Prolonged production of hydroxyl radical in rat hippocampus after brain ischemia-reperfusion is decreased by 21-aminosteroids.

Transient global ischemia may lead to persistent production of reactive oxygen species in selected brain regions thereby contributing to selective vulnerability to ischemia. Using cerebral microdialysis, we assessed the production of the highly reactive hydroxyl radical (OH.) in rat hippocampus during global ischemia and reperfusion (IR). During IR, perfusate containing salicylic acid was collected and analyzed for non-enzymatic hydroxylation of salicylate to 2,3-DHBA. Since 21-aminosteroids can attenuate excitatory amino acid-mediated OH. production in the brain, we repeated the experiments a

Hydroxyl radical generation and nitric oxide synthase activity in Fisher 344 x brown-Norway F1 rat brain.

Hydroxyl radical production in the cerebral cortex, striatum and hippocampus of young (6 months), middle-aged (15 months) and old (28 months) Fisher 344 x Brown-Norway F1 rats was quantitated by measuring the salicylate hydroxyl radical-trapping product, 2,5-dihydroxybenzoic acid. The levels of hydroxyl radical between different age groups in each region examined were not statistically different. In all regions, with the exception of hippocampus from old rats which had a lower content of hydroxyl radical, the levels of hydroxyl radical were higher for middle-aged and old rats than young ones.

BSA-mediated proton-coupled electron transfer to unlock the photogeneration of hydroxyl radicals in cancer cells.

The unique attributes of proteins-including their confined nanoscale cavities, abundant redox-active amino acid residues, and excellent biocompatibility-offer significant advantages for constructing protein-based enzymatic systems. However, their potential remains underexplored in photosensitization systems for shifting the photosensitization pathways. Here, we report a bovine serum albumin (BSA)-based photosensitizing system, termed TC2@BSA, in which the BSA nanoreactor mediated a proton-coupled electron transfer (PCET) process to unlock the photogeneration of hydroxyl radicals (˙OH) in cance

Rice Root Fe Plaque-Induced Hydroxyl Radicals Increase Paddy Soil CO2 Emissions.

Poorly crystalline iron (Fe) oxides often accumulate on rice root surfaces, forming Fe plaque. While this plaque has been implicated in soil organic carbon (SOC) transformation, its specific role in regulating SOC mineralization is still insufficiently understood. In this study, we hypothesize that Fe plaque induces the generation of hydroxyl radicals (•OH), which disrupt the "enzyme latch" mechanism and promote CO2 emissions. The results showed that seedlings with Fe plaque showed a 150% higher •OH concentration, 50% higher laccase activity, 25% higher SOC mineralization rate, and 20% lower p

In situ generation of hydroxyl radicals by B-doped TiO2 for efficient photocatalytic degradation of acetaminophen in wastewater.

Acetaminophen (AP) is a widely used antipyretic analgesic belonging to the class of PPCPs, which is difficult to be effectively degraded by traditional water treatment processes. However, photocatalytic technology may be an effective approach. Herein, B-doped TiO2 photocatalytic materials were synthesized by sol-gel method, calcinated at 600℃ for 2 h, investigated by XRD, TEM, XPS, and other characterization methods. The photocatalytic efficiency and factors affecting the photocatalytic activity were assessed by degradation of AP under 365 nm UV light. Compared with undoped TiO2, 4%B-TiO2 nano

A Circular Dichroism and Photoacoustic Dual-Mode Probe for Detection In Vitro and Imaging In Vivo of Hydroxyl Radicals.

The drug toxicity is a long-term concern in contemporary medical research. Serious drug toxicity may cause acute liver failure or even patient death. Currently, the pathogenesis of drug-induced liver injury (DILI) is not entirely clear, and there is still no specific treatment for patients with DILI. Accordingly, improving the diagnosis and treatment level of DILI is a major challenge facing the basic and clinical research in relevant fields. To address these, an ·OH-activated circular dichroism (CD) and photoacoustic (PA) dual-mode nanoprobe was here designed and synthesized. The probe was pr

Combustion Processes as a Source of High Levels of Indoor Hydroxyl Radicals through the Photolysis of Nitrous Acid.

