Association of Hyperoxia During Cardiopulmonary Bypass and Postoperative Delirium in the Pediatric Cardiac ICU.

OBJECTIVE: ICU delirium commonly complicates critical illness associated with factors such as cardiopulmonary bypass (CPB) time and the requirement of mechanical ventilation (MV). Recent reports associate hyperoxia with poorer outcomes in critically ill children. This study sought to determine whether hyperoxia on CPB in pediatric patients was associated with a higher prevalence of postoperative delirium. DESIGN: Secondary analysis of data obtained from a prospective cohort study. SETTING: Twenty-two-bed pediatric cardiac ICU in a tertiary children's hospital. PATIENTS: All patients (18 yr

The Impact of Short-Term Hyperoxia on Cerebral Metabolism: A Systematic Review and Meta-Analysis.

BACKGROUND: Cerebral ischemia due to hypoxia is a major cause of secondary brain injury and is associated with higher morbidity and mortality in patients with acute brain injury. Hyperoxia could improve energetic dysfunction in the brain in this setting. Our objectives were to perform a systematic review and meta-analysis of the current literature and to assess the impact of normobaric hyperoxia on brain metabolism by using cerebral microdialysis. METHODS: We searched Medline and Scopus, following the recommendations of the Preferred Reporting Items for Systematic Reviews and Meta-Analyses st

Observations on the perfusion recovery of regenerative angiogenesis in an ischemic limb model under hyperoxia.

This study combines two well-known vascular research models, hyperoxia and hind limb ischemia, aiming to better characterize capacities of the hyperoxia challenge. We studied two groups of C57/BL6 male mice, a control (C) and a hind limb ischemia (HLI) group. Perfusion from both limbs was recorded in all animals by laser Doppler techniques under an oxygen (O2 ) saturated atmosphere, once for control and, during 35 days for the HLI group. We used a third set of normoxic animals for HLI morphometric control. The expected variability of responses was higher for the younger animals. In the HLI gro

Aerobic efficiency is associated with the improvement in maximal power output during acute hyperoxia.

This study investigated the relationship between aerobic efficiency during cycling exercise and the increase in physical performance with acute hyperoxic exposure (FiO2 ~31%) (HOX) and also tested the hypothesis that fat oxidation could be increased by acute hyperoxia. Fourteen males and four females were recruited for two sessions, where they exercised for 2 × 10 min at 100 W to determine efficiency. HOX and normoxia (NOX) were administered randomly on both occasions to account for differences in nitrogen exchange. Thereafter, a progressive ramp test was performed to determine VO2max and maxi

Chronic intermittent hyperoxia alters the development of the hypoxic ventilatory response in neonatal rats.

Chronic exposure to sustained hyperoxia alters the development of the respiratory control system, but the respiratory effects of chronic intermittent hyperoxia have rarely been investigated. We exposed newborn rats to short, repeated bouts of 30% O2 or 60% O2 (5 bouts h(-1)) for 4-15 days and then assessed their hypoxic ventilatory response (HVR; 10 min at 12% O2) by plethysmography. The HVR tended to be enhanced by intermittent hyperoxia at P4 (early phase of the HVR), but it was significantly reduced at P14-15 (primarily late phase of the HVR) compared to age-matched controls; the HVR recove

Hypoxia-hyperoxia paradigms in the development of oxygen-induced retinopathy in a rat pup model.

BACKGROUND: Retinopathy of prematurity [ROP] continues to be a significant clinical problem in preterm infants. There is a need for animal models to better understand the roles of hypoxia/hyperoxia in the pathogenesis and management of ROP. OBJECTIVES: To test the hypothesis that multiple daily cycles of intermittent hypoxia, followed by brief hyperoxia, would provide a clinically relevant protocol for generation of ROP in a rat pup. METHODS: Rat pups were exposed for the first 14 days to one of three protocols: room air [RA], sustained cycles of hyperoxia/hypoxia [SHH] as previously employe

"Oxygen supply" as modulator of aging processes: hypoxia and hyperoxia models for aging studies.

Cell growth is regulated by several factors, including oxygen supply, which influence cell metabolism. Aging is characterized by decreased oxygen supply to tissue, a reduction of tissue PO(2) and of the activity of several enzymes and metabolic factors. The oxygen-gradient diffusion at capillary tissue level is essential for the cellular survival, while the homeostasis of the oxygen in the arterial blood is mediated by reflexes sensitive to oxygen decrease and by release of several factors. Aging is correlated with a reduction of cells' oxygen supply concomitant to a parallel decrease in oxyge

Stability of housekeeping gene expression in the rat retina during exposure to cyclic hyperoxia.

Recent evidence suggests a genetic component to oxygen-induced retinopathy (OIR), a robust experimental model of human retinopathy of prematurity. OIR lends itself well to quantitative analysis of gene expression in rodents with well-defined genetic backgrounds. Such analysis by real-time reverse transcription polymerase chain reaction (RT-PCR) requires the use of reference genes as internal standards for purposes of normalization. We sought to identify housekeeping genes showing stable retinal expression across different rat strains and developmental stages, that were not regulated by oxygen

Structural and functional consequences of trolox C treatment in the rat model of postnatal hyperoxia.

PURPOSE: Previous studies have shown that newborn rats exposed to hyperoxia within the first 2 weeks of life develop vasculopathy in addition to permanent changes in retinal structure and function. It has also been suggested that free radicals may be the source of these pathologic effects. Trolox C, a water-soluble analogue of vitamin E, was previously shown to limit the vascular consequences of exposure to postnatal hyperoxia. The aim of this study was to investigate whether trolox C could also help prevent the functional (electroretinography) and structural (retinal histology) consequences a

Poly(ADP-ribose) polymerase activity in the cat carotid body in hypoxia and hyperoxia.

