoses contribute to 60% of emerging human infections. Wet markets, wildlife hunting, intensive wildlife farming, and interactions between domestic animals and humans are key transmission sources. Historical examples like the bubonic plague and English Sweats illustrate the longstanding impact of zoonotic diseases. With new transmission patterns emer

Multispecies perspectives on dementia in the poetry of Jenna Butler.

Jenna Butler's Wells (2012) is a collection of poetry in which the poet mourns the loss of her grandmother, a life-long birdwatcher who can still recognize the familiar calls of birds that she is no longer able to name due to cognitive issues associated with dementia. Treating this scenario as one that invites interpretation from a posthuman perspective, this paper conducts a close analysis that is situated at the intersection of age studies and animal studies and highlights the possibilities and the perils of linking these two fields of inquiry. Balanced on the divide between animalizing peop

Human iPSC-Derived Brain Organoids: A Disease-Oriented Evaluation of Modeling Fidelity.

Two-dimensional (2D) cell culture systems and conventional animal models have been crucial in the study of central nervous system (CNS) pathology. However, they are limited in their abilities to elucidate human-specific neurobiology and disease etiology. Human induced pluripotent stem cell (iPSC)-derived brain organoids have emerged as a complementary platform that offers a three-dimensional (3D) structure and human-specific environment. They exhibit cellular heterogeneity, apical-basal polarity, and the ability to perform tissue-specific functions such as absorption, secretion, and electrophy

A review of risk factors at the human-animal-environmental interface of garbage dumps that are driving current and emerging zoonotic diseases.

An increasing trend in zoonotic and emerging infectious diseases (EIDs) has been observed worldwide. Most EID outbreaks originate from wildlife, and these outbreaks often involve pathogen-host-environment interaction. Garbage dumps act as an interface between humans, animals, and the environment, from which EIDs could arise. Therefore, this review considers the presence of important pathogens associated with animals and vectors at garbage dumps from a One Health perspective, looking at animal, human, and environmental factors that play a role. A narrative review was performed focusing on four