Hydroxyl radicals (OH) are known to control the oxidative capacity of the atmosphere but their influence on reactivity within indoor environments is believed to be of little importance. Atmospheric direct sources of OH include the photolysis of ozone and nitrous acid (HONO) and the ozonolysis of alkenes. It has been argued that the ultraviolet light fraction of the solar spectrum is largely attenuated within indoor environments, thus, limiting the extent of photolytic OH sources. Conversely, the ozonolysis of alkenes has been suggested as the main pathway of OH formation within indoor settings

Potential mechanism for pentachlorophenol-induced carcinogenicity: a novel mechanism for metal-independent production of hydroxyl radicals.

The hydroxyl radical ((*)OH) has been considered to be one of the most reactive oxygen species produced in biological systems. It has been shown that (*)OH can cause DNA, protein, and lipid oxidation. One of the most widely accepted mechanisms for (*)OH production is through the transition metal-catalyzed Fenton reaction. Pentachlorophenol (PCP) was one of the most widely used biocides, primarily for wood preservation. PCP is now ubiquitously present in our environment and even found in people who are not occupationally exposed to it. PCP has been listed as a priority pollutant by the U.S. Env

Accumulation of amino acids and hydroxyl free radicals in brain and retina of gerbil after transient ischemia.

The mongolian gerbil was introduced as a stroke model because of its incomplete circle of Willis. Unilateral carotid ligation ischemia produced in such a fashion was not effective in all gerbils. We have selected gerbils by examination of the ocular fundus to study the level of amino acids and hydroxyl free radicals (OH0 formation of DHBA, dihydroxybenzoic acid, from salicylate) in gerbil cerebral ischemia. Only gerbils with absence of retinal blood after ligation were selected as sensitive. One group (sham operated) served as control. The other group was subjected to unilateral left carotid o

Site-specific and bulk-phase generation of hydroxyl radicals in the presence of cupric ions and thiol compounds.

Addition of a thiol compound to a solution containing Cu2+ and H2O2 resulted in the generation of hydroxyl radicals (OH.). These radicals were able to oxidize salicylic acid and tryptamine in a reaction that was strongly inhibited by the OH.-scavenger mannitol. Covalent coupling of the thiol compound to tryptamine did not significantly influence the degradation of the indole moiety subsequent to addition of H2O2 and Cu2+. The inhibiting effect of mannitol, however, was strongly reduced, indicating that the scavenger could not interfere with site-specific reactions of OH..

Pyrogenic carbon drives abiotic methane formation: Hydroxyl radicals and reductive capacity mediate demethylation.

Methane (CH4) oversaturation in oxygenated waters has challenged the long-held view that biogenic CH4 production is strictly anoxic. Abiotic CH4 formation has been observed in oxidative, photochemical, and radical-driven systems. However, the potential role of pyrogenic carbon (PyC) remains largely unknown. PyC is a fire‑derived material that is widely transported into and suspended within lakes, rivers, and coastal waters. Here, we investigated the abiotic methanogenic activity of PyC derived from rice straw, pine twig, and peanut shell at pyrolysis temperatures of 300, 500, and 700 °C under

Unveiling the Critical Pathways of Hydroxyl Radical Formation in Breakpoint Chlorination: The Role of Trichloramine and Dichloramine Interactions.

Chlorination of ammonia or chloramine-containing waters induces breakpoint chlorination reactions, producing a hydroxyl radical (•OH), but enhances the formation of undesirable N-nitrosamines. The prevailing view attributes •OH formation to a nitrosyl intermediate derived from the hydrolysis of dichloramine, but this pathway is unlikely at neutral or acidic pH. This study reveals a novel mechanism where •OH is generated via interactions between trichloramine (NCl3) and dichloramine (NHCl2), which also form nitrosation agents. Our experiments demonstrated that the NCl3-NHCl2 interaction degrade

New Barrier Role of Iron Plaque: Producing Interfacial Hydroxyl Radicals to Degrade Rhizosphere Pollutants.

Iron plaque, as a natural barrier between rice and soil, can reduce the accumulation of pollutants in rice by adsorption, contributing to the safe production of rice in contaminated soil. In this study, we unveiled a new role of iron plaque, i.e., producing hydroxyl radicals (·OH) by activating root-secreted oxygen to degrade pollutants. The ·OH was produced on the iron plaque surface and then diffused to the interfacial layer between the surface and the rhizosphere environment. The iron plaque activated oxygen via a successive three-electron transfer to produce ·OH, involving superoxide and h

Multiple Effects of Humic Components on Microbially Mediated Iron Redox Processes and Production of Hydroxyl Radicals.