Reactive oxygen species (ROS) induce DNA damage with the ensuing activation of the chromosomal repair enzyme poly(ADP-ribose) polymerase (PARP). ROS also interact with the function of carotid body chemoreceptor cells. The possibility arises that PARP is part of the carotid chemosensing process. This study seeks to determine the presence of PARP and its changes in response to contrasting chemical stimuli, hypoxia and hyperoxia, both capable of generating ROS, in cat carotid bodies. The organs were dissected from anesthetized cats exposed in vivo to acute normoxic (PaO2 approximately 90 mmHg), h

Exposure to alternating hypoxia and hyperoxia causes severe proliferative retinopathy in the newborn rat.

Exposure to variable hyperoxia has recently been shown to be much more effective at producing proliferative retinopathy in the newborn rat than exposure to constant hyperoxia. To incorporate a more clinically relevant oxygen-exposure paradigm in our studies, we have now used a cycle between 50 and 10% oxygen and have compared its effects with those found using new exposures to the previously used 80/40% cycle. Starting at birth and continuing for 14 d, rats were exposed to environments that cycled between 50 and 10% oxygen or 80 and 40% oxygen every 24 h. After exposure, some rats were killed

Oxygen toxicity in rats. Varied effect of dexamethasone treatment depending on duration of hyperoxia.

The low rate of survival in patients with the adult respiratory distress syndrome (ARDS) may in part reflect a failure to consider that the lung's response to applied therapies may not be constant throughout the course of illness. To test this notion, we used hyperoxia to produce progressive lung damage in rats and administered dexamethasone at different times during O2 exposures of various lengths. Dexamethasone improved survival and decreased lung damage if given when exposure to hyperoxia was to be soon terminated; pulmonary inflammation was marked at the time at which the administration of

Acute and long-term ventilatory effects of hyperoxia in the adult sleep apnea syndrome.

Eight adult patients with sleep apnea were studied to evaluate the ventilatory and cardiac effects of hyperoxia on an acute basis. Five patients then used low-flow nighttime oxygen for 30 to 90 days. The results of the acute study showed that for 30-min study periods, the total number of apneas and per cent apnea time (duration of apneas divided by sleep time) decreased significantly from the room air to the oxygen period (207 to 68, p less than 0.05; 41.1 +/- 18.3% SD to 20.5 +/- 14.4%, p less than 0.05, respectively). Also, the apnea-associated slowing in heart rate is blocked by the supplem

[Experiences with the hyperoxia test in neonates and infants with congenital heart malformations (author's transl)].

Hyperoxia test with arterialized capillary blood was performed in 63 neonates and infants suffering from congenital heart defect with shunt. The lowest values were found in neonates with transposition of the great arteries without major systemic-pulmonary shunt. In cases with left to right shunt requiring surgical intervention the pO2 increased less than in those not needing operation. Observations with this non-invasive mentod proved valuable in early recogniton of congenital heart defect of neonates and infants requiring urgent surgical treatment.

Advances in research on hyperoxia-induced acute lung injury: from pathogenesis to therapy.

Hyperoxia-induced acute lung injury (HALI) is a serious complication of excessive oxygen exposure, yet its pathogenesis and treatment remain incompletely understood. A growing body of evidence suggests that HALI results from the interplay of oxidative stress, inflammation, immune dysregulation, organelle dysfunction, alveolar barrier disruption, and regulated cell death. In this review, we summarize recent advances in our understanding of these processes and discuss therapeutic strategies that have been investigated in experimental models. These approaches include antioxidant and anti-inflamma

Antioxidant suppression of hyperoxia-induced reactive oxygen species in cultured rat hippocampal neurons.

Central nervous system oxygen toxicity (CNS-OT) is driven, in part, by excess reactive oxygen species (ROS) generated during hyperoxic exposure. Herein, we developed an in vitro screening platform using embryonic rat hippocampal neurons to evaluate antioxidant candidates under hyperoxic stress (1.34 ATA) and quantify ROS by Dihydrorhodamine (DHR)123 fluorescence imaging and flow cytometry. Neuronal cultures remained viable throughout the assay, with approximately 87% anti-βIII tubulin staining efficiency and 88% neurite outgrowth-associated viability. Among the compounds tested, sulforaphane a

Is hyperoxia in early pediatric veno-veno extracorporeal life support associated with mortality?

BACKGROUND: Data is limited regarding the effects of supraphysiologic blood oxygen tension (hyperoxia) in patients requiring extracorporeal life support (ECLS). We sought to evaluate the association between hyperoxia and outcomes in pediatric patients requiring veno-venous (VV) ECLS. METHODS: Retrospective single-center study at an academic children's hospital, Children's Healthcare of Atlanta, Emory University School of Medicine that included all patients 0-18 years who required VV-ECLS between 01/2014 and 12/2019. RESULTS: During the study period 110 VV-ECLS runs occurred in 110 patients.

Gelsolin Attenuates Neonatal Hyperoxia-Induced Inflammatory Responses to Rhinovirus Infection and Preserves Alveolarization.

Prematurity and bronchopulmonary dysplasia (BPD) increase the risk of asthma later in life. Supplemental oxygen therapy is a risk factor for chronic respiratory symptoms in infants with BPD. Hyperoxia induces cell injury and release of damage-associated molecular patterns (DAMPs). Cytoskeletal filamentous actin (F-actin) is a DAMP which binds Clec9a, a C-type lectin selectively expressed on CD103+ dendritic cells (DCs). Co-stimulation of Clec9a and TLR3 induces maximal proinflammatory responses. We have shown that neonatal hyperoxia (a model of BPD) increases lung IL-12+Clec9a+CD103+ DCs, pro-
