Protective effects of passively transferred immune T- or B-lymphocytes in mice infected with Salmonella typhimurium.

Purified populations of bone marrow-derived (B-) lymphocytes and thymus-derived (T-) lymphocytes were obtained from C3D2F1 hybrid mice shown to be immune to Salmonella typhimurium. These subpopulations of lymphocytes were injected into normal mice; four days later the animals were challenged with 50 50% lethal doses of S. typhimurium, and viable bacteria in livers, spleens, and blood were counted at various intervals after challenge. On day 8 after challenge, the mice supplemented with B-lymphocytes showed a significant decrease in the number of organisms recovered from all three sites, compar

Effects of prepartum zinc-methionine supplementation on feed digestibility, rumen fermentation patterns, immunity status, and passive transfer of immunity in dairy cows.

The aim of this study was to determine the effects of prepartum supplementation of zinc-methionine (Zn-Met) on feed digestibility, rumen fermentation patterns, and immunity status in dams and passive immunity transfer in their calves. A randomized complete design was used in this study. Forty multiparous Holstein dairy cows in late pregnancy (60 d before the expected calving date) were blocked by parity (2.1 ± 0.3), body weight (651 ± 52 kg), and expected calving date, and randomly assigned to 1 of 4 treatments. Cows were supplemented with Zn as Zn-Met at 0, 20, 40, or 60 mg/kg of dry matter (

Immunization of Mice with a Live Transconjugant Shigella Hybrid Strain Induced Th1 and Th17 Cell-Mediated Immune Responses and Confirmed Passive Protection Against Heterologous Shigellae.

An avirulent, live transconjugant Shigella hybrid (LTSHΔstx) strain was constructed in our earlier study by introducing a plasmid vector, pPR1347, into a Shiga toxin gene deleted Shigella dysenteriae 1. Three successive oral administrations of LTSHΔstx to female adult mice produced comprehensive passive heterologous protection in their offspring against challenge with wild-type shigellae. Production of NO and different cytokines such asIL-12p70, IL-1β and IL-23 in peritoneal mice macrophages indicated that LTSHΔstx induced innate and adaptive immunity in mice. Furthermore, production of IFN-γ,

Passive immunization with Leptospira LPS-specific agglutinating but not non-agglutinating monoclonal antibodies protect guinea pigs from fatal pulmonary hemorrhages induced by serovar Copenhageni challenge.

Leptospira interrogans serovar Copenhageni causes pulmonary hemorrhages with respiratory failure, a major cause of death in leptospirosis patients. Protective immunity to Leptospira is known to correlate with the production of leptospiral lipopolysaccharide (L-LPS)-specific agglutinating antibodies. We generated L-LPS-specific mouse monoclonal antibodies (MAbs) and investigated if these MAbs can protect guinea pigs against fatal pulmonary hemorrhages caused by serovar Copenhageni. The MAbs L8H4 and L9B11 against 22kDa L-LPS agglutinated leptospires and completely protected guinea pigs from the

Protection against 17D yellow fever encephalitis in mice by passive transfer of monoclonal antibodies to the nonstructural glycoprotein gp48 and by active immunization with gp48.

The protective capacity of antiviral antibodies has generally been considered to depend on their interactions with structural components of the virion. Here we report protection against lethal 17D yellow fever virus (YF) encephalitis of mice by passive administration of nonneutralizing monoclonal antibodies to a 17D YF-specified nonstructural glycoprotein, gp48, and by active immunization with purified gp48. Among five anti-gp48 monoclonal antibodies tested, two with high titer complement-fixing (CF) activity were protective, whereas three antibodies with little or no CF activity were not. The

Early-Onset Network Hyperexcitability in Presymptomatic Alzheimer's Disease Transgenic Mice Is Suppressed by Passive Immunization with Anti-Human APP/Aβ Antibody and by mGluR5 Blockade.

Cortical and hippocampal network hyperexcitability appears to be an early event in Alzheimer's disease (AD) pathogenesis, and may contribute to memory impairment. It remains unclear if network hyperexcitability precedes memory impairment in mouse models of AD and what are the underlying cellular mechanisms. We thus evaluated seizure susceptibility and hippocampal network hyperexcitability at ~3 weeks of age [prior to amyloid beta (Aβ) plaque deposition, neurofibrillary pathology, and cognitive impairment] in a triple transgenic mouse model of familial AD (3xTg-AD mouse) that harbors mutated hu

Microneedle patch delivery of influenza vaccine during pregnancy enhances maternal immune responses promoting survival and long-lasting passive immunity to offspring.

Influenza virus causes life-threatening infections in pregnant women and their newborns. Immunization during pregnancy is the most effective means of preventing maternal and infant mortality/morbidity; however, influenza vaccination rates of pregnant women remain under 50%. Furthermore, the availability of vaccines in low-resource populations is limited. Skin immunization with microneedle patches (MN) is a novel and safe vaccination platform featuring thermostable vaccine formulations. Cold-chain independence and the potential for self-administration can expand influenza vaccination coverage i

The effect of herd-level passive immunity in dairy farms on survival probability during the first year of life.

This study examined the impact of passive immunity at the herd level on survival probability during the first year of life for successful passive immunity (SPI) farms (n=19), <20% of the proportion of newborn calves with serum immunoglobulin (Ig) G concentrations of <10.0 g/L, and failure of passive immunity (FPI) farms (n=20) with ≥20%. The proportions of serum IgG concentrations on SPI farms were clearly shifted toward the high-concentration strata compared with FPI farms. SPI farms showed significantly (P<0.001) higher survival probability than FPI farms. These results suggest that having t

Influence of age range of dairy Gir cows on the composition of colostrum and milk, and on calves' passive immunity transfer.