Microbially mediated iron redox processes are of great significance in the biogeochemical cycles of elements, which are often coupled with soil organic matter (SOM) in the environment. Although the influences of SOM fractions on individual reduction or oxidation processes have been studied extensively, a comprehensive understanding is still lacking. Here, using ferrihydrite, Shewanella oneidensis MR-1, and operationally defined SOM components including fulvic acid (FA), humic acid (HA), and humin (HM) extracted from black soil and peat, we explored the SOM-mediated microbial iron reduction and

Self-Immolative Dye-Doped Polymeric Probe for Precisely Imaging Hydroxyl Radicals by Avoiding Leakage.

For sensing low abundance of biomarkers, utilizing nanocarriers to load dyes is an efficient method to amplify the detected signal. However, the non-specific leak of the internal dyes in this approach is accompanied by false positive signals, resulting in inaccurate signal acquirement. To address this issue, in this work, we reported a novel signal amplification strategy with dye as a scaffold to construct a self-immolative dye-doped polymeric probe (SDPP). In our proposed approach, the dyes were covalently integrated into the main chain of a polymer, which can avoid the non-specific leak of t

Effects of tetrathiomolybdate and penicillamine on brain hydroxyl radical and free copper levels: a microdialysis study in vivo.

Wilson disease is an inherited disorder of excessive copper accumulation. The commonly used drug d-penicillamine (PA) or trientine both cause a high incidence (10-50%) of neurological worsening, which rarely occurs with tetrathiomolybdate (TM) treatment. To investigate the mechanisms of neurologic deterioration after the initiation of chelation therapy, brain hydroxyl radical and free copper were assessed in vivo in this study. On days 3, 7, 14, and 21 after PA or TM administration, striatal hydroxyl radical levels of both TX mice and controls were assessed by terephthalic acid (TA) combined w

Degradation pathways of lamotrigine under advanced treatment by direct UV photolysis, hydroxyl radicals, and ozone.

Lamotrigine is recently recognized as a persistent pharmaceutical in the water environment and wastewater effluents. Its degradation was studied under UV and ozone advanced oxidation treatments with reaction kinetics of lamotrigine with ozone (≈4 M(-1)s(-1)), hydroxyl radical [(2.1 ± 0.3) × 10(9)M(-1)s(-1)] and by UV photolysis with low and medium pressure mercury vapor lamps [quantum yields ≈0 and (2.7 ± 0.4)× 10(-4) respectively] determined. All constants were measured at pH 6 and at temperature ≈20°C. The results indicate that lamotrigine is slow to respond to direct photolysis or oxidation

Fluorescence and HPLC Detection of Hydroxyl Radical by a Rhodamine-Nitroxide Probe and its Application in Cell Imaging.

A rhodamine nitroxide probe was designed to detect the hydroxyl radical (·OH), which presented high selectivity for ·OH over other reactive oxygen species (ROS) and linear fluorescence response to ·OH produced by Fenton reaction. The product was detected by HPLC-MS, indicating that the main product of the reaction was O-methylhydroxylamine and the product peak areas measured by HPLC-UV/vis and HPLC-FLD both enhanced proportionally with the increase of ·OH concentration. The application of the probe in biological system was explored to trace the production of ·OH in cells under oxidative stress

DNA cleavage by hydroxyl radicals generated in the Cu,Zn-superoxide dismutase and hydrogen peroxide system.

To study the catalytic activity of Cu,Zn-superoxide dismutase (SOD) in the generation of hydroxyl radical (.OH) from H2O2, we investigated the mechanism of DNA cleavage mediated by human Cu,Zn-SOD and H2O2. When plasmid DNA was incubated with 5 microM Cu, Zn-SOD and 1.0 mM H2O2, DNA cleavage occurred within 15 min. A spectrophotometric study using a 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulfonate) (ABTS) showed that free .OH formation was proportional to concentrations of Cu,Zn-SOD and H2O2. .OH formation and DNA cleavage were inhibited by hydroxyl radical scavengers such as cyanide, azide, a