Colostrogenesis and lactogenesis influenced by age require further understanding in Zebu herds, due to the longevity of breeding cows. Thus, we aimed to compare colostrum and milk physicochemical and proteinogram of Dairy Gir cows and calves' passive immunity transfer by grouping 28 cows and their calves by maternal age: Young (n=6; aged 24-36 months), Adult (n=15; 37-91 months) and Senior (n=7; 96-137 months) groups. Colostrum and milk were collected at first milking (immediately after calving), transition milk (3rd milking, after 24h of calving) and full milk (10th milking, after 5 days of c

Influence of Capsaicin Supplementation on the Enhancement of Passive Immunity Transfer Through Modulation of Immunoglobulin Absorption in Neonatal Calves.

In cattle, the transfer of passive immunity from mother to calf across the placenta is limited. Therefore, providing quality colostrum or supplementing with additives that enhance this attribute is crucial to optimise the transfer of passive immunity (TPI). The objective of the present study was to investigate the effect of capsaicin on the absorption of immunoglobulins, metabolites, and enzymes to optimise TPI in neonatal calves, and to assess its impact on selected physiological parameters. Two experimental groups were established: a control group (CON; n = 8), which received colostrum in th

Synergistic modeling of hemorrhagic dengue fever: Passive immunity dynamics and time-delay neural network analysis.

Dengue fever poses a formidable epidemiological challenge, particularly for vulnerable groups such as infants. This research paper establishes a mathematical model to describe the dynamics of secondary immunity in infants against dengue hemorrhagic fever, who acquired primary immunity through maternal antibodies. The effect of passive immunity in the form of dengue immunoglobulin is analyzed for high-risk patients for different scenarios, including standard dengue infections, host with pre-existing immunity, delayed diagnosis or treatment, and end-stage dengue cases. Convergence analysis of th

Active and passive immunizations with Bordetella colonization factor A protect mice against respiratory challenge with Bordetella bronchiseptica.

Bordetella colonization factor A (BcfA) is an outer membrane immunogenic protein, which is critical for efficient colonization of the murine respiratory tract. These properties of BcfA prompted us to examine its utility in inducing a protective immune response against Bordetella bronchiseptica in a mouse model of intranasal infection. Mice vaccinated with BcfA demonstrated reduced pathology in the lungs and harbored lower bacterial burdens in the respiratory tract. Immunization with BcfA led to the generation of BcfA-specific antibodies in both the sera and lungs, and passive immunization led

Active and passive immunizations with the streptococcal esterase Sse protect mice against subcutaneous infection with group A streptococci.

The human pathogen group A Streptococcus (GAS) produces many secreted proteins that play important roles in GAS pathogenesis, including hydrolases that degrade proteins and nucleic acids. This study targets another kind of hydrolase, carboxylic esterase, with the objectives of identifying GAS esterase and determining whether it is a protective antigen. The putative esterase gene SPy1718 was cloned, and the recombinant protein (Sse) was prepared. Sse was detected in GAS culture supernatant, and patients with streptococcal pharyngitis seroconverted to Sse, indicating that Sse was produced in viv

Protection of Junín virus-infected marmosets by passive administration of immune serum: association with late neurologic signs.

Argentine hemorrhagic fever (Junín virus) is a human viral disease for which immune therapy proves effective, though a late neurologic syndrome is occasionally associated with the treatment. We attempted to determine in the infected marmoset Callithrix jacchus whether immune therapy leads to protection and/or CNS damage. Fifteen C jacchus were inoculated with 10(3) tissue culture infectious dose 50% (TCID50) of the XJ strain of Junín virus. On day 6 post infection (pi), 12 primates were treated with homologous immune serum. Animals were observed daily; and hematologic, serologic, virologic, an

Passive maternal immunity in children born to women with systemic autoimmune rheumatic disease - A case-control study.

INTRODUCTION: The transplacental transfer of maternal antibodies is essential for neonatal immunity but can be affected by maternal health conditions and pregnancy complications. In women with systemic autoimmune rheumatic diseases (SARD) this transfer may be influenced by the autoimmune condition itself and/or the immunosuppressive therapies administered during pregnancy. OBJECTIVE: This study aimed to assess the transplacental transfer and efficacy of vaccine-induced antibodies in pregnant women with SARD compared to healthy controls. METHODS: We enrolled pregnant women with and without SA

IgG-cleavage protein allows therapeutic AAV gene delivery in passively immunized MPS IIIA mice.

The widespread pre-existing αAAV-Abs in humans pose a critical challenge in translation of AAV gene therapy. The IgG degrading enzyme of Streptococci (IdeS) is demonstrated to specifically cleave IgG of humans and other species (not mouse). This study developed a modified new modified IdeS protein product (IdeSop). When incubated in vitro, IdeSop was shown to completely cleave human and rabbit IgGs within 6 h. To test IdeSop in a disease setting, we established a rabbitized αAAV9-Ab+ mouse by an IV infusion of purified acute αAAV9-Ab+ rabbit IgG into MPS IIIA mice, resulting in serum αAAV9-IgG

Passive immune therapy and other immunomodulatory agents for the treatment of severe influenza: Systematic review and meta-analysis.

BACKGROUND: A range of immunomodulatory therapies have been proposed as adjuncts to conventional antivirals to suppress harmful inflammation during severe influenza infection. We conducted a systematic review to assess available data of the effect of adjunctive non-corticosteroid immunomodulatory therapy and potential adverse effects. METHOD: We searched MEDLINE, Embase, Web of Science and clinical trial databases for published and unpublished studies, and screened the references of included articles. We included RCTs, quasi-RCTs and observational studies of virologically confirmed influenza
